Significance: Sickle-cell leg ulcers (SCLUs) are a severe, chronic, and recurrent complication of sickle-cell disease (SCD). There are no official recommendations for treatment. Recent Advances: Only a few studies with a high level of evidence have been conducted to evaluate treatment of SCLUs. However, several studies have been conducted with a high level of evidence to evaluate the efficacy of treatments in venous leg ulcers, and SCLUs could benefit from these treatments, especially when a venous incompetence or an edema is associated. Pathophysiology of SCLUs includes a vasculopathy related

Dysbiosis of the microbiome on the airway mucosa leads to the development of chronic inflammatory and allergic disorders. The aim of this study was to consider the potential diagnostic criteria for allergic fungal rhinosinusitis (AFRS) and nonallergic fungal rhinosinusitis (FRS), and the role of fungal presence in an environment for the development of AFRS. In this study, 136 patients were divided into two groups: patients with positive specific immunoglobulin E (sIgE) and fungal finding (AFRS group), and patients with negative sIgE and positive fungal finding (FRS group). The study design inc

Forensic analysis of biological traces generally encompasses the investigation of both the person who contributed to the trace and the body site(s) from which the trace originates. For instance, for sexual assault cases, it can be beneficial to distinguish vaginal samples from skin or saliva samples. In this study, we explored the use of microbial flora to indicate vaginal origin. First, we explored the vaginal microbiome for a large set of clinical vaginal samples (n = 240) by next generation sequencing (n = 338,184 sequence reads) and found 1,619 different sequences. Next, we selected 389 ca

BACKGROUND: Seborrheic dermatitis (SD) is characterized by limited therapeutic options and high recurrence rates, and the efficacy and mechanism of ozone hydrotherapy for SD remain unclear, especially its potential role in regulating metabolic pathways of keratinocytes.
METHODS: We performed a single-arm, open-label, self-controlled before-and-after study of ozone hydrotherapy in 24 SD patients, evaluating clinical symptoms and analyzing scalp microbiome. A Malassezia-induced guinea pig model of SD-like lesions and a Malassezia-infected HaCaT cell model were used to explore barrier repair, an

In an increasingly urbanized world, environmental pollution is recognized for its adverse effects on both systemic and skin health. While its role in conditions such as atopic dermatitis and psoriasis is well documented, its impact on acne vulgaris remains less clear. This review aims to evaluate existing literature examining the association between environmental pollutants - such as particulate matter (PM2.5, PM10), nitrogen oxides (NO2, NOx) and traffic-related emissions - and the development, severity or exacerbation of acne. A systematic search of peer-reviewed English-language studies pub

BACKGROUND: Treponema pallidum subsp pertenue and Haemophilus ducreyi are causative agents of cutaneous ulcer (CU) in yaws-endemic regions in the tropics. However, a significant proportion of CU patients remain polymerase chain reaction (PCR) negative for both bacterial agents. We aimed to identify potential additional etiological agents of CU in a yaws-endemic region.
METHODS: This population-based cohort study included children in Lihir Island (Papua New Guinea) examined during a yaws eradication campaign in October 2013-October 2014. All consenting patients with atraumatic exudative ulcers

BACKGROUND: Vulvar lichen sclerosus (VLS) is a chronic inflammatory skin disorder that severely impairs women’s physical and psychological well-being. Topical glucocorticoids are the first-line treatment; however, their long-term efficacy is limited due to frequent symptom relapse after discontinuation and incomplete resolution of lesions. Therefore, effective adjunctive strategies are urgently needed to achieve sustained disease control. METHODS: This study aimed to first characterize disease-associated alterations in vulvar skin microbiota by comparing patients with VLS and healthy volunteer

The gut-skin axis plays a pivotal role in psoriasis pathogenesis, yet the precise metabolic crosstalk by which intestinal commensals regulate cutaneous immunity remains elusive. Here, we identify a functional "microbe-enzyme-metabolite-immune" axis that orchestrates skin homeostasis. Through multi-omics analyses of psoriatic patients and imiquimod-induced murine models, we reveal a significant depletion of the gut commensal Parabacteroides distasonis (P. distasonis) and its metabolite, genistein. We demonstrate that P. distasonis utilizes its inherent β-glucosidase activity to convert dietary

