Local Protein Synthesis at Synapses: A Driver for Synapse Diversification.

Local protein synthesis within neuronal processes seems to be crucial for the rapid and dynamic remodeling of the proteome at synaptic compartments. Indeed, this capability enables neurons to swiftly adapt their synaptic functions in response to activity. In this review, we first explore the diverse mechanisms that allow the targeted transport of mRNAs into both dendrites and axons. Then, we report evidence that local mRNAs are actively recruited for protein synthesis during plasticity. Finally, we highlight how this molecular complexity contributes to the establishment and stabilization of me

Aquaporin-4 Localization in Terminal Glial Cells of Human Mechanosensory Corpuscles: An Immunohistochemical Study of Penile Tissues.

Aquaporin-4 (AQP4), the predominant water channel in the central nervous system, has been extensively characterized in astroglia, but its presence in peripheral mechanoreceptive end-organs remains unknown. We investigated AQP4 localization in human mechanosensory corpuscles using formalin-fixed, paraffin-embedded penile tissues from 11 adult cadavers and 12 surgical prepuces. Single and double immunohistochemistry was performed in serial sections with a comprehensive battery of antibodies targeting axons (neurofilaments, neuron-specific enolase [NSE], protein gene product 9.5 [PGP9.5], synapto

Inhibition of pathogenic tau signaling via blocking of the phosphatase-activating domain by novel small molecules.

INTRODUCTION: Tau pathology is a major feature of Alzheimer's disease (AD) and multiple other adult-onset neurodegenerative diseases. Aberrant exposure of an N-terminal phosphatase-activating domain (PAD) is characteristic of pathological tau, representing a toxic gain of function. Exposure of the PAD in pathological tau leads to dysregulation of protein phosphatase 1/glycogen synthase kinase 3 (PP1/ GSK3β) signaling, inhibition of fast axonal transport, synaptic dysfunction, and altered transcription, along with other pathological consequences. Previous studies showed that TNT1, an antibody a…

Genetically induced impairment of retinal ganglion cells at the axonal level is linked to extrastriate cortical plasticity.

Leber hereditary optic neuropathy (LHON) is a maternally inherited mitochondrial disorder, which leads to initially silent visual loss due t … nsatory cortical plasticity occurring in V2/V3 is predicted by the swelling (indicating deficits of axonal transport and intracellular edema) of the macular RGC axonal layer. Increased cortical thickness (CT) in V2 and V3 was observed in peripheral regions, like visual field loss, in these mutation carriers. CT was a very discriminative measure between carriers an …

Transcriptional control of visual neural circuit development by GS homeobox 1.

As essential components of gene expression networks, transcription factors regulate neural circuit assembly. The homeobox transcription factor encoding gene, gs homeobox 1 (gsx1), is expressed in the developing visual system; however, no studies have examined its role in visual system formation. In zebrafish, retinal ganglion cell (RGC) axons that transmit visual information to the brain terminate in ten arborization fields (AFs) in the optic tectum (TeO), pretectum (Pr), and thalamus. Pretectal AFs (AF1-AF9) mediate distinct visual behaviors, yet we understand less about their development com

Significance of the plasma membrane for the nerve cell function, development and plasticity.

Lipoid character of plasma membrane namely the presence of polyenic fatty acids enables to interact with membrane proteins and in certain extent also to modulate their function. During the development, molecules of membrane fatty acids become more and more complex, and the ratio of polyenic fatty acids/saturated fatty acids in the brain rises, while the concentration of monoenic fatty acids remained relatively stable. This phenomenon is apparent also in the ratio of unsaturated fatty acids OMEGA-3 in plasma of newborns which correlates with the birth weight. Plasma membrane reflects local spec

Expression of raf serine/threonine protein kinases in the cell bodies of primary sensory neurons of the adult rat.

We have used immunocytochemistry to examine the distribution of raf protein kinases in sensory neurons of the adult rat. In lumbar and trigeminal sensory ganglia, cells of all size ranges appeared to be raf immunoreactive and this was confirmed by double labeling using subpopulation specific markers. Almost all cells labeled with Griffonia simplicifolia IB4 (a small cell marker) or immunostained by using a large cell marker (RT97) showed raf immunoreactivity. These two markers label cells known to differ in their expression of neurotrophin receptors (trk). Thus raf kinases are not confined to

Enhancing Motor and Sensory Axon Regeneration after Peripheral Nerve Injury Using Bioluminescent Optogenetics.

Introduction-Recovery from peripheral nerve injuries is poor even though injured peripheral axons can regenerate. Novel therapeutic approaches are needed. The most successful preclinical experimental treatments have relied on increasing the activity of the regenerating axons, but the approaches taken are not applicable to many nerve-injured patients. Bioluminescent optogenetics (BL-OG) is a novel method of increasing the excitation of neurons that might be similar to that found with activity-dependent experimental therapies. We investigated the use of BL-OG as an approach to promoting axon reg

Activity-driven sharpening of the regenerating retinotectal projection: effects of blocking or synchronizing activity on the morphology of individual regenerating arbors.

