Taurine Alleviates Sepsis-Induced Myocardial Injury by Inhibiting NF-κB Pathway-Mediated Inflammation, Mitochondrial Dysfunction, and Apoptosis.

Sepsis-induced cardiomyopathy (SICM) is a life-threatening complication characterized by cardiac dysfunction, inflammation, oxidative stress, and mitochondrial impairment, with limited therapeutic options. Taurine, a naturally occurring amino acid with known anti-inflammatory and mitochondrial-stabilizing properties, may offer therapeutic benefits. However, its role in SICM is not well defined. This study aimed to evaluate the protective effects of taurine against SICM and elucidate its underlying mechanisms. A murine model of SICM was established via cecal ligation and puncture (CLP), and AC1

Chronic allergic lung inflammation is associated with brain mast cell-mediated suppression of neuronal activity and behavioral changes in mice.

BACKGROUND: We have previously observed depressive-like behaviors in mice following the development of chronic allergic lung inflammation that potentially involved brain mast cell activation. However, the specific brain regions that might be responsible for the observed behavioral changes were not identified. The anterior cingulate cortex (ACC) has been implicated in depression, and hypoactivity of this region is seen in the electroencephalogram of patients with depression. In the current study, we tested our hypothesis that chronic allergic lung inflammation causes mast cell activation that a

Galectin-3 mediates lysosome-related inflammation within monocyte-derived macrophages in a mouse model of ischemic brain injury.

Circulating monocyte-derived macrophages (MDMø) rapidly invade the brain after stroke, exerting both detrimental and beneficial effects. Elucidating mechanisms that mediate detrimental properties of MDMø may identify therapeutic strategies to divert MDMø from destructive phenotypes, while preserving their favorable effects. Toward this goal, the current study explores the function of Galectin-3 (GAL3) in MDMø and elucidates mechanisms whereby MDMø-derived GAL3 exacerbates stroke injury. In the acutely injured brain, GAL3 expression was upregulated primarily within MDMø. Global KO of GAL3 reduc

Sirtuin 7-mediated deacetylation of hypoxia-inducible factor 1-alpha facilitates glycolytic reprogramming and inflammation of keratinocytes in psoriasis.

INTRODUCTION: Psoriasis is a chronic inflammatory disease characterized by epidermal hyperplasia and dermal T cell infiltration. Epidermal keratinocytes function not only as the primary targets of the inflammatory response but also play a pivotal role in initiating and maintaining the inflammatory state. Sirtuin 7 (SIRT7), an NAD+-dependent protein deacetylase, is critical for various cellular processes, including proliferation, metabolic homeostasis, and inflammation. However, its role in psoriasis remains elusive. OBJECTIVES: To investigate the role of SIRT7 in psoriasis pathogenesis and it

Nanoparticle-Loaded Injectable Hydrogel Alleviates Titanium Particle-Induced Osteolysis by Disrupting GATA6/DDX3X-Mediated Macrophage Inflammation.

Aseptic loosening (AL) represents the primary cause of joint arthroplasty failure, which is predominantly triggered by chronic inflammatory reactions to prosthetic wear particles, with macrophages serving as the central effector cells. Accumulating evidence indicates that the crosstalk between endoplasmic reticulum stress and mitochondrial stress exacerbates macrophage-mediated inflammation; however, the core molecular regulators orchestrating this pathological cascade remain elusive. Herein, we investigated the functional role of the GATA6/DDX3X (DEAD-box helicase 3 X-linked) axis in titanium

Targeting NFATc1-regulated MTHFD2 one-carbon metabolism to suppress sustained T-cell-mediated inflammation in rheumatoid arthritis.

T cells are central drivers of inflammation across autoimmune and inflammatory diseases, yet current therapies inadequately target pathogenic T-cell pathways, limiting durable disease control. Here, we identified a novel, targetable transcriptional-metabolic axis that sustains inflammatory T-cell responses, characterized by NFATc1-regulated activation of MTHFD2-dependent one-carbon metabolism. We demonstrate that NFATc1 directly binds the MTHFD2 promoter region, driving metabolic reprogramming in activated T cells from rheumatoid arthritis (RA) patients as well as in experimental arthritis mod

Intelligent nanoliposome ameliorate chronic intermittent hypoxia-mediated neuronal injury via a dual regulation microglial inflammation strategy.

The etiology of obstructive sleep apnea syndrome (OSAS)-associated cognitive dysfunction is unclear and complex. There is growing evidence demonstrate that hyper-activated neuroinflammation, M1 phenotypic microglia polarization, and subsequent neuronal inflammatory damage induced by chronic intermittent hypoxia (CIH) pose a crucial role in OSAS-related cognitive dysfunction. However, the regulatory mechanisms remain unclear, and during treatment, there are inevitable issues with small molecule drugs such as hydrophobicity, lack of targeting, and uncontrolled dosages, especially their inability

β-carotene attenuates calcium oxalate crystal-induced renal injury through suppression of NF-κB-mediated inflammation.

