Pollen is becoming an increasingly important subject for molecular researchers in genetic engineering, plant breeding, and environmental monitoring. To broaden the scope of these studies, it is essential to develop accessible methods for scientists who are not specialized in palynology. The article presents a simplified technical procedure for preparing pollen grains for scanning electron microscopy (SEM). The protocol is convenient for any molecular laboratory due to its small set of reagents, ease of execution, low cost, does not require special equipment, and takes only one hour to complete…

Six veterinary active ingredients (imidacloprid, albendazole, fenbendazole, praziquantel, fipronil and permethrin) were extracted and quantified by liquid chromatography with diode array detection in water samples from a wetland system used for the treatment of waste from a dog breeding plant. Response surface methodology, based on least-squares and artificial neural networks modelling, was applied for the optimization of a dispersive liquid-liquid microextraction (DLLME) procedure. Firstly, two experimental designs were built for screening and optimization, respectively. Then, the desirabilit

Physiological leaf spotting, or flecking, is a mild-lesion phenotype observed on the leaves of several commonly used maize (Zea mays) inbred lines and has been anecdotally linked to enhanced broad-spectrum disease resistance. Flecking was assessed in the maize nested association mapping (NAM) population, comprising 4,998 recombinant inbred lines from 25 biparental families, and in an association population, comprising 279 diverse maize inbreds. Joint family linkage analysis was conducted with 7,386 markers in the NAM population. Genome-wide association tests were performed with 26.5 million si

Elucidating the molecular mechanisms that govern seed dormancy is important for developing crops with improved resistance to preharvest sprouting (PHS). A series of seed dormancy genes have been cloned, but the connections among different seed dormancy genes remain poorly understood. Here, we have revealed the relationship between two important dormancy quantitative trait loci (QTLs): SDR3.1 and Sdr4. SDR3.1 formed complexes with ABI3 and ABI5 and suppressed their activation of Sdr4 transcription. Haplotype analysis showed that the germination rate of SDR3.1 L-type and Sdr4 n- or K-type combin

The PIN-LIKES (PILS) gene family is crucial for regulating auxin homeostasis and stress adaptation in plants; nevertheless, a comprehensive study on this family in alfalfa (Medicago sativa) remains insufficient. This research found 46 MsPILS genes within the tetraploid alfalfa genome and categorized them into four subfamilies. The genes are irregularly allocated throughout 16 chromosomes, with tandem duplications acting as a primary catalyst for family expansion. Analysis indicated that all MsPILS proteins contain the conserved Mem_trans domain. The promoter study revealed that MsPILS genes ha

Internode morphology critically determines bamboo wood quality. Phyllostachys edulis 'Heterocycla' (GJ) is a natural dwarf mutant of P. edulis (WT), yet regulatory mechanisms remain unclear. Here, we show that impaired parenchyma cell elongation is the primary cellular basis for dwarfism in GJ. This phenotype correlates with a marked reduction in gibberellin (GA) and an increase in abscisic acid (ABA) levels. Through weighted gene co-expression network analysis, we identified PeGA20ox1 as a critical regulator of internode elongation. In GJ, PeGA20ox1 undergoes alternative splicing (AS), produc

Sodium azide is a strong mutagen that has been successfully employed in mutation breeding of crop plants. In biological systems, it is metabolically converted to the proximate mutagen azidoalanine, which requires further bioactivation to a putative ultimate mutagen that remains elusive. The nature of the DNA modifications induced by azides leading to mutations is also unknown. Other mutagenic organic azido compounds seem to share the same bioactivation pathway to the ultimate mutagenic species as they induce point mutations dependent on the same DNA repair pathways. We investigated mutations i

The possible role of pigs as arbovirus maintenance hosts and their importance as amplifier hosts was studied. Blood samples from 464 pigs of all ages collected in 1962 and 1964 were tested against 10 arboviruses. Antibodies to Japanese encephalitis and Getah viruses were particularly prevalent and their calculated monthly infection rates were 19-5% and 13-3% respectively. In 1969, 447 pigs were bled monthly throughout the year and the infection rates for Japanese encephalitis virus were calculated in pigs during the first year of life. Infection rates were not uniform throughout the year; the

BACKGROUND: Amorpha fruticosa L. is a leguminous shrub with high tolerance to drought, poor soil, and saline-alkali stress conditions. As a member of the family of transcription factors in higher plants, the ethylene response factor AP2/ERF plays a crucial role in both plant adaptation to abiotic stress and in growth and development. In this study, based on genes identified from the transcriptomic sequencing of Amorpha fruticosa L. under drought stress, the upregulated gene AfRAP2 was isolated from its seedlings, with the aim of elucidating its stress-response function using molecular biologic

