Morphiceptin ( beta-casomorphin (1-4) amide): a peptide opioid antagonist in the field stimulated rat vas deferens.

The effect of Morphiceptin (H-Tyr-Pro-Phe-Pro-NH2) on opioid inhibition of field stimulated rat vas deferens (RVD) was studied. The morphiceptin was found to produce a weak inhibition of the twitch response of the RVD. In addition morphiceptin antagonized full agonists acting on the same receptor system (i.e. beta-endorphin selective antagonist for the delta receptor system of RVD by virtue of being a weak partial agonist.

SPE-55 and VHA-18 are essential for FB-MO complex assembly and transport during spermatogenesis in C. elegans.

The spatiotemporal control of asymmetric positioning of cellular cargoes, such as macromolecules and organelles, during spermatogenesis is essential for producing functional sperm in most animal species. However, the regulatory mechanisms that regulate cargo sorting and transporting remain largely unknown. Here, we demonstrate that the casein-like kinase SPE-55 localized on the surface of a membranous organelle (MO) is required for the SPE-18 localization/puncta formation and facilitates the assembly of major sperm protein (MSP) condensate, named the fibrous body (FB), in spermatocytes during

Cellular Nanoparticle-Probiotic Biohybrid for Multimodal Oral Therapy of Food Allergy.

Food allergies, driven by abnormal immune responses to dietary allergens, can cause severe and potentially life-threatening reactions. Current treatments remain limited by partial efficacy and prolonged regimens. Here, we present a biohybrid formulation composed of macrophage membrane-derived nanoparticles and Escherichia coli Nissle 1917 probiotics for targeted, multimodal oral therapy of food allergies. The nanoparticles are loaded with an allergen-degrading enzyme and conjugated onto probiotics via click chemistry. This design enables the biohybrid to neutralize pro-inflammatory cytokines,

In Vitro Evaluation of Thymus serpyllum Essential Oil Against Paracoccus marcusii IBR3: A Potential Natural Protection Strategy for Historic Mural Paintings.

Historic mural paintings are particularly vulnerable to biodeterioration caused by microbial colonization, which can lead to the degradation of the organic and inorganic components of wall surfaces. This study presents, to the best of our knowledge, the first identification of the Paracoccus marcusii IBR3 strain on a 19th-century mural painting from a private Italian villa. In vitro analyses revealed that strain IBR3 exhibits significant enzymatic activities, such as casein hydrolysis and calcium carbonate dissolution. These activities implicate the strain in the degradation of proteinaceous b

Picomolar bivalent inhibitors of protein kinase CK2 active against β-coronavirus replication.

The protein casein kinase 2 (CK2) has been shown to be upregulated in SARS-CoV-2 infections. However, few inhibitors have been tested for antiviral activity against coronaviruses. In this study, highly potent and selective bivalent inhibitors targeting the protein kinase CK2 were developed. The chemical structure of the well-known inhibitor CX-4945 was used as an anchor for the ATP-binding site and was linked by polar and aliphatic linkers to chemical structures that bind the cryptic αD pocket of CK2. The bivalent inhibitor Biv5, which demonstrated a sub-nanomolar kinase inhibition, showed pot

Characterization of the effects of high voltage pulsed electric field treatment on milk fat globule membrane proteins and phospholipid based on 31P NMR and 4D-DIA quantitative proteomics.

The milk fat globule membrane (MFGM) promotes brain development in newborns, although its extraction and recovery have been challenging. This study investigated the effects of high-voltage pulsed electric field (PEF) treatment on MFGM proteins and phospholipids. The results demonstrated that PEF treatment significantly reduced the particle size of milk fat globules, increased the absolute zeta potential value, and improved microstructure stability. Compared with pasteurization, PEF (35 kv/cm for 40 times) treatment significantly increased protein (XO/XDH, BTN, PAS6/7, and ADRP) and phospholipi

Preparation, Conformational Structure, and Proteolytic Activity of Papain Covalently Conjugated to Poly(ethylene glycol)-Tethered Lipid Bilayer Membranes.

Conjugation of enzymes to lipid membranes is a key approach to reconstitute fascinating features of cell organelles and to deduce the nature of membrane-involved biological events. In this work, papain was covalently conjugated via a cross-linker to phospholipid vesicles (liposomes) tethered with poly(ethylene glycol) (PEG) at 25 °C and pH = 7.0, followed by chromatographic purification. The size of the PEG moiety and the type of cross-linker were optimized to obtain PEG-tethered liposome-conjugated papain (liposome-PEG-papain). Slight conformational changes of the membrane-conjugated papain i

Volumetric printed biomimetic scaffolds support in vitro lactation of human milk-derived mammary epithelial cells.

The human breast is remarkably plastic and remodels with each birth to produce milk optimally suited for the changing demands of the newborn. This dynamic nature of lactation makes it challenging to study under controlled conditions. Given the health benefits of human milk, models of secretory mammary tissue would offer opportunities to study factors that influence this important food source. First, 3D models of the mammary duct/alveoli (D/A) were designed inspired by shapes found in vivo. Photoresins based on mammary decellularized extracellular matrix (dECM) were optimized to match the mecha

Diet modulates Vibrio cholerae colonization and competitive outcomes with the gut microbiota.

