Genetic polymorphism and natural selection of merozoite surface protein 1 C-terminal 42 kDa in Vietnamese Plasmodium vivax isolates.

Plasmodium vivax merozoite surface protein-1 (PvMSP-1) is a major surface protein of merozoites and plays crucial roles in erythrocyte invasion by merozoites. PvMSP-1 undergoes sequential proteolytic cleavages during erythrocyte invasion, resulting in four fragments of 82, 30, 38, and 42 kDa. The carboxy-terminal 42 kDa fragment (PvMSP-142) is further cleaved into two smaller fragments of 33 kDa (PvMSP-133) and 19 kDa (PvMSP-119). PvMSP-142 is an attractive vaccine candidate since it induces antibody production in naturally exposed humans, and specific antibodies against this region inhibit me

Emergence of multidrug-resistant E. coli ST8346 isolates carrying three distinct plasmids with NDM-5, KPC-2, and OXA-181.

BACKGROUND: E. coli ST8346 is a unique strain associated with the potential carriage of multiple carbapenemases. Three unique E. coli ST8346 isolates were identified, each concurrently harboring multiple carbapenemases, including blaNDM-5, blaKPC-2, and/or blaOXA-181. This study aimed to characterize the genetic and plasmid structures of these isolates to understand their transmission and resistance mechanisms. METHODS: Antibiotic resistance profiles, genetic relatedness, and plasmid structures were determined using antibiotic susceptibility testing, multilocus sequence typing (MLST), pulsed-

De novo balanced reciprocal translocation mosaic t(1;3)(q42;q25) detected by prenatal genetic diagnosis: a fetus conceived using preimplantation genetic testing due to a t(12;14)(q22;q13) balanced paternal reciprocal translocation.

INTRODUCTION: De novo balanced reciprocal translocations mosaicism in fetus conceived using preimplantation genetic testing from a different balanced translocation carrier parent has been rarely reported. METHODS: Chromosomal microarray analysis, karyotype analysis and fluorescent in situ hybridization were performed to verify the type and heredity of the rearrangement. STR analysis was conducted to identify potential contamination and verify kinship. In addition, a local BLAST engine was performed to locate potentially homologous segments which might contribute to the translocation in breakp

Genetic Manipulation of Non-pneumophila Legionella: Protocols Developed for Legionella longbeachae.

Current biomedical research into Legionnaires' disease is dominated by studies of Legionella pneumophila, largely because this pathogen is responsible for approximately 90% of clinical disease worldwide. However, in certain geographical regions, infections with non-pneumophila species are responsible for a significant proportion of diagnosed Legionnaires' disease. Understanding the pathogenesis of these non-pneumophila species of Legionella is an important step toward clinical intervention. The capacity to genetically manipulate these pathogens is essential in order to understand the genetic f

Generation and characterization of Chd7-iCreERT2-tdTomato mice.

Heterozygous mutation of CHD7 gene causes a severe developmental disorder called CHARGE syndrome. In order to further explore the expression and function of Chd7 in vivo, we generated a Chd7-P2A-iCreERT2-P2A-tdTomato (in short, Chd7-CT-tdT) knockin mouse line using the CRISPR/Cas9 technology. The specificity and efficiency of two knockin genetic elements were validated. The Chd7-CT-tdT reporter gene could accurately reflect both the dynamic expression pattern of endogenous Chd7 during neurodevelopment and cell-type specific expression in the brain and eye. The recombination efficiency of Chd7-

A major locus for myoclonus-dystonia maps to chromosome 7q in eight families.

Myoclonus-dystonia (M-D) is an autosomal dominant disorder characterized by myoclonic and dystonic muscle contractions that are often responsive to alcohol. The dopamine D2 receptor gene (DRD2) on chromosome 11q has been implicated in one family with this syndrome, and linkage to a 28-cM region on 7q has been reported in another. We performed genetic studies, using eight additional families with M-D, to assess these two loci. No evidence for linkage was found for 11q markers. However, all eight of these families showed linkage to chromosome 7 markers, with a combined multipoint LOD score of 11

Mapping of a locus for a familial autosomal recessive idiopathic myoclonic epilepsy of infancy to chromosome 16p13.

