As sessile organisms, plants rely on intricate molecular regulatory networks to navigate complex, fluctuating environments and diverse stress conditions. Nitrogen Limitation Adaptation (NLA), a RING-type ubiquitin ligase harboring an SPX domain, has emerged as a central hub integrating plant responses to nutrient availability, pathogen attack, and cold stress. This review provides a systematic overview of the structural features of NLA, with a particular focus on the regulatory role of inositol pyrophosphates (PP-InsPs) in plant phosphorus signaling. We highlight the core functions of NLA in m

Carrot is an important root vegetable crop for its richness in carotenoids and other nutrients. The presence of terpenoids gives carrot a unique flavor and texture. Nudix hydrolases (NUDX) are widely present in living organisms. In plants, the NUDX members were involved in carotenoid biosynthesis, terpenoid metabolism, and plant immune responses. Till now, the little-known function of NUDX members in carrot. In this study, 33 carrot DcNUDX genes were identified and categorized into 9 subgroups based on the preferred substrates. The occurrence of tandem duplication and segmental duplication eve

Bacterial leaf spot disease is among the most common diseases affecting tomato production. It is caused by Pseudomonas syringae pv. tomato (Pst) and severely affects tomato plant growth, development, and yield. In this study, we identified a fructose-1,6-diphosphate aldolase gene, SlFBA3, whose expression was significantly increased in the tomato pathway for resistance to Pst. Overexpression of SlFBA3 enhanced tomato plant resistance, whereas knockout of SlFBA3 reduced the resistance of tomato plants. Further analysis revealed that SlSGR is an interacting protein of SlFBA3. Functional analysis

A significant global health crisis is predicted to emerge due to antimicrobial resistance by 2050, with an estimated 10 million deaths annually. Increasing antibiotic resistance necessitates continuous therapeutic innovation as conventional antibiotic treatments become increasingly ineffective. The naturally occurring antibacterial, antifungal, and antiviral compounds offer a viable alternative to synthetic antibiotics. This review presents bacterial resistance mechanisms, nanocarriers for drug delivery, and plant-based compounds for nanoformulations, particularly nanoantibiotics (nAbts). Gree

Polyphenols are secondary plant metabolites or organic compounds synthesized by them. In other words, these are molecules that are found in plants. Due to the wide variety of polyphenols and the plants in which they are found, these compounds are divided according to the source of origin, the function of the polyphenols, and their chemical structure; where the main ones are flavonoids. All the beneficial properties of polyphenols have not yet been studied, since this group of substances is very extensive and diverse. However, most polyphenols are known to be powerful antioxidants and have anti

Nonspecific lipidtransfer proteins (nsLTPs), which are small, cysteine-rich proteins, belong to the pathogenesis-related protein family, and several of them act as positive regulators during plant disease resistance. However, the underlying molecular mechanisms of these proteins in plant immune responses are unclear. In this study, a typical nsLTP gene, StLTP10, was identified and functionally analysed in potato. StLTP10 expression was significantly induced by Phytophthora infestans, which causes late blight in potato, and defence-related phytohormones, including abscisic acid (ABA), salicylic

Transient infiltrations in tobacco are commonly used in plant studies, but the host response to different disarmed Agrobacterium strains is not fully understood. The present study shows that pretreatment with disarmed Agrobacterium tumefaciens GV3101 primes the defense response to subsequent infection by Pseudomonas syringae in Nicotiana tabacum. The presence of a trans-zeatin synthase (tzs) gene in strain GV3101 may be partly responsible for the priming response, as the tzs-deficient Agrobacterium sp. strain LBA4404 only weakly imparts such responses. Besides inducing the expression of defens

A DNA encoding the 27-kDa domain I of anthrax lethal factor protein (LF), was linked to the carboxyl terminus of the cholera toxin B-subunit (CTB-LF). The CTB-LF fusion gene was transferred into Solanum tuberosum cells by Agrobacterium tumefaciens-mediated in vivo transformation methods and antibiotic-resistant plants were regenerated. The CTB-LF fusion gene was detected in transformed potato leaf genomic DNA by polymerase chain reaction (PCR)-mediated DNA amplification. Immunoblot analysis with anti-CTB and anti-LF primary antibodies verified the synthesis and assembly of biologically active

