Neural mechanisms of joint pain.

Joint pain is a common symptom in various forms of arthritis. Unfortunately, the mechanisms involved in the pathogenesis of joint pain are not well understood, but probably include peripheral and central neural mechanisms. The sympathetic system appears to interact with sensory afferents under pathological conditions, and this may be mediated directly via receptors on sensory neurons, or indirectly via inflammatory mediators. Classical inflammatory mediators such as serotonin and bradykinin appear to activate some nociceptive afferents and serotonin may sensitise these afferents to non-noxious

Somatic afferent fibers which continuously discharge after being isolated from their receptors.

A previous study in our laboratory has shown that some afferent axons produce prolonged discharges after the axons have been completely isolated. We have attempted to identify the type of afferent fibers displaying such activity. Single unit activity was recorded from a filament dissected from the distal stump of the cut sural or plantar nerve of an anesthetized rat. After thorough identification of the receptor type, the nerve was cut at a site between the recording electrode and the receptive field, completely isolating the fiber being recorded. Unit activity was recorded up to 1 h after sec

Morphology, distribution and innervation of the lateral-line receptors of the Florida gar, Lepisosteus platyrhincus.

The lateral-line system of gars consists of superficial neuromasts, which are arranged in lines termed pit lines, canal neuromasts and spiracular organs, which are located within diverticula of the hyoid gill pouch. Both canal and superficial neuromasts possess polarized hair cells whose directional sensitivity parallels the long axis of their respective lines. However, the apical surfaces of canal neuromasts are larger and possess far more hair cells than do those of superficial neuromasts, but superficial neuromasts have longer kinocilia and, presumably, longer cupulae. The receptors of the

The Bötzinger complex as the pattern generator for retching and vomiting in the dog.

To clarify the location of the pattern generator for the emetic act, the bulb was systematically stimulated and partially cut in decerebrate, paralyzed dogs. Stimulation of the following bulbar structures elicited the activities which could be recognized as retching and vomiting in the following muscle nerves. The bulbar structures were: the intra-bulbar bundle of the vagal afferents, the solitary tract and the medial subdivision of its nucleus (NTS), the area postrema, the commissural nucleus, the raphe area at the obex level, and the longitudinal reticular column which consists of 3 areas--t

An electrophysiological and anatomical study of afferents reaching the cerebellar uvula in the rabbit.

Stimulation of a medial region of the cerebellar uvula cortex produces sympathetically mediated cardiovascular effects in the decerebrate rabbit. In the present study the afferents to this cerebellar area have been determined using electrophysiological and neuroanatomical methods. Field potentials of prevalent mossy origin evoked by single-shock stimulation of different peripheral nerves have been recorded from sublobules IXa and IXb. No response to vagus and aortic nerves stimulation has been found. A modulation of Purkinje cell discharge following natural vestibular stimulation has been foun

Correlative contribution of carotid and aortic afferences to the ventilatory chemosensory drive in steady-state normoxia and to the ventilatory chemoreflexes induced by transient hypoxia.

The contributions of the peripheral arterial chemoreceptors to the tonic and phasic reflex ventilatory regulation were studied in spontaneously breathing pentobarbitone anesthetized adult cats. The chemosensory drive during eucapnic normoxia was inferred from the transient ventilatory effects induced by anesthetic blockade of the buffer nerves. Aortic nerves block did not modify ventilation. Carotid nerves block provoked transient ventilatory depression, decreasing VT by 46% and fR by 26%, followed by recovery to steady-state values in VT, fR and PETCO2. Changes in PETCO2 were correlated with

Reflex activation of the adrenal medulla during hypoglycemia and circulatory dysregulations is regulated by capsaicin-sensitive afferents.