Myoclonic epilepsies with onset in infancy and childhood are clinically and etiologically heterogeneous. Although genetic factors are thought to play an important role, to date very little is known about the etiology of these disorders. We ascertained a large Italian pedigree segregating a recessive idiopathic myoclonic epilepsy that starts in early infancy as myoclonic seizures, febrile convulsions, and tonic-clonic seizures. We typed 304 microsatellite markers spanning the 22 autosomes and mapped the locus on chromosome 16p13 by linkage analysis. A maximum LOD score of 4.48 was obtained for

Transfection of malaria parasites.

The stable genetic transformation of three phylogenetically diverse species of Plasmodium, the parasitic etiological agent of malaria, is now possible. The parasite is haploid throughout the vast majority of its life cycle. Therefore with the single selectable marker activity and protocols currently available, it is possible not only to express introduced transgenes but also to study the effects of site-specific homologous recombination such as gene knockout. Transgene expression will allow the detailed study of many aspects of the cellular biology of malaria parasites, for example, the mechan

Genetic linkage analysis in familial benign (hypocalciuric) hypercalcemia: evidence for locus heterogeneity.

Familial benign hypercalcemia (FBH, or hypocalciuric hypercalcemia) is characterized by inheritance, in an autosomal dominant pattern, of lifelong hypercalcemia without hypercalciuria, which is often mistaken for classical primary hyperparathyroidism. Recently, the FBH trait was linked, in four families, to chromosome 3q. We report genetic linkage analysis in 140 persons from five additional families having FBH (65 affected, 67 unaffected, and 8 unclassifiable). In four families, FBH mapped to chromosome 3q, between D3S1215 and D3S20, maximum multipoint lod score 12.9. By contrast, in the fift

Genetic diversity and evolutionary dynamics of Senecavirus A in Brazil.

Senecavirus A (SVA) is a picornavirus that was first isolated in the USA in 2002; however, there is evidence that the virus was circulating in swine herds since 1988. Despite frequent reports of vesicular disease outbreaks caused by SVA infection in swine in Brazil since 2014, there is limited data on the genetic diversity and evolution of the virus in the country. SVA was isolated from swine exhibiting vesicular lesions, with samples originating from farms or slaughterhouses across 57 municipalities in 8 Brazilian states between 2018 and 2022. We obtained 501 SVA genomes through Sanger and Ox

Detection, virus isolation, and phylogenetic analysis of PRRSV2 in China from 2022 to 2025.

BACKGROUND: Porcine reproductive and respiratory syndrome (PRRS) is one of the most important diseases threatening the global pig industry. It has been reported that PRRSV predominantly circulating in clinical practice in China belongs to the genotype 2 (PRRSV2), comprising four lineages: lineages 1, 3, 5, and 8. Among these, lineage 1 strains are further divided into sublineage 1.5 (NADC34-like strains) and sublineage 1.8 (NADC30-like strains). To further understand the prevalence and variation characteristics of PRRSV2 in China, this study conducted detection, virus isolation, and genetic evo

Contrasting Genomic Outcomes of Secondary Contact in Bornean Bird Lineages: Insights From Barrier Loci, Demographic History and Genomic Features.

Secondary contact before the completion of reproductive isolation can alter speciation trajectories by impeding divergence through gene flow or accelerating it via reinforcement. Using genomic data, we investigated secondary contact between two pairs of Copsychus (Muscicapidae) lineages in Borneo: subspecies of magpie-robins (C. saularis) and two species of shamas (C. malabaricus and C. stricklandii). Although eastern and western lineages in both groups have similar divergence times, they show markedly different phenotypic and genomic outcomes following contact. Magpie-robins exhibit extensive

Genomic and cytogenetic evidence for divergence and speciation in geladas.