The amount or type of dietary macronutrients dramatically changes the microbiota and physicochemical environment of the gut. Because the microbiota plays key roles in pathogen susceptibility, macronutrients could impact infection outcomes. We show that dietary protein sources differentially restrict colonization of Vibrio cholerae (V. cholerae) as well as impact pathogen-dependent changes in microbiota composition. Specifically, dietary proteins, notably casein, alter levels of the V. cholerae central flagellar regulator, FlrA, which controls expression of the type VI secretion system (T6SS),

NUCKS1, a LINC00629-upregulated gene, facilitated osteosarcoma progression and metastasis by elevating asparagine synthesis.

Nuclear ubiquitous casein and cyclin-dependent kinase substrate 1 (NUCKS1) has been reported to play an oncogenic role in several cancers. However, the biological functions and regulatory mechanism of NUCKS1 in osteosarcoma have not been fully understood. In this study, we reported that NUCKS1 was significantly increased in osteosarcoma. Depletion of NUCKS1 decreased osteosarcoma cell proliferation and metastasis in vivo and in vitro. Overexpression of NUCKS1 accelerated osteosarcoma cell aggressiveness. Mechanistically, NUCKS1 facilitated asparagine (Asn) synthesis by transcriptionally upregu

Characterization and rational design for substrate specificity of a prolyl endopeptidase from Stenotrophomonas maltophilia.

A novel prolyl endopeptidase from Stenotrophomonas maltophilia, SmPEP, was discovered and characterized. The specific activity of the recombinant SmPEP expressed by Escherichia coli BL21 (DE3), was 68.3 U/mg at pH 8.0 and 37 °C. In order to improve the substrate specificity for long-chain peptide, rational design was applied based on the structure constructed by homology modeling. Inter-domain sites within the β-propeller domain were chosen for the mutation to weaken the inter-domain interaction and form an open conformation for long-chain substrate entering into the active site. The substrate

FTO Demethylates Cyclin D1 mRNA and Controls Cell-Cycle Progression.

N6-Methyladenosine (m6A) modification is the major chemical modification in mRNA that controls fundamental biological processes, including cell proliferation. Herein, we demonstrate that fat mass and obesity-associated (FTO) demethylates m6A modification of cyclin D1, the key regulator for G1 phase progression and controls cell proliferation in vitro and in vivo. FTO depletion upregulates cyclin D1 m6A modification, which in turn accelerates the degradation of cyclin D1 mRNA, leading to the impairment of G1 progression. m6A modification of cyclin D1 oscillates in a cell-cycle-dependent manner;

Targeting High Dynamin-2 (DNM2) Expression by Restoring Ikaros Function in Acute Lymphoblastic Leukemia.

Dynamin-2 (DNM2) is a GTPase essential for intracellular vesicle formation and trafficking, cytokinesis and receptor endocytosis. Mutations in DNM2 are common in early T-cell precursor acute lymphoblastic leukemia. However, DNM2 expression in other types of ALL are not reported. We studied DNM2 mRNA level in adults with B- and T-cell ALL. We found DNM2 is more highly expressed compared with normals in both forms of ALL. High DNM2 expression is associated with some clinical and laboratory features, inferior outcomes and with leukaemia cell proliferation. We also found Ikaros directly binds the

New anti-hepatitis B virus drugs under development and evaluation.

PURPOSE OF REVIEW: Although available therapies can effectively inhibit hepatitis B virus (HBV) replication in patients with active chronic hepatitis B (CHB) infection, therapeutic efficacy is limited because of potential drug resistance, and an inability to mediate viral clearance and to rectify immune impairment in CHB patients. This review will summarize the state-of-the-art for anti-HBV drugs and focus on potential drugs and targets under development and evaluation. RECENT FINDINGS: New developing drugs are evaluated for their antiviral effects in the areas of interference with the viral

Cloning, characterization, and bacterial over-expression of an osmotin-like protein gene from Solanum nigrum L. with antifungal activity against three necrotrophic fungi.

A new osmotin-like protein gene from Solanum nigrum L. var indica (SindOLP) was cloned and overexpressed in Escherichia coli. The full-length intron-less gene is 744 bp, encoding a mature protein of 247 amino acids with a molecular mass of 26 kDa. The protein has an N-terminal cleavable signal sequence of 21 amino acids. There is the Thaumatin family signature pattern, with one each of amidation, N-myristoylation, casein kinase II phosphorylation, tyrosine kinase phosphorylation, and protein kinase C phosphorylation sites. Hydropathy plot showed that it has six transmembrane helices. It has an

PER2 Differentially Regulates Clock Phosphorylation versus Transcription by Reciprocal Switching of CK1ε Activity.

Casein kinase 1ε (CK1ε) performs key phosphorylation reactions in the circadian clock mechanism that determine period. We show that the central clock protein PERIOD2 (PER2) not only acts as a transcriptional repressor but also inhibits the autoinactivation of CK1ε, thereby promoting CK1ε activity. Moreover, PER2 reciprocally regulates CK1ε's ability to phosphorylate other substrates. On output pathway substrates (e.g., P53), PER2 inhibits the activity of CK1ε. However, in the case of central clock proteins (e.g., CRYPTOCHROME2), PER2 stimulates the CK1ε-mediated phosphorylation of CRY2. CK1ε a
