Impairment of oxidative metabolism compromises Rad51 recruitment and potentiates PARP inhibitor effectiveness in ovarian cancer.

BACKGROUND: The treatment of ovarian cancer has significantly improved since the introduction of PARP inhibitors (PARPi), small molecules designed to directly target and kill cancer cells with deficiencies in homologous recombination (HR) pathway. However, nearly half of patients present with HR-proficient tumors, rendering them not eligible for PARPi-based therapies and underscoring the urgent need for alternative treatment strategies. METHODS: Oxidative metabolism has been altered either by silencing the mitochondria regulator PGC-1β or by using the OXPHOS inhibitor IACS-010759. The metaboli

Genomic features of liquid biopsies from patients with prostate cancer with and without ductal adenocarcinoma.

Ductal adenocarcinoma of the prostate (DA) is relatively rare and highly co-existent with prostate adenocarcinoma (AC). This study aimed to investigate the distinctive genomic profiles of patients with DA compared to those without. Blood samples were obtained from 144 patients (36 with DA and 108 without DA) who were diagnosed from 2017 to 2023 at West China Hospital. We performed cell-free DNA sequencing to investigate the genomic differences between patients with DA [DA(+)] and those without [DA(-)], and explored the potential associations between their mutational status and prognosis. Patho

ATR inhibition increases reliance on PARP-mediated DNA repair revealing an improved therapeutic strategy for cervical cancer.

OBJECTIVE: Cervical cancer results from persistent infection with high-risk human papillomavirus (HR-HPV) and the expression of E6 and E7 oncoproteins. E6 and E7 compromise the activity of p53 and Rb, the G1-S cell cycle checkpoint, and ATM-mediated DNA damage repair (DDR), which in turn increases reliance on ATR- and PARP-mediated DDR at the G2 cell cycle checkpoint. This study aimed to determine the effects of an ATR inhibitor (ATRi, AZD6738) and a PARP-inhibitor (PARPi, AZD2281) on HR-HPV+ cervical cancer cell lines. METHODS: The effects of ATRi and PARPi, alone and in combination, on meta

Functional Restoration of BRCA1 Nonsense Mutations by Aminoglycoside-Induced Readthrough.

BRCA1 is a major tumor suppressor that functions in the accurate repair of DNA double-strand breaks via homologous recombination (HR). Nonsense mutations in BRCA1 lead to inactive truncated protein products and are associated with high risk of breast and ovarian cancer. These mutations generate premature termination codons (PTCs). Different studies have shown that aminoglycosides can induce PTC suppression by promoting stop codon readthrough and restoring full-length (FL) protein expression. The use of these compounds has been studied in clinical trials for genetic diseases such as cystic fibr

Functional analysis of XRCC4 mutations in reported microcephaly and growth defect patients in terms of radiosensitivity.

Non-homologous end joining is one of the main pathways for DNA double-strand break (DSB) repair and is also implicated in V(D)J recombination in immune system. Therefore, mutations in non-homologous end-joining (NHEJ) proteins were found to be associated with immunodeficiency in human as well as in model animals. Several human patients with mutations in XRCC4 were reported to exhibit microcephaly and growth defects, but unexpectedly showed normal immune function. Here, to evaluate the functionality of these disease-associated mutations of XRCC4 in terms of radiosensitivity, we generated stable

Wwox Deletion in Mouse B Cells Leads to Genomic Instability, Neoplastic Transformation, and Monoclonal Gammopathies.

WWOX (WW domain containing oxidoreductase) expression loss is common in various cancers and characteristic of poor prognosis. Deletions, translocations, and loss of expression affecting the WWOX gene are a common feature of various B cell neoplasms such as certain B cell lymphomas and multiple myeloma. However, the role of this common abnormality in B cell tumor initiation and/or progression has not been defined. In this study, we conditionally deleted Wwox early in B cell development by means of breeding Cd19-Cre transgenic mice crossed to Wwox floxed mice (Cd19 Wwox KO). We observed a signif

Comprehensive BRCA1/2 mutation landscape in prostate cancer in the UAE and Arab population.

BACKGROUND: Prostate cancer (PCa) is the most common malignancy among men in the United Arab Emirates (UAE) and is often diagnosed at advanced stages with aggressive features. Germline mutations in DNA-repair genes, especially BRCA1 and BRCA2, increase PCa risk, with BRCA2 conferring up to 8.6-fold and BRCA1 a 3.7-fold risk. The objective is to determine the prevalence, zygosity, and clinical significance of BRCA1/2 mutations in high-grade PCa among UAE and Arab patients and describe their potential role in disease aggressiveness. METHODS: A retrospective analysis was performed on 40 archived

RAD51 Predicts Poor Prognosis and Serves as a Therapeutic Target in Intrahepatic Cholangiocarcinoma.

INTRODUCTION: Intrahepatic cholangiocarcinoma (ICC) is a highly aggressive malignancy with limited responsiveness to chemotherapy. RAD51, a key component of homologous recombination repair, has been implicated in tumor progression and therapeutic resistance in several cancers; however, its clinical and biological role in ICC is unclear. METHODS: RAD51 expression was evaluated by immunohistochemistry in 96 patients having upfront surgery for ICC. Clinicopathological correlations and survival outcomes were analyzed. Functional studies were conducted in the ICC cell lines, HuCCT1 and TKKK, using

Non-catalytic Roles for XPG with BRCA1 and BRCA2 in Homologous Recombination and Genome Stability.

