From Acellular Matrices to Smart Polymers: Degradable Scaffolds that are Transforming the Shape of Urethral Tissue Engineering.

Several congenital and acquired conditions may result in severe narrowing of the urethra in men, which represent an ongoing surgical challenge and a significant burden on both health and quality of life. In the field of urethral reconstruction, tissue engineering has emerged as a promising alternative to overcome some of the limitations associated with autologous tissue grafts. In this direction, preclinical as well as clinical studies, have shown that degradable scaffolds are able to restore the normal urethral architecture, supporting neo-vascularization and stratification of the tissue. Whi…

Blocking of matrix metalloproteinases-13 responsive peptide in poly(urethane urea) for potential cartilage tissue engineering applications.

The matching of scaffold degradation rate with neotissue growth is required for tissue engineering applications. Timely provision of proper spaces especially for cartilage tissue engineering plays a pivotal role in chondrocyte cluster formation. In this study, poly(urethane urea) was synthesized using conventional two-stage method by extending the isocyanate group terminated prepolymers with different amounts of GPLGLWARK peptide, which responses the degrading induced by matrix metalloproteinase 13, the main proteinase for cartilage matrix degradation. The Fourier transform infrared spectromet…

Fabrication of PEOT/PBT Nanofibers by Atmospheric Pressure Plasma Jet Treatment of Electrospinning Solutions for Tissue Engineering.

This study focuses on the enhanced electrospinning of 300-Polyethylene oxide-polyethylene oxide terephthalate/polybutylene terephthalate (PEOT/PBT). An atmospheric pressure plasma jet for liquid treatment is applied to a solution with 9 w/v% PEOT/PBT dissolved in either chloroform (CHCl3 ), CHCl3  + N,N-dimethylformamide (DMF), CHCl3  + methanol (MeOH), or CHCl3  + hexafluoroisopropanol (HFIP). For all conditions, the plasma-treated samples present better-quality fibers: less or no-beads and uniform fiber diameter distribution. Except for CHCl3  + DMF, no significant changes to the material bu

A Gelatin Hydrogel-Containing Nano-Organic PEI⁻Ppy with a Photothermal Responsive Effect for Tissue Engineering Applications.

The introduction and designing of functional thermoresponsive hydrogels have been recommended as recent potential therapeutic approaches for biomedical applications. The development of bioactive materials such as thermosensitive gelatin-incorporated nano-organic materials with a porous structure and photothermally triggerable and cell adhesion properties may potentially achieve this goal. This novel class of photothermal hydrogels can provide an advantage of hyperthermia together with a reversibly transformable hydrogel for tissue engineering. Polypyrrole (Ppy) is a bioorganic conducting polym…

Improving chondrocyte harvests with poly(2-hydroxyethyl methacrylate) coated materials in the preparation for cartilage tissue engineering.

Remarkable advances have been made in cartilage regenerative medicine to cure congenital anomalies including microtia, tissue defects caused by craniofacial injuries, and geriatric diseases such as osteoarthritis. However, those procedures require a substantial quantity of chondrocytes for tissue engineering. Previous studies have required several passages to obtain sufficient cell numbers for three-dimensional and monolayer cultures. Thus, our objective was to improve the quantity of chondrocytes that can be obtained by examining an anti-fouling polyhydrophilic chemical called poly(2-hydroxye…

Fabrication & Characterization of 3D Electrospun Biodegradable Nanofibers for Wound Dressing, Drug Delivery and Other Tissue Engineering Applications.

BACKGROUND: The use of electrospinning technology (ET) in fabrication of threedimensional biodegradable electrospun nanofibers scaffolds (BENS) has recently gained considerable attention in tissue engineering. BENS are superior to other existing scaffolds in tissue regeneration as they provide high surface area-to-volume ratio, possess high porosity, and offer a biomimetic environment in a nanometer scale. OBJECTIVES: To fabricate & characterize BENS using Poly (ethylene glycol) (PEG35000) as a biodegradable polymer loaded with Amoxicillin Trihydrate (AMX) for use as a wound dressing. METHOD…

Transplantation of tissue engineering neural network and formation of neuronal relay into the transected rat spinal cord.

Severe spinal cord injury (SCI) causes loss of neural connectivity and permanent functional deficits. Re-establishment of new neuronal relay circuits after SCI is therefore of paramount importance. The present study tested our hypothesis if co-culture of neurotrophin-3 (NT-3) gene-modified Schwann cells (SCs, NT-3-SCs) and TrkC (NT-3 receptor) gene-modified neural stem cells (NSCs, TrkC-NSCs) in a gelatin sponge scaffold could construct a tissue engineering neural network for re-establishing an anatomical neuronal relay after rat spinal cord transection. Eight weeks after transplantation, the …

High-fidelity tissue engineering of patient-specific auricles for reconstruction of pediatric microtia and other auricular deformities.

