Calcium intake and disease prevention.

While the fundamental metabolic function of calcium is to serve as a second messenger, coupling intracellular responses to extracellular signals, nutritional deficiency of calcium is manifested at a higher level of organization: 1) depletion of the calcium nutrient reserve; 2) inadequate complexation of digestive byproducts; and 3) collateral effects of hormones produced primarily to compensate for low calcium intake. The first mechanism contributes to the osteoporosis problem, the second to kidney stones and colon cancer, and the third to hypertension, preeclampsia, obesity, and insulin resis

The activity of cholinergic neurons in the basal forebrain interferes with anesthesia-arousal process of propofol.

Previous research has demonstrated that basal forebrain (BF) regulates arousal during propofol anesthesia. However, as the BF comprises cholinergic neurons alongside two other types of neurons, the specific role of cholinergic neurons has not been definitively elucidated. In our study, calcium signal imaging was utilized to monitor the real-time activities of cholinergic neurons in the BF during propofol anesthesia. Additionally, we selectively stimulated these neurons to investigate EEG and behavioral responses during propofol anesthesia. Furthermore, we specifically lesioned cholinergic neur

Calcium-induced PII oligomerization enhances physiological performance in the cyanobacterium Synechococcus elongatus PCC 7942.

Calcium (Ca²⁺) functions as a universal signaling ion across biological systems, yet its role in cyanobacterial metabolic regulation remains … ss through reduced superoxide accumulation. These findings position PII as a key integrator linking calcium signaling to energy balance, photosynthetic performance, and stress resilience. Together, these findings uncover a previously unrecognized regulatory axis wherein Ca2+ enhances stability of PII, thereby coupling ionic signals to metabolic and physiological pr …

Tolylfluanid induces oxidative stress-associated calcium dysregulation and mitochondrial dysfunction in porcine trophectoderm and uterine luminal epithelial cells.

Tolylfluanid is an aryl sulfamide fungicide widely used in agriculture and marine industries, but its effects on reproductive processes remain poorly understood. This study aimed to evaluate the cytotoxic effects of tolylfluanid and to investigate the mechanisms underlying its action via porcine trophectoderm (pTr) and uterine luminal epithelial (pLE) cells. These cells are essential for early embryonic development and implantation. Tolylfluanid reduced cell viability in monolayer cultures and impaired spheroid formation in 3D cultures, accompanied by cell-cycle dysregulation and apoptosis. It

The Role of MRGPRX1 in the Melanogenesis of Human Primary Epidermal Melanocytes.

MRGPRX1 (mas-related G protein-coupled receptor X1) is an orphan receptor, and its function in the skin cells remains unclear. In this study, we demonstrated MRGPRX1 expression in skin cells, including melanocytes. We also found that MRGPRX1 knockdown using short hairpin RNA increased melanin content, cellular tyrosinase activity, and the expression of melanogenic proteins, including tyrosinase, tyrosinase-related protein 2, and MITF in human epidermal melanocytes. In addition, MRGPRX1 knockdown increased CRE-binding protein and p38 phosphorylation and decreased p44/42 phosphorylation. Further

Decrease in Caveolae-Gαq Interaction Mediates Pressure Overload-Induced Cardiac Remodeling in Rats.

Despite its significant clinical implications, pressure overload-induced cardiac remodeling is poorly understood. This study aimed to investigate the role of Caveolae-Gαq interaction in the pathophysiology of pressure overload-induced cardiac remodeling. We used the abdominal aortic constriction (AAC) rat model and the angiotensin II-treated cell model to simulate pressure overload-induced cardiac remodeling. Histological changes were assessed using hematoxylin-eosin staining, immunofluorescence staining, and transmission electron microscopy. The expression, colocalization, and calcium respons

Sophorolipid inhibits histamine-induced itch by decreasing PLC/IP3R signaling pathway activation and modulating TRPV1 activity.