This review examines the effects of cannabinoids on immune function, with a focus on effects on T-cells, as well as on resistance to infection. The paper considers the immune modulating capacity of marijuana, of ∆(9)-THC extracted from the marijuana plant, and synthetic cannabinoids. Of particular interest are synthetic compounds that are CB2 receptor (CB2R) selective agonists. As the CB2R is principally expressed on cells of the immune system, agonists that target this receptor, and not CB1 (which is mainly expressed on neurons), have the possibility of altering immune function without psycho

Porcine reproductive and respiratory syndrome virus (PRRSV) is a highly infectious RNA virus that severely affects swine herds, causing respiratory and reproductive disorders with major global economic impact. Current first-generation vaccines based on inactivated or attenuated viruses offer limited protection and may pose safety risks. Given the virus's high mutation rate and genetic variability, there is a growing need for safer and more effective vaccines. Plant molecular farming represents a promising alternative production system for second-generation subunit vaccines, offering advantages

Lettuce necrotic yellows virus (LNYV) is a plant rhabdovirus which has a type-1 transmembrane glycoprotein. Here, we describe the generation of murine anti-sera to the glycoprotein. Rational design, expression, and purification of recombinant glycoprotein, termed rLGe, was undertaken using SignalP4.1 and camSol servers to predict signal peptide cleavage and protein solubility. In order to successfully obtain expression in mammalian cells, LNYV glycoprotein native signal peptide was substituted with that of Rabies virus glycoprotein. In addition, rather than expression of the entire molecule, r

Salicylic acid (SA) is an important phytohormone that is involved in the regulation of plant defence, growth and development. A large number of proteins have been shown to have the ability to interact with SA, and NPR4 has been demonstrated to be a receptor of SA that plays significant roles in the innate immune response of plants. In this study, Spodoptera frugiperda (Sf9) cells were used to express full-length AtNPR4 from Arabidopsis thaliana. To facilitate crystallization, T4 lysozyme (T4L) was added to the N-terminus of the AtNPR4 protein. The recombinant T4L-AtNPR4 protein was expressed,

Flavonoids present in plants and fruits have been used in traditional medicine to reduce inflammation under various inflammatory conditions, including colitis. The pharmacological mechanisms that regulate intestinal inflammation are associated with the colonic microbiota, protection against oxidative stress, preservation of epithelial barrier function, and immune homeostasis. This review describes the main flavonoids present in the diet and examines their role in mitigating colon inflammation, as well as their impact when chronic inflammation progresses to colitis-associated colorectal cancer

Recently, the concept of local allergic rhinitis (LAR) was established, namely rhinitis symptoms with local IgE production and negative serum antigen-specific IgE. However, the natural course of LAR development and the disease pathogenesis is poorly understood. This study investigated the pathophysiology of mice with allergic rhinitis that initially sensitized with ragweed pollen through the nasal route. Mice were nasally administrated ragweed pollen over consecutive days without prior systemic immunization of the allergen. Serial nasal sensitization of ragweed pollen induced an allergen-speci

PURPOSE OF REVIEW: To review and discuss recent studies on molecular diagnosis of peanut and other legume allergy.
RECENT FINDINGS: Studies from the UK and France suggest that quantification of Ara h 2-specific IgE may accurately discriminate peanut allergy from tolerance. However, the pattern of allergenic component recognition in peanut-sensitized patients from different populations or geographical areas varies, reflecting different pollen and dietary exposures. In the USA, peanut-allergic patients are commonly sensitized to Ara h 1-3, in Spain to Ara h 9 and in Sweden to Ara h 8. Patients

Mycobacterium tuberculosis is responsible for over 8 million cases of tuberculosis (TB) annually. Natural products may play important roles in the chemotherapy of TB. The antimycobacterial activity and the innate immune response of methanol (METH) and dichloromethane (DCM) extracts of Indigofera suffruticosa Miller (Fabaceae) were evaluated. We observed that the minimum inhibitory concentrations (MICs) for METH and DCM extracts were 125 and 1000 microg/mL, respectively. However, they were able to induce the innate immune response through the production of high levels of NO and TNF-alpha (p < 0

BACKGROUND: An extract of the tropical fern Polypodium leucotomos (PL) administered orally to mice inhibits ultraviolet B (UVB) radiation-induced skin cancer formation. UVB-induced murine skin cancers occur, in part, because of UVB-induced immunosuppression. Thus, we examined whether PL inhibits UVB-suppression of the induction of contact hypersensitivity (CHS) locally or systemically.
METHODS: C57BL/6 mice received standard drinking water or water-containing PL. In the local model, mice were shaved on the dorsum and exposed to 3500 J/m(2) of UVB radiation daily for 4 days. Control mice were
