Neuromyelitis optica IgG stimulates an immunological response in rat astrocyte cultures.

Neuromyelitis optica (NMO) is a primary astrocyte disease associated with central nervous system inflammation, demyelination, and tissue injury. Brain lesions are frequently observed in regions enriched in expression of the aquaporin-4 (AQP4) water channel, an antigenic target of the NMO IgG serologic marker. Based on observations of disease reversibility and careful characterization of NMO lesion development, we propose that the NMO IgG may induce a dynamic immunological response in astrocytes. Using primary rat astrocyte-enriched cultures and treatment with NMO patient-derived serum or purif…

Image features of retinal astrocytic hamartoma in a patient with tuberous sclerosis complex.

PURPOSE: To describe the image features of retinal astrocytic hamartoma in a 35-year-old male patient with tuberous sclerosis complex (TSC). METHODS: Fundus photography, fundus autofluorescence (FAF), fundus fluorescein angiography (FFA), and spectral domain optical coherence tomography (SD-OCT) were performed for this retinal astrocytic hamartoma. RESULTS: Fundus photography showed that the retinal astrocytic hamartoma presented as a well-circumscribed, mulberry-like lesion consisting of glistening yellowish spherules of calcification. FAF demonstrated dense hyper-autofluorescence spots cor

Form follows function: astrocyte morphology and immune dysfunction in SIV neuroAIDS.

Cortical function is disrupted in neuroinflammatory disorders, including HIV-associated neurocognitive disorders (HAND). Astrocyte dysfunction includes retraction of foot processes from the blood-brain barrier and decreased removal of neurotransmitters from synaptic clefts. Mechanisms of astrocyte activation, including innate immune function and the fine neuroanatomy of astrocytes, however, remain to be investigated. We quantified the number of glial fibrillary acidic protein (GFAP)-labeled astrocytes per square millimeter and the proportion of astrocytes immunopositive for Toll-like receptor …

Astrocytic phosphorylation of PDK1 on Tyr9 following an excitotoxic lesion in the mouse hippocampus.

3-phosphoinositide-dependent kinase-1 (PDK1) is suggested to play important roles in the regulation of synaptic plasticity and neuronal cell … le immunohistochemistry further evaluated that these phosphorylated forms of PDK1 were localized in astrocytes not other cells. Overexpression of unphosphorylatable mutant, PDK1-Y9F leads to inhibit Protein kinase B (PKB/Akt) activation and cAMP responsive element binding protein (CREB) phosphorylation. In conclusion, our results suggested for the first time …

Phagocytized corpora amylacea as a histological hallmark of astrocytic injury in neuromyelitis optica.

Neuromyelitis optica (NMO) is an inflammatory demyelinating and necrotizing disorder of the CNS that mainly affects the optic nerve and spin … sent in most lesions assessed. These findings suggest the following pathophysiological process: the astrocytes are affected at an early phase in NMO, CA are expelled from the astrocytes and phagocytized by macrophages finally leading to clearance. A phagocytized figure and subsequent loss of CA can be a histological hallmark of astrocytic injury of NMO.

Astrocyte signaling controls spike timing-dependent depression at neocortical synapses.

Endocannabinoid mediated spike timing-dependent depression (t-LTD) is crucially involved in the development of the sensory neocortex. t-LTD at excitatory synapses in the developing rat barrel cortex requires cannabinoid CB(1) receptor (CB(1)R) activation, as well as activation of NMDA receptors located on the presynaptic terminal, but the exact signaling cascade leading to t-LTD remains unclear. We found that astrocytes are critically involved in t-LTD. Astrocytes gradually increased their Ca(2+) signaling specifically during the induction of t-LTD in a CB(1)R-dependent manner. In this way, as…

Pramipexole has astrocyte-mediated neuroprotective effects against lactacystin toxicity.

Pramipexole, a dopamine D2/D3 receptor agonist used in the treatment of Parkinson's disease, has been reported to have neuroprotective poten … t of pramipexole against the lactacystin-toxicity was not direct but a secondary effect mediated by astrocytes. Therefore, we investigated the efficacy of conditioned medium collected from mecencephalic astrocytes treated with pramipexole. The conditioned medium increased the viability of SH-SY5Y cells against the toxicity of lactacystin. Pramipexole increas …

Developmental expression of FMRP in the astrocyte lineage: implications for fragile X syndrome.

One of the most common causes of mental retardation in humans, Fragile X syndrome, results from the absence of FMRP, the protein product of … us. Prominent coexpression was also observed in ependymal cells surrounding the third ventricle and astrocytes of the glia limitans. No double-labeled cells were evident in the brains of young adult mice. Cells coexpressing FMRP and the oligodendrocyte precursor marker NG2 were also identified in the hippocampus and corpus callosum of the early postnatal bra …

Astrocyte-leucocyte interactions and the mechanisms regulating matrix degradation in CNS tuberculosis.

