Bioreducible polyethylenimine nanoparticles for the efficient delivery of nucleic acids.

Recently, non-viral vectors for nucleic acid delivery have received considerable attention. Among the various non-viral vectors, branched polyethylenimine (bPEI, 25 kDa) has been one of the most widely used carrier systems due to its high transfection efficiency, however, it imparts high cytotoxicity. In this study, we have crosslinked bPEI with a bioreducible linker, 3,3'-dithiodipropionic acid (DTPA), via electrostatic interactions to obtain DTPA crosslinked bPEI (DP) nanoparticles. The crosslinking significantly reduced the cytotoxicity of the nanoparticles. To arrive at the best formulatio

Monodisperse boronate polymeric particles synthesized by a precipitation polymerization strategy: particle formation and glycoprotein response from the standpoint of the Flory-Huggins model.

The development of a highly specific recognition system for glycoprotein capture from complex biological samples is a prerequisite to the success of mass spectra-based glycoproteomics analysis. To achieve this purpose, a one-pot precipitation polymerization (PP) strategy with a novel solvent system composed of water/ethanol (4:1, v/v) is described for preparing boronate-affinity polymeric micro/nano particles using 4-vinylphenylboronic acid (VPBA) as the functional monomer and N,N'-methylenebis(acrylamide) (MBA) alone or together with divinylbenzene (DVB) as the cross-linker(s). The proposed p

Structural characterization of the Borrelia burgdorferi outer surface protein BBA73 implicates dimerization as a functional mechanism.

Borrelia burgdorferi, which is the causative agent of Lyme disease, is transmitted from infected Ixodes ticks to a mammalian host following a tick bite. Upon changing the host organism from an Ixodes tick to a warm-blooded mammal, the spirochete must adapt to very different conditions, which is achieved by altering the expression of several genes in response to a changing environment. Recently, considerable attention has been devoted to several outer surface proteins, including BBA73, that undergo dramatic upregulation during the transmission of B. burgdorferi from infected Ixodes ticks to mam

In silico, in vitro and in vivo analysis of binding affinity between N and C-domains of Clostridium perfringens alpha toxin.

Clostridium perfringens alpha toxin/phospholipase C (CP-PLC) is one of the most potent bacterial toxins known to cause soft tissue infections like gas gangrene in humans and animals. It is the first bacterial toxin demonstrated to be an enzyme with phospholipase, sphingomyelinase and lecithinase activities. The toxin is comprised of an enzymatic N-domain and a binding C-domain interconnected by a flexible linker. The N-domain alone is non-toxic to mammalian cells, but incubation with C-domain restores the toxicity, the mechanism of which is still not elucidated. The objectives of the current s

The structure of the XPF-ssDNA complex underscores the distinct roles of the XPF and ERCC1 helix- hairpin-helix domains in ss/ds DNA recognition.

Human XPF/ERCC1 is a structure-specific DNA endonuclease that nicks the damaged DNA strand at the 5' end during nucleotide excision repair. We determined the structure of the complex of the C-terminal domain of XPF with 10 nt ssDNA. A positively charged region within the second helix of the first HhH motif contacts the ssDNA phosphate backbone. One guanine base is flipped out of register and positioned in a pocket contacting residues from both HhH motifs of XPF. Comparison to other HhH-containing proteins indicates a one-residue deletion in the second HhH motif of XPF that has altered the hair

Accelerated gene transfer through a polysorbitol-based transporter mechanism.

Here we report an accelerated gene transfer through a polysorbitol-based osmotically active transporter (PSOAT) that shows several surprising results through interesting mechanisms. The nano-sized and well-complexed PSOAT/DNA particles are less toxic, stable at serum and show no aggregation after lyophilization due to their polysorbitol backbone. The transfection is remarkably accelerated both in vitro and in vivo, presumably due to a transporter mechanism of PSOAT in spite of possibility of reduction of transfection by many hydroxyl groups in the transporter. PSOAT possesses a transporter mec

Two novel mutations in the ED1 gene in Japanese families with X-linked hypohidrotic ectodermal dysplasia.

