Molecular markers of paragangliomas/pheochromocytomas.

Paragangliomas/pheochromocytomas comprise rare tumors that arise from the extra-adrenal paraganglia, with an incidence of about 2 to 8 per million people each year. Approximately 40% of cases are due to genetic mutations in at least one out of more than 30 causative genes. About 25-30% of pheochromocytomas/paragangliomas develop under the conditions of a hereditary tumor syndrome a third of which are caused by mutations in the VHL gene. Together, the gene mutations in this disorder have implicated multiple processes including signaling pathways, translation initiation, hypoxia regulation, prot

The routes for transformation of follicular lymphoma.

PURPOSE OF REVIEW: Aggressive transformation, a frequent event in the natural history of follicular lymphoma, is associated with increased lymphoma-related mortality and yet the underlying biology remains poorly defined. This review outlines recent advances in our understanding of the genetic basis and evolutionary process leading to transformation. RECENT FINDINGS: Both the antecedent indolent and transformed follicular lymphoma (tFL) arise through branched divergent evolution with tumors emerging from a founder precursor population, the common progenitor cell. Although the majority of tFLs

New aspects of Helicobacter pylori infection involvement in gastric oncogenesis.

Gastric adenocarcinoma not located in the cardia still remains second only to lung cancer as the leading cause of cancer-related mortality worldwide, whereas adenocarcinoma of the cardia and gastroesophageal junction has been rapidly rising over the past two decades. Gastric malignancy can be subdivided into diffuse and intestinal pathologic entities that have different epidemiological and prognostic features. Various genetic and environmental factors lead to either abnormal gene overexpression or inappropriate expression of normal genes, whose products confer the malignant phenotype. Advances

[Penis cancer. The etiological importance of papilloma viruses].

Few studies on molecular biological and genetical studies on penile carcinomas have been published, owing to the low incidence of such tumours. Potential aetiological factors of a viral nature have, however, been the subject of a variety of investigation: DNA sequences of human papillomaviruses HPV 16 and (to a lesser extent) HPV 18 were found in the majority of cases. HPV 16 sequences were found to be integrated into the genome of primary and metastatic penile tumours, suggesting an aetiological significance of HPV 16 in the development of penile carcinomas.

The expression of the sarco/endoplasmic reticulum Ca2+-ATPases in thyroid and its down-regulation following neoplastic transformation.

Maintaining a high Ca(2+) concentration in the lumen of the endoplasmic reticulum (ER), by the action of sarco/endoplasmic reticulum Ca(2+)-ATPases (SERCAs), is important in many cellular processes, such as Ca(2+)-mediated cytosolic signaling in response to extracellular stimuli, cell growth and proliferation, and synthesis, processing and folding of ER-translated proteins. In the thyroid gland, SERCAs have not been studied yet, and there is little information available on general problems such as the expression of SERCAs following neoplastic transformation. In this study we investigated the e

Analysis of High-Risk Neuroblastoma Transcriptome Reveals Gene Co-Expression Signatures and Functional Features.

Neuroblastoma represents a neoplastic expansion of neural crest cells in the developing sympathetic nervous system and is childhood's most common extracranial solid tumor. The heterogeneity of gene expression in different types of cancer is well-documented, and genetic features of neuroblastoma have been described by classification, development stage, malignancy, and progression of tumors. Here, we aim to analyze RNA sequencing datasets, publicly available in the GDC data portal, of neuroblastoma tumor samples from various patients and compare them with normal adrenal gland tissue from the GTE

CCDC6, a ferroptosis-related gene, modulates stemness features of pancreatic cancer cells in vitro.

OBJECTIVE: Pancreatic cancer (PC) poses a significant threat to patient quality of life. Ferroptosis, a novel form of non-apoptotic cell death, is implicated in pancreatic tumorigenesis and therapy resistance. This study aims to identify key ferroptosis-related genes (FRGs) as prognostic biomarkers, to construct a risk model for predicting patient outcomes, and to explore the underlying biological mechanisms. METHODS: We integrated transcriptomic and single-cell RNA sequencing (scRNA-seq) data to develop a prognostic model. We used cross-validation across three machine learning algorithms to

Single-cell profiling reveals distinct populations of tumor-associated macrophages and metastatic tumor cells in breast cancer brain metastasis.

