Evidence for a direct arteriovenous connection (A-V shunt) between the testicular artery and pampiniform plexus in the spermatic cord of the tree shrew (Tupaia glis).

In the tree shrew Tupaia glis, 5 or 6 small ramifying arterioles arose directly from the testicular artery and then gave off numerous small … to the testis. The capillaries were covered by vesiculated cells which were located adjacent to the pericytes. The vesiculated cells contained abundant mitochondria, rough endoplasmic reticulum, a well developed Golgi complex and cytoplasmic vesicles. Their cellular processes were long and surrounded more than one capillary. The morphological features of th …

Electron microscopic observations of hepatic and subcutaneous hemangiosarcomas induced in mice exposed to vinyl chloride monomer.

Both subcutaneous and hepatic hemangiosarcomas were induced in mice exposed to vinyl chloride monomer, and for the first time, the ultrastructure of hemangiosarcomas related to vinyl chloride exposure has been described. The subcutaneous hemangiosarcoma developed in a mouse's ear 29 weeks after exposure to 10 ppm vinyl chloride for 4 weeks, and the hepatic hemangiosarcoma was found in a mouse's liver at necropsy 56 weeks after exposure to 600 ppm vinyl chloride for 4 weeks. Both tumors showed a localized, cystic nodular appearance with a dark red tone. Histologically, an angiomatous architectu

Segmental differentiations of cell junctions in the vascular endothelium. The microvasculature.

Small vascular units consisting of an arteriole, its capillaries, and the emerging venule (ACV units) were identified in the rat omentum and mesentery. They were fixed in situ and processed for electron microscopy either as whole units or as dissected segments. Systematic examination of the latter (in thin sections, as well as in freeze-cleaved preparations) showed that the intercellular junctions of the vascular endothelium vary characteristically from one segment to another in the microvasculature. In arterioles, the endothelium has continuous and elaborate tight junctions with interpolated

Multiomic data integration unveils molecular mechanisms underlying chestnut and skin tissue differentiation in Dezhou donkeys.

BACKGROUND: Donkey chestnuts are hairless, darkly pigmented keratinized structures located on the medial forelimbs, anatomically distinct fr … sion, including KRT1, KRT5, KRT10, and KRT14 (TPM > 13,000), and was enriched in endothelial cells, pericytes, T cells, and mast cells. Proteomic analysis revealed significant upregulation of collagen I (COL1A1/COL1A2), indicative of active extracellular matrix remodeling. Pathway analysis identified enrichment in peroxisome activity, fatty acid metabolism, …

Characterization of IFN-γ-mediated cellular responses in the HMC3 cell line.

Microglia, the primary immune cells of the brain parenchyma, play critical roles in neurodevelopment and homeostasis. The human microglia clone 3 (HMC3) cell line has long been a staple in vitro tool for exploring microglia dynamics. However, recent evidence suggests that HMC3 cells behave less like natural microglia and more like pericytes and astrocytes. Until a large-scale shift in replacing HMC3 cells with alternative in vitro models occurs, it is important to standardize characterization techniques used to study HMC3 polarization dynamics to enhance repeatability of findings. Here, we cha…

TGF-β-induced fibrotic scar formation limits recovery of spinal cord injury.

Spinal cord injury (SCI) often causes long-term disability. But effective means to promote proper regeneration after SCI has so far failed t … ifferentiation, thus promoting scar formation. We also found that activated TGF-β1 acts on resident pericytes in the endothelial niche of the blood-spinal cord barrier to promote their differentiation into fibroblasts, which also contributes to scarring. Interrupting these pathways by selective genetic KOs or treatment with a TGF-β-neutralizing antibody inh …

Vascular Endothelial Barrier in Salivary Glands: From Physiological Regulation to Pathological Impairment of Secretion.

Although salivary glands are highly vascularized, the microvascular endothelial barrier has only recently emerged as a pivotal determinant of glandular homeostasis and disease. This review synthesizes current understanding of the salivary gland endothelial barrier, with particular emphasis on the regulation of tight junctions (TJs). Structurally, the barrier comprises endothelial cells interconnected by TJs and adherens junctions, supported by a basement membrane and pericytes. Among TJ components, claudin-5 serves as a key endothelial-specific regulator of paracellular permeability, and is dy…

A human iPSC-based perfusable and membrane-free neurovascular unit-on-chip connecting brain organoids to the blood-brain barrier.

The blood-brain barrier (BBB) protects the central nervous system by restricting entry of harmful blood-borne factors, but this selectivity actively limits delivery of therapeutics to the brain. Because BBB function is shaped by dynamic interactions within the neurovascular unit (NVU), particularly between brain endothelial cells, pericytes, and astrocytes, there is a need for human-relevant models that capture both barrier properties and neurovascular crosstalk in a controlled setting. Here, we present a novel human induced pluripotent stem cell (iPSC)-based NVU-on-chip that couples a perfusa…

Blood-Brain Barrier: Structure, Function, Diseases, and Drug Delivery Systems.

The blood-brain barrier (BBB) is a highly selective and dynamic neurovascular interface essential for maintaining central nervous system homeostasis. This specialized barrier comprises brain microvascular endothelial cells interconnected by tight junctions, supported by pericytes and astrocytic end-feet within the neurovascular unit. While protecting the brain from circulating pathogens and toxins, the BBB presents formidable obstacles to drug delivery, restricting approximately 98% of small-molecule therapeutics and nearly all large biomolecules from reaching the brain parenchyma. BBB dysfunc…

Establishment of vascularized human retinal organoids from induced pluripotent stem cells.

Pluripotent stem cell-derived retinal organoids (ROs) have been investigated for applications in regenerative medicine, retinal disease mode … reated from ROs by co-culturing them with vascular organoid (VO)-derived vascular endothelial cells/pericytes. The expression of mature neuronal markers is markedly higher in the vROs than in the ROs. When vROs are cultured under diabetic conditions, their size and the number of retinal ganglion cells are significantly decreased. In conclusion, the co-cultu …

KATP channels in cerebral hemodynamics: a systematic review of preclinical and clinical studies.

Cumulative evidence suggests that ATP-sensitive potassium (KATP) channels act as a key regulator of cerebral blood flow (CBF). This implication seems to be complicated, since KATP channels are expressed in several vascular-related structures such as smooth muscle cells, endothelial cells and pericytes. In this systematic review, we searched PubMed and EMBASE for preclinical and clinical studies addressing the involvement of KATP channels in CBF regulation. A total of 216 studies were screened by title and abstract. Of these, 45 preclinical and 6 clinical studies were included. Preclinical data…
