Targeting the AGE-RAGE/GSK-3β axis in diabetic retinopathy: wound-healing effects of trans-anethole and FPS-ZM1 and putative GSK-3β inhibitory activity of FPS-ZM1.

OBJECTIVES: Diabetic retinopathy (DR) causes retinal detachment, capillary degeneration, and diabetic macular edema. Chronic hyperglycemia leads to the formation of advanced glycation end products (AGEs). This activates the receptor for advanced glycation end-products (RAGE) signaling pathway, resulting in inflammation, oxidative stress, and cellular issues in the retina. The AGE-RAGE axis and signaling pathways are attractive therapeutic targets. METHODS: This study investigated the protective effects of trans-anethole (TA) and the RAGE inhibitor FPS-ZM1 in an in vitro model of high glucose-

Quantitative analysis of molecular transport in the extracellular space using physics-informed neural network.

The brain extracellular space (ECS), an irregular, extremely tortuous nanoscale space located between cells or between cells and blood vessels, is crucial for nerve cell survival. It plays a pivotal role in high-level brain functions such as memory, emotion, and sensation. However, the specific form of molecular transport within the ECS remain elusive. To address this challenge, this paper proposes a novel approach to quantitatively analyze the molecular transport within the ECS by solving an inverse problem derived from the advection-diffusion equation (ADE) using a physics-informed neural ne

Probing the aggregation behavior of collagen molecules regulated by dibenzaldehyde-terminated-PEG with varying molecular weights in solution.

Rigid and fragile nature of collagen-based materials cross-linked with biocompatible aldehyde-functionalized polysaccharides remains a challenge. Drawing inspiration from the pangolins' protective barrier, we introduce a novel cross-linker with a flexible chain to impart a "rigid-flexible coupled structure" to the collagen-based matrix. Successful integration of dibenzaldehyde-terminated-PEG (DF-PEG) into collagen molecules was confirmed by XRD and FTIR analyses. CD measurements demonstrated that the intact triple-helical structure was preserved in all samples. Distinct effects of DF-PEG with

Multiscale modeling of keratin, collagen, elastin and related human diseases: Perspectives from atomistic to coarse-grained molecular dynamics simulations.

Scleroproteins are an important category of proteins within the human body that adopt filamentous, elongated conformations in contrast with typical globular proteins. These include keratin, collagen, and elastin, which often serve a common mechanical function in structural support of cells and tissues. Genetic mutations alter these proteins, disrupting their functions and causing diseases. Computational characterization of these mutations has proven to be extremely valuable in identifying the intricate structure-function relationships of scleroproteins from the molecular scale up, especially i

Olfactory receptors: transduction, diversity, human psychophysics and genome analysis.

The emerging understanding of the molecular basis of olfactory mechanisms allows one to answer some long-standing questions regarding the complex recognition machinery involved. The ability of the olfactory system to detect chemicals at sub-nanomolar concentrations is explained by a plethora of amplification devices, including the coupling of receptors to second messenger generation through GTP-binding proteins. Specificity and selectivity may be understood in terms of a diverse repertoire of olfactory receptors of the seven-transmembrane-domain receptor superfamily, which are probably dispose

Molecular insights into SEN177 binding to human glutaminyl cyclase: a combined MD and DFT study.

Aberrant upregulation of human glutaminyl cyclase (hQC) is associated with the onset and progression of neurodegenerative disorders, notably Alzheimer's disease and Huntington's disease. Consequently, inhibition of hQC activity represents a potential avenue for treatment. SEN177, a potent small-molecule hQC inhibitor, has demonstrated notable preclinical efficacy but its binding mechanism remains incompletely understood. Herein, an integrated computational investigation was performed to elucidate the molecular basis of SEN177-hQC recognition. Notably, we implemented an integrated MD-energy dec

Computational Insights into the Regioselective Hydroxylation of Nirmatrelvir Metabolized by Cytochrome P450 3A4.

Nirmatrelvir, a covalent inhibitor of SARS-CoV-2 main protease (Mpro), is the active pharmaceutical ingredient in Paxlovid─the first oral antiviral drug granted emergency use authorization by the U.S. Food and Drug Administration (FDA) for the treatment of COVID-19. Previous studies revealed that the hepatic metabolism of nirmatrelvir was predominantly mediated by cytochrome P450 3A4 (CYP3A4) and that nirmatrelvir can be metabolized to generate multiple products. However, the precise molecular mechanism underlying the regioselective metabolism of nirmatrelvir by CYP3A4 remains to be disclosed.