Ocimum, a widely used medicinal and culinary herb, is known for its health-promoting properties, yet most research has focused on its volatile oils, leaving the therapeutic potential and biosynthesis of its coumarins largely unexplored. Despite reports of coumarin bioactivity in other plants, a comprehensive understanding of coumarin diversity, pharmacology, and genetic regulation within Ocimum remains lacking-limiting both its breeding potential and pharmaceutical applications. Here, we applied an integrated multi-omics approach-including UPLC-MS/MS metabolomics, transcriptomics, network phar

BACKGROUND: Domesticated pigs are the main source of Trichinella sp. infections for humans, particularly when reared in backyards or free-ranging. In temperate areas of southern Europe, most pigs are farmed under controlled housing conditions, but sows and sometimes fattening pigs have access to outdoors to improve animal welfare. The aim of the present study was to investigate whether outdoor access of breeding pigs farmed under controlled housing conditions can represent a risk for Trichinella sp. transmission when the farm is located in an agricultural area interspersed with wooded areas an

Cadmium (Cd) in agricultural soils inhibits potato growth, disrupts tuber development, and causes Cd accumulation in edible tubers, thereby threatening food safety. Here, we determined the EC50 of Cd in potatoes, which induced oxidative stress and chloroplast damage. Transcriptomic and metabolomic coanalysis identified StAPR3, an APS reductase involved in sulfur metabolism, as a Cd-responsive candidate. Heterologous yeast expression and potato overexpression demonstrated that StAPR3 enhanced Cd tolerance. Under Cd treatment, StAPR3-overexpressing lines maintained superior growth, while wild-ty

Phoebe bournei is an economically and ecologically important woody species native to China. As a core component of colchicine-triggered polyploid breeding, the tubulin genes (TUA and TUB) have been identified and functionally analyzed in many plants, but not yet in P. bournei. Here, tubulin family members in P. bournei were identified through sequence alignment and subsequently characterized using comprehensive bioinformatic analyses. In particular, a total of six PbTUA and ten PbTUB members were identified and grouped into two and five subfamilies, respectively, according to phylogenetic rela

Alfalfa spring black stem, caused by Ascochyta medicaginicola, severely affects global alfalfa production. This study isolates and identifies brefeldin A (BFA) from pathogen fermentation products and demonstrates its strong phytotoxicity. BFA inhibits leaf photosynthesis and seed germination in a dose-dependent manner at concentrations ≥10-4 mol. Significant variation in BFA sensitivity is observed among cultivars, with 'Longdong' showing the highest susceptibility and 'Spyder' displaying relative resistance. LC-MS/MS analysis detects BFA in naturally infected plants, confirming its presence d

Rising atmospheric CO2 exposes plants to high-CO2 environments, while excessive nitrogen fertilizer use degrades soil, highlighting the need to reduce nitrogen input and cultivate vigorous cucumber seedlings under HC-LN conditions. Calcineurin B-like proteins (CBLs) sense calcium signals and regulate carbon/nitrogen metabolism via CBL-interacting protein kinases (CIPKs), though their roles in cucumber under HC-LN conditions are unclear. Here, we identified seven CsCBL and 19 CsCIPK genes. Transcriptome analysis of two cucumber varieties with differing HC-LN sensitivities revealed that CsCBL4.2

Introduction: Sugar beets are an important crop for global sugar production. Intense drought and the increasing lack of water resources pose a great threat to sugar beet cultivation. It is a priority to investigate favourable germplasms and functional genes to improve the breeding of drought tolerant plants. Methods: Thus, in this study, 328 sugar beet germplasms were used in a genome-wide association study (GWAS) to identify single nucleotide polymorphism (SNP) markers and candidate genes associated with drought tolerance. Results: The results showed that under drought stress (9% PEG-6000), t

In crops, exposure to salt stress triggers pronounced water shortage, disruption of nutrient balance, and metabolic dysfunction, which collectively curtail biomass accumulation and cause substantial yield losses. Thus, identifying the genetic basis and underlying mechanisms that confer salt tolerance is therefore critical for crop improvement. However, functional validation of loci associated with salinity resilience remains limited to a small subset of genes in soybean. In this study, we comparatively evaluated plant height, root length, and fresh weight under salt stress using soybean germpl

Fruit shape is a key agronomic trait influencing harvest efficiency, post-harvest handling, and market value. Therefore, creating diverse fruit morphologies and elucidating their regulatory networks are major objectives in tomato breeding. Currently, although multiple genes regulating fruit shape (such as SUN, OVATE, FAS, etc.) have been identified, the molecular network governing tomato fruit shape remains incomplete. SHAGGY-like kinases (SKs)/Glycogen synthase kinase3 (GSK3) are evolutionarily conserved regulators of plant development and have been implicated in diverse processes. However, t
