Effect of the TIM-3/Gal-9 signaling pathway on macrophage polarization in peri-implantitis.

INTRODUCTION: Peri-implantitis (PI) is a common complication of oral implant surgeries. Understanding the role of T-cell immunoglobulin and mucin domain-containing protein 3 (TIM-3) and its ligand galectin-9 (Gal-9) in PI is essential for advancing treatment strategies. This study investigated the effect of the TIM-3/Gal-9 signaling pathway on macrophage polarization in PI. METHODS: We included 31 PI patients and 30 controls with healthy implants. Peripheral blood and peri-implant tissue samples were collected. Serum Gal-9 and cytokine levels were measured using ELISA. CD86, CD206, and TIM-3

The Role of Myocardial Fibrosis and Bioenergetic Dysfunction in Heart Failure Prognosis: A Study Protocol.

Introduction The blockage of the renin-angiotensin-aldosterone and sympathetic nervous systems is fundamental in heart failure (HF) treatment; nonetheless, other mechanisms related to myocardial fibrosis and bioenergetic dysfunction also have a relevant role in the genesis and progression of this syndrome. We aim to stratify the short-term outcome of patients hospitalized with acute decompensated heart failure (ADHF) in New York Heart Association (NYHA) class III or IV based on myocardial fibrosis biomarkers galectin-3 (Gal-3), suppression of tumorigenicity 2 (ST2) and growth differentiation f

Circulating Biomarkers and Recurrence of Persistent Atrial Fibrillation After Electrical Cardioversion.

BACKGROUND: Rhythm control therapy is recommended for individuals with symptomatic atrial fibrillation (AF) to reduce symptoms and improve quality of life. Electrical cardioversion (ECV) is used to restore sinus rhythm (SR), but AF recurrence is common. OBJECTIVE: We aimed to investigate if a selection of circulating biomarkers can predict rhythm outcomes in individuals with persistent AF treated with ECV. DESIGN: This was an observational cohort study. METHODS: We included 200 individuals aged ⩾ 18 years referred for ECV of AF from November 2017 to March 2022. We obtained blood samples 0 t

Noninvasive biomarkers identify eosinophilic esophagitis: A prospective longitudinal study in children.

BACKGROUND: Esophageal histology is critical for diagnosis and surveillance of disease activity in eosinophilic esophagitis (EoE). A validated noninvasive biomarker has not been identified. We aimed to determine the utility of blood and urine eosinophil-associated proteins to diagnose EoE and predict esophageal eosinophilia. METHODS: Blood and urine were collected from children undergoing endoscopy with biopsy. Absolute eosinophil count (AEC), plasma eosinophil-derived neurotoxin (EDN), eosinophil cationic protein (ECP), major basic protein-1 (MBP-1), galectin-10 (CLC/GAL-10), Eotaxin-2 and E

Role of Lgals9 Deficiency in Attenuating Nephritis and Arthritis in BALB/c Mice in a Pristane-Induced Lupus Model.

OBJECTIVE: In systemic lupus erythematosus (SLE), an autoimmune disease associated with multiple organ involvement, the development of lupus nephritis determines prognosis, and arthritis impairs quality of life. Galectin 9 (Gal-9, Lgals9) is a β-galactoside-binding lectin that has been used for clinical application in autoimmune diseases, since recombinant Gal-9, as a ligand for T cell immunoglobulin and mucin domain-containing protein 3 (TIM-3), induces apoptosis of activated CD4+TIM-3+ Th1 cells. This study was undertaken to investigate whether deficiency of Lgals9 has beneficial or deleteri

Absence of mutations in four genes encoding for congenital cataract and expressed in the human brain in Tunisian families with cataract and mental retardation.

BACKGROUND: To identify the genetic defect associated with autosomal recessive congenital cataract (ARCC), mental retardation (MR) and ARCC, MR and microcephaly present in most patients in four Tunisian consanguineous families. METHODS: We screened four genes implicated in congenital cataract by direct sequencing in two groups of patients; those affected by ARCC associated to MR and those who presented also microcephaly. Among its three genes PAX6, PITX3 and HSF4 are expressed in human brain and one gene LIM2 encodes for the protein MP20 that interact with the protein galectin-3 expressed in

Lymphocyte activation gene 3 is increased and affects cytokine production in rheumatoid arthritis.

BACKGROUND: Lymphocyte activation gene-3 (LAG-3) inhibits T cell activation and interferes with the immune response by binding to MHC-II. As antigen presentation is central in rheumatoid arthritis (RA) pathogenesis, we studied aspects of LAG-3 as a serological marker and mediator in the pathogenesis of RA. Since Galectin-3 (Gal-3) is described as an additional binding partner for LAG-3, we also aimed to study the functional importance of this interaction. METHODS: Plasma levels of soluble (s) LAG-3 were measured in early RA patients (eRA, n = 99) at baseline and after 12 months on a treat-to-

Probiotics Ameliorate Atrial Fibrillation-Associated Biomarkers and Inflammation in Rats via Modulation of the Gut Microbiota and NLRP3 Inflammasome Pathway.