Breast cancer brain metastasis (BCBM) is a major cause of breast cancer-related mortality, but the molecular mechanisms underlying its progression remain poorly understood. Here, we profiled the tumor immune microenvironment of BCBM at single-cell resolution and identified candidate regulators associated with brain metastatic progression. Single-cell RNA sequencing (scRNA-seq) was performed on brain metastatic tissue, adjacent tumor tissue, cerebrospinal fluid, and circulating tumor cells (CTCs) from seven patients, and bulk DNA sequencing was conducted on primary breast tumors, peripheral blo

Single-Cell Molecular Profiling of Head and Neck Squamous Cell Carcinoma Reveals Five Dysregulated Signaling Pathways Associated With Circulating Tumor Cells.

OBJECTIVES: To determine the dysregulated signaling pathways of head and neck squamous cell carcinoma associated with circulating tumor cells (CTCs) via single-cell molecular characterization. INTRODUCTION: Head and neck squamous cell carcinoma (HNSCC) has a significant global burden and is a disease with poor survival. Despite trials exploring new treatment modalities to improve disease control rates, the 5 year survival rate remains low at only 60%. Most cancer malignancies are reported to progress to a fatal phase due to the metastatic activity derived from treatment-resistant cancer cells

circRNA-miRNA-mRNA network in age-related macular degeneration: From construction to identification.

The aim of the present study was to investigate the pathogenesis of age-related macular degeneration (AMD) by constructing a regulatory circRNA-miRNA-mRNA network. By adjusting the P value to <0.05 and the absolute log value of fold change to >0.25, 2920 and 1057 differentially expressed mRNAs were identified from GSE50195 and GSE29801, respectively. Based on a literature review, Starbase database analysis, and RNA hybrid assays, we obtained 77 miRNA-mRNA and 331 circRNA-miRNA pairs. After combining these pairs, we constructed a circRNA-miRNA-mRNA network possessing 303 circRNA nodes, 4 miRNA

CALB2 is a Mechanoresistance Gene in Metastatic Prostate Cancer.

It remains unclear how circulating tumor cells (CTCs) adapt to and survive mechanical forces in the bloodstream, including fluid shear stress (FSS), making it critical to define the mechanisms that enable their survival and promote metastasis. To model these forces, we generated "mechanoresistant" (MR) LNCaP and PC3 prostate cancer (PCa) cell lines by repeatedly exposing cells to high‑intensity (HI) FSS (3950 dyn/cm2). LNCaP MR cells acquired marked resistance to HI FSS, exhibiting significantly reduced apoptosis, while PC3 cells demonstrated innate resistance. Bulk RNA‑sequencing revealed tha

Biomechanical profiles and molecular regulation of clinical cancer stem cells and tumor cells: Implications for tumorigenicity and metastasis of lung cancer.

The biomechanics of clinical cancer stem cells (CSCs) and tumor cells (TCs) play a crucial role in cancer progression. We compared the biomechanical profiles of clinical CSCs and TCs from untreated patients with lung benign (LB) and malignant (LM) tumors, including lung adenocarcinoma (LADC) and lung squamous cell carcinoma (LSCC). The results showed that TCs were mechanically stiffer than CSCs, with a greater stiffness increase in LB tumors compared to LM tumors., indicating that LB tumors have softer CSCs and stiffer TCs than those of LM tumors. The mechanical properties of TCs and CSCs from

SPI1-mediated MIR222HG transcription promotes proneural-to-mesenchymal transition of glioma stem cells and immunosuppressive polarization of macrophages.

Background: Glioma stem cells (GSCs) are a key factor in glioblastoma (GBM) development and treatment resistance. GSCs can be divided into the mesenchymal (MES) and proneural (PN) subtypes, and these two subtypes of GSCs can undergo interconversion under certain conditions. MES GSCs have higher malignancy and radioresistance and are closely associated with an immunosuppressive microenvironment. Long noncoding RNAs (lncRNAs) play a broad role in GBM, while the role of GSCs subtype remains unknown. Methods: We performed RNA sequencing to explore the lncRNA expression profile in MES- and PN-subty
