Compact polyethylenimine-complexed mRNA vaccines.

Here we describe formulations comprising individual, polymer-complexed self-amplifying RNA (saRNA) molecules, designed for vaccination against infectious diseases and other prophylactic and therapeutic applications. When exposed to a large excess of the cationic polymer polyethylenimine (PEI), the single saRNA molecules in solution reorganize from an extended to a globular organization, characterized by a high packing density, low polymer mass fraction and, consequently, a very small size of the polyplex nanoparticles of about 30 nm. This format of PEI-complexed saRNA exhibits enhanced biologi

Functional insight into a neurodevelopmental disorder caused by missense variants in an RNA-binding protein, RBM10.

The RNA-binding motif protein 10, RBM10, is an RNA splicing regulator essential for development. Loss-of-function RBM10 variants are associated with TARP syndrome, a severe X-linked recessive condition in males. We report a 3-year-old male with a mild phenotype, consisting of cleft palate, hypotonia, developmental delay, and minor dysmorphisms, associated with a missense RBM10 variant, c.943T>C, p.Ser315Pro, affecting the RRM2 RNA-binding domain. His clinical features were similar to a previously reported case associated with a missense variant. The p.Ser315Pro mutant protein was expressed nor

METTL3-mediated M6A methylation modification is involved in colistin-induced nephrotoxicity through apoptosis mediated by Keap1/Nrf2 signaling pathway.

Colistin is a cationic polypeptide antibiotic. Despite its nephrotoxicity, it is still widely used as a last-line antibiotic against infection worldwide with the emergence of multi-drug resistant Gram-negative bacilli. N-methyladenosine (m6A) methylation-mediated degradation of RNA is essential for kidney development. However, m6A methylation impacts not only RNA stability, but also other RNA metabolism processes. How RNA decay affects the nephrotoxicity of colistin is largely unknown. Therefore, in this study, we verified that colistin could induce mouse kidney apoptosis through some apoptoti…

Crystal structure of the Borna disease virus matrix protein (BDV-M) reveals ssRNA binding properties.

Borna disease virus (BDV) is a neurotropic enveloped RNA virus that causes a noncytolytic, persistent infection of the central nervous system in mammals. BDV belongs to the order Mononegavirales, which also includes the negative-strand RNA viruses (NSVs) Ebola, Marburg, vesicular stomatitis, rabies, mumps, and measles. BDV-M, the matrix protein (M-protein) of BDV, is the smallest M-protein (16.2 kDa) among the NSVs. M-proteins play a critical role in virus assembly and budding, mediating the interaction between the viral capsid, envelope, and glycoprotein spikes, and are as such responsible fo

A role for VICKZ proteins in the progression of colorectal carcinomas: regulating lamellipodia formation.

VICKZ proteins are a highly conserved family of RNA binding proteins, implicated in RNA regulatory processes such as intracellular RNA localization, RNA stability, and translational control. During embryogenesis, VICKZ proteins are required for neural crest migration and in adults, the proteins are overexpressed primarily in different cancers. We hypothesized that VICKZ proteins may play a role in cancer cell migration. In patients, VICKZ expression varies with tumour type, with over 60% of colon, lung, and ovarian tumours showing strong expression. In colorectal carcinomas (CRCs), expression …

Emerging roles of circRNAs in leukemia and the clinical prospects: An update.

BACKGROUND: Circular RNAs (circRNAs) are a new category of endogenous non-protein coding RNAs (ncRNAs), and show the characteristics of high conservation, stability, and tissue specificity. Due to rapid advances in next-generation sequencing and transcriptome profiling technologies, circRNAs have been widely discovered in many organisms and participated in the development and progress of a variety of diseases. As a type of molecular sponge, circRNAs mainly absorb micro RNAs competitively and interplay with RNA-binding proteins to modulate the splicing as well as transcription of target genes.

A chemically optimized, GalNAc-conjugated siRNA targeting HSD17B13 demonstrates efficacy in a human 3D organoid model of MASH, showcasing an alternative to animal testing.