Biosurfactants are attracting much interest due to their potential application as therapeutic agents in the medical and cosmetic field. Previous studies have demonstrated that biosurfactant such as sophorolipid (SL) exhibits immunomodulatory effects. In this article, we found the potential of sophorolipid for inhibiting histamine-induced itch and preliminarily explored its molecular basis. First, behavioral tests indicated that SL can remit the histamine-induced scratching behaviors of mice. Second, SL can suppress the the calcium influx induced by histamine, HTMT and VUF8430 in HaCaT cells. R

Transcriptome Sequencing of Peripheral Blood Mononuclear Cells from Elite Controller-Long Term Non Progressors.

The elite controller (EC)-long term non-progressor (LTNP) phenotype represent a spontaneous and advantageous model of HIV-1 control in the absence of therapy. The transcriptome of peripheral blood mononuclear cells (PBMCs) collected from EC-LTNPs was sequenced by RNA-Seq and compared with the transcriptomes from other phenotypes of disease progression. The transcript abundance estimation combined with the use of supervised classification algorithms allowed the selection of 20 genes and pseudogenes, mainly involved in interferon-regulated antiviral mechanisms and cell machineries of transcripti

Chronic Calcium Channel Inhibitor Verapamil Antagonizes TNF-α-Mediated Inflammatory Reaction and Protects Against Inflammatory Arthritis in Mice.

It is well established that the tumor necrosis factor-α (TNF-α) plays a dominant role in rheumatoid arthritis (RA). Calcium channel is recently reported to be closely associated with various inflammatory diseases. However, whether chronic calcium channel blocker verapamil plays a role in RA still remains unknown. To investigate the role of verapamil in antagonizing TNF-α-mediated inflammation reaction and the underlying mechanisms, bone marrow-derived macrophages (BMDM) cells were cultured with stimulation of TNF-α, in the presence or absence of verapamil. Inflammation-associated cytokines, in

Porcine Circovirus Type 2 Activates CaMMKβ to Initiate Autophagy in PK-15 Cells by Increasing Cytosolic Calcium.

Porcine circovirus type 2 (PCV2) induces autophagy via the 5' adenosine monophosphate-activated protein kinase (AMPK)/extracellular signal-regulated kinase (ERK)/tuberous sclerosis complex 2 (TSC2)/mammalian target of rapamycin (mTOR) pathway in pig kidney PK-15 cells. However, the underlying mechanisms of AMPK activation in autophagy induction remain unknown. With specific inhibitors and RNA interference (RNAi), we show that PCV2 infection upregulated calcium/calmodulin-dependent protein kinase kinase-beta (CaMKKβ) by increasing cytosolic Ca(2+) via inositol 1,4,5-trisphosphate receptor (IP3R

Opioid receptor agonists and Ca2+ modulation in human B cell lines.

Opiates and opioid peptides have been shown to modulate lymphocyte functions; however, little attention has been given to the type of receptors or receptor signaling mechanisms that are involved. Receptor-mediated signaling via ionized free Ca2+ is an event thought to be important in the triggering of lymphocyte activities. We report use of the calcium indicator dye, indo-1, and flow cytometry to identify B lymphocyte calcium responses to physiologic concentrations of opioid peptides. The human B cell lines Nalm 6 and JY responded to the naturally occurring opioid pentapeptide methionine-enkep

Disrupted Ca2+ homeostasis and immunodeficiency in patients with functional IP3 receptor subtype 3 defects.

Calcium signaling is essential for lymphocyte activation, with genetic disruptions of store-operated calcium (Ca2+) entry resulting in severe immunodeficiency. The inositol 1,4,5-trisphosphate receptor (IP3R), a homo- or heterotetramer of the IP3R1-3 isoforms, amplifies lymphocyte signaling by releasing Ca2+ from endoplasmic reticulum stores following antigen stimulation. Although knockout of all IP3R isoforms in mice causes immunodeficiency, the seeming redundancy of the isoforms is thought to explain the absence of variants in human immunodeficiency. In this study, we identified compound het…

Analyzing Gene Expression Profiles from Ataxia and Spasticity Phenotypes to Reveal Spastic Ataxia Related Pathways.