1. Catecholamines were measured in the adrenal venous outflow during hypoglycemia, hypotension, hypovolaemia and efferent splanchnic nerve stimulation in anaesthetized capsaicin-pretreated rats and their vehicle controls. In control rats, efferent splanchnic nerve stimulation caused a marked rise in adrenaline and noradrenaline levels. In contrast, the other stimuli mainly elicited adrenaline release. 2. The release of adrenaline and nor-adrenaline evoked by splanchnic nerve stimulation was of the same magnitude in capsaicin-pretreated rats (whose afferent C-fibres were destroyed by this pretr

Inhibition of antral and pyloric electrical activity by vagal afferent stimulation in the rabbit.

The effects of stimulation of cervical vagal afferents on the electromyographic activity (e.m.g.) of the gastric antrum and pylorus have been studied both in conscious and decorticate, curarized rabbits. In the latter group, stimulation at 10-15 Hz (0.3-0.4 mA; 5-7 ms duration) inhibited both the antral and pyloric e.m.g. in 21 rabbits, elicited an excitatory response in two more but produced no change in the remaining 13 animals. The inhibitory response was not affected by guanethidine (1 mg/kg, i.v.), phentolamine (1 mg/kg, i.v.) or propranolol (0.1 mg/kg, i.v.) and persisted in rabbits in w

Olfactory marker protein is present in olfactory receptor cells of human fetuses.

Light microscopic observations indicated that cellular differentiation of the primary olfactory pathway in human fetuses was relatively advanced by the end of the first trimester. However, immunohistochemical staining showed that olfactory marker protein was present in the receptor cells only at about 28 weeks post-conception. At about 32 weeks, modest amounts of the protein could be found in the peripheral olfactory nerve layer and a few glomeruli, with increased staining observed in subsequent weeks. The results show conclusively that human fetal olfactory neurons synthesize olfactory marker

Peripheral nerve injury in developing rats reorganizes representation pattern in motor cortex.

We investigated the effect of neonatal nerve lesions on cerebral motor cortex organization by comparing the cortical motor representation of normal adult rats with adult rats that had one forelimb removed on the day of birth. Mapping of cerebral neocortex with electrical stimulation revealed an altered relationship between the motor cortex and the remaining muscles. Whereas distal forelimb movements are normally elicited at the lowest threshold in the motor cortex forelimb area, the same stimuli activated shoulder and trunk muscles in experimental animals. In addition, an expanded cortical rep

[Somatosensory evoked potentials and their dynamics among trigeminal neuralgia patients during reflexotherapy].

The study of somatosensory evoked potentials (SSEP) in patients with trigeminal neuralgia made it possible to elucidate their characteristic changes as compared to those in healthy subjects. After a course of acupuncture 75% of the patients showed correlation between some parameters of SSEP and positive clinical results. The authors discuss pathophysiological mechanisms of impairment of cortico-subcortical interactions in patients with trigeminal neuralgia and analyze the possibility of using SSEP for dynamic evaluation of the brain status in the process of treatment.

Postsynaptic dorsal column pathway of the rat. II. Evidence against an important role in nociception.

The response characteristics of neurons at the origin of the postsynaptic dorsal column (PSDC) pathway were determined in unanesthetized, decerebrated, spinalized rats. Sixty-four percent of PSDC neurons responded only to innocuous mechanical stimuli. Thirty-six percent responded to innocuous stimuli but were more powerfully activated by noxious pinch. Ninety-three percent of the tested PSDC neurons were not activated by any of several intensities of sustained, repeated noxious heating of their receptive fields. The failure of pinch-responsive PSDC cells to respond to thermal stimulation, even

Review of the literature supporting a scientific basis for the chiropractic subluxation complex.

A review of the literature reveals strong evidence for both the mechanical model of disease production (structural) and the neurobiological model (functional). Outdated models which attempt to describe a scientific basis for chiropractic theory are inadequate and indeed harmful to the progress and acceptance of chiropractic. Pragmatic or empirical arguments that "Chiropractic works and that's what counts" have served a useful purpose, but now must be augmented by extant research findings. The "paradigm shift" is on. Research investigators around the world are focusing on the multiple component

The effects of naloxone, morphine and methionine enkephalinamide on Ia afferent terminations in the cat spinal cord.