X-linked hypohidrotic ectodermal dysplasia (XLHED), which is characterized by hypodontia, hypotrichosis, and hypohidrosis, is caused by mutations in ED1, the gene encoding ectodysplasin-A (EDA). This protein belongs to the tumor necrosis factor ligand superfamily. We analyzed ED1 in two Japanese patients with XLHED. In patient 1, we identified a 4-nucleotide insertion, c.119-120insTGTG, in exon 1, which led to a frameshift mutation starting from that point (p.L40fsX100). The patient's mother was heterozygous for this mutation. In patient 2, we identified a novel missense mutation, c.1141G>C, i

The molecular mechanism of toxin-induced conformational changes in a potassium channel: relation to C-type inactivation.

Recently, a solid-state NMR study revealed that scorpion toxin binding leads to conformational changes in the selectivity filter of potassium channels. The exact nature of the conformational changes, however, remained elusive. We carried out all-atom molecular dynamics simulations that enabled us to cover the complete pathway of toxin approach and binding, and we validated our simulation results by using solid-state NMR data and electrophysiological measurements. Our structural model revealed a mechanism of cooperative toxin-induced conformational changes that accounts both for the signal chan

Identification of the substrate-binding exosites of two snake venom serine proteinases: molecular basis for the partition of two essential functions of thrombin.

The venom of the South American snake Bothrops jararaca contains two serine proteinases, bothrombin and the platelet-aggregating enzyme PA-BJ, which share 66% sequence identity. Each of these proteinases possesses one of the two essential procoagulant functions of thrombin-the clotting of fibrinogen and platelet aggregation. Thus, bothrombin clots fibrinogen but has no direct effect on platelets, unless in the presence of exogenous fibrinogen. PA-BJ induces platelet aggregation by interacting with the protease-activated platelet receptor without clotting fibrinogen. On the other hand, thrombin

Defective membrane interactions of familial Parkinson's disease mutant A30P alpha-synuclein.

alpha-Synuclein (alpha-Syn) is an abundant presynaptic protein of unknown function, which has been implicated in the pathogenesis of Parkinson's disease. Alpha-Syn has been suggested to play a role in lipid transport and synaptogenesis, and growing evidence suggests that alpha-Syn interactions with cellular membranes are physiologically important. In the current study, we demonstrate that the familial Parkinson's disease-linked A30P mutant alpha-Syn is defective in binding to phospholipid vesicles in vitro as determined by vesicle ultracentrifugation, circular dichroism spectroscopy, and low-a

Brownian dynamics simulations of interaction between scorpion toxin Lq2 and potassium ion channel.

The association of the scorpion toxin Lq2 and a potassium ion (K(+)) channel has been studied using the Brownian dynamics (BD) simulation method. All of the 22 available structures of Lq2 in the Brookhaven Protein Data Bank (PDB) determined by NMR were considered during the simulation, which indicated that the conformation of Lq2 affects the binding between the two proteins significantly. Among the 22 structures of Lq2, only 4 structures dock in the binding site of the K(+) channel with a high probability and favorable electrostatic interactions. From the 4 candidates of the Lq2-K(+) channel b

Protease inhibition and absorption enhancement by functional nanoparticles for effective oral insulin delivery.

Complexing agents such as diethylene triamine pentaacetic acid (DTPA) are known to disrupt intestinal tight junctions and inhibit intestinal proteases by chelating divalent metal ions. This study attempts to incorporate these benefits of DTPA in functional nanoparticles (NPs) for oral insulin delivery. To maintain the complexing agent concentrated on the intestinal mucosal surface, where the paracellular permeation enhancement and enzyme inhibition are required, DTPA was covalently conjugated on poly(γ-glutamic acid) (γPGA). The functional NPs were prepared by mixing cationic chitosan (CS) wit

Dietary phosphate restriction prevents the appearance of sarcopenia signs in old mice.

