Piezo channels for skeletal development and homeostasis: Insights from mouse genetic models.

Piezo1 and Piezo2 are recently discovered mechanosensory ion channels. Piezo channels transduce mechanical stimulation into cellular signaling in a variety of tissues and organ systems. The functional roles of Piezo1 and Piezo2 have been revealed in both developmental and physiological scenarios by using mouse genetic models. Mechanotransduction by Piezo1 channels regulates osteoblast/osteocyte activity and, thus, strengthens the skeleton enabling it to adapt to a wide range of mechanical loadings. Deletion of the Piezo1 gene in the developing skeleton causes bone malformations that lead to sp

Immunopathology of BCG infection in genetically resistant and susceptible mouse strains.

Natural resistance to Mycobacterium bovis (BCG) is under the control of a single gene, designated Bcg. Resistant (Bcgr) mice prevent multiplication of an i.v. injected inoculum of congruent to 10(4) dispersed BCG cells, whereas progressive multiplication of this pathogen occurs in the first 3 wk of infection in spleens and livers of susceptible (Bcgs) mice. Striking differences in the development of cellular immunity, as measured by granuloma formation in the liver and spleen, delayed-typed hypersensitivity, and a resistance to the challenge with homologous (BCG) and heterologous (Listeria mon

Novel GnRH analogue decorated paclitaxel and CQDs co-loaded theranostic zein nanoparticles for treatment of breast cancer: Formulation optimization, in vitro and in vivo evaluation.

Breast cancer is a diverse disease with intertumoral as well as intratumoral heterogeneity, which results in different treatment regimens, disease prognosis, and treatment outcomes. This heterogeneity generally arises due to genetic and epigenetic changes. This requires a delivery system that can treat the tumor as well as provide real-time monitoring of the tumor, i.e., size and location. Hence, current research work is focused on the development of biodegradable and cost-effective ligand-decorated theranostic zein nanoparticles (NPs) prepared by co-loading of an imaging agent, i.e., carbon q

Transmission of hepatitis B virus from an orthopedic surgeon with a high viral load.

BACKGROUND: During the evaluation of a needle-stick injury, an orthopedic surgeon was found to be unknowingly infected with hepatitis B virus (HBV) (viral load >17.9 million IU/mL). He had previously completed two 3-dose series of hepatitis B vaccine without achieving a protective level of surface antibody. We investigated whether any surgical patients had acquired HBV infection while under his care. METHODS: A retrospective cohort study of all patients who underwent surgery by the surgeon was conducted. Patients were notified of their potential exposure and need for testing, and samples with

Articular cartilage and biomechanical properties of the long bones in Frzb-knockout mice.

OBJECTIVE: Ligands and antagonists of the WNT pathway are linked to osteoporosis and osteoarthritis. In particular, polymorphisms in the FRZB gene, a secreted WNT antagonist, have been associated with osteoarthritis. The aim of this study was to examine cartilage and bone in Frzb(-/-) mice. METHODS: The Frzb gene in mice was inactivated using a Cre/loxP strategy. Three models of osteoarthritis were used: collagenase, papain, and methylated bovine serum albumin induced. Bone biology was studied using density measurements and microfocal computed tomography. Bone stiffness and mechanical loading

On the pathogenesis of osteogenesis imperfecta: some insights of the Utah paradigm of skeletal physiology.

The pathogenesis of osteogenesis imperfecta (OI) baffled physiologists and physicians for over a century. Most past efforts to explain it depended heavily on cell and molecular biology and on changes in the material properties of affected bones (an old idea that OI patients could not make enough bone erred). To such views the still-evolving Utah paradigm of skeletal physiology can add a model for bone and bones that depends on errors in three genetically-determined features. The errors include, 1,2) elevated 'set points' of the strain-dependent thresholds that help to control how lamellar bone

The distribution of the parasitic fauna dictates the distribution of the haemochromatosis genes.

No satisfactory explanation has been offered, to date, to account for the prevalence of the haemochromatosis genes in the European population and yet relative paucity of the gene in the tropics. Traditional wisdom suggests that, in antiquity, the haemochromatosis gene, which promotes iron absorption, would have protected ancient man from iron loss resulting from injury either during hunting or through war. However, such an advantage would be equally desirable for other populations where the incidence of the alleles is negligible. Others have tackled the polemic from the another view, postulati

Impact of PICALM and CLU on hippocampal degeneration.

PICALM and CLU are two major risk genes of late-onset Alzheimer's disease (LOAD), and there is strong molecular evidence suggesting their interaction on amyloid-beta deposition, hence finding functional dependency between their risk genotypes may lead to better understanding of their roles in LOAD development and greater clinical utility. In this study, we mainly investigated interaction effects of risk loci PICALM rs3581179 and CLU rs11136000 on hippocampal degeneration in both young and elderly adults in order to understand their neural mechanism on aging process, which may help identify rob

HLA-DQA1/B1 alleles as putative susceptibility markers in congenital toxoplasmosis.

Host and parasite genotypes are among the factors associated with congenital toxoplasmosis pathogenesis. As HLA class II molecules play a key role in the immune system regulation, the aim of this study was to investigate whether HLA-DQA1/B1 alleles are associated with susceptibility or protection to congenital toxoplasmosis. One hundred and twenty-two fetuses with and 103 without toxoplasmosis were studied. The two study groups were comparable according to a number of socio-demographic and genetic variables. HLA alleles were typed by PCR-SSP. In the HLA-DQA1 region, the allele frequencies show

Genotypic and phenotypic analysis of antimicrobial resistance in Salmonella enterica isolates from retail chicken products in Kuwait.