INTRODUCTION: Autologous techniques for the reconstruction of pediatric microtia often result in suboptimal aesthetic outcomes and morbidity at the costal cartilage donor site. We therefore sought to combine digital photogrammetry with CAD/CAM techniques to develop collagen type I hydrogel scaffolds and their respective molds that would precisely mimic the normal anatomy of the patient-specific external ear as well as recapitulate the complex biomechanical properties of native auricular elastic cartilage while avoiding the morbidity of traditional autologous reconstructions. METHODS: Three-di

Biosilica: Molecular Biology, Biochemistry and Function in Demosponges as well as its Applied Aspects for Tissue Engineering.

Biomineralization, biosilicification in particular (i.e. the formation of biogenic silica, SiO(2)), has become an exciting source of inspiration for the development of novel bionic approaches following 'nature as model'. Siliceous sponges are unique among silica-forming organisms in their ability to catalyze silica formation using a specific enzyme termed silicatein. In this study, we review the present state of knowledge on silicatein-mediated 'biosilica' formation in marine demosponges, the involvement of further molecules in silica metabolism and their potential applications in nano-biotech

Electrospinning and evaluation of PHBV-based tissue engineering scaffolds with different fibre diameters, surface topography and compositions.

While electrospinning is an effective technology for producing poly(hydroxybutyrate-co-hydroxyvalerate) (PHBV) micrometre-scale fibrous scaffolds for tissue regeneration, electrospinning of PHBV fibrous scaffolds composed of sub-micrometre fibres, surface-porous fibres or nanocomposite fibres is rarely explored. In this study, the average PHBV fibre diameter was successfully reduced to the sub-micrometre scale by dissolving a conductivity-enhancing salt in the polymer solution for electrospinning. Surface-porous fibres were made using a mixture of solvents, and carbonated hydroxyapatite (CHA)

The use of phospholipase A(2) to prepare acellular porcine corneal stroma as a tissue engineering scaffold.

This study was to develop a method using phospholipase A(2) (PLA(2)) to prepare acellular porcine corneal stroma (APCS) for tissue engineering. The APCS was prepared from native porcine cornea (NPC) that was treated with 200 U/ml PLA(2) and 0.5% sodium deoxycholate (SD). The removal of DNA content, representing decellularization efficiency, reached to 91%, while all hydroxyproline and 80% of glycosaminoglycan were retained in the APCS when compared with NPC. The residual PLA(2) and SD were 0.35+/-0.04 U/mg dry weight and 4.3+/-0.8 ng/mg dry weight respectively. The extracts of APCS had no inhi…

Liver tissue engineering at extrahepatic sites in mice as a potential new therapy for genetic liver diseases.

Liver tissue engineering using hepatocyte transplantation has been proposed as an alternative to whole-organ transplantation or liver-directed gene therapy to correct various types of hepatic insufficiency. Hepatocytes are not sustained when transplanted under the kidney capsule of syngeneic mice. However, when we transplanted hepatocytes with the extracellular matrix components extracted from Engelbreth-Holm-Swarm cells, hepatocytes survived for at least 140 days and formed small liver tissues. Liver engineering in hemophilia A mice reconstituted 5% to 10% of normal clotting activity, enough …

Engineering Functional Skeletal Muscle Tissues from Human Induced Pluripotent Stem Cells.

In vitro cell culture models of human skeletal muscle offer a valuable platform for studies of pathology and discovery of new therapeutics. However, traditional two-dimensional cell culture models suffer from limited durability and challenges with assessment of skeletal muscle contractile function. Furthermore, finite proliferation potential and scarce availability of patient-derived primary myoblasts limit experimental throughput when using these cells. Here, we describe a method for using human induced pluripotent stem cells (hiPSCs) to generate functional three-dimensional skeletal muscle t

Dystrophin Loss in Engineered Heart Tissues Recapitulates Clinically Relevant Aspects of Dystrophic Cardiomyopathy.

Heart failure is the leading cause of death in patients with Duchenne muscular dystrophy (DMD), but the mechanisms underlying the associated dilated cardiomyopathy (DCM) are not fully understood. To address this gap, we generated engineered heart tissues (EHTs) using CRISPR-edited human induced pluripotent stem cell-derived cardiomyocytes that lack dystrophin. These dystrophic EHTs reproduced aspects of systolic and diastolic dysfunction seen in DMD-related DCM as they showed impaired contractile function and slower kinetics. Increased beat rate variability was also observed in dystrophic EHTs

Long-term remodeling of compliance matched tissue engineered vascular grafts is sex specific.

Compliance matching of tissue engineered vascular grafts (TEVGs) is a promising technique to combat common failure modes seen clinically in small diameter vascular procedures like coronary artery bypass grafting (CABG). However, despite the influence of sex on vascular response to injury, the role of biological sex on the success of TEVG innovations such as compliance matching, namely in long-term remodeling, has been understudied. In this study we fabricated compliance matched TEVGs (CM TEVGs) and hypocompliant TEVGs (Hypo TEVGs) by adjusting the thicknesses of a high polyester urethane urea