Naloxone, morphine, Met5-enkephalinamide (MENKA) and procaine were administered microelectrophoretically near extracellularly stimulated extensor muscle group Ia afferent fibres and terminations in the lumbar spinal cord of cats anaesthetized with pentobarbitone sodium. Observations were made of effects on the electrical threshold, on the depolarizing action of GABA or piperidine-4-sulphonate (P4S), and on bicuculline-sensitive primary afferent depolarization (PAD) generated by tetanic stimulation of flexor muscle low threshold afferents. All 4 agents reversibly elevated the threshold of Ia fi

Afferent connections of the cerebellum in various types of reptiles.

The origin of cerebellar afferents was studied in various types of reptiles, viz., the turtles Pseudemys scripta elegans and Testudo hermanni, the lizard Varanus exanthematicus, and the snake Python regius, with retrograde tracers (the enzyme horseradish peroxidase and the fluorescent tracer "Fast Blue"). Projections to the cerebellum were demonstrated from the nucleus of the basal optic root, the interstitial nucleus of the fasciculus longitudinalis medialis, the vestibular ganglion, and the vestibular nuclear complex, two somatosensory nuclei, viz., the descending nucleus of the trigeminal n

Spinal neurones with long projections activated from the abdominal viscera of the cat.

1. Recordings have been made from seventy-three neurones responding to electrical stimulation of pelvic, hypogastric or lumbar colonic nerves, in decerebrate or anaesthetized cats. Fifty-two of the units had long projections that ascended to the first cervical segment, and no units with visceral inputs were found to belong to the spino-cervical tract. Twenty-one units had long descending projections.2. Twenty percent (i.e. 11/46) responded to parasympathetic (pelvic) nerve stimulation (group 1) whilst 80% (35/46) responded to stimulation of hypogastric and/or lumbar colonic nerves (group 2). N

Role of the carotid body and of afferent vagal stimuli in the arousal response to airway occlusion in sleeping dogs.

We studied the independent effects of bilateral carotid body denervation and bilateral cold blockade of the vagus nerves on the arousal response to airway occlusion during sleep. Dogs breathed through a cuffed endotracheal tube inserted via a chronic tracheostomy, and airway occlusion was imposed at end-expiration and maintained until the moment of arousal. Arterial O2 saturation (SaO2) was measured with an ear oximeter, and sleep stage was determined by electroencephalographic and behavioral criteria. Mean +/- SE SaO2 values at arousal were 88.7 +/- 2.7% during slow-wave sleep, and 84.9 +/- 3

The activity of human muscle spindle endings in normal motor behavior.

In none of the above instances has the fusimotor system been found to be responsible for the described changes in stretch reflex gain. These findings do not prove that the fusimotor system is incapable of altering the gain of the stretch reflex; they do, however, establish that there are quite adequate mechanisms of gain control completely independent of the fusimotor system. One could reasonably question whether the fusimotor system ever operates primarily as a gain-controlling mechanism. That "more in" would probably result in "more out" cannot be disputed, but this may be of little real fun

Effects of cervical vagus nerve stimulation on hypothalamic neuronal activity.

The effects of cervical vagus nerve stimulation on the activity of 56 neurons recorded in various parts of the rat brain were determined. Recordings were made from neurons in both the ipsilateral and contralateral hemispheres during vagus nerve stimulation. Both frequency, 10 to 100 Hz, and voltage, 1 to 20 V, of 0.5 msec pulses were applied to the nerve in a random manner over a 4 sec period while monitoring ongoing single neuronal activity. Frequency response relationships were established for 64% of the lateral preoptic-lateral hypothalamic-medial forebrain bundle (LPA-LH-MFB) neurons which