Both blocking activity with intraocular tetrodotoxin (TTX) and synchronizing activity with a xenon strobe light (1 Hz) prevent retinotopic sharpening of regenerating optic projection in goldfish (Meyer, 1983; Schmidt, 1985; Cook and Rankin, 1986). In this study, we tested, in both normal and regenerating projections, the effects of these two treatments on individual optic arbors. Arbors were stained via anterograde transport of HRP, drawn in camera lucida from tectal whole mounts, and analyzed for spatial extent in the plane of the retinotopic map, order of branching, number of branch endings,

CSF proteomic pathway alterations and caudate dopamine transporter deficits predict cognitive vulnerability in early Parkinson's disease.

PURPOSE: To investigate the relationships among CSF proteomic pathway alterations, dopamine transporter single-photon emission computed tomography (DAT-SPECT)-derived dopamine transporter uptake measures, and cognitive vulnerability, longitudinal cognitive decline, and incident mild cognitive impairment (MCI) in early PD. METHODS: We analyzed data from the Parkinson's Progression Markers Initiative, integrating CSF proteomics, DAT uptake measures, and longitudinal cognitive assessments. Cross-sectional analyses included 318 PD patients and 152 healthy controls with baseline clinical, DAT upta

Vesicular glutamate transporters VGLUT1 and VGLUT2 define two subsets of unipolar brush cells in organotypic cultures of mouse vestibulocerebellum.

Different isoforms of a vesicular glutamate transporter (VGLUT) mediate glutamate uptake into synaptic vesicles of excitatory neurons. There is agreement that the VGLUTs are differentially expressed in brain, and that two isoforms, VGLUT1 and VGLUT2, are localized to excitatory axon terminals in the cerebellar cortex. While granule cells express solely VGLUT1, there is no report about the VGLUT(s) of the unipolar brush cell (UBC), the second type of glutamatergic interneuron residing in the cerebellar granular layer. In the mouse, UBCs are particularly numerous in the uvula (lobule IX) and nod

Imaging progenitor cell differentiation during central nervous system remyelination using an MRI gene reporter.

Demyelination, the loss of the myelin sheath from around otherwise intact axons, occurs in several diseases, most notably multiple sclerosis (MS). Demyelinated axons that are not remyelinated are vulnerable to irreversible degeneration and therefore therapies that enhance remyelination have been sought. However, there remains a paucity of suitable outcome measures to assess their efficacy. Early and selective localization of tau filaments to glutamatergic subcellular domains within the human anterodorsal thalamus.

Widespread cortical accumulation of misfolded pathological tau proteins (ptau) in the form of paired helical filaments is a major hallmark of Alzheimer's disease. Subcellular localization of ptau at various stages of disease progression is likely to be informative of the cellular mechanisms involving its spread. Here, we found that the density of ptau within several distinct rostral thalamic nuclei in post-mortem human tissue (n = 25 cases) increased with the disease stage, with the anterodorsal nucleus (ADn) consistently being the most affected. In the ADn, ptau-positive elements were present

Energy Metabolism in Mouse Sciatic Nerve A Fibres during Increased Energy Demand.

The ability of sciatic nerve A fibres to conduct action potentials relies on an adequate supply of energy substrate, usually glucose, to maintain necessary ion gradients. Under our ex vivo experimental conditions, the absence of exogenously applied glucose triggers Schwann cell glycogen metabolism to lactate, which is transported to axons to fuel metabolism, with loss of the compound action potential (CAP) signalling glycogen exhaustion. The CAP failure is accelerated if tissue energy demand is increased by high-frequency stimulation (HFS) or by blocking lactate uptake into axons using cinnema

Functional differences of short and long isoforms of spastin harboring missense mutation.

Mutations of the SPG4 (SPAST) gene encoding for spastin protein are the main causes of hereditary spastic paraplegia. Spastin binds to micro … w that only mutated M1 decorates microtubules of proximal axons and dendrites, and strongly impairs axonal transport in cortical neurons through a mechanism likely independent of the microtubule-severing activity of this protein.

Spastin couples microtubule severing to membrane traffic in completion of cytokinesis and secretion.

Mutations in the gene encoding the microtubule (MT)-severing protein spastin are the most common cause of hereditary spastic paraplegia, a genetic condition in which axons of the corticospinal tracts degenerate. We show that not only does endogenous spastin colocalize with MTs, but that it is also located on the early secretory pathway, can be recruited to endosomes and is present in the cytokinetic midbody. Spastin has two main isoforms, a 68 kD full-length isoform and a 60 kD short form. These two isoforms preferentially localize to different membrane traffic pathways with 68 kD spastin bein