INTRODUCTION: Animal models have limited ability to predict human metabolically dysfunction-associated steatohepatitis (MASH), creating a major obstacle in drug development. To address this challenge, we introduce an integrated drug discovery approach that combines rational small-interfering RNA (siRNA) design and chemical modifications to improve stability, validated in a complex human 3D NAC-organoid MASH model. METHODS: Using this system, we created new GalNAc-conjugated siRNAs targeting the 17β-hydroxysteroid dehydrogenase 13 (HSD17B13). The lead candidates were evaluated for stability in

WTAP-Mediated m6A Modification Targets the LRP1-Lipid Metabolism Axis to Regulate Joint Cartilage Regeneration.

Osteoarthritis (OA) arises from impaired epigenetic coordination of inflammatory and metabolic cues, leading to compromised cartilage homeostasis. Such coordination is partly governed by ribonucleic acid (RNA) epigenetic mechanisms, however, the role of the predominant RNA modification N6-methyladenosine (m6A) in this process remains unclear. Herein, we identify an epigenetic-metabolic pathway in which Wilms' Tumor 1-Associating Protein (WTAP)-mediated m6A modification stabilizes low-density lipoprotein receptor-related protein 1 (LRP1) and redirects lipid metabolism toward chondrogenesis. Los

An epitranscriptomic program maintains skeletal stem cell quiescence via a METTL3-FEM1B-GLI1 axis.

Skeletal stem cells (SSCs) maintain the skeletal system via pluripotency and differentiation capacity. However, it remains largely unknown h … capacity and differentiation potential. Mechanistically, Mettl3-mediated m6A modification reduced mRNA stability of the Cul2-RING E3 ligase complex subunit Fem1b, which subsequently stabilizes Gli1 protein, a key transcription factor of Hedgehog pathway for maintaining SSC identity and function. Thus, we present a comprehensive RNA m6A modification landscape o …

Novel hybrid silicon-lipid nanoparticles deliver a siRNA to cure autosomal dominant osteopetrosis in mice. Implications for gene therapy in humans.

Rare skeletal diseases are still in need of proper clinically available transfection agents as the major challenge for first-in-human translation relates to intrinsic difficulty in targeting bone without exacerbating any inherent toxicity due to used vector. SiSaf's silicon stabilized hybrid lipid nanoparticles (sshLNPs) constitute next-generation non-viral vectors able to retain the integrity and stability of constructs and to accommodate considerable payloads of biologicals, without requiring cold-chain storage. sshLNP was complexed with a small interfering RNA (siRNA) specifically designed

Therapeutic gene silencing delivered by a chemically modified small interfering RNA against mutant SOD1 slows amyotrophic lateral sclerosis progression.

Inherited neurodegenerative diseases, such as Huntington disease and subset of Alzheimer disease, Parkinson disease, and amyotrophic lateral sclerosis, are caused by the mutant genes that have gained undefined properties that harm cells in the nervous system, causing neurodegeneration and clinical phenotypes. Lowering the mutant gene expression is predicted to slow the disease progression and produce clinical benefit. Administration of small interfering RNA (siRNA) can silence specific genes. However, long term delivery of siRNA to silence the mutant genes, a requirement for treatment of these

A stress granule-associated RNA-binding protein FAM120A drives cisplatin resistance in non-small cell lung cancer.

Cisplatin-based chemotherapy is a standard treatment for non-small cell lung cancer (NSCLC), but drug resistance poses a major clinical challenge. Stress-adaptive mechanisms, such as stress granule (SG) formation, are increasingly recognized alternative pathways that facilitate cancer cell survival. Here, we identify the RNA-binding protein, family with sequence similarity 120A (FAM120A), as an SG-associated factor that drives cisplatin resistance in NSCLC. FAM120A expression was markedly elevated in cisplatin-resistant NSCLC cell lines and clinical tumor specimens and was essential for SG for

hnRNP F: An emerging orchestrator of RNA processing and gene expression.

Heterogeneous nuclear ribonucleoprotein F (hnRNP F), a member of the hnRNP family, is an essential regulator of RNA metabolism, influencing splicing, translation, stability, and telomere maintenance. This review provides a comprehensive overview of the regulation of expression and multifunctional roles of hnRNP F. Owing to the high degree of sequence homology and functional similarity between hnRNP F and hnRNP H/H', this review also integrates discussions on hnRNP H/H' to facilitate a comparative and holistic understanding of their biological significance.

Quantifying Label-Free Protein-RNA Binding Using Microscale Thermophoresis.

