Regulation of chondrocyte apoptosis in osteoarthritis by endoplasmic reticulum stress.

Osteoarthritis (OA), a common degenerative joint disease, is characterized by the apoptosis of chondrocytes as a primary pathophysiological change, with endoplasmic reticulum stress (ERS) playing a crucial role. It has been demonstrated that an imbalance in endoplasmic reticulum (ER) homeostasis can lead to ERS, activating three cellular adaptive response pathways through the unfolded protein response to restore ER homeostasis. Mild ERS exerts a protective effect on cells, while prolonged ERS that disrupts the self-regulatory balance of the ER activates apoptotic signaling pathways, leading to

Pib2 is a cysteine sensor involved in TORC1 activation in Saccharomyces cerevisiae.

Target of rapamycin complex 1 (TORC1) is a master regulator that monitors the availability of various amino acids to promote cell growth in Saccharomyces cerevisiae. It is activated via two distinct upstream pathways: the Gtr pathway, which corresponds to mammalian Rag, and the Pib2 pathway. This study shows that Ser3 was phosphorylated exclusively in a Pib2-dependent manner. Using Ser3 as an indicator of TORC1 activity, together with the established TORC1 substrate Sch9, we investigated which pathways were employed by individual amino acids. Different amino acids exhibited different dependenc

Mitochondrial Regulatory Pathways in the Pathogenesis of Alzheimer's Disease.

Alzheimer's disease (AD) is an age-associated neurodegenerative brain disorder with progressive cognitive decline that leads to terminal dementia and death. For decades, amyloid-beta (Aβ) and neurofibrillary tangle (NFT) aggregation hypotheses have dominated studies on the pathogenesis and identification of potential therapeutic targets in AD. Little attention has been paid to the mitochondrial molecular/biochemical pathways leading to AD. Mitochondria play a critical role in cell viability and death including neurons and neuroglia, not only because they regulate energy and oxygen metabolism b

Cellular iron uptake, trafficking and metabolism: Key molecules and mechanisms and their roles in disease.

Iron is a crucial transition metal for virtually all life. Two major destinations of iron within mammalian cells are the cytosolic iron-storage protein, ferritin, and mitochondria. In mitochondria, iron is utilized in critical anabolic pathways, including: iron-storage in mitochondrial ferritin, heme synthesis, and iron-sulfur cluster (ISC) biogenesis. Although the pathways involved in ISC synthesis in the mitochondria and cytosol have begun to be characterized, many crucial details remain unknown. In this review, we discuss major aspects of the journey of iron from its initial cellular uptake

T cell exhaustion and Interleukin 2 downregulation.

T cells reactive to tumor antigens and viral antigens lose their reactivity when exposed to the antigen-rich environment of a larger tumor bed or viral load. Such non-responsive T cells are termed exhausted. T cell exhaustion affects both CD8+ and CD4+ T cells. T cell exhaustion is attributed to the functional impairment of T cells to produce cytokines, of which the most important may be Interleukin 2 (IL2). IL2 performs functions critical for the elimination of cancer cells and virus infected cells. In one such function, IL2 promotes CD8+ T cell and natural killer (NK) cell cytolytic activiti

Integrated analysis of microRNAs and their disease related targets in the brain of mice infected with West Nile virus.

To determine whether cellular miRNAs play a role in West Nile virus (WNV) neuropathogenesis, we evaluated WNV-infected mice brain for the expression profile of miRNAs, their potential functions and their correlation with genes involved in inflammatory pathways. A total of 528 miRNAs and 168 mRNA genes were examined. One hundred thirty-nine miRNAs were significantly differentially expressed in WNV-infected mice brain. Ingenuity pathway analysis demonstrated that these miRNAs and their target genes are involved in pathways related to inflammatory response, immune-cell trafficking and cell death.

Hedgehog in the Drosophila testis niche: what does it do there?

Stem cell niche is a specialized microenvironment crucial to self-renewal. The testis in Drosophila contains two different types of stem cells, the germline stem cells and the somatic cyst stem cells that are sustained by their respective niche signals, thus is a good system for studying the interaction between the stem cells and their hosting niche. The JAK-STAT and BMP pathways are known to play critical roles in the self-renewal of different kinds of stem cells, but the roles of several other pathways have emerged recently in a complex signaling network in the testis niche. Reports of indep

Acquired neuromuscular weakness and early mobilization in the intensive care unit.

Survival from critical illness has improved in recent years, leading to increased attention to the sequelae of such illness. Neuromuscular weakness in the intensive care unit (ICU) is common, persistent, and has significant public health implications. The differential diagnosis of weakness in the ICU is extensive and includes critical illness neuromyopathy. Prolonged immobility and bedrest lead to catabolism and muscle atrophy, and are associated with critical illness neuromyopathy and ICU-acquired weakness. Early mobilization therapy has been advocated as a mechanism to prevent ICU-acquired w

TF and TFRC regulate ferroptosis in swine testicular cells through the JNK signaling pathway.