XPG is a structure-specific endonuclease required for nucleotide excision repair, and incision-defective XPG mutations cause the skin cancer-prone syndrome xeroderma pigmentosum. Truncating mutations instead cause the neurodevelopmental progeroid disorder Cockayne syndrome, but little is known about how XPG loss results in this devastating disease. We identify XPG as a partner of BRCA1 and BRCA2 in maintaining genomic stability through homologous recombination (HRR). XPG depletion causes DNA double-strand breaks, chromosomal abnormalities, cell-cycle delays, defective HRR, inability to overcom

Increased oxidative stress in AOA3 cells disturbs ATM-dependent DNA damage responses.

Ataxia telangiectasia (AT) is caused by a mutation in the ataxia-telangiectasia-mutated (ATM) gene; the condition is associated with hyper-radiosensitivity, abnormal cell-cycle checkpoints, and genomic instability. AT patients also show cerebellar ataxia, possibly due to reactive oxygen species (ROS) sensitivity in neural cells. The ATM protein is a key regulator of the DNA damage response. Recently, several AT-like disorders have been reported. The genes responsible for them are predicted to encode proteins that interact with ATM in the DNA-damage response. Ataxia with oculomotor apraxia type

AKT2 Counteracts Hypoxia-Induced Suppression of Homologous Recombination in Triple-Negative Breast Cancer: A Targeted Approach to Sensitize Tumors to PARP Inhibition.

Triple-negative breast cancer (TNBC) is characterized by hypoxia and impaired fidelity to DNA repair and the activation of survival pathways. However, the role of specific AKT isoforms, particularly AKT2, in regulating residual homologous recombination (HR) activity and PARP inhibitor sensitivity under hypoxic stress remains unclear. This study aimed to establish the effect of AKT2 on hypoxia-induced defects in HR repair, DNA damage accretion, and therapeutic outcomes of PARP inhibition in TNBC. TNBC cells were cultured under chronic hypoxia (1% O2) or normoxia. Western blotting was used to me

Ceralasertib-Mediated ATR Inhibition Combined With Olaparib in Advanced Cancers Harboring DNA Damage Response and Repair Alterations (Olaparib Combinations).

UNLABELLED: Poly (ADP-ribose) polymerase (PARP) inhibitors have emerged as promising therapy in cancers with homologous recombination repair deficiency. However, efficacy is limited by both intrinsic and acquired resistance. The Olaparib Combinations basket trial explored olaparib alone and in combination with other homologous recombination-directed targeted therapies. Here, we report the results of the arm in which olaparib was combined with the orally bioavailable ataxia telangiectasia and RAD3-related inhibitor ceralasertib in patients with relapsed or refractory cancers harboring DNA damag

Rb1 deficiency induces synthetic lethality with ATR and PKMYT1 coinhibition in breast cancer cell lines and patient-derived xenografts.

Targeting cell cycle checkpoints has emerged as a promising strategy in cancer therapy, yet single-agent inhibitors often fail because of compensatory mechanisms. Here, we demonstrate that coinhibition of ATR (RP-3500) and PKMYT1 (RP-6306) induces synthetic lethality in Rb1-deficient breast cancers by disrupting both S-G2 and G2-M checkpoints. This leads to replication stress, premature mitotic entry, and accumulation of DNA damage. In vitro, Rb1-deficient breast cancer cells exhibited marked apoptosis and loss of clonogenic survival, whereas Rb1-proficient models remained resistant to combina

RAD51C germline mutations in breast and ovarian cancer cases from high-risk families.

BRCA1 and BRCA2 are the most well-known breast cancer susceptibility genes. Additional genes involved in DNA repair have been identified as predisposing to breast cancer. One such gene, RAD51C, is essential for homologous recombination repair. Several likely pathogenic RAD51C mutations have been identified in BRCA1- and BRCA2-negative breast and ovarian cancer families. We performed complete sequencing of RAD51C in germline DNA of 286 female breast and/or ovarian cancer cases with a family history of breast and ovarian cancers, who had previously tested negative for mutations in BRCA1 and BRCA

HRR gene promoter methylation-guided nomogram predicts PARP inhibitor efficacy and progression-free survival in high-grade serous ovarian cancer.

BACKGROUND: High-grade serous ovarian cancer (HGSOC) remains a lethal gynecologic malignancy with limited tools for accurately predicting PARP inhibitor (PARPi) efficacy. While promoter methylation of homologous recombination repair (HRR) genes is a potential biomarker, its clinical utility remains underexplored. This study aimed to systematically evaluate HRR gene promoter methylation profiles to predict PARPi sensitivity in HGSOC patients. METHODS: We conducted a retrospective analysis of 96 HGSOC patients receiving PARPi maintenance therapy (olaparib or niraparib). The promoter methylation

A yeast excision-repair gene is inducible by DNA damaging agents.

Plasmids containing various RAD-lacZ gene fusions were integrated into the chromosome of haploid yeast cells. These integrant strains were tested for expression of Escherichia coli beta-galactosidase after treatment with agents that damage DNA or interfere with normal DNA replication. We did not observe induction of single-copy RAD1-lacZ or RAD3-lacZ fusion genes under the experimental conditions used. However, exposure of cells containing an integrated RAD2-lacZ fusion gene to UV-radiation, gamma-radiation, 4-nitroquinoline 1-oxide, or nalidixic acid resulted in 4- to 6-fold enhanced expressi

Processing of directly and indirectly ultraviolet-induced DNA damage in human cells.

Mutations caused by ultraviolet (UV)-induced DNA damage represent the initial genetic changes in the tumorigenesis of UV-induced skin cancer. Different wavelengths of UV radiation cause different kinds of DNA damage and mutations. UVB (290-320 nm) generates pyrimidine dimers by direct excitation of the DNA molecule. UVA (320-400 nm) can damage the DNA only indirectly through a photosensitized reaction. This indirect action is mediated mainly by singlet oxygen, which generates purine base modifications, and has been implicated in the carcinogenic effects of UVA. In order to study the processing
