Genetic association studies can be made more cost-effective by exploiting linkage disequilibrium patterns between nearby single-nucleotide polymorphisms (SNPs). The International HapMap Project now offers a dense SNP map across the human genome in four population samples. One question is how well tag SNPs chosen from a resource like HapMap can capture common variation in independent disease samples. To address the issue of tag SNP transferability, we genotyped 2,783 SNPs across 61 genes (with a total span of 6 Mb) involved in DNA repair in 466 individuals from multiple populations. We picked t

The advent of large-scale, population genetic databases (PGDs) in several countries around the world marks a significant development in human DNA banking and genetic research. The European countries that have led the way in the development of PGDs are Iceland, Sweden, Estonia and the U.K. In legal terms, the emergence of PGDs has been far from straightforward as such projects pose a range of difficult and complex issues for the law to address. This article canvasses the current law in Iceland, Estonia, Sweden and the U.K. on four fundamental issues of principle pertaining to PGDs, in order to

Research on the utilization of genetic testing services for mutations in BRCA1 and BRCA2 has focused on women with a strong family history of breast and ovarian cancer. We conducted a population-based case-control-family study of Australian women diagnosed with invasive breast cancer before age 40 years, unselected for family history, and tested for germ line mutations in BRCA1 and BRCA2. Case subjects found to carry a deleterious mutation and their relatives who had given a research blood sample were informed by mail that the study had identified "genetic information" and were offered the opp

There is some evidence showing that genetic factors are involved in human susceptibility to parasitic diseases such as schistosomiasis and malaria. Studies have shown that the Nramp1 and H-2 genes are implicated in the control of Leishmania donovani infection in mice. We sought genetic loci involved in the control of susceptibility to visceral disease caused by L. donovani in humans. We studied 37 families with at least two affected sibs living in a village in eastern Sudan, where an outbreak of visceral leishmaniasis occurred between 1995 and 2000. The genetic markers located in five chromoso

OBJECTIVE: To evaluate a screening protocol using advanced maternal age, triple-marker screening, and genetic sonography.
METHODS: We compared adverse chromosomal outcomes of pregnancy in 1556 women referred for increased risk of aneuploidy because of either advanced maternal age or triple-marker test results. Patients were counseled about the results of the triple-marker test and subsequent sonography, which led to a patient decision of whether to pursue amniocentesis. Fetal measurements and structural abnormalities were compared with chromosomal findings. When patients elected amniocentesis

Culex pipiens pipiens is a common mosquito found in North Africa and is the primary vector of Bancroftian filariasis in Egypt. Experimental infections with C. p. pipiens have demonstrated that this species supports Brugia pahangi development (parasitic yield 6.3%). In addition, the genetic variation among C. p. pipiens populations from Ben Arrous (in the suburb of Tunis) estimated from isoenzyme polymorphism of five neutral genes (Pgm, Gpi, Hk1, Got-1, and Got-2) was slight (Fst = +0.083). C. p. pipiens from Tunis seems to consist of a homogeneous population that is susceptible to Brugia infec

Serum and/or plasma samples from 360 Whites and 126 Blacks were assayed for activity of the lysosomal hydrolase alpha-N-acetyl-D-glucosaminidase (NAG). The samples from the Blacks had an increased mean (0.50 nm/ml/min) and standard deviation (0.30 nm/ml/min) compared to those from the Whites (0.29 nm/ml/min and 0.10 nm/ml/min, respectively). After loge transformation and admixture analysis, it was possible to demonstrate the presence of 3 distributions of NAG activity in Blacks and at least 2 in Whites. Segregation analysis of the NAG activity of 29 White half-sib twin families indicated that

In order to clear out the role of genetico-automatic processes in Gd- distribution the investigation of population structure of three settlements in the Shekii district was carried out. Sex and age ratios and the average number of living children on one marriage (> 5.7) bear evidence of expanded reproduction in this population. The high frequency of endogamous marriages (approximately 80%) and 14 cases of consaguinous marriage for 183 analysed pedigree were revealed. The allele frequencies in 6 polymorphic systems were determined: ABO, Rh, Hp, PGM, Gc and AP; in the first three there were foun