BACKGROUND: The gut-heart axis, particularly the role of gut microbiota and the NOD-like receptor family pyrin domain containing 3 (NLRP3) inflammasome, is increasingly implicated in the pathogenesis of atrial fibrillation (AF). While probiotics show therapeutic potential, their mechanism in AF remains unclear. OBJECTIVES: This study investigates whether probiotics mitigate AF pathology by restoring gut microbiota homeostasis and inhibiting NLRP3 activation. METHODS: An AF model was induced in rats via acetylcholine (ACH)/CaCl₂ infusion. Animals were divided into five groups: Sham, AF, AF+Pr

A disintegrin and metalloproteinase with thrombospondin motifs 4-targeted molecular magnetic resonance imaging for early detection of extracellular matrix remodeling in a porcine model of abdominal aortic aneurysm.

BACKGROUND: Abdominal aortic aneurysm (AAA) progression is driven by extracellular matrix (ECM) proteolysis and vascular smooth muscle cell loss, processes insufficiently captured by diameter-based surveillance. A disintegrin and metalloproteinase with thrombospondin motifs 4 (ADAMTS4) is upregulated during early ECM remodeling, making it a compelling target for molecular imaging. METHODS: Eighteen female German Landrace swine were enrolled. Eight animals underwent AAA induction and four served as controls; six animals were excluded due to procedural complications. Molecular 3 T MRI was perfo

Mentha spicata and Its Bioactive Compound Carvone Ameliorate Metabolic Disturbance and Ovarian Dysfunction in Experimental Rat Model of PCOS by Suppression of SREBP1/TLR4-Dependent Pathway.

PROBLEM: Polycystic ovarian syndrome (PCOS) is a common endocrine and metabolic disorder in women of child-bearing age, often linked to ovarian dysfunction and complications beyond infertility. However, its management remains suboptimal due to unclear etiology. Sterol Regulatory Element Binding Protein 1 (SREBP1) and Toll-like Receptor 4 (TLR4) have been implicated in lipid metabolism and immune responses, but their interaction with ovarian function in PCOS remains unclear. Mentha spicata (spearmint) traditionally recognized for its hormonal and metabolic benefits, and its bioactive component,

Mineralocorticoid Receptors in Vascular Smooth Muscle: Blood Pressure and Beyond.

After half a century of evidence suggesting the existence of mineralocorticoid receptors (MR) in the vasculature, the advent of technology to specifically knockout the MR from smooth muscle cells (SMCs) in mice has elucidated contributions of SMC-MR to cardiovascular function and disease, independent of the kidney. This review summarizes the latest understanding of the molecular mechanisms by which SMC-MR contributes to (1) regulation of vasomotor function and blood pressure to contribute to systemic and pulmonary hypertension; (2) vascular remodeling in response to hypertension, vascular inju

Phenotypic characterization of testicular immune cells expressing immune checkpoint molecules in wild-type and pituitary adenylate cyclase-activating polypeptide-deficient mice.

PROBLEM: Pituitary adenylate cyclase-activating polypeptide (PACAP) is a neuropeptide having several regulatory functions in the nervous system and in peripheral organs including those of the reproductive system. PACAP-deficient male mice have several morphological, biochemical, behavioral defects and show disturbed signaling in spermatogenesis affecting fertility in PACAP KO mice. Reproductive functions such as fertility, mating, and maternal behaviors have been widely investigated, but no immune analyses are available regarding the testicular immune-privileged environment in male PACAP-defic

Galectin-9 as a Predictive Marker for the Onset of Immune-Related Adverse Effects Associated with Anti-CCR4 MoAb Therapy in Patients with Adult T Cell Leukemia.

Adult T-cell leukemia/lymphoma (ATL/ATLL) is one of the most malignant lymphomas with poor prognosis. ATL/ATLL cells express CC chemokine receptor 4, and mogamulizumab (anti-CCR4 monoclonal antibody) exhibits strong cytotoxicity for ATL/ATLL cells. We analyzed plasma samples of 6 patients with ATL/ATLL treated with chemotherapy followed by mogamulizumab therapy (mogatherapy) for changes in the levels of biomarkers in relation to immune-related adverse effects. As treatment is often associated with skin eruptions, we investigated the profiles of inflammatory cytokines, including galectin-9 (Gal

Expression of galectin-1, a new component of slit diaphragm, is altered in minimal change nephrotic syndrome.

Nephrin is an essential structural component of the glomerular slit diaphragm (SD), a highly organized intercellular junction that constitutes the ultrafiltration barrier of the kidney. Recent studies have identified two additional nephrin-interacting SD proteins (NEPH1 and NEPH2), suggesting that the zipper-like pattern of the SD is formed through complex intra- and intermolecular interactions of these proteins. However, the complexity of the SD structure suggests that additional SD components remain to be discovered. In this study, we identified galectin-1 (Gal-1) as a new component of the S