Spastic ataxia (SA) is a group of rare neurodegenerative diseases, characterized by mixed features of generalized ataxia and spasticity. The pathogenetic mechanisms that drive the development of the majority of these diseases remain unclear, although a number of studies have highlighted the involvement of mitochondrial and lipid metabolism, as well as calcium signaling. Our group has previously published the GBA2 c.1780G > C (p.Asp594His) missense variant as the cause of spastic ataxia in a Cypriot consanguineous family, and more recently the biochemical characterization of this variant in pat…

MXene-chitosan photo-responsive conduit for wireless optogenetic stimulation to enhance neural regeneration and functional recovery after optic nerve injury.

Optic nerve injury triggers progressive degeneration of retinal ganglion cells (RGCs) and axonal loss, driven by inhibitory microenvironmental factors such as glial scarring, myelin debris, and growth-inhibitory signaling. Physical stimuli such as photothermal and photoelectric stimulations have gained attention, yet little is known about their potential on normal cells or the optic nerve due to setbacks from over-exposure. Photothermal stimulus presents photoelectric cues and, at the same time, energy conversion for heat generation. Herein, a bio-functional platform was designed by incorporat

μ-Opioid inhibition of Ca2+ currents and secretion in isolated terminals of the neurohypophysis occurs via ryanodine-sensitive Ca2+ stores.

μ-Opioid agonists have no effect on calcium currents (I(Ca)) in neurohypophysial terminals when recorded using the classic whole-cell patch-clamp configuration. However, μ-opioid receptor (MOR)-mediated inhibition of I(Ca) is reliably demonstrated using the perforated-patch configuration. This suggests that the MOR-signaling pathway is sensitive to intraterminal dialysis and is therefore mediated by a readily diffusible second messenger. Using the perforated patch-clamp technique and ratio-calcium-imaging methods, we describe a diffusible second messenger pathway stimulated by the MOR that inh

Hydrogen peroxide activates endothelial nitric-oxide synthase through coordinated phosphorylation and dephosphorylation via a phosphoinositide 3-kinase-dependent signaling pathway.

Endothelial nitric-oxide synthase (eNOS) is an important component of vascular homeostasis. During vascular disease, endothelial cells are exposed to excess reactive oxygen species that can alter cellular phenotype by inducing various signaling pathways. In the current study, we examined the implications of H(2)O(2)-induced signaling for eNOS phosphorylation status and activity in porcine aortic endothelial cells. We found that H(2)O(2) treatment enhanced eNOS activity and NO bioactivity as determined by the conversion of l-[(3)H]arginine to l-[(3)H]citrulline and cellular cGMP content. Concom

Low-Temperature Decomposition of CaCO3 in NiO/CaCO3 and CuO/CaCO3 Powder Under H2 at Industrial Waste Heat Temperatures.

Calcium looping (CaL) is a CO2 capture technology based on the reversible carbonation of calcium salts. Calcium-based materials are highly reactive with CO2 and are generated as by-products or waste across a wide range of industries-such as cement, steel, and fisheries-leading to expectations that the calcium looping (CaL) process can be implemented at a low cost. However, a key challenge remains in the regeneration step: releasing CO2 from CaCO3, the carbonated form of the sorbent, and returning it to a reusable state conventionally requires calcination at around 700 °C, which imposes a subst

Lead inhibits meso-2,3-dimercaptosuccinic acid induced calcium transients in cultured rhesus monkey kidney cells.

Previously we have shown that meso-2,3-dimercaptosuccinic acid (DMSA, 15-500 microM) elicits concentration-dependent increases in intracellular calcium levels ([Ca2+]i) in untreated rhesus monkey kidney cells (LLC-MK2) (Pokorski et al., 1997, unpublished results). Little is known about the restorative effects of the chelating agent 2,3-dimercaptosuccinic acid on intracellular calcium homeostasis in the presence of lead. Lead interacts at numerous sites in Ca2+ homeostasis and may mimic Ca2+ to interfere with Ca2+-mediated intracellular signaling. To examine the effects of lead on [Ca2+]i and D
