Vascular endothelium as a regulator of granulopoiesis: production of colony-stimulating activity by cultured human endothelial cells.

Colony-stimulating activity is a regulatory factor(s) that promotes differentiation of hemopoietic stem cells to mature granulocytes and macrophages; in man it has been found that blood monocytes, lymphocytes, and tissue macrophages produce it. In an effort to identify other potenitally physiologic tissue sources of colony-stimulating activity, we have studied the capacity of primary cultures of human vascular endothelial cells to produce colony-stimulating activity. Medium conditioned by incubation with endothelial cultures contained activity that promoted granulocyte-macrophage colony format

Pathogenesis and immunology of experimental gonococcal infection: virulence of colony types of Neisseria gonorrhoeae for chicken embryos.

The virulence of gonococcal strains and colony types was evaluated in embryonated hen eggs of various ages inoculated by different routes. Striking differences in virulence of colony types were revealed by intravenous inoculation of 11-day embryos. T1 and T2 colony types were found to have high virulence for embryos, whereas T3 and T4 colony types were relatively avirulent. These observations are in accord with previous studies in volunteers. The differences in virulence were not related to differences in toxicity of killed gonococci, sonic lysates, or to the susceptibility of gonococci to cid

"They talk about it like it's an overdose crisis when in fact it's basically genocide": The experiences of Indigenous peoples who use illicit drugs in Vancouver's Downtown Eastside neighbourhood.

Indigenous Peoples who use illicit drugs (IPWUID) are disproportionately represented among toxic drug poisoning deaths in Canada. These drug-related harms are framed by the historical and ongoing trauma related to settler colonialism and are acutely visible in Vancouver, Canada's Downtown Eastside - a low-income neighbourhood that is an epicenter of the drug poisoning crisis and characterized by entrenched poverty, substance use, violence, and homelessness. This study was undertaken to examine the experiences and perspectives of IPWUID in the Downtown Eastside regarding the drug poisoning cris…

Exploring the Quellerflächen: Knowledge production on Danish internal colonial expeditions to Schleswig's west coast, 1859-1863.

Between 1859 and 1863, Danish government official Arthur Earl of Reventlow traveled to North Frisia several times to explore possible ways to develop new agricultural areas in the Wadden Sea. Although expeditions have become a focus in the history of science, little research has addressed the journeys of officials into state peripheries. While these did not travel to "exotic" landscapes like conventional expeditions, their goals were nearly identical: to expand knowledge and increase control over areas where state authority was weak. Reventlow's expeditions into the borderlands were driven by

Continuous infusion of granulocyte colony-stimulating factor is associated with an advantage in neutrophil recovery in pediatric oncologic disorders.

INTRODUCTION: The use of intensified chemotherapy has significantly improved overall survival (OS) and relapse-free survival (RFS) in cancer patients. However, it also increases the risk of chemotherapy-induced neutropenia (CIN). Granulocyte colony-stimulating factor (G-CSF) is commonly used to hasten neutrophil recovery, but limited research has focused on the superiority of different routes of G-CSF administration. MATERIALS AND METHODS: This was a randomized, prospective, single-institution study. Twenty-eight participants were enrolled and randomly assigned to experimental and control arm

Fluorine-18-labeled PET radiotracers for imaging the macrophage colony-stimulating factor 1 receptor (CSF1R).

BACKGROUND: Positron emission tomography is a useful tool for quantitative imaging of neuroinflammation in human subjects; however, more suitable radiotracers are sought. One of the promising targets for imaging neuroinflammation is macrophage colony-stimulating factor 1 receptor (CSF1R) because of its expression on activated microglia. Several PET radiotracers for imaging CSF1R have been developed, however, a suitable CSF1R radiotracer radiolabeled with 18F is not yet available. We synthesized and performed preclinical evaluation of a new series of high-affinity, 18F-labeled PET radiotracers

Efficacy of the Granulocyte Colony-Stimulating Factor in Sepsis-Associated Immunosuppression: An Open-Label Randomized Controlled Trial.

