Harnessing greater statistical power: Comprehensive evaluation of disease modifying treatment effects across all or multiple post-baseline visits compared to the last visit for Alzheimer's disease clinical trials.

BackgroundIn Alzheimer's disease (AD) clinical trials, efficacy inference is traditionally based on the last visit (e.g., 18 months). However, recent studies suggest that disease-modifying treatment effects may emerge as early as 3 months post-baseline.ObjectiveTo explore this further, our study aimed to assess the increased statistical power achieved by incorporating all or multiple post-baseline visits to estimate treatment effect, compared to relying solely on the last visit.MethodsWe developed explicit formulas for the basis functions of the natural cubic spline model, ensuring compatibili

Taxometric investigation of the latent structure of nicotine dependence: an epidemiological sample.

A considerable number of daily tobacco users do not fulfill the DSM-IV and ICD-10 diagnostic criteria for nicotine dependence (ND). This suggests that such a diagnostic boundary may be arbitrary. This paper addresses this question empirically by comparing the viability of two models, respectively hypothesizing a dimensional and a categorical latent structure of ND. An epidemiological sample of 6,926 individuals was selected from a cross-sectional probabilistic stratified sampling design. All participants having smoked in the past 30 days were included in the study. Half of this sample was used

Joint analysis of bivariate longitudinal ordinal outcomes and competing risks survival times with nonparametric distributions for random effects.

We propose a semiparametric joint model for bivariate longitudinal ordinal outcomes and competing risks failure time data. The association between the longitudinal and survival endpoints is captured by latent random effects. This approach generalizes previous joint analysis that considers only one response variable at the longitudinal endpoint. One unique feature of the proposed model is that we relax the commonly used normality assumption for random effects and leave the distribution completely unspecified. We use a modified version of the vertex exchange method in conjunction with an expecta

Defining statistical perceptions with an empirical Bayesian approach.

Extracting statistical structures (including textures or contrasts) from a natural stimulus is a central challenge in both biological and engineering contexts. This study interprets the process of statistical recognition in terms of hyperparameter estimations and free-energy minimization procedures with an empirical Bayesian approach. This mathematical interpretation resulted in a framework for relating physiological insights in animal sensory systems to the functional properties of recognizing stimulus statistics. We applied the present theoretical framework to two typical models of natural i

Mad cows and computer models: the U.S. response to BSE.

The proportion of slaughtered cattle tested for BSE is much smaller in the U.S. than in Europe and Japan, leaving the U.S. heavily dependent on statistical models to estimate both the current prevalence and the spread of BSE. We examine the models relied on by USDA, finding that the prevalence model provides only a rough estimate, due to limited data availability. Reassuring forecasts from the model of the spread of BSE depend on the arbitrary constraint that worst-case values are assumed by only one of 17 key parameters at a time. In three of the six published scenarios with multiple worst-ca

Exploration and demonstration of explainable machine learning models in prosthetic rehabilitation-based gait analysis.

Quantitative gait analysis is important for understanding the non-typical walking patterns associated with mobility impairments. Conventional linear statistical methods and machine learning (ML) models are commonly used to assess gait performance and related changes in the gait parameters. Nonetheless, explainable machine learning provides an alternative technique for distinguishing the significant and influential gait changes stemming from a given intervention. The goal of this work was to demonstrate the use of explainable ML models in gait analysis for prosthetic rehabilitation in both popu

Critical-Network Lesion-Symptom Mapping.

Complex datasets used for lesion-symptom mapping (LSM) present serious challenges for interpretable statistical analysis. We introduce Critical Network Lesion-Symptom Mapping (CN-LSM), a neuroanatomical atlas-based method that uses low-dimensional regression models and out-of-sample prediction to identify brain regions that contribute significantly to the prediction of clinical behavioral scores. Rather than focusing on a single most-predictive model, CN-LSM identifies a set of plausible models analyzed as an ensemble. CN-LSM offers three primary advantages over existing approaches: (1) greate

Low-burden speech-based screening of depressive symptoms using ultra-short speech samples.

BACKGROUND: Speech holds promise for depressive symptom screening, but practical applications require clearer guidance on effective segmentation, optimal speech tasks, minimum duration, and feature-model matching. OBJECTIVE: To enable low-burden screening for depressive symptoms, this study aims to evaluate the effectiveness of short speech samples and systematically investigate how to identify the optimal practical combination of different speech tasks, time-window lengths, and the matching between features and models. METHODS: Using the HMU-DDAC-24 dataset of 143 participants aged 18-22 ye

Korean panic disorder severity scale: construct validity by confirmatory factor analysis.