Antimicrobial resistance in non-typhoidal Salmonella is recognized as an important threat to food safety and public health. The objectives of this study were to characterize the antimicrobial resistance patterns, molecular epidemiology and agreement between genetic resistance markers and phenotypic susceptibility profiles among Salmonella enterica isolates found in chicken products in Kuwait. Seventy-two Salmonella enterica isolates were tested for susceptibility to eleven antibiotics using the disk-diffusion method. Whole genome sequencing was performed to test the genotypic antibiotic resist

Genetic Determinants of Spontaneous Hepatitis B Surface Antigen Seroclearance and Its Associated Viral Markers.

BACKGROUND: Chronic hepatitis B virus (HBV) rarely leads to spontaneous hepatitis B surface antigen (HBsAg) seroclearance. However, host genetics contributing to this outcome remain poorly understood. We investigate genetic variants associated with HBsAg seroclearance and related viral markers. METHODS: We conducted GWAS in 3240 REVEAL-HBV participants to identify SNPs/HLA variants associated with HBV DNA load, hepatitis B e antigen (HBeAg) seropositivity, and HBsAg level. Variants with suggestively pleiotropic effects (p < 1 × 10-5) were tested for association with spontaneous HBsAg seroclea

Donor-specific pathological features associate with genetic background, lesion type distribution, and clinical heterogeneity in multiple sclerosis.

Multiple sclerosis (MS) shows pronounced pathological and clinical variability between individuals, reflecting differences in genetic susceptibility, inflammatory activity, and tissue repair. This variability complicates efforts to relate lesion pathology to clinical trajectories. In previous work in the Netherlands Brain Bank MS autopsy cohort (NBB-MS), we showed that relative proportions of different lesion types, lesion load, and microglia/macrophage activity score, associate with clinical severity, while also revealing marked inter-individual variability. Here, we extend these observations

Brain and serum lipidomic profiles implicate Lands cycle acyl chain remodeling association with APOEε4 and mild cognitive impairment.

INTRODUCTION: At least one-third of the identified risk alleles from Genome-Wide Association Studies (GWAS) of Alzheimer's disease (AD) are involved in lipid metabolism, lipid transport, or direct lipid binding. In fact, a common genetic variant (ε4) in a cholesterol and phospholipid transporter, Apolipoprotein E (APOEε4), is the primary genetic risk factor for late-onset AD. In addition to genetic variants, lipidomic studies have reported severe metabolic dysregulation in human autopsy brain tissue, cerebrospinal fluid, blood, and multiple mouse models of AD. METHODS: We aimed to identify an

Low proviral load in the Kumamoto strain of Japanese Brown cattle infected with the bovine leukemia virus.

BACKGROUND: The Kumamoto strain of Japanese Brown (JBRK) cattle is a sub-breed of Wagyu and has a different genetic background than that of Japanese Black (JB) cattle. Bovine leukemia virus (BLV) is the pathogen causing enzootic bovine leukosis (EBL), the predominant type of bovine leukosis (BL). EBL is one of the most common bovine infectious diseases in dairy countries, including Japan. Some host genetic factors, including the bovine leukocyte antigen (BoLA)-DRB3 gene, have been associated with the proviral load (PVL) of BLV and/or onset of EBL. Here, we determined the number of BL cases by

Peutz-Jeghers syndrome without STK11 mutation may correlate with less severe clinical manifestations in Chinese patients.

BACKGROUND: Peutz-Jeghers syndrome (PJS) is an autosomal dominant genetic disease with skin mucosal pigment spots and gastrointestinal (GI) multiple hamartoma polyps as clinical characteristics. At present, it is considered that the germline mutation of STK11 gene is the genetic cause of PJS. However, not all PJS patients can be detected STK11 germline mutations. The specific clinical characteristics of these PJS patients without STK11 mutation is an interesting clinical question. Or, like wild type GI stromal tumor, whether these PJS without STK11 mutation are also called PJS is worth discuss

Male reproductive ageing: a radical road to ruin.

In modern post-transition societies, we are reproducing later and living longer. While the impact of age on female reproductive function has been well studied, much less is known about the intersection of age and male reproduction. Our current understanding is that advancing age brings forth a progressive decline in male fertility accompanied by a reduction in circulating testosterone levels and the appearance of age-dependent reproductive pathologies including benign prostatic hypertrophy and erectile dysfunction. Paternal ageing is also associated with a profound increase in sperm DNA damage

Highly parallel introduction of nucleic acids into mammalian cells grown in microwell arrays.

High-throughput cell-based screens of genome-size collections of cDNAs and siRNAs have become a powerful tool to annotate the mammalian genome, enabling the discovery of novel genes associated with normal cellular processes and pathogenic states, and the unravelling of genetic networks and signaling pathways in a systems biology approach. However, the capital expenses and the cost of reagents necessary to perform such large screens have limited application of this technology. Efforts to miniaturize the screening process have centered on the development of cellular microarrays created on micros

A gE/gI/US9-triple deletion inactivated vaccine provides complete protection against emerging PRV variant infection.

Since 2011, pseudorabies virus (PRV) variant strains have emerged in China, against which conventional vaccines confer inadequate protection, resulting in substantial economic losses. Consequently, there is an urgent need to develop a novel vaccine capable of effectively addressing this challenge. Here, the homology-directed repair (HDR)-CRISPR/Cas9 technology was employed to specifically target and knockout the gI, gE, and US9 genes, which were further developed into inactivated vaccines. Subsequently, these three recombinant viruses, along with a previously constructed gE/gI/US9/US2 deleted
