Invasive chronic pulmonary aspergillosis masquerading as lung malignancy in a patient with asthma and ABPA overlap.

Chronic pulmonary aspergillosis (CPA) is a slowly progressive fungal infection that commonly complicates underlying structural lung disease and is frequently misdiagnosed in tuberculosis-endemic regions. Invasive forms of CPA can closely mimic lung malignancy.A man in his late 30s with a history of repeated empirical antitubercular therapy presented with progressive dyspnoea, chronic cough with mucoid sputum, seasonal variation of symptoms and recurrent haemoptysis. Imaging revealed a large right upper lobe mass with mediastinal extension, vascular encasement and complete bronchial obstruction

Design, synthesis, antifungal activity, and molecular docking studies of benzothiazole, S-benzyl-2,4-isodithiobiuret, and thiourea derivatives of 1-hepta-O-benzoyl-β-d-maltose nanoparticles.

The surge in fungal infection-related mortality worldwide is being caused by drug resistance to well-established antifungals such azole derivatives (bifonazole, fluconazole, miconazole, and clotrimazole). Finding novel molecules that are structurally different to these could be a useful tactic to get over medication resistance that is currently on the market. In an effort to develop highly potent non-resistance antifungal agents, we reported a series of compounds with benzothiazole, S-benzyl-2,4-isodithiobiuret and thiourea derivatives of 1-hepta-O-benzoyl-β-d-maltose NPs and their antifungal

Deciphering cell wall integrity signalling in Aspergillus fumigatus: identification and functional characterization of cell wall stress sensors and relevant Rho GTPases.

The fungal cell wall, a conserved and highly dynamic structure, is essential for virulence and viability of fungal pathogens and is an important antifungal drug target. The cell wall integrity (CWI) signalling pathway regulates shape and biosynthesis of the cell wall. In this work we identified, localized and functionally characterized four putative CWI stress sensors of Aspergillus fumigatus, an airborne opportunistic human pathogen and the cause of invasive aspergillosis. We show that Wsc1 is specifically required for resistance to echinocandin antifungals. MidA is specifically required for

Synthesis and antiproliferative evaluation of L-amino acid-6-gliotoxin ester trifluoroacetates.

This study aimed to develop gliotoxin derivatives with high yield and low toxicity through fermentation optimisation and structural modification. First, the culture conditions were optimised, resulting in a yield of 208 mg L-1. Subsequently, 17 new salt-type derivatives were synthesised by introducing amino acid esters at the 6-position. These compounds retained the essential disulphide pharmacophore while exhibiting significantly reduced toxicity towards normal GES-1 cells. Representative compounds 3b and 3e exhibited good inhibitory activity against breast cancer MCF-7 and oesophageal cancer

Biofilm Formation in Aspergillus fumigatus: A Comparative Study of Strains from Different Origins.

One of the most notable aspects of Aspergillus fumigatus, and related to its dynamic adaptation, is its ability to form biofilm and produce a wide variety of secondary metabolites. The aim of this study is to advance the characterization of biofilms generated by different A. fumigatus strains across their developmental stages and analytically evaluate their structure and composition and their relationship with secondary metabolism activation. An in vitro biofilm model was standardized to investigate structural and analytical differences among strains isolated from distinct clinical settings an

Minimal domain peptides derived from enterocins exhibit potent antifungal activity.

The antimicrobial peptide (AMP) circularized bacteriocin enterocin AS-48 produced by Enterococcus sp. exhibits broad-spectrum antibacterial activity via dimer insertion into the plasma membrane to form membrane pore structures, compromising membrane integrity and leading to bactericidal activity. A specific alpha-helical region of enterocin AS-48 has been shown to be responsible for the membrane-penetrating activity of the peptide. The canon syn-enterocin peptide library, generated using rational design techniques to have ninety-five synthetic peptide variants from the truncated, linearized, m

Biosynthesis and functions of a melanoid pigment produced by species of the sporothrix complex in the presence of L-tyrosine.

Sporothrix schenckii is the etiological agent of sporotrichosis, the main subcutaneous mycosis in Latin America. Melanin is an important virulence factor of S. schenckii, which produces dihydroxynaphthalene melanin (DHN-melanin) in conidia and yeast cells. Additionally, l-dihydroxyphenylalanine (l-DOPA) can be used to enhance melanin production on these structures as well as on hyphae. Some fungi are able to synthesize another type of melanoid pigment, called pyomelanin, as a result of tyrosine catabolism. Since there is no information about tyrosine catabolism in Sporothrix spp., we cultured

Aspergillus fumigatus as a causative agent of a number of dangerous allergic diseases.

