Macrophage migration inhibitory factor in the human epididymis and semen.

During epididymal transit, mammalian spermatozoa acquire new proteins involved in the acquisition of motility and of male gamete fertilising ability. We have previously shown that membranous vesicles called epididymosomes are involved in the transfer of epididymal-originating proteins to spermatozoa. The cytokine macrophage migration inhibitory factor (MIF) is one of these proteins but the role played by MIF in relation to epididymal sperm maturation still remains unclear. As this protein has already been shown to bear different functions depending on its location, we investigated its distribu…

Identification of epididymal stones in diverse rooster populations.

The epididymal region of the male reproductive tract is essential for sperm maturation, and dysfunction of this region results in infertility. Adult roosters have been observed to develop epididymal stones and consequently have reduced fertility. Efferent ductule cysts were first observed in White Leghorn roosters ages 18 to 26 wk. By 26 wk of age, the cysts had become solid, irregularly shaped, yellow-green stones primarily containing calcium (48%). The number and size of stones (9 to 160 microm, largest diameter) increased with age in affected males. Incidence ranged from 0 to 94% within roo…

Royal jelly supplementation in semen extender enhances post-thaw quality and fertility of Nili-Ravi buffalo bull sperm.

Two experiments were conducted to evaluate the effect of royal jelly (RJ) on post-thaw sperm quality, in vitro and in vivo fertility rate of cryopreserved buffalo bull sperm. The semen was collected from three mature regular donor buffalo bulls, ejaculates were pooled and semen evaluated initially. In Experiment 1, the ejaculates were extended in tris-citric acid diluter supplemented with different RJ concentrations (0, 0.05, 0.1, 0.2, 0.3 or 0.4%). The diluted semen was cooled to 4°C, packaged into 0.5 mL straws and frozen using standard procedure. The straws were thawed and assessed for sper

Expression of matrix metalloproteinases (MMP-2, MMP-9) and their inhibitors (TIMP-1, TIMP-2) in canine testis, epididymis and semen.

This study aims to investigate the role of matrix metalloproteinases (MMPs) in determining semen quality and to evaluate the expression and cellular localization of MMP-2, MMP-9, tissue inhibitor of metalloproteinase-1 (TIMP-1) and TIMP-2 in the testes, epididymis and ejaculated spermatozoa. Gelatinase activities between normal (n = 21) and abnormal (n = 25) semen samples showed a significant, sixfold increase in proMMP-2 and MMP-2 activity in high than low sperm concentration samples (p < 0.001). ProMMP-9 and MMP-9 levels were significantly elevated in samples with low sperm counts compared t

Improving ICSI: A review from the spermatozoon perspective.

UNLABELLED: Intracytoplasmic sperm injection (ICSI) is the most frequently applied method for fertilization making the process of identifying the perfect spermatozoon fundamental. Herein we offer a critical and thorough presentation on the techniques reported regarding (i) handling and preparing semen samples, (ii) identifying and 'fishing' spermatozoa, and (iii) improving key factors, such as motility for a successful ICSI practice. These approaches are suggested to make the process easier and more effective especially in atypical and challenging circumstances. Furthermore, we present an epig

[Peroxidative damage induced by cumene hydroperoxide in testis and epididymis of rats in vivo].

OBJECTIVE: To establish an oxidative stress model induced by cumene hydroperoxide (cHP) in testis and epididymis of rats in vivo, and to understand the peroxidative damage of oxidative stress in testis, epididymal sperm and its propensity to induce nuclear DNA damage during spermatogenesis and sperm maturation in vivo. METHODS: An organic hydroperoxide, cHP, 70% aqueous, diluted by 0.9% NaCl, was employed as model prooxidant. Ninety-day-old male Wistar rats were divided into a control and three cHP groups, and were administered intraperitoneally 0, 1/10, 1/6 and 1/4 LD50 cHP per day respectiv…

Altered G-Protein Transduction Protein Gene Expression in the Testis of Infertile Patients with Nonobstructive Azoospermia.

Recent studies have shown that several members of the G-protein-coupled receptors (GPCR) superfamily play crucial roles in the maintenance of ion-water homeostasis of the sperm and Sertoli cells, development of the germ cells, formation of the blood barrier, and maturation of sperm. The GPCR, guanyl-nucleotide exchange factor, membrane traffic protein, and small GTPase genes were analyzed by microarray and bioinformatics (3513 sperm and Sertoli cell genes). In the microarray analyses of three human cases with different nonobstructive azoospermia sperm, the expression of GOLGA8IP, OR2AT4, PHKA1

Ameliorative effect of L-carnitine on chronic lead-induced reproductive toxicity in male rats.

Lead toxicity is one of the causative agents of male infertility that raised concern from environmental contamination worldwide. L-carnitine, a biologically active amino acid, present in high concentration in the reproductive organs such as the epididymis, is involved in sperm maturation. The possible protective effect of L-carnitine in experimentally lead-induced male reproductive toxicity in rats was evaluated in this study. Thirty adult male Wistar rats were divided into three groups. Group 1: the negative control group was treated with normal saline; Group 2: exposed to 50 mg/kg lead aceta…

Optimizing Assisted Reproductive Outcomes using a Sperm Vitrification Device in severe Oligozoospermia: A Breakthrough in Male Infertility Treatment.

This case study highlights the effective management of severe oligozoospermia using innovative sperm vitrification techniques. A 32-year-old woman and her 38-year-old husband sought treatment for primary infertility after three years of unsuccessful attempts to conceive. The male partner was diagnosed with severe oligozoospermia, which means having a low sperm count of 2 million/ml progressive motility and abnormal morphology. Advanced vitrification technology was employed to preserve 13 viable spermatozoa. The female partner's hormonal profile was normal with no reproductive issues, and she u

The importance of both an early orchidopexy and germ cell maturation for fertility.