BACKGROUND: Chromosomal rearrangements can play an important role in shaping patterns of genomic divergence and speciation, yet their evolutionary context is still largely unclear, as such events are rare and challenging to identify in recent evolutionary history. Geladas (Theropithecus gelada) provide a unique system to investigate this process, as a novel fission on chromosome 7 has resulted in polymorphic karyotypes among gelada populations. RESULTS: We integrate new cytogenetic data from ten animals together with whole-genome sequencing from 149 individuals spanning all three extant gelad

A Chimeric Airway Model Enables Evaluation of Essential Genes In Vivo.

A pseudostratified epithelium lines the upper airways and is maintained by stem cells that express a basal cell phenotype. These airway stem cells share molecular and functional similarities with other tissue stem cells, particularly those that are involved in development or postnatal maintenance of the esophagus and epidermis. Thus, analysis of gene function in the airway is complicated by the potential for lethal non-respiratory phenotypes. Conditional genetic approaches employing cell type-specific and/or temporally controlled expression systems provide some options, but limitations remain.

Frequent HIV-1 recombination among MSM in Beijing revealed by subtyping and near full-length genome analyses.

The high mutation and recombination rates of HIV-1 drive extensive genetic diversity. Co-infection with different subtypes or recombinant forms facilitates the emergence of new recombinants, complicating transmission networks and posing challenges for molecular surveillance. Although antiretroviral therapy (ART) coverage is high among diagnosed individuals in China, ongoing HIV-1 transmission-including from undiagnosed infections-likely contributes to the continued circulation and accumulation of recombinant lineages. We retrospectively analyzed 1006 plasma samples from 993 individuals diagnos

Bioprospecting potential genetic biomarkers of gallbladder cancer.

BACKGROUND: Gallbladder cancer (GBC) is a rare and aggressive cancer of the biliary tract with a very low survival rate. The availability of diagnostic biomarkers and targeted therapies for its management is limited. The study identifies potential genetic biomarkers of GBC by analyzing differentially expressed genes (DEGs) through microarray profiling and constructing regulatory networks using systems biology techniques. METHODS: We used Clariom™ D Array in gallbladder cancer, cholelithiasis, and normal tissues (10 cases in each group), identifying DEGs and key biological pathways. Functional

Isolation and Pathogenicity of a Natural Recombinant Pig Reproductive and Respiratory Syndrome Virus in Northeast China.

First reported in 1987, the porcine reproductive and respiratory syndrome virus (PRRSV) has significantly disrupted the major regions affected by PRRSV in the pig breeding industry. Recently, outbreaks of disease caused by recombinant PRRSV strains in China have raised serious concerns. Effective immunization and infection control in pig populations is critical, as the virus frequently undergoes mutation and recombination. This study characterized a novel recombinant PRRSV strain, BX/CH/22, isolated from Northeast China. Genetic analysis revealed that BX/CH/22 is a recombinant of JXA1, NADC 30

XRCC4-related microcephalic primordial dwarfism: description of a clinical series of 7 cases, phenotype expansion and new diagnostic approaches.

The non-homologous end joining (NHEJ) pathway is essential to repair DNA double-strand breaks. XRCC4 acts as a stabilizer of the DNA ligase LIG4 in the NHEJ process. In humans, XRCC4 pathogenic variants are responsible for a microcephalic primordial dwarfism syndrome (MPD). Currently, 17 patients have been reported with XRCC4-related MPD and we report 7 new patients from 6 different families, including one fetus. The patients present with short stature, severe microcephaly, neurodevelopmental disorder and additional features, such as transient increase in nuchal translucency, congenital glauco

Population genomic analyses reveal high diversity, recombination and nosocomial transmission among Candida glabrata (Nakaseomyces glabrata) isolates causing invasive infections.

Candida glabrata is a commensal yeast of the gastrointestinal tract and skin of humans. However, it causes opportunistic infections in immunocompromised patients, and is the second most common Candida pathogen causing bloodstream infections. Although there are many studies on the epidemiology of C. glabrata infections, the fine- and large-scale geographical nature of C. glabrata remain incompletely understood. Here we investigated both the fine- and large-scale population structure of C. glabrata through genome sequencing of 80 clinical isolates obtained from six tertiary hospitals in Qatar an