Aspergillus fumigatus is a clinically important airborne fungal pathogen capable of causing a broad spectrum of respiratory disorders. Persistent exposure or airway colonization can provoke severe allergic diseases, particularly in patients with asthma or cystic fibrosis, whereas impaired immunity or pre-existing structural lung damage may allow the development of chronic or life-threatening invasive pulmonary aspergillosis. Accordingly, the aim of this review was to synthesize current evidence on the role of A. fumigatus in the development of allergic and non-allergic diseases, with particula

Persistent presumed allergic fungal rhinosinusitis during tezepelumab therapy with improvement after revision surgery: a case report.

Allergic fungal rhinosinusitis (AFRS) is a clinically distinct subset of type 2 chronic rhinosinusitis with nasal polyps (CRSwNP), characterized by markedly elevated total IgE, fungal sensitization, and expansile disease with bony remodeling. Definitive diagnosis requires all five Bent and Kuhn criteria, including histopathological demonstration of eosinophilic mucin without tissue invasion and a positive fungal stain. Tezepelumab, a monoclonal antibody against thymic stromal lymphopoietin (anti-TSLP), has phase 3 evidence in severe CRSwNP but has not been evaluated specifically in AFRS. We re

Exploring the Arsenal of Novel Antifungal Drug Targets for Combating Fungal Infections.

Fungal infections contribute to over 1.5 million fatalities each year, with cutaneous mycoses standing as prominent global infections. The spectrum of these mycoses varies widely, encompassing enduring afflictions like ringworm, localized infections such as tinea capitis, recurrent instances like vaginal candidiasis, and potentially fatal systemic infections impacting multiple organ systems. The escalating recognition of the health and socioeconomic ramifications associated with fungal pathogens underscores their importance in contemporary discourse. On a global scale, projections indicate tha

Invasive Fungal Infection.

BACKGROUND: The incidence of invasive fungal infection is approximately 6 cases per 100 000 persons per year. It is estimated that only half of such infections are detected during the patient's lifetime, making this one of the more common overlooked causes of death in intensive-care patients. The low detection rate is due in part to the complexity of the diagnostic work-up, in which the clinical, radiological, and microbiological findings must be considered. Fungi with resistance to antimycotic drugs have been found to be on the rise around the world. METHODS: This review is based on pertinen

Neutral Processes Drive Seasonal Assembly of the Skin Mycobiome.

The importance of microorganisms to human skin health has led to a growing interest in the temporal stability of skin microbiota. Here we investigated the dynamics and assembly of skin fungal communities (mycobiomes) with amplicon sequencing of samples collected from multiple sites on 24 healthy Chinese individuals across four seasons (in the order of winter, spring, summer, and autumn in a calendar year). We found a significant difference in community compositions between individuals, and intrapersonal community variation increased over time at all body sites. Within each season, the frequenc

Design, synthesis, and antifungal activity of novel amide imidazole CYP51 inhibitors with aromatic fused ring hydrophobic side chains.

Invasive fungal infections caused by Candida albicans are becoming increasingly severe, creating an urgent need to explore new antifungal drugs. Compound YW01 is a structurally novel CYP51 inhibitor that was screened by our research group in the preliminary stage. To enhance its activity, three rounds of structural optimization and modification were conducted in this study. Through in vitro antifungal activity testing and time-kill curve analysis, it was found that compound B3 exhibited potent antifungal activity, which was superior to that of the positive control drug fluconazole. Further res

Efficiency of newly prepared thiazole derivatives against some cutaneous fungi.

A series of fourteen novel synthesized arylazothiazole and arylhydrazothiazole derivatives were tested for their antifungal activity and structure-activity relationship. The activity of the compounds depends mainly on the side chains of the nucleus compound. The antifungal activity was more significant when both side chains are aromatic > one aromatic and one aliphatic and substituted aromatic with CH3 or OCH3 > non-substituted > substituted aromatic with chloro- or nitro-groups. Thiazole derivatives 7a, 7c, 7e, 7f, 7 g, 7i, 7 m, and 11a showed the most effective as antifungal compounds and we

A Designed Tryptophan- and Lysine/Arginine-Rich Antimicrobial Peptide with Therapeutic Potential for Clinical Antibiotic-Resistant Candida albicans Vaginitis.

New therapeutic agents for Candida albicans vaginitis are urgently awaiting to be developed because of the increasing antibiotic resistance of C. albicans. Antimicrobial peptides (AMPs) are one of the most promising choices for next-generation antibiotics. In this study, novel peptides were designed based on snake venom antimicrobial peptide cathelicidin-BF to promote anti-C. albicans activity and decrease side-effects. The designing strategies include substitutions of charged or hydrophobic amino acid residues for noncharged polar residues to promote antimicrobial activity and insertion of a
