Bio-isopropanol production in Corynebacterium glutamicum: Metabolic redesign of synthetic bypasses and two-stage fermentation with gas stripping.

Isopropanol is a commodity chemical widely used as a biofuel, fuel additive, rubbing alcohol and intermediate in various fields. Here, an engineered Corynebacterium glutamicum overproducing isopropanol was developed. To our knowledge, despite a representative industrial host to produce valuable chemicals, the high-level production of isopropanol in C. glutamicum has never been reported. First, the problem of the inability to produce isopropanol was solved by finding a key factor in its metabolism. The consolidation and modular optimization of synthetic bypasses including succinate and mevalona

[Estimation of efficiency reamberin in the correction of endogenous intoxication syndrome at the patients with the phlegmonous and gangrenous forms of erysipelas on background diabetes mellitus at their surgical treatment].

The influence of modern detoxic preparation reamberin on indicators of the of endogenous intoxication syndrome (EIS)--the level of average molecules, malon dialdehyde, cyrcukatiry immune complexes and its fractional composition in blood serum of the patients with destructive forms of erysipelas--phlegmonous and gangrenous on background diabetes mellitus (DM). It is established that the inclusion of reamberin in the complex surgical treatment of destructive forms of erysipelas on background DM accelerates the elimination of SEI. From this we can recommend the use of reamberin in the complex sur

Morphology and enzyme histochemistry of dorsal muscles of the cat neck.

An examination has been made of the five dorsal muscles of the cat neck which insert into the lamboidal crest. The three larger muscles, splenius, biventer cervicis, and complexus, are characterized by the presence of tendinous inscriptions which serve as the insertion points of shorter muscle fibers which do not run the length of the muscle. Longer fibers are bound by the inscriptions and, thus can transmit tension developed by shorter fibers. These three muscles are multiply innervater by nerves emerging from upper cervical spinal roots. Multiple innervation is not associated with multiple e

Computational study of multicharged cyclodextrin derivatives with deep cavities for high-affinity host-guest recognition.

Cyclodextrin (CD) derivatives bearing multicharged side chains exhibit enhanced host-guest binding, with higher selectivity and affinity compared with natural CDs, due to their deep cavities, polar binding sites and electrostatic interactions. To further understand the relationship between their structures and molecular recognition, molecular dynamics simulations were performed on a series of multicharged CD hosts with per-6-substituted side chains and their oppositely charged guests. In the case of rocuronium bromide (ROC) as the guest, umbrella sampling identified glutamic acid-substituted γ

Fluopyram in agricultural ecosystems: environmental fate, microbial transformation, ecotoxicity, and resistance risks of an SDHI fungicide.

Fluopyram is a next-generation succinate dehydrogenase inhibitor (SDHI) fungicide, which is very popular in the management of economically significant fungal conditions in cereals, fruits, vegetables, and ornamental crops. It works by inhibiting the complex II present in the mitochondria by blocking succinate dehydrogenase, which electron transport and energy generation, eventually hindering fungal development leading to death of the pathogen. Besides having good fungicidal effects, fluopyram has also demonstrated a nematicidal effect, which has also enhanced its utility in various agricultura

Iron-Sulfur Cluster Targeting: A Novel Perspective for Treating Alcohol-Related Lipid Metabolic Diseases.

Iron-sulfur (Fe-S) clusters are pivotal molecular cofactors bridging inorganic chemistry and organic life processes, with their tightly regulated biosynthesis and function underpinning cellular metabolic homeostasis. Chronic alcohol-induced metabolic stress impairs Fe-S cluster stability and biosynthesis via reactive oxygen species (ROS)-mediated oxidative damage and direct acetaldehyde toxicity, culminating in functional failure of this core cofactor. Current evidence is synthesized herein to delineate the hierarchical pathogenic cascade triggered by Fe-S cluster dysfunction. At the cellular

The Effect of Anti-Inflammatory Dimethylmalonic Acid on the Neurobehavioral Phenotype of a Neonatal ASD Model Induced by Antiepileptic Valproic Acid.