Microscale thermophoresis (MST) is an innovative technique for quantitatively analyzing molecular interactions in free solution at the microliter scale, requiring only minimal sample volumes. It relies on the thermophoretic movement of molecules or molecule-ligand complexes along temperature gradients, enabling the sensitive detection of changes in size, charge, hydration shell, or conformation associated with binding events. This high sensitivity allows the determination of equilibrium dissociation constants in the nanomolar to micromolar range. In this study, MST was used to characterize int

Curcumin-based nanomaterials for dual-target inhibition of SARS-CoV-2 omicron main protease and HMPV RNA polymerase: A computational study.

Since the emergence of SARS-CoV-2 in 2020, the world has faced a global pandemic, underscoring the urgent need for next-generation antiviral therapies to address the growing threat of respiratory RNA viruses, including human metapneumovirus (HMPV). In this study, we investigated four green-synthesized carbon-based nanomaterials derived from curcumin, amino (CD1), graphitic (CD2), pyridinic (CD3), and pyrrolic (CD4) forms, as potential dual-target inhibitors of the SARS-CoV-2 Omicron main protease (Mpro) and HMPV RNA-dependent RNA polymerase (RdRp). Molecular docking analyses revealed that thes

Acyclic serinol nucleic acid modification of siRNAs overcomes seed region mediated off-target effects while maintaining potency.

Similar in structure to both glycol nucleic acid (GNA) and peptide nucleic acid (PNA), serinol nucleic acid (SNA) combines the acyclic flexibility of GNA with the amide bond found in PNA and can form stable heteroduplexes with DNA and RNA. While the thermal and metabolic stability of SNA and SNA-modified heteroduplexes has been previously reported, we here describe the application of SNA in RNA interference (RNAi) using GalNAc-conjugated small interfering RNAs (siRNAs). Single incorporations were evaluated at each position across both the guide and passenger strands and subsequently evaluated

The Role of Non-coding RNAs in Connective Tissue Diseases: Diagnostic and Therapeutic Potential of miRNAs, lncRNAs and circRNAs.

Connective tissue diseases (CTDs) are a heterogeneous group of disorders characterized by structural and functional impairment of the extracellular matrix, manifesting primarily as autoimmune, heritable or degenerative conditions such as systemic lupus erythematosus (SLE), osteogenesis imperfecta (OI) and abdominal aortic aneurysm (AAA). In SLE, lupus nephritis remains a leading cause of morbidity and a major determinant of long-term outcomes. Recent advances in transcriptomics and epigenomics have revealed non-coding RNAs (ncRNAs), including microRNA (miRNAs), long non-coding RNA (lncRNAs) an

Disease-driven post-transcriptional alterations and alternative splicing in podocytes in focal segmental glomerulosclerosis.

Focal segmental glomerulosclerosis (FSGS) is a major cause of nephrotic syndrome and progression to end-stage renal disease, yet its molecular pathogenesis remains still incompletely defined. While transcriptional alterations in podocytes have been extensively characterized, the contribution of post-transcriptional regulatory mechanisms is poorly understood. Here, we combined a zebrafish podocyte-specific injury model with glomerulus-resolved transcriptomic profiling to dissect RNA regulatory alterations during FSGS progression. Integrated analyses of bulk RNA sequencing, small RNA profiling,

m6A-SPP: Identification of RNA N6-methyladenosine modification sites through multi-source biological features and a hybrid deep learning architecture.

The N6-methyladenosine(m6A) modification plays crucial regulatory roles in various biological processes including gene expression regulation, RNA stability, splicing, and translation. Accurate prediction of m6A modification sites is essential for understanding their biological functions and implications in diseases. To address this, we introduce m6A-SPP, a novel deep learning framework for predicting m6A modification sites effectively. The model integrates both sequence features and physicochemical properties of RNA through two specialized modules. The sequence feature module leverages a pretr…

Emerging roles of RNA-binding proteins in fatty liver disease.

A rampant and urgent global health issue of the 21st century is the emergence and progression of fatty liver disease (FLD), including alcoholic fatty liver disease and the more heterogenous metabolism-associated (or non-alcoholic) fatty liver disease (MAFLD/NAFLD) phenotypes. These conditions manifest as disease spectra, progressing from benign hepatic steatosis to symptomatic steatohepatitis, cirrhosis, and, ultimately, hepatocellular carcinoma. With numerous intricately regulated molecular pathways implicated in its pathophysiology, recent data have emphasized the critical roles of RNA-bindi
