Inactivating mutations and overexpression of BCL10, a caspase recruitment domain-containing gene, in MALT lymphoma with t(1;14)(p22;q32).

Mucosa-associated lymphoid tissue (MALT) lymphomas most frequently involve the gastrointestinal tract and are the most common subset of extranodal non-Hodgkin lymphoma (NHL). Here we describe overexpression of BCL10, a novel apoptotic signalling gene that encodes an amino-terminal caspase recruitment domain (CARD), in MALT lymphomas due to the recurrent t(1;14)(p22;q32). BCL10 cDNAs from t(1;14)-positive MALT tumours contained a variety of mutations, most resulting in truncations either in or carboxy terminal to the CARD. Wild-type BCL10 activated NF-kappaB but induced apoptosis of MCF7 and 29

Modulation of caspase-8 and FLICE-inhibitory protein expression as a potential mechanism of Epstein-Barr virus tumorigenesis in Burkitt's lymphoma.

Ligation of the Fas receptor induces death-inducing signaling complex (DISC) formation, caspase activation, and subsequent apoptotic death of several cell types. Epstein-Barr virus (EBV)-positive group III Burkitt's lymphoma (BL) cell lines have a marked resistance to Fas-mediated apoptosis, although expressing each of the DISC components, Fas/ APO-1-associated death domain protein (FADD), and caspase-8 (FLICE/MACH/Mch5). The apoptotic pathway distal to the DISC is intact because ceramide analogs, staurosporine, and granzyme B activate caspase-3 and induce apoptosis. Fas resistance was not exp

Synergistic Neurotoxicity of environmental Cadmium and Paraquat in Parkinsonism: Unveiling the Mito-ROS/OPA1/Caspase-3/GSDME-driven Apoptosis Axis.

The increasing environmental presence of cadmium (Cd) and paraquat (PQ), driven by industrial emissions and overuse of herbicide, poses heightened risks for neurodegenerative disorders. Although each of these toxins can independently induce neuronal damage, the synergistic neurotoxic effects resulting from chronic, low-dose co-exposure to Cd and PQ remain inadequately understood. This study demonstrates that exposure to subtoxic levels of Cd and PQ concurrently induces neuronal cell death and contributes to Parkinson's disease (PD)-like symptoms. Mechanistically, chronic co-exposure to Cd and

Melatonin protects keratinocytes from diesel-derived PM2.5-induced damage by suppressing JNK/p38 MAPK signaling and caspase activation.

Melatonin is a naturally produced hormone in the human body that primarily regulates the sleep-wake cycle. This study examined the role of melatonin in protecting human keratinocytes from diesel-derived particulate matter 2.5 (DPM2.5)-induced oxidative injury. HaCaT keratinocytes and male HR-1 hairless mice were exposed to DPM2.5 to induce oxidative injury and treated with various concentrations of melatonin, followed by various analyses, including reactive oxygen species (ROS) detection, mitochondrial membrane potential analysis, western blotting, and cell cycle analysis. Melatonin exhibited

CD73 overexpression in ADSCs accelerates bladder repair by regulating the NFκB/NLRP3/caspase-1 signaling axis in neurogenic bladder rats.

Neurogenic bladder (NB) is a disabling condition lacking effective therapies. This study investigated whether CD73-expressing adipose-derived stem cells (ADSCs) promote bladder repair in a rat model of NB and explored the underlying mechanisms. ADSCs were sorted into CD73⁺ and CD73⁻ populations, and CD73⁺ cells were further modified to generate CD73⁺/ev ADSCs and CD73-overexpressing CD73⁺/⁺ADSCs, while CD73 inhibition was achieved using APCP. Conditioned media were applied to rat bladder smooth muscle cells in vitro, and ADSCs were injected into the bladder wall of rats subjected to bilateral

Cauloside A interacts with TLR4 to induce JNK/Caspase-3/GSDME-dependent pyroptosis in non-small cell lung cancer cells.

With non-small cell lung cancer (NSCLC) accounts for the majority of diagnoses, lung cancer continues to be the most common and deadly type of cancer worldwide. While treatment options have advanced, therapeutic efficacy remains constrained by drug resistance and side effects, highlighting the demand for novel therapeutic agents. Cauloside A, a natural compound isolated from Fructus Akebiae, has shown anti-tumor potential; however, its precise mechanism of action in NSCLC has remained unclear. This study demonstrates that Cauloside A inhibits tumor growth by interacting with TLR4 to initiate a

Antioxidant efficacy of N-acetylcysteine against acrylamide-induced toxicity in Leydig cells by modulating p53/Bax/Caspase-3 pathway.

Acrylamide (Acr) is a widely encountered environmental and dietary toxicant known to induce oxidative stress and disrupt male reproductive function. Leydig cells, due to their high metabolic activity and mitochondrial dependence, are particularly vulnerable to redox imbalance. N-acetylcysteine (Nac), a thiol-containing antioxidant and glutathione precursor, has been extensively studied for its cytoprotective properties. However, its modulatory effects on Acr-induced toxicity in Leydig cells and its pharmacodynamic interaction profile remain incompletely characterized. In this study, TM3 Leydig

NF-κB/Apaf1/caspase-9 axis suppresses autophagy to drive tubular inflammation and apoptosis in septic acute kidney injury.

Septic acute kidney injury (AKI) is a life-threatening complication of systemic infection, characterized by rapid loss of renal function and high mortality. Despite its clinical significance, the underlying molecular mechanisms remain incompletely elucidated. In this study, we identify the nuclear factor-κB (NF-κB)/apoptotic protease-activating factor 1 (Apaf1)/caspase-9 signaling axis as a central regulator of tubular apoptosis and inflammation through suppression of autophagy. Using proximal tubule-specific Apaf1 knockout mice, we demonstrate that Apaf1 deficiency significantly mitigates lip

Caspase-Independent Cell Death Induced by a Nitric Oxide Donor Derived from Valproic Acid in Human Pancreatic Cancer Cells.