The CNS (central nervous system) has a unique pattern of immune response to infection. TB (tuberculosis) of the CNS is devastating with widespread tissue destruction. In TB, astrocyte-leucocyte interactions are key in regulating MMP (matrix metalloproteinase) activity and are regulated by complex signalling pathways. A synergistic interaction between interferon gamma and monocyte-derived mediators drives high-level astrocyte MMP-9 secretion; this and other networking effects are inhibited by steroids. Better understanding of regulatory mechanisms may identify potential switch points that could

Morphologic change and astrocyte response to unilateral spinal cord compression in rabbits.

In myelopathy, unilateral compression of the spinal cord in cases of disc herniation would be expected to produce Brown-Séquard syndrome. However, a transverse lesion syndrome occurs in most clinical cases. In order to reveal the mechanism by which unilateral compression induces transverse damage to the spinal cord, damage of the gray and white matter in each half of the spinal cord were evaluated quantitatively to determine the density of GFAP-positive astrocytes. The cervical spinal cord in rabbits was unilaterally compressed with a small screw. The area of each half of the damaged cord and …

Expression of mos in astrocytic tumors and its potential role in neoplastic progression.

The c-mos gene and its protein product mos, components of the mitogen-activated protein kinase transduction pathway, are known to be involved in the control of meiosis and mitosis. Apart from a study on lung carcinomas, there is little information about its role in human neoplasia. The aim of this study was to investigate expression of mos in astrocytic tumors and to correlate it with accumulation of p53. We studied expression of mos in 62 cases of supratentorial astrocytic tumor. Intracytoplasmic immunostaining for mos was found in 28 (45%) cases: 3 of 20 (15%) grade 2 astrocytomas, 9 of 20 (

Astrocyte-derived factors modulate the inhibitory effect of ethanol on dendritic development.

Numerous studies in vivo and in vitro have demonstrated that ethanol disrupts neuromorphogenesis. However, it has not been determined what role, if any, is played by non-neuronal cells in mediating this effect. We recently reported that ethanol inhibits dendritic development in low-density cultures of fetal rat hippocampal pyramidal neurons (Yanni and Lindsley, 2000: Dev Brain Res 120:233-243). In this culture system, cortical astrocytes precondition neuronal culture media for 2 days before the addition of neurons, which then develop on a separate substrate in coculture with the astrocytes. To…

Calcium/calmodulin-dependent protein kinase activity in primary astrocyte cultures.

Calcium, calmodulin-dependent protein kinase (Ca/CaM kinase) is an important component of calcium signalling mechanisms in the brain, but little is known about the properties of this protein phosphorylation system in astrocytes. Addition of calcium and calmodulin to supernatant or membrane fractions obtained from rat astrocytes in primary culture increased phosphate incorporation into an exogenously added substrate, casein, and into endogenous protein substrates; this increase was greater than that observed with either calcium alone or calmodulin alone. The calcium, calmodulin-stimulated incre…

Astrocytic reaction predominance in chronic encephalitis of Junin virus-infected rats.

Junin virus antigen distribution and astrocytic reaction to prolonged infection were characterized in rat brain by the PAP technique. During the acute stage of neurologic disease following intracerebral inoculation, Junin antigen was detected in 100% of animals, strongly in most neurons but also to a much lesser degree in scattered astrocytes, dropping to 20% of rats at 540 days postinfection. Initially labeled in all brain areas, viral antigen gradually disappeared from hippocampus but persisted irregularly in cerebral cortex, basal ganglia, Purkinje cells, pons, and medulla oblongata. Such a…

Astrocyte differentiation induced by Junín virus in rat brain cell cultures.

Morphological and immunocytochemical differentiation was observed in astroglial cell cultures of the rat infected with Junín virus. From days 3 to 6 postinoculation (p.i.), GFAP immunostaining was observed in both the perikaryon and processes of maturated astrocytes, whereas it was limited to the perikaryon in less differentiated cells. The rather slow spontaneous differentiation usually occurring in astroglial cell cultures was seen to be accelerated by viral infection, mimicking the astrocytic reaction formerly described in Junín virus-inoculated mice. Infected cell monolayers showed orderly…

Dopamine-Induced L-Lactate Production in Cortical Astrocytes Cross-Reacts with β1-Adrenoceptor-Mediated cAMP Signalling.

Astrocytes maintain homeostasis of the central nervous system and supply neurons with metabolic substrates essential for synaptic plasticity and learning. Upon stimulation by noradrenaline (NA), released primarily by locus coeruleus (LC) neurons and acting via volume transmission, astrocytes enhance aerobic glycolysis and produce L-lactate, a process critical for energy support yet dependent on β-adrenoceptor-mediated cyclic adenosine monophosphate (cAMP) signalling. In neurodegenerative diseases such as Alzheimer's disease and Parkinson's disease, early degeneration of the LC diminishes norad…
