Engineering of functional auditory neurons from human induced pluripotent stem cells.

BACKGROUND: Spiral ganglion neurons (SGNs) relay auditory sensory information from the cochlea to the brain. Their loss results in permanent hearing impairment in humans due to their limited regenerative capacity. Progress in hearing restoration has been constrained by the inaccessibility of human inner ear tissue and challenges in generating functionally mature human SGN-like neurons from stem cells in vitro. METHODS: To generate human SGN-like neurons from human induced pluripotent stem cells (hiPSCs), we recapitulated key signaling pathways involved in human inner ear development. On day (

Nanomaterial-Enhanced Corneal Cross-Linking: Engineering Strategies for Transforming Keratoconus Management.

Keratoconus, a progressive corneal ectasia, remains a major cause of irreversible visual impairment worldwide. Conventional corneal cross-linking (CXL) with riboflavin/ultraviolet A (UVA) has revolutionized clinical management, yet its efficacy is still constrained by epithelial barriers, oxygen dependence, and safety concerns in thin corneas. Emerging nanotechnology provides a transformative opportunity to overcome these bottlenecks. This review highlights the enhancement of riboflavin delivery efficiency by nanocarriers, the photodynamic optimization of nano-enhanced cross-linking agents, an

Engineering Protein-Based HIV Entry Inhibitors: Advances, Challenges, and Translational Strategies.

Human immunodeficiency virus (HIV) is an enveloped virus with a remarkable capacity for genetic diversification, enabling rapid escape from host immune responses and therapeutic interventions. Despite extensive global efforts, the development of an effective vaccine has remained elusive owing to the virus's high genetic variability and antigenic diversity. Consequently, considerable effort has been directed toward the development of therapeutic agents targeting viral entry, reverse transcriptase, integrase, protease, and more recently, capsid. Although antiretroviral therapy (ART) remains the

Engineering the Vero Cell Lineage: Toward a Programmable Vaccine Manufacturing Platform.

Vero cells remain an indispensable continuous substrate for human viral vaccine manufacturing. Despite decades of empirical process optimization, intrinsic genomic instability, including segmental aneuploidy and dynamic chromatin rearrangements, continues to limit the durability of engineered phenotypes under sustained viral burden and bioreactor stress. Here, we review the expanding engineering toolkit for the Vero lineage across a three-layered functional framework: the membrane interface, cytoplasmic foundry, and nuclear blueprint, evaluating translational prospects at each level. Receptor …

Metabolic engineering of Saccharomyces cerevisiae for de novo biosynthesis of isoorientin.

The flavonoid glycoside isoorientin has demonstrated antitumor, cardioprotective, and gut-protective activities, as well as potential applications as a natural biopesticide and antimicrobial agent. However, large-scale production and industrial development of isoorientin are limited by its low abundance in plants. In this study, we used metabolic engineering to construct an isoorientin-producing budding yeast (Saccharomyces cerevisiae) cell factory. To enhance production of the intermediate naringenin, we overexpressed key genes in the shikimic acid and naringenin pathways, reinforced the cyto…

Engineering Chinese Hamster Ovary Cells to Mitigate Polysorbate Degradation in Biotherapeutics.

A recurring root cause for polysorbate degradation and associated particle observations in liquid biotherapeutics is the presence of residual host cell proteins (HCPs). Some of these HCPs can hydrolyze the polysorbate surfactant to release poorly-soluble fatty acids that accumulate over time and form particles. To tackle this industry-wide issue, we disrupted the genes for a plurality of potential polysorbate-degrading HCPs in CHO cells via two CHO cell engineering approaches. First, we sequentially knocked out six hydrolase genes in a stable recombinant mAb-expressing cell line and substantia…

Unlocking the Room Temperature Phosphorescence through Halogen Engineering in Carbazole Dimer.

Room-temperature phosphorescence (RTP) in metal-free organic materials offers immense potential for advanced optoelectronic applications. However, the rational design and fine-tuning of RTP remain challenging due to the complex correlation between molecular structure and photophysical processes. Herein, we explored the impact of intersystem crossing (ISC), spin-orbit coupling (SOC), and halogen interactions to promote RTP in crystalline brominated carbazole dimer (BrCz-D). In contrast, the unsubstituted analogue, carbazole dimer (Cz-D) exhibits thermally activated delayed fluorescence (TADF) u

Engineering of Polyisoprene Networks Enabled by Host-Guest Thiol-Ene Reaction.

Regulating cross-linking of polymers is critical for optimizing the physical properties of polymer networks. Herein, we present a strategic approach for designing polymer networks using dithiol-functionalized metal-organic frameworks (MOFs) with both one- and three-dimensional pore architectures. Upon thermal treatment, thiyl radicals were generated from the MOFs through the dissociation of S-H bonds, as confirmed by electron spin resonance measurements. Unlike in solution and bulk phases, the confinement of these radicals within the MOFs effectively suppressed homocoupling reactions, thus ena

Observation of giant dipole moments of interlayer excitons via layer engineering.

Interlayer excitons in van der Waals (vdW) heterostructures (HSs) have garnered significant attention due to their unique properties, including prolonged lifetimes and long-range transport. While extensive studies have been conducted on interlayer excitons in HSs composed of different monolayers, research on HSs formed by multilayer constituents remains limited, particularly regarding dipole moments, which play a crucial role in light-matter interactions. In this study, we investigate the dipole moments of interlayer excitons in multilayer WS₂ and InSe HSs using the quantum-confined Stark effe

Surface-State Engineering for Generation of Nonlinear Charge and Spin Photocurrents.

We systematically explore the generation of nonlinear charge and spin photocurrents using spin-orbit split surface states. This mechanism enables net DC flow along the surface plane even in centrosymmetric bulk environments like the Rashba prototype Au(111), where we characterize the main quadratic contributions by combining model predictions with density functional calculations. We further identify the Tl/Si(111) surface as a prime scenario for experimental verification; with slight doping, it develops metallic surface states featuring remarkable relativistic properties deviating from the Ras

Engineering Bacopa monnieri for improved bacoside content and its neurological evaluation.

Bacosides are triterpenoidal saponins with numerous pharmacological benefits. One of the significant drawbacks is the low availability of these bacosides. The bacoside pathway is not well elucidated, and there is no prior report of a metabolic engineering approach in this plant. In this study, we have over-expressed the active isoform of Bacopa monnieri squalene synthase (BmSQS1-OE) and silenced the B. monnieri G10H (BmG10H-1-KD), the competitive metabolic pathway, to divert the flux towards triterpene biosynthesis. Absolute quantification of bacosides in these BmSQS1(OE)-BmG10H1(KD) lines has…

Isolation, engineering and ecology of temperate phages from the human gut.

Large-scale metagenomic and data-mining efforts have revealed an expansive diversity of bacteriophages (phages) within the human gut1-3. However, functional understanding of phage-host interactions within this complex environment is limited, largely due to a lack of cultured isolates available for experimental validation. Here we characterize 134 inducible prophages originating from 252 human gut bacterial isolates using 10 different induction conditions to expand the experimentally validated temperate phage-host pairs originating from the human gut. Importantly, only 18% of computationally pr