Transferrin (TF) is a serum glycoprotein that plays a critical role in iron metabolism and typically functions through binding to its transferrin receptor (TFRC). TF is also considered a key indicator of sperm quality and, together with TFRC, plays a critical role in regulating spermatogenesis. This study aimed to explore the effects of increased TF and TFRC expression on ferroptosis in swine testicular cells (ST cells). Our findings revealed that the overexpression of either TF or TFRC diminishes ST cell viability, increases cytotoxicity, intensifies oxidative stress damage, decreases mitocho

Profiling Initial Thrombin Generation in Cardiovascular Disease Using a High Sensitivity Coagulation Assay.

Initial thrombin (FIIa) generation is a critical trigger for the amplification and propagation phases of coagulation, driven by two distinct pathways: the tissue factor (TF)-driven and the FVIIIa/FIXa-dependent pathways. However, the clinical utility of measuring initial thrombin generation (ITG) as a marker of thrombogenicity or as a tool to monitor the efficacy of oral anti-FXa therapy remains uncertain.ITG driven by TF and the FVIIIa/FIXa complex was first measured in plasma samples from healthy adults (n = 40). This was followed by an analysis of ITG profiles in 771 consecutive patients wi

Bioinformatics Analysis of Differentially Expressed Genes in Carpal Tunnel Syndrome Using RNA Sequencing.

BACKGROUND: Carpal tunnel syndrome (CTS) is a common disease resulting from the median nerve entrapment at the wrist. Although CTS (prevalence=5%-10% in the general population) is the most common neuropathy, its molecular mechanisms need elucidation. We used bioinformatics to detect genes with differential expressions in CTS and introduce the molecular regulatory noncoding RNAs and signaling pathways involved. METHODS: The raw files of the RNA sequencing of CTS patients and controls were obtained from GEO (accession: GSE108023), and the samples were analyzed. Differentially expressed genes we

Eriodictyol in Cancer Therapy: Reviewing Mechanistic Insights and Translational Opportunities.

Eriodictyol, a naturally occurring flavanone, has appeared as a biologically versatile compound with increasing relevance in biomedical research, especially in cancers. Evidence over the past few decades indicates that eriodictyol influences cancer cell fate through coordinated modulation of cell-cycle control, survival, and regulated cell death pathways. Eriodictyol appears to reshape oncogenic signaling networks, including PI3K/Akt/mTOR and associated kinase cascades, thereby restricting proliferative capacity and lowering resistance thresholds. Studies consistently report cell-cycle arrest

MicroRNA-Mediated Autophagy and Cardiovascular Diseases: Exploring the Crossroads of Atherosclerosis and Beyond.

BACKGROUND: Atherosclerosis (AS) is a complex cardiovascular disease characterized by the accumulation of lipids, inflammatory cells, and fibrous materials within arterial walls. Autophagy plays a critical role in maintaining cellular homeostasis and regulating lipid metabolism in vascular cells. MicroRNAs (miRNAs), small non-coding RNA molecules involved in post-transcriptional gene regulation, have emerged as key modulators of autophagy and contributors to AS progression. METHODS: This review synthesizes current evidence on the interplay between miRNAs and autophagy in the context of AS and

Nano-layered tendon patch to orchestrate timely interactions between tendon stem/progenitor cells and macrophages.

Tendon regeneration remains limited by the unmet challenge of precisely controlling the intensity and duration of local inflammation, often resulting in scarred healing. Here, we present a novel, micro-thin, nano-layered patch that employs Layer-by-Layer (LbL) self-assembly for the precisely controlled release of small molecules, Oxo-M and 4-PPBP, to address this technical challenge. This novel combination of small molecules exhibits distinct functions in modulating the polarization of macrophages and the tenogenic differentiation of tendon stem/progenitor cells (TSCs). Defining PI3K Signaling as a Key Regulator of Molecular Heterogeneity and Inflammation in Glomerular Lupus Nephritis.

Lupus nephritis (LN) is a critical manifestation in systemic lupus erythematosus. However, there has been little progress in treatment outcomes, and the discovery of robust and clinically relevant molecular subtypes would be informative for guiding stratified therapy for LN. Glomerular transcriptomic datasets of LN were obtained from publicly repositories. We investigate the molecular heterogeneity of LN by analyzing renal glomerular transcriptomic data from 135 patients with LN using integrated optimal algorithms and propose a refined two-molecular subtype scheme for LN (designated GC1 and GC