Background: Valproic acid (VPA) is a medication used to treat epilepsy, bipolar disorder, and migraine. If taken during pregnancy, it can cause neural tube defects (NTDs) and leads to offspring ASD behavioral phenotype. It has recently been found that early postnatal VPA exposure can also induce the ASD phenotype, but the details of model production and intervention still need further investigation. Dimethylmalonic acid (DMM), a competitive inhibitor of succinate dehydrogenase, blocks the key element succinate of OXPHOS, decreasing the secretion of anti-inflammatory cytokines and ROS productio

Self-neutralizing tannin-citric acid wood adhesives.

Tannin-based adhesives have historically relied on formaldehyde or hexamine hardeners, but recent research has focused on sustainable alternatives, such as tannin-citric acid adhesives, which avoid harmful chemicals and show prospect to provide a viable eco-friendly solution for wood bonding applications. However, these adhesives typically suffer from low pH, leading to long-term degradation of wood substrates. This study aims to regulate the pH of tannin-citric acid adhesives by introducing morpholine complexes with weak acids (succinic and malic acids), thereby improving the adhesive's mecha

Metabolomic and microbiota profiles in cervicovaginal lavage fluid of women with high-risk human papillomavirus infection.

The presence of high-risk human papillomavirus (HR-HPV) contributes to the development of cervical lesions and cervical cancer. Recent studies suggest that an imbalance in the cervicovaginal microbiota might be a factor in the persistence of HR-HPV infections. In this study, we collected 156 cervicovaginal fluid (CVF) of women with HR-HPV infection, which were divided into three groups (negative for intraepithelial lesions = 78, low/high-grade squamous intraepithelial lesions = 52/26). We performed metabolomics and 16 S rRNA sequencing to identify changes in metabolites and cervicovaginal micr

Structural Basis of Redox-Dependent Affinities of Dihydroorotate Dehydrogenase for Its Substrates and Products.

Dihydroorotate dehydrogenase (DHODH), containing a cofactor, flavin mononucleotide (FMN), catalyzes the oxidation of dihydroorotate to orotate. In Trypanosoma brucei, the causative parasite of African trypanosomiasis, the DHODH enzyme (TbDHODH) couples this reaction with the reduction of fumarate to succinate. These reactions occur successively in the same catalytic site through a ping-pong bi-bi mechanism. FMN is reduced in the first step and re-oxidized in the second one. Here, we investigated how the redox state of FMN influences the binding of substrates and products to TbDHODH by NMR anal

STING regulates metabolic reprogramming in macrophages via HIF-1α during Brucella infection.

Macrophages metabolic reprogramming in response to microbial insults is a major determinant of pathogen growth or containment. Here, we reveal a distinct mechanism by which stimulator of interferon genes (STING), a cytosolic sensor that regulates innate immune responses, contributes to an inflammatory M1-like macrophage profile upon Brucella abortus infection. This metabolic reprogramming is induced by STING-dependent stabilization of hypoxia-inducible factor-1 alpha (HIF-1α), a global regulator of cellular metabolism and innate immune cell functions. HIF-1α stabilization reduces oxidative pho

Use of GC-MS based metabolic fingerprinting for fast exploration of fungicide modes of action.

BACKGROUND: The widespread occurrence of fungicide resistance in fungal plant pathogens requires the development of new compounds with different mode(s) of action (MOA) to avoid cross resistance. This will require a rapid method to identify MOAs. RESULTS: Here, gas chromatography-mass spectrometry (GC-MS) based metabolic fingerprinting was used to elucidate the MOAs of fungicides. Botrytis cinerea, an important pathogen of vegetables and flowers, can be inhibited by a wide range of chemical fungicides with different MOAs. A sensitive strain of B. cinerea was exposed to EC50 concentrations of

Altered Hypoxic-Adenosine Axis and Metabolism in Group III Pulmonary Hypertension.

Group III pulmonary hypertension (PH) is a highly prevalent and deadly lung disorder with limited treatment options other than transplantation. Group III PH affects patients with ongoing chronic lung injury, such as idiopathic pulmonary fibrosis (IPF). Between 30 and 40% of patients with IPF are diagnosed with PH. The diagnosis of PH has devastating consequences to these patients, leading to increased morbidity and mortality, yet the molecular mechanisms involved in the development of PH in patients with chronic lung disease remain elusive. Our hypothesis was that the hypoxic-adenosinergic sys