BACKGROUND: Sarcopenia is defined by the progressive and generalized loss of muscle mass and function associated with aging. We have previously proposed that aging-related hyperphosphataemia is linked with the appearance of sarcopenia signs. Because there are not effective treatments to prevent sarcopenia, except for resistance exercise, we propose here to analyse whether the dietary restriction of phosphate could be a useful strategy to improve muscle function and structure in an animal model of aging. METHODS: Five-month-old (young), 24-month-old (old) and 28-month-old (geriatric) male C57B

Case Report: Electrical impedance tomography-guided ultra-protective lung ventilation during veno-venous extracorporeal membrane oxygenation support for fat embolism syndrome complicated by severe acute respiratory distress syndrome.

BACKGROUND: Fat embolism syndrome (FES) is a rare complication of severe multiple trauma. Electrical impedance tomography (EIT) is a noninvasive, real-time, bedside tool for monitoring regional pulmonary ventilation. Veno-venous extracorporeal membrane oxygenation (VV-ECMO) may improve outcomes in critically ill patients in whom conventional mechanical ventilation fails. Ultra-protective lung ventilation strategy (ULPVS) uses very low tidal volumes to markedly reduce lung stress and mitigate ventilator-induced lung injury (VILI). A Full Polymer Piezoelectric Flextensional Energy Harvester.

This study presents a full polymer piezoelectric flextensional energy harvester (FPPFEH) comprising a single-layer poly(vinylidene fluoride) (PVDF) film bonded to a 3D-printed polylactic acid (PLA) flextensional frame. For an arm inclination angle of θ=10°, the free-body model gives a theoretical geometric force-amplification factor of MF=cotθ≈5.67; this value represents an ideal upper bound and was not independently validated by local force or strain measurements. During assembly, the film was tensioned only to remove visible slack and maintain a flat configuration. No intentional pretension

Two-dimensional dynamics and microstructure of dense suspensions of ideally polarizable particles in an electric field: The nontrivial effect of confinement.

The dynamics and microstructure of two-dimensional suspensions of ideally polarizable particles in an electric field are studied using large-scale numerical simulations. The particles are assumed to carry no net charge and thus known to undergo a nonlinear electrokinetic phenomenon termed dipolophoresis-the combination of dielectrophoresis and induced-charge electrophoresis. For ideally polarizable particles, the effect of induced-charge electrophoresis is dominant, which results in chaotic and diffusive motions. Up to an area fraction ϕ≈50%, the diffusive particle dynamics get hindered due to

Cardiovascular damage, arrhythmogenesis and overlapping Alzheimer and Parkinson's diseases and TDP-43 pathology start in pediatric and young adult urban populations: Magnetic ultrafine particulate matter and industrial particles with motion behavior play a key role.

Cardiac damage and neurodegeneration simultaneously evolve in children and young adults, exposed to environmental oxidative, magnetic, motion capable, ultrafine particulate matter (UFPM) and industrial nanoparticles (NPs). Particles move in situ when exposed to DC/AC magnetic 30-50 μT fields, and are documented in fetal hearts, brains and placentas. We identify magnetic NPs and measured motion behavior under AC and DC fields in ventricles, septum, atrioventricular (AV) conduction axis, auricles and ventricles, in healthy hearts of 18 Metropolitan Mexico City (MMC) 30.4 ± 9.4y olds with Alzheim

Dynamic Navigation in Endodontics: Guided Access Cavity Preparation by Means of a Miniaturized Navigation System.

In the case of teeth with pulp canal calcification (PCC) and apical pathology or pulpitis, root canal treatment can be very challenging. PCC are common sequelae of dental trauma but can also occur with stimuli such as caries, bruxism, or after placing a restoration. In order to access the root canal as minimally invasive as possible in case of a necessary root canal treatment, dynamic navigation has recently been introduced in endodontics in addition to static navigation. The use of a dynamic navigation system (DNS) requires pre-operative cone-beam computed tomography (CBCT) imaging and a digi