Calcium oxalate (CaOx) nephrolithiasis represents the most prevalent form of kidney stone disease and is characterized by persistent renal inflammation and tissue damage. β-Carotene, a naturally occurring carotenoid with well-documented anti-inflammatory properties, has demonstrated beneficial effects in a variety of inflammatory conditions; however, its potential role in CaOx crystal-associated renal injury remains largely unexplored. In this study, the effects of β-carotene on CaOx-induced renal damage were evaluated using both cellular and animal models. In human renal proximal tubular epit

Aconitine promotes injured peripheral nerve recovery through restraining the activation of inflammasome-mediated cell pyroptosis and pathological inflammation.

Peripheral nerve injury (PNI) represents a common neurological condition with significant social and economic implications. Aconitine, a diterpenoid alkaloid derived from Aconitum species, exhibits potent anti-cancer, anti-viral, anti-inflammatory, analgesic, and immunomodulatory activities against malignancies, rheumatic disorders, arthralgia, and select endocrine pathologies. However, the neuroprotective potential of aconitine in PNI repair remains unclear. Here, we revealed that aconitine treatment at the optimal dose significantly improved SFI values, electrophysiological conduction, axon

Putative role of NF-κB-mediated inflammation in acute cardiac injury induced by S-band microwave radiation in rats.

With the widespread adoption of wireless communication technology, the issue of environmental exposure to microwave radiation has become increasingly prominent. In the field of cardiovascular research, it has been observed that microwave radiation may have potential effects on target organs such as the heart. However, the dynamic evolution characteristics of this radiation over time and its relationship with inflammation still require further exploration. This study aimed to establish a rat model of uniform whole-body exposure to S-band microwaves. A range of methods were used, including detec

[ERβ-mediated estrogen signaling promotes the alleviation of inflammation and myofiber repair in cardiotoxin-induced acute skeletal myositis in mice].

Traumatic stress often leads to abnormalities in sex hormone levels, among which elevated estrogen levels are associated with improved patient prognosis. However, the mechanism by which estrogen affects the inflammatory response and repair process in acute skeletal myositis remains unclear. This study aimed to investigate the effects of estradiol (E2) signaling on the resolution of acute skeletal myositis and muscle fiber repair in mice induced by cardiotoxin (CTX). An acute skeletal myositis model was established in C57BL/6 mice. Endogenous E2 levels were reduced by ovariectomy (OVX) in femal

miR-92a Agomir protects against ischemic stroke by suppressing oxidative stress, inflammation, excitotoxicity, and p53-mediated apoptosis.

Various microRNAs (miRs) have long served as therapeutic targets for ischemic stroke, so we aimed to assess whether miR-92a-3p protects against cerebral ischemia and identify downstream targets in an animal model. For this purpose, rats were randomly divided into sham, middle cerebral artery occlusion (MCAO), miR-92a Agomir + MCAO, and miR-92a Antagomir + MCAO groups. Twenty-four hours before MCAO surgery, rno-miR-92a-3p Agomir or Antagomir (100 µM) were injected into the right striatum of rats. Neurological deficit, infarction, blood-brain barrier (BBB), cerebral edema, oxidative stress and i

Micheliolide alleviates myocardial ischemia/reperfusion injury in rats via KEAP1/NRF2-mediated suppression of oxidative stress and inflammation.

BACKGROUND: After acute myocardial infarction (AMI), reperfusion therapy can help restore blood flow and nutritional support to the ischemic myocardium, thereby limiting ongoing myocardial injury. However, its effectiveness is increasingly challenged by myocardial ischemia-reperfusion (I/R) injury. In this regard, micheliolide (MCL) has been reported to exert beneficial effects in cardiovascular disease. Herein, we aimed to determine the mechanism underlying the cardioprotective effects of MCL in a rat I/R model. METHOD: Rats were randomly divided into a control group, an I/R group, and an I/

Subchronic Alcohol Exposure Exacerbates Acetaminophen-Induced Acute Liver Injury via Macrophage-Mediated Inflammation and Mitochondrial Dysfunction.

Alcohol use is a major coexposure in patients taking acetaminophen (APAP), yet how subchronic alcohol exposure reshapes hepatic vulnerability to APAP remains incompletely defined. In this study, we examined the effects of chronic alcohol intake on APAP-induced liver injury in mice. Our results indicated that subchronic alcohol exposure significantly increased the mortality rate in APAP-overdosed mice. Alcohol pretreatment also exacerbated APAP-induced hepatic damage, as evidenced by elevated serum ALT and AST levels, enhanced inflammatory responses, and aggravated oxidative stress. Quantitativ