The Panic Disorder Severity Scale (PDSS) is a seven-item instrument used to rate the overall severity of panic disorder. This scale has previously evidenced excellent psychometric properties in a sample of patients with panic disorder, with or without agoraphobia. However, despite several factor-analytic PDSS studies, the number of factors has not been entirely consistent. Our objective in this study, then, was to evaluate the fit of two competing models to data collected with a sample of 176 patients with panic disorder. Patients (N = 176) who fulfilled DSM-IV criteria for panic disorder, wit

A review of dynamic network models with latent variables.

We present a selective review of statistical modeling of dynamic networks. We focus on models with latent variables, specifically, the latent space models and the latent class models (or stochastic blockmodels), which investigate both the observed features and the unobserved structure of networks. We begin with an overview of the static models, and then we introduce the dynamic extensions. For each dynamic model, we also discuss its applications that have been studied in the literature, with the data source listed in Appendix. Based on the review, we summarize a list of open problems and chall

The More, the Better? Evaluating the Role of EEG Preprocessing for Deep Learning Applications.

The last decade has witnessed a notable surge in deep learning applications for electroencephalography (EEG) data analysis, showing promising improvements over conventional statistical techniques. However, deep learning models can underperform if trained with bad processed data. Preprocessing is crucial for EEG data analysis, yet there is no consensus on the optimal strategies in deep learning scenarios, leading to uncertainty about the extent of preprocessing required for optimal results. This study is the first to thoroughly investigate the effects of EEG preprocessing in deep learning appli

Spatial connectivity in mosquito-borne disease models: a systematic review of methods and assumptions.

Spatial connectivity plays an important role in mosquito-borne disease transmission. Connectivity can arise for many reasons, including shared environments, vector ecology and human movement. This systematic review synthesizes the spatial methods used to model mosquito-borne diseases, their spatial connectivity assumptions and the data used to inform spatial model components. We identified 248 papers eligible for inclusion. Most used statistical models (84.2%), although mechanistic are increasingly used. We identified 17 spatial models which used one of four methods (spatial covariates, local

Development and validation of a climate-based ensemble prediction model for West Nile Virus infection rates in Culex mosquitoes, Suffolk County, New York.

BACKGROUND: West Nile Virus (WNV) is an endemic public health concern in the United States that produces periodic seasonal epidemics. Underlying these outbreaks is the enzootic cycle of WNV between mosquito vectors and bird hosts. Identifying the key environmental conditions that facilitate and accelerate this cycle can be used to inform effective vector control. RESULTS: Here, we model and forecast WNV infection rates among mosquito vectors in Suffolk County, New York using readily available meteorological and hydrological conditions. We first validate a statistical model built with surveill

Interpretable Multi-Class Lung Disease Classification from Chest X-Ray Images Using Attention-Enhanced Deep Learning.

Accurate and interpretable multi-class recognition of lung diseases from chest X-ray images remains challenging because different pulmonary conditions can present with overlapping radiographic patterns, making reliable automated diagnosis difficult in clinical screening and decision support. This study aims to develop an interpretable chest X-ray-based multi-class lung disease classification framework using attention-enhanced deep learning and to compare its performance with standard transfer learning and baseline CNN models. The work contributes a comparative benchmark of attention mechanisms

Modeling zoonotic and vector-borne viruses.

The 2013-2016 Ebola virus disease epidemic and the coronavirus disease 2019 pandemic galvanized tremendous growth in models for emerging zoonotic and vector-borne viruses. Therefore, we have reviewed the main goals and methods of models to guide scientists and decision-makers. The elements of models for emerging viruses vary across spectrums: from understanding the past to forecasting the future, using data across space and time, and using statistical versus mechanistic methods. Hybrid/ensemble models and artificial intelligence offer new opportunities for modeling. Despite this progress, chal

Predicting rotator cuff tears using data mining and Bayesian likelihood ratios.

OBJECTIVES: Rotator cuff tear is a common cause of shoulder diseases. Correct diagnosis of rotator cuff tears can save patients from further invasive, costly and painful tests. This study used predictive data mining and Bayesian theory to improve the accuracy of diagnosing rotator cuff tears by clinical examination alone. METHODS: In this retrospective study, 169 patients who had a preliminary diagnosis of rotator cuff tear on the basis of clinical evaluation followed by confirmatory MRI between 2007 and 2011 were identified. MRI was used as a reference standard to classify rotator cuff tears
