Identification of Burkholderia pseudomallei Near-Neighbor Species in the Northern Territory of Australia.

Identification and characterization of near-neighbor species are critical to the development of robust molecular diagnostic tools for biothreat agents. One such agent, Burkholderia pseudomallei, a soil bacterium and the causative agent of melioidosis, is lacking in this area because of its genomic diversity and widespread geographic distribution. The Burkholderia genus contains over 60 species and occupies a large range of environments including soil, plants, rhizospheres, water, animals and humans. The identification of novel species in new locations necessitates the need to identify the true…

Elevated root retention of hexahydro-1,3,5-trinitro-1,3,5-triazine (RDX) in coniferous trees.

For decades, the explosive RDX (hexahydro-1,3,5-trinitro-1,3,5-triazine) has been used for military and industrial applications. Residues of … . However, it also opens the opportunity for microbial mineralisation in the litter layer or in the rhizosphere of coniferous forests and offers a chance for repeated uptake of RDX by the tree roots.

Banana-associated microbial communities in Uganda are highly diverse but dominated by Enterobacteriaceae.

Bananas are among the most widely consumed foods in the world. In Uganda, the country with the second largest banana production in the world, bananas are the most important staple food. The objective of this study was to analyze banana-associated microorganisms and to select efficient antagonists against fungal pathogens which are responsible for substantial yield losses. We studied the structure and function of microbial communities (endosphere, rhizosphere, and soil) obtained from three different traditional farms in Uganda by cultivation-independent (PCR-SSCP fingerprints of 16S rRNA/ITS ge…

This organic mercury compound … and coastal marshes, but also the anoxic hypolimnion of lakes and anaerobic microhabitats like the rhizosphere of floating macrophytes. Prime suspects for methylation are sulfate-reducing bacteria, although also iron reducers have lately been identified as capable mercury methylators. Toxicological effects of selective herbicides on plant growth promoting activities of phosphate solubilizing Klebsiella sp. strain PS19.

This study examines the effect of four herbicides, quizalafop-p-ethyl, clodinafop, metribuzin and glyphosate, on plant growth promoting activities like phosphate solubilization, siderophores, indole acetic acid, exo-polysaccharides, hydrogen cyanide and ammonia production by herbicide tolerant Klebsiella sp. strain PS19. The strain was isolated from mustard rhizosphere. The selected herbicides were applied two to three times at the recommended rates. Klebsiella sp. strain PS19 tolerated a concentration of 1600 μg/ml each of quizalafop-p-ethyl and clodinafop, and 3200 and 2800 μg/ml of metribuz…

Genetic diversity and biocontrol potential of fluorescent pseudomonads producing phloroglucinols and hydrogen cyanide from Swiss soils naturally suppressive or conducive to Thielaviopsis basicola-mediated black root rot of tobacco.

Pseudomonas populations producing the biocontrol compounds 2,4-diacetylphloroglucinol (Phl) and hydrogen cyanide (HCN) were found in the rhizosphere of tobacco both in Swiss soils suppressive to Thielaviopsis basicola and in their conducive counterparts. In this study, a collection of Phl+ HCN+Pseudomonas isolates from two suppressive and two conducive soils were used to assess whether suppressiveness could be linked to soil-specific properties of individual pseudomonads. The isolates were compared based on restriction analysis of the biocontrol genes phlD and hcnBC, enterobacterial repetitive…

Laboratory assessment of novel endophytic Trichoderma-based bioformulations for the biological control of sorghum leaf spot and stalk rot diseases.

Stalk rot and leaf spot, caused by Fusarium verticillioides and Curvularia lunata, respectively, are among the most destructive seed-borne f … ive, persistent biofungicide development. Adapted to the seed-soil interface, they can colonize the rhizosphere, outcompete pathogens, and remain active under field conditions. Thus, they are strong candidates for sustainable biofungicides, pending greenhouse/field validation and large-scale production optimization.

Pseudomonas cistanchae sp. nov., a novel plant-associated bacterium isolated from diseased Cistanche soil in Xinjiang, China.

A novel bacterial strain, designated JK66T, was isolated from the rhizosphere soil of diseased Cistanche plants collected in Turpan, Xinjiang, China. Strain JK66T grew at 20-42 °C (optimum, 37 °C), pH 5.0-10.0 (optimum, pH 7.0), and with 0-3.0% (w/v) NaCl (optimum, 2.0% (w/v)). The strain was selected for taxonomic characterization because of its antagonistic activity against Fusarium acuminatum and its potential to produce indole-3-acetic acid. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain JK66T belongs to the genus Pseudomonas, showing the highest sequence simi…

Deciphering of a bioactive molecule with a three-domain inhibitory activity from a halophilic bacterium Arhodomonas sp. AD133.

Pyoluteorin, a biological pesticide, is mainly produced by growth promoting rhizosphere bacteria such as Pseudomonas species showing strong antifungal and antibacterial activities. Little is known about pyoluteorin from halophiles living in hypersaline environments. The halophilic bacterium Arhodomonas sp. strain AD133, which was isolated from a hypersaline environment, demonstrated clear inhibitory activity against Bacteria, Eukaryotes and Archaea. Strain AD133 not only can inhibit halophilic archaea and bacteria that live in the same niche, but also can suppress other microorganisms, such as…

The native soil microbiome is critical for early root-associated microbiota assembly and canola growth.

BACKGROUND: The native soil microbiome contributes to regulating the root-associated microbiota, root morphology, and plant growth. Using two canola (Brassica napus L.) genotypes contrasting in root size (small-rooted NAM23 and large-rooted NAM37), we investigated how the native soil microbiome influences canola establishment. Plants were grown in rhizoboxes containing gamma-irradiated (microbiome dysbiosis) or untreated (healthy microbiome) soils for 14 days. We evaluated plant growth and profiled bacterial and fungal communities in unplanted soil, rhizosphere soil, and root samples via DNA a…