Nitric oxide (NO) plays diverse roles in tumor biology, including modulation of blood flow and induction of cell death at high concentrations. In this study, we synthesized a novel NO donor derived from valproic acid (NVA) and investigated its cytotoxic mechanism in human pancreatic cancer cells. NVA released approximately 40% of its total nitrate/nitrite (NOx) immediately after dissolution in phosphate-buffered saline and then remained almost unchanged for 72 h, indicating a rapid initial NO release followed by stabilization. NVA significantly decreased the viability of BxPC-3 cells, whereas

Heat Shock Proteins 60 and 70, Ki67 and Caspase 3 Are Differentially Expressed in the Canine Pregnant and Non-Pregnant Uterus and Ovaries.

Heat shock proteins (HSPs) fulfil protective tasks in the whole organism; in pregnant dogs, they are expressed in the ovary, placenta and preimplantation embryo. Our objective was to compare the expression of HSP60 and -70, along with indicators of proliferation and apoptosis, in the non-pregnant and pregnant uterus/placenta and ovaries. Tissues were obtained after ovariohysterectomy and examined by means of immunohistochemistry. There were differences between pregnant and non-pregnant tissues: the expression level of HSP70 during preimplantation in superficial cells was significantly lower th

(2R,6R)-HNK improved LPS-induced depression-like behavior by inhibiting Vcam1/Caspase-1/IL-1β pathway.

Depression-like behavior is closely associated with neuroinflammation. Lipopolysaccharide (LPS) can activate N-methyl-d-aspartate receptors and induce cellular pyroptosis and neuroinflammation, thereby triggering depression-like behavior in animals. The ketamine metabolite (2R,6R)-hydroxynorketamine (HNK) exerts significant antidepressant-like effects without the adverse effects associated with ketamine, such as addiction and abuse liability. However, its underlying mechanisms remain largely unclear. In this study, C57BL/6 J male mice and PC12 cells were used to establish an in vivo depression

Caspase-11-mediated hyperinflammation impairs CD8+ T cell immunity and viral clearance in severe SARS-CoV-2 infection.

Severe SARS-CoV-2 infection is characterized by lung hyperinflammation, impaired IFN responses, and defective T cell activation, yet the molecular drivers of these immune dysregulations remain incompletely understood. Caspase-11 (CASP11), a key effector of the noncanonical inflammasome, has been shown to mediate an innate hyperinflammatory response and cytokine release in a mild SARS-CoV-2 infection model. However, the role played by CASP11 in severe SARS-CoV-2 disease and how it affects adaptive immunity has not been identified. Here, we found that CASP11 exacerbates severe SARS-CoV-2 pathoge

Structure-based design of a 1H-Tetrazole-containing scaffold for Caspase-1 inhibition: Synthesis, biological evaluation, and preliminary in Vivo studies.

Targeting caspase-1 with small-molecule inhibitors represents a promising strategy for the treatment of inflammatory diseases. Herein, we report the structure-based design, synthesis, and biological evaluation of a new 1H-tetrazole-containing scaffold for caspase-1 inhibitors. This scaffold incorporates a 1H-tetrazole as a carboxylic acid bioisostere and lacks electrophilic functionalities commonly employed to promote covalent modification of the catalytic cysteine. The resulting molecules inhibited caspase-1 with IC50 values ranging from the mid-nanomolar to low-micromolar range, with compoun

Suilysin of Streptococcus suis Serotype 2 Drives Astrocyte Pyroptosis via Activation of the ROS/NLRP3/Caspase-1/GSDMD Pathway.

Emerging evidence indicates that pyroptosis contributes to neuroinflammation and central nervous system (CNS) injuries. However, its role in Streptococcus suis (S. suis)-induced brain damage remains unclear. Here, we demonstrate that S. suis strain P1/7 infection triggers pyroptosis in astrocytes, specifically characterized by activating NLRP3/caspase-1/GSDMD axis. Suilysin (SLY), a key virulence factor of S. suis, dose- and time-dependently induced pyroptosis in U251 cells. Notably, genetic ablation of NLRP3 or GSDMD significantly attenuated SLY-induced pyroptosis, and caspase-1 inhibition si

A CRISPR-HITI strategy approach to improve CHO cell viability by modifying the 3'UTR of Caspase 8 Associated Protein 2.

Chinese Hamster Ovary (CHO) cells are essential in biopharmaceutical manufacturing. Scientists use CRISPR to enhance productivity. mRNAs contain UTRs that regulate gene expression, affecting protein abundance. Targeting these regions creates desirable knockout cells. The Caspase 8 Associated Protein 2 (CASP8AP2) gene is a promising target for improving host cell viability. This study used the CRISPR-Homology-Independent Targeted Integration (HITI) strategy to modify the 3'UTR region of the CASP8AP2 gene in CHO cells. The aim was to evaluate the effects of CASP8AP2 silencing on cell proliferati

Photobiomodulation regulates inflammation and autophagy in spinal cord injury through NLRP3/Caspase-1/IL-1β pathway by targeting TLR2.

After spinal cord injury (SCI), peripherally derived macrophages infiltrated the injury area to exert inflammatory effects, causing barriers to the repair of spinal cord injury. Our previous study confirmed that photobiomodulation (PBM) could promote the motor function recovery and inhibit the secretion of inflammatory cytokines after SCI, moreover, PBM also has a role in promoting autophagy, but the mechanism is not clear. Therefore, we aimed to investigate whether PBM promotes autophagy by regulating the inflammatory response of macrophages, which in turn regulates functional repair after SC