Delayed orchidopexy for undescended testes has been associated with abnormal testicular histology, but the effect on later fertility is unknown. We aimed to establish the importance of the first postnatal maturational step-ie, the transformation of gonocytes into Ad (dark) spermatogonia on fertility. We matched histological findings of the testes from 31 patients who had undergone an early orchidopexy with their total number of sperm. If Ad spermatogonia were present, 17 (94%) of 18 (95% CI 72.7-99.9) of the men had a total sperm count of 40 x 10(6)/ejaculate or greater. By contrast, despite s

Semen Thresholds of Normality Established by the WHO Do Not Reveal Genome Instability-A Potential Occult Male Factor.

Semen parameters are unable to inform on the function or fertilizing capacity of the male gamete. Standardized methods are provided by the WHO but, the lower reference limits have reduced sensitivity to predict chances of conception. Subfertile men may be falsely classified as "normal" and a male factor contributing to genome instability may be overlooked. Semen parameters, sperm DNA fragmentation (SDF), sperm chromatin maturity and stability, and sperm aneuploidy were assessed in fertile (F), subfertile normozoospermic (SN) and subfertile non-normozoospermic males (SN-N). Standardized assays

Enhanced fat consumption potentiates acrylamide-induced oxidative stress in epididymis and epididymal sperm and effect spermatogenesis in mice.

Acrylamide (ACR) and high contents of fat could be found co-existent in many foods processed by high temperature, such as deep-frying and roasting. This study investigated the effect of enhanced fat consumption on deficits of spermatogenesis induced by ACR, and explored potential mechanisms of oxidative damage involved in this pathology in mice. Results show that enhanced feeding of corn oil and pork fat on mice potentiated the decreases of spermatogonia along with mature sperms after treatment of ACR, and that spermatozoa quality is significantly reduced as a result of enhanced feeding of cor

Defects in secretory pathway trafficking during sperm development in Adam2 knockout mice.

Adam2-null and Adam3-null male mice exhibit reduced levels of one or more ADAM proteins on mature sperm, in addition to the loss of the genetically targeted protein. ADAM protein loss was believed to occur posttranslationally, although the timing of loss and the mechanism by which the loss occurred were not explored. In this study we have found that in Adam3-null mice, fertilin beta (also known as ADAM2) is lost during the formation of testicular sperm. In Adam2-null males, most cyritestin (ADAM3) protein is also lost at this stage, but 25% of cyritestin is lost later, during sperm passage thr

Rapid protamine evolution suppresses meiotic drive in Drosophila.

Many animal species replace histones with protamines during spermatogenesis. Although essential for sperm function, protamines evolve rapidly for reasons that remain unknown. Using in vivo gene replacement, we examined the causes and consequences of the rapid evolution of Mst77F, a protamine essential for male fertility in Drosophila melanogaster (D. melanogaster). Replacing Mst77F with divergent orthologs caused DNA compaction defects in X-bearing sperm, resulting in fewer mature X-bearing sperm and male-biased progeny. Reducing D. melanogaster Mst77F dosage caused the same bias, whereas incr

A comprehensive review about sperm-oocyte interactions and key activation factors of fertilization in mammals.

Mammalian fertilization involves the migration of spermatozoa through the female reproductive system. Early embryonic development is a consequence of several steps and signaling pathways being activated, as well as biochemical and morphological modifications of spermatozoa that enable them to penetrate the membrane of mature oocytes. There are some crucial steps known to clearly explain the process of fertilization, starting with hyperactivation of spermatozoa, mutual recognition, and binding of gametes mediated by receptors located on the surface membranes of both gametes. The final step is f

POU4F2/Brn-3b is Essential for Spermatogenesis and its Disruption is Linked to Male Infertility in Mice and Humans.

Male infertility is rising globally, yet its causes remain unclear. This study identifies the transcription factor Brn-3b (POU4F2) as essential for spermatogenesis and sperm function. Brn-3b is highly expressed in mature spermatids and infertility in constitutive male Brn-3b knockout (KO) mice is characterised by structural and functional testicular changes such as reduced sperm counts, impaired motility and ultrastructural defects including disrupted acrosomes and defects in the mitochondria and flagella. RNA-seq analyses reveal significant changes in Brn-3b-dependent regulation of genes esse

Airway exposure to TiO2 nanoparticles and quartz and effects on sperm counts and testosterone levels in male mice.

Several types of engineered nanoparticles (ENP) have been shown to adversely affect male reproduction in rodent studies, but the airway route of exposure has been little investigated. This precludes adequate risk assessment of ENP exposure in occupational settings. Titanium dioxide nanoparticles (TiO2 NP) have been shown to affect total sperm count in adult male mice after intravenous and oral administration. This study aimed to investigate whether also airway exposure would affect sperm counts in male mice. Mature C57BL/6J mice were intratracheally instilled with 63 μg of rutile nanosized TiO

Reinvestigation of the sperm ultrastructure of Hypoderaeum conoideum (Digenea: Echinostomatidae).

Spermatological characteristics of the digenean Hypoderaeum conoideum (Echinostomatidae) collected from Anas platyrhynchos in the Lac d'Annecy (France) were reinvestigated using transmission electron microscopy. The previous study on this species only describes the presence of two axonemes of unequal lengths, a mitochondrion, a posterior nucleus, and the disposition of cortical microtubules. The present ultrastructural study reveals that the mature spermatozoon of H. conoideum is a filiform cell tapered at both extremities. The sperm cell exhibits the characteristics of a digenean spermatozoa