The current study aimed to investigate the effect of different dietary supplemental oils on the immune status of broilers. One-day-old Cobb 500 broiler chicks were randomly distributed into eight batteries and fed eight experimental diets. There were 680 broilers, 85 birds per battery. The experimental oils were all used at 10% of the total diet. Each dietary treatment (TRT) contained one of the following essential oils: TRT 1 = control group that received a basal diet + soybean oil (SO); TRT 2 = basal diet as in TRT 1 + sunflower oil (SFO); TRT 3 = basal diet as in TRT 1 + canola oil (CO); TR

INTRODUCTION: Atopic dermatitis (AD) is a chronic inflammatory skin disorder resulting from the interplay of genetic and environmental factors, with a dysregulated type-2 immune response. The association between AD onset and intestinal dysbiosis supports research into nutritional interventions such as fermentable fibers intake. Agave-derived fructans (AFs) display prebiotic activity, modulating gut microbial communities that may positively influence immune functions. In this study, we evaluated the anti-inflammatory and immunomodulatory effects of oral AFs in a rat AD model.
METHODS: AD-like

Short-chain fatty acids (SCFAs) are critical metabolites produced by gut microbiota that play a key role in modulating inflammation and regulating systemic immunity, including against cancer. Decreases in SCFAs can foster a permissive tumor immune environment. Recent studies have shown that cutaneous T-cell lymphoma (CTCL) patients exhibit increasing gut dysbiosis and loss of bacteria predicted to produce SCFAs with increasing disease severity. To investigate this functional connection, we collected stool swab samples from 15 individuals- 8 mycosis fungoides (MF) patients and 7 matched healthy

Ticks are arachnid ectoparasites that rank second only to mosquitoes in the transmission of human diseases including bacteria responsible for anaplasmosis, ehrlichiosis, spotted fevers, and Lyme disease among other febrile illnesses. Due to the paucity of data on bacteria transmitted by ticks in Kenya, this study undertook a bacterial metagenomic-based characterization of ticks collected from Isiolo, a semi-arid pastoralist County in Eastern Kenya, and Kwale, a coastal County with a monsoon climate in the southern Kenyan border with Tanzania. A total of 2,918 ticks belonging to 3 genera and 10

Aberrant activation of the stimulator of interferon genes (STING) pathway is a hallmark of autoinflammatory disorders such as STING-associated vasculopathy with onset in infancy (SAVI), characterized by systemic inflammation affecting blood vessels, skin, and lungs. Despite its clinical significance, the mechanisms linking STING activation to disease pathology remain poorly defined. In this study, we demonstrated that SAVI mice harboring the N153S STING mutation exhibit diverse disease phenotypes, with a subset developing severe colitis and diarrhea alongside exacerbated systemic inflammation.

Microplastics (MPs) transfer from the environment to living organisms is a nonignorable global problem. As a complete metamorphosis insect, the larvae and adult Culex quinquefasciatus mosquito live in aquatic and terrestrial environments, respectively, where they easily access MPs. However, little is known about mosquitoes' potential role in MPs accumulation throughout ecosystems. Therefore, we conducted a study with different MPs particle sizes (0.1/1/10 μm) and concentrations (0.5/5/50 μg/mL) on Cx. quinquefasciatus to address this issue. Once exposed at the young larval stage, MPs could acc

BACKGROUND: Although several mouse models of exogenous-agent-induced atopic dermatitis (AD) are currently available, the lack of certainty regarding their similarity with human AD has limited their scientific value. Thus, comprehensive evaluation of the characteristics of mouse models and their similarity with human AD is essential.
OBJECTIVE: To compare six different exogenous-agent-induced AD mouse models and find out the optimum models for study.
METHODS: Female BALB/c mice underwent induction of AD-like dermatitis by MC903 alone or in combination with ovalbumin (OVA), dinitrofluorobenzen

Gastrointestinal bacteria are known to have an impact on local and systemic immunity, and consequently either promote or suppress cancer development. Following the notion that perinatal bacterial exposure might confer immune system competency for life, we investigated whether early-life administration of cholera-toxin (CT), a protein exotoxin of the small intestine pathogenic bacterium Vibrio cholerae, may shape local and systemic immunity to impart a protective effect against tumor development in epithelia distantly located from the gut. For that, newborn mice were orally treated with low non
