Association between immune recovery uveitis and a diverse intraocular cytomegalovirus-specific cytotoxic T cell response.

Cytomegalovirus (CMV) causes serious infection in individuals with deficient T cell immunity. In acquired immunodeficiency syndrome, the retina is a major site of progressive infection, despite the availability of therapy that targets CMV. The administration of highly active antiretroviral therapy to suppress human immunodeficiency virus frequently results in resolution of CMV retinitis, but this may be complicated by ocular inflammation termed "immune recovery uveitis" (IRU). To provide insight into the pathogenesis of IRU, the phenotype and specificity of intraocular T cells in a single pati

Host cell actin remodeling in response to Cryptosporidium.

Cryptosporidium exhibits a complex strategy to invade and establish productive infection sites, involving complimentary parasite and host cell processes. While the work regarding host cell actin remodeling has greatly enhanced our understanding of the molecular pathways involved in the parasite induced actin reorganization, the specific function of host cell actin remodeling is still equivocal. We contend that host cell actin polymerization contributes to the development of productive C. parvum infection sites by generating membrane protrusion events, which may assist in the retention of the p

Broad-host-range Kayvirus phages preferentially infect Staphylocococcus aureus cells with β-glycosylated WTA, the most common bacterial form in the human host.

OBJECTIVES: In Staphylococcus aureus, WTA glycosylation participates in the interaction with the immune system and bacteriophage adsorption. Indeed, virulent phages with podovirus morphology and some myoviruses require β-glycosylation (catalized by TarS), and are hindered by α-glycosylation (carried out by TarM) of WTA. In contrast, other myophages, like phiIPLA-RODI and phage K, exhibit a broad-host range and infect strains without either glycosylation according to the spot test. Here, we set out to determine the impact of glycosylation type on susceptibility to the Kayvirus phiIPLA-RODI. ME

Evolution of generalism under Muller's ratchet.

Host-range expansion in parasites is usually framed as an adaptive response to heterogeneous environments and trade-offs. In this opinion article, I propose Ratchet-Driven Generalism (RDG): in some parasites, broad host range may instead arise non-adaptively after the loss of sexual recombination. Under Muller's ratchet, deleterious mutations accumulate in asexual or rarely recombining lineages. Over time, genomic erosion can degrade host-specific adaptations, including host-interaction genes. In parallel, reduced competitive filtering among parasite genotypes can weaken selection for host-spe

Host Range Breadth Correlates with Genic Diversity in Honeybee Phages.

Bacteriophages can evolve rapidly. Mutation and recombination via horizontal gene transfer allow them to counter adaptive responses by microbial hosts. However, little is known about the genomic processes underlying phage evolution within an ecological context-especially within natural microbial communities. This is due in part to the difficulty in resolving aspects of phage ecology, such as host range. To better understand the interplay of phage ecology and evolution within natural microbial communities, we combined measures of phage host range in vivo with measures of genome evolution in ord

European elms as potential hosts of Saperda tridentata (Coleoptera: Cerambycidae).

The elm borer, Saperda tridentata (Olivier, 1795) (Coleoptera: Cerambycidae), is a North American longhorn beetle that has been repeatedly intercepted in Europe on imported elm logs. Although climatic conditions are considered suitable for its establishment, uncertainty remains regarding the suitability of European tree species as hosts. We evaluated the potential host range of S. tridentata by testing 4 elm species (Ulmus glabra, Ulmus laevis, Ulmus minor, and Ulmus pumila), 3 maple species (Acer campestre, Acer platanoides, and Acer pseudoplatanus), and 3 poplar species (Populus alba, Populu

Giant endogenous viral elements in the genome of the model protist Euglena gracilis reveal past interactions with giant viruses.

Genomes of giant viruses (phylum Nucleocytoviricota) have increasingly been found integrated into the genomes of diverse eukaryotes. Here, we report eight giant endogenous viral elements (GEVEs) in the genome of the microalgae Euglena gracilis. The GEVEs in E. gracilis bear signatures of genomic erosion, including invasion of transposable elements and duplications, suggesting that they are incapable of reactivation and virion production. Most of the GEVEs exhibit high average amino acid identity and cluster near each other in phylogenies of viral marker genes, suggesting that they are derived

Ecological range and host-biome associations of pathogenic Leptospira in Brazil: A One Health perspective from a tropical area.