Clinical, hematological and genetic studies were carried out on 40 patients with symptomatic sickle-cell disease, selected on the basis of a predominant HbS fraction and absence of other abnormal hemoglobin variants. Family studies showed they included 26 homozygotes for the sickle-cell gene (SS) and 14 double heterozygotes for both the sickle-cell and the (0)beta-thalassemia genes ((S)(0)beta-thalassemia). Comparison of the two groups revealed the more common occurrence of splenomegaly, lower MCV and mCH, and higher HbA2 in (S)(0)beta-thalassemia. Total hemoglobin was slightly lower in SS dis

BACKGROUND: Alagille syndrome (ALGS) is a rare multisystem disorder primarily caused by pathogenic variants in JAG1 and NOTCH2. Although its genetic basis is well established, existing variant repositories often lack comprehensive clinical annotation, sufficient ethnic representation, and open accessibility. These limitations reduce their translational relevance and hinder genotype-phenotype correlation and evidence-based molecular diagnosis. To address this gap, we developed AGEX (Alagille Genetics Exploration Database), a curated, open-access resource integrating genetic variants with phenot

PREMISE: Calls to adopt seed-sourcing strategies in biological restoration that preserve local adaptation while maximizing genetic variation (e.g., regional admixture provenancing) are increasingly common. Heterosis, the increased fitness of progeny from crosses between populations relative to progeny from crosses within populations, could provide an added benefit under such strategies.
METHODS: We quantified heterosis in crosses between three small remnant populations of the rare prairie forb Silene regia. We measured early fitness components (seed number per fruit, germination, and juvenile

BACKGROUND: Telomere length (TL), a biomarker of biological aging, but its association with Alzheimer’s disease (AD) remains unclear. METHODS: We estimated TL in whole-genome sequencing data from 35,014 Alzheimer’s Disease Sequencing Project participants using TelSeq, which after quality control yielded a dataset including 6,973 persons of European ancestry (EA), 4,188 African Americans (AA), 4,005 Caribbean Hispanics (CH), and 4,170 Native American Hispanics (NAH). TL was log-transformed, adjusted for age and blood cell counts, and z-scaled. Scaled TL was dichotomized into long (TL > 0) and s

BACKGROUND: Refractive errors are the leading cause of visual impairment and blindness globally. High myopia (HM) poses significant risks of severe ocular complications and blindness. 12 SNPs, including rs580839, have been associated with refractive errors in European and Asian populations, but their roles in Chinese cohorts remain unexplored. Their genetic association with HM is investigated in this study.
METHODS: Genotyping of specific SNPs was conducted using multiplexed SNP/exome capture sequencing in a cohort consisting of HM patients and emmetropic individuals. The 3DSNP and GTEx datab

We sought to clarify the inter-relationships among a range of psychiatric and substance use disorders by employing two-dimensional genetic maps, with axes defined by family genetic risk for major depression (MD) and drug use disorder (DUD). Using comprehensive Swedish national register data, we located cases of 12 disorders within this space and examined how their genetic risk profiles varied as a function of sex, age at onset, and level of recurrence. Disorders segregated robustly into two clusters. Internalizing disorders were characterized by moderate genetic liability for MD and low liabil

Urban-driven landscape fragmentation is a major factor contributing to amphibian population decline yet its genetic consequences remain incompletely understood. We examined the genetic structure and variability of the common toad (Bufo bufo) in Warsaw, Poland, using seven nuclear microsatellite loci and mitochondrial cytochrome b sequences (800 bp). Samples (N = 97) were collected from six breeding sites representing contrasting urban habitats on both banks of the Vistula River. Genetic analyses revealed low but significant population differentiation, elevated inbreeding coefficients, and a he

Ancestry-informative SNPs are widely used for biogeographical ancestry inference, yet relatively few studies have explored strategies to maximize ancestry inference potential of existing AISNP panels. Here, we evaluated patterns of genetic differentiation among Asian populations and the utility of a single panel of 164 AISNPs for group-level regional ancestry inference using machine learning (ML) approaches. Genotype data from 2285 individuals representing 16 Asian populations were analyzed using conventional population genetic methods and six supervised ML algorithms. Population genetic analy

BACKGROUND: In India, indigenous camel populations have evolved under diverse ecological conditions, leading to the emergence of several distinct breeds with specialised adaptive traits. Understanding the genomic architecture and population structure of such camel populations is essential for their conservation and sustainable utilisation. In the present study, whole-genome sequencing data were generated and analysed for Indian dromedary camels representing nine recognised breeds, along with Indian Bactrian (double-humped) and Arabian Peninsula dromedaries with the primary objective to investi