OBJECTIVE: To determine the efficacy of filgrastim (G-CSF) in reducing the mortality in children with multi-organ failure syndrome (MOFS) persisting for three consecutive days.  Methods: Children aged 1 month to 18 years with two or more organ failures persisting for three days according to Goldstein's criterion were included and randomized to receive either standard of care and filgrastim (G-CSF) at a dose of 4 mcg/kg/day subcutaneously for seven days or standard of care at a 1:1 ratio. The stored blood samples were estimated for TNF-α, A Disintegrin and Metalloproteinase Motifs 13 (ADAMTS13)

Crystallized colony-stimulating factor-1 receptor inhibitor protects immunoisolated allo but not xeno transplants in primates.

Immunoisolation devices containing therapeutic protein-secreting cells offer potential for long-term therapy without immune suppression. However, scar tissue formation driven by the foreign body response (FBR) hinders nutritional exchange and ultimately leads to graft failure. We previously showed that inhibiting the colony-stimulating factor-1 receptor (CSF1R) pathway in monocytes and macrophages can block the FBR to implanted materials. Here, we demonstrate that coencapsulation of slow-releasing CSF1R inhibitor (GW2580) crystals with human stem cell-derived β cells (SC-β) in alginate spheres

Absence of pesticide avoidance during chronic colony-level exposure modifies locomotor activity in bumble bees.

Exposure to pesticides partly depends on the foraging behavior of bees, which may exhibit indifference, deterrence, or attraction to contaminated food. In the present study, we conducted laboratory experiments to test the foraging avoidance of Bombus terrestris for honey syrup contaminated with field-realistic concentrations of the neonicotinoid acetamiprid (ACE), herbicide glyphosate (GLY), and fungicide metalaxyl-M (MET). Tests were also conducted with the recommended field concentration of a biopesticide, the sweet orange essential oil (EOE). Bees’ behavior, and lethal and sublethal effects

Proinflammatory monocyte-derived granulocyte-macrophage colony-stimulating factor fuels airway inflammation in bronchopulmonary dysplasia.

BACKGROUND: Bronchopulmonary dysplasia (BPD) remains a leading cause of morbidity and long-term respiratory complications in preterm neonates. Effective strategies for moderate-severe BPD are currently lacking. METHODS: Tracheal aspirate (TA) samples were collected from 62 preterm neonates, and the immune cell composition was analyzed using multicolor flow cytometry. Subsequently, CD14+ monocytes were isolated and purified from the TA samples for transcriptomic sequencing analysis. Cytokine arrays and enzyme-linked immunosorbent assays were utilized in the cytokine level detection from TA sam

Cardiac autolysosome dysfunction associated with reverse osmosis water in a breeding colony of prairie voles (Microtus ochrogaster).

Prairie voles (Microtus ochrogaster) from 1 of the 3 laboratory animal facilities experienced increased unexpected deaths with no clinical signs. There was a total of 30 deaths in the affected facility and 16 deaths in the other 2 unaffected facilities in the same period. Significant gross findings included cardiomegaly (6/30, 20%) and pleural effusion (4/30, 13%). Heart weights (P = .0049) and widths (P = .0047) were significantly higher in M. ochrogaster from the affected facility than the other 2. Histology revealed cardiomyocytes with intracytoplasmic basophilic granular material in most M

Stochastic models indicate rapid smallpox spread and mass mortality of Indigenous Australians after colonial exposure.

The impact of smallpox (variola) on Aboriginal communities in Australia beginning in 1789 was catastrophic and continues to cause intergenerational trauma. Historically biased perspectives and contemporary misinformation of the disease's introduction and spread impede modern-day truth-telling and efforts towards reconciliation and national healing. Understanding whether the disease entered and spread from pre-colonial Makassan (Indonesian) trade along the northern coast, or from the British First Fleet's arrival in southeastern Australia in 1788, is necessary to estimate the demographic impact

Eradicating syphilis? Alternative projects, failures, and infectious disease control in the French colonial empire (1916-1940).

The inter-war period was a time of mobilisation against syphilis in France and its colonial empire. The spread of the disease was perceived as a threat to the economic development of its colonies, particularly because of the labour shortages it might cause. In 1916, a new player appeared on the scene of the French efforts to control the disease: the Institut Prophylactique, founded by Arthur Vernes. Its project was nothing less than to eradicate the disease, and its activities in the colonies expanded significantly during the 1920s and 1930s. However, the Institut Prophylactique has been large