Pathogenic Leptospira spp. cause a globally distributed zoonosis with complex transmission cycles involving multiple hosts and environments. Brazil's vast ecological diversity and high Leptospira spp. circulation demand integrative approaches to understand the pathogen's genetic diversity and distribution. We compiled an extensive molecular dataset of pathogenic Leptospira spp. in Brazil, encompassing sequences (n = 825) from several genetic markers and WGS data. A molecular landscape was provided based on associated metadata, including host species, geographic origin, and source type (clinica

How host species and body part determine the microbial communities of five ambrosia beetle species.

The ambrosia beetles are farming insects that feed mainly on their cultivated fungi, which in some occasions are pathogens from forest and fruit trees. We used a culture-independent approach based on 16S and 18S rRNA gene metabarcoding analysis to investigate the diversity and composition of the bacterial and fungal communities associated with five ambrosia beetle species: four species native to America (Monarthrum dimidiatum, Dryocoetoides capucinus, Euwallacea discretus, Corthylus consimilis) and an introduced species (Xylosandrus morigerus). For the bacterial community, the beetle species h

Mapping environmental suitability of Haemagogus and Sabethes spp. mosquitoes to understand sylvatic transmission risk of yellow fever virus in Brazil.

BACKGROUND: Yellow fever (YF) is an arboviral disease which is endemic to Brazil due to a sylvatic transmission cycle maintained by infected mosquito vectors, non-human primate (NHP) hosts, and humans. Despite the existence of an effective vaccine, recent sporadic YF epidemics have underscored concerns about sylvatic vector surveillance, as very little is known about their spatial distribution. Here, we model and map the environmental suitability of YF's main vectors in Brazil, Haemagogus spp. and Sabethes spp., and use human population and NHP data to identify locations prone to transmission

Reptiles and Amphibians as Potential Reservoir Hosts of Chikungunya Virus.

Chikungunya virus is an emerging arbovirus of significant human-health concern. Little is known about its sylvatic cycle, including whether ectothermic vertebrates are permissive to infection. In this study, individuals from ten species of reptiles and amphibians were inoculated with chikungunya virus and samples of blood were tested to characterize viremia and seroconversion. Viremia was not detected in cane toads, house geckos, or American alligators, but most of the green iguanas, red-eared sliders, ball and Burmese pythons, leopard frogs, Texas toads, and garter snakes developed viremia. First intermediate hosts of Paragonimus spp. in Vietnam and identification of intramolluscan stages of different Paragonimus species.

BACKGROUND: Members of the genus Paragonimus require at least three hosts in their life-cycles. The obligatory first intermediate hosts are freshwater snails. In Vietnam, although seven Paragonimus species have been recorded, the natural first intermediate hosts of almost all species have not been confirmed. The aim of this study was, therefore, to investigate snail hosts of Paragonimus species in Vietnam, and to identify Paragonimus species at intramolluscan stages. METHODS: Freshwater snails were collected from streams in Yen Bai and Quang Tri Provinces, where high prevalences of Paragonimu

Genetic differentiation of the oriental rat flea, Xenopsylla cheopis, from two sympatric host species.

BACKGROUND: The oriental rat flea (Xenopsylla cheopis), which infests several mammals, primarily rats (Rattus spp.), is the most notorious vector of human plague. In this study, we measured the genetic differentiation among populations of fleas from the Asian house rat (Rattus tanezumi) and the brown rat (R. norvegicus) using microsatellite markers in order to investigate the extent of host-switching in this parasite. RESULTS: We developed 11 polymorphic microsatellite loci for our study, nine of which showed high potential for inbreeding. AMOVA showed that the majority (84.07%, P < 0.001) of

Species identification, host range and diversity of Cecidophyopsis mites (Acari: Trombidiformes) infesting Ribes in Latvia.

Cecidophyopsis mites are important pests in all cultivation regions of Ribes causing bud galls and sterility. Despite their economic importance, the knowledge on Cecidophyopsis species infesting Ribes in various areas of the world is still deficient. The present study was carried out to identify Cecidophyopsis species occurring in Latvia on cultivated and wild Ribes, to assess their host range and gain insight into the genetic diversity of these insufficiently studied pests by use of multiplex PCR, rDNA sequences and morphological characters. Cecidophyopsis alpina, C. aurea, C. spicata and C.
