CD70/CD27 signaling promotes the pathogenesis of multiple myeloma and represents a promising therapeutic target.

The increasing therapeutic options for multiple myeloma (MM) have significantly improved long-term survival for many patients. However, disease progression remains inevitable due to the emergence of drug-resistant myeloma cell populations, often culminating in extramedullary disease manifestations. In this study, we identified CD70-expressing myeloma cells as critical drivers of disease progression and propagation. CD70 expression in MM cells is an independent negative prognostic factor for overall survival. Mechanistically, CD70/CD27 signaling activates the MAPK/ERK and Wnt signaling pathways

Emerging molecular mechanism of neuroinflammation in Parkinson's disease: A JAK/STAT and TGFβ/SMAD signaling crosstalk.

Neuroinflammation has emerged as a convergent pathological signature of Parkinson's disease (PD), shaping both its onset and progression. Pe … restraint. This review synthesizes current understanding of the mechanistic interplay between these critical pathways, proposing that the imbalance between JAK/STAT activation and TGF-β/SMAD suppression represents a key candidate regulatory interface governing the neuroinflammatory trajectory of PD. Unraveling this crosstalk provides new insight into how signaling …

Glycyrrhizic Acid Attenuates Aβ42-Induced Neurodegeneration Through Coordinated Regulation of Oxidative Stress, Synaptic Markers, and Key Alzheimer's Signaling Pathways.

Alzheimer's disease (AD) is a catastrophic neurodegenerative disorder marked by progressive decline of cognitive function, memory loss, and neuronal death. Its pathology is characterized by the formation of extracellular amyloid-beta (Aβ) plaques and intracellular neurofibrillary tangles from tau hyperphosphorylation. Despite extensive research, current treatments are limited to symptomatic relief and are associated with significant side effects. This accentuates the critical need for alternative therapeutic strategies with potent neuroprotective effects and minimal toxicity. This study invest

Impact of Secondhand Smoke and E-Cigarette Exposure on Placental Apoptotic and Growth-Regulatory Proteins in Mouse Pregnancy.

Apoptosis is critical in placental development, and its dysregulation is linked to pregnancy complications such as intrauterine growth restriction (IUGR) and preeclampsia (PE). Environmental exposures, particularly secondhand smoke (SHS) and e-cigarettes (eCigs), may contribute to placental dysfunction through apoptotic pathways. This study examined the effects of SHS and eCig exposure on placental apoptosis and growth-regulatory proteins in a murine model. C57BL/6 pregnant mice were exposed to SHS or eCigs at two critical gestational time points: early trophoblast invasion (E12.5 to E18.5) an

Exploration of key markers driving ferroptosis in the progression of non-alcoholic fatty liver disease.

Non-alcoholic fatty liver disease (NAFLD) is a prevalent condition strongly linked to obesity, diabetes, and metabolic syndrome. Its global incidence is steadily increasing, placing a significant health burden on both patients and society. This study aims to identify ferroptosis-related differentially expressed genes (DEGs) through bioinformatics analysis and to explore their roles in NAFLD. By comparing samples from NAFLD patients and healthy controls, we identified 1,770 significant DEGs, with 1,073 being upregulated and 697 downregulated. Pathway analysis revealed a marked decrease in expre

Visual Neuroplasticity: Modulating Cortical Excitability with Flickering Light Stimulation.

The balance between cortical excitation and inhibition (E/I balance) in the cerebral cortex is critical for cognitive processing and neuroplasticity. Modulation of this balance has been linked to a wide range of neuropsychiatric and neurodegenerative disorders. The human visual system has well-differentiated magnocellular (M) and parvocellular (P) pathways, which provide a useful model to study cortical excitability using non-invasive visual flicker stimulation. We present an Arduino-driven non-image forming system to deliver controlled flickering light stimuli at different frequencies and wav

HOXA10 and HOXA11 in Human Endometrial Benign Disorders: Unraveling Molecular Pathways and Their Impact on Reproduction.

HOX genes, a family of conserved transcription factors, are critical for reproductive tract development and endometrial functionality. This review highlights the molecular underpinnings of HOXA10/HOXA11 in reproductive health and their dysregulation in benign pathologies associated with infertility, such as endometriosis, adenomyosis, and endometrial polyps. These genes are dynamically regulated by estrogen and progesterone, with peak expression during the secretory phase of the menstrual cycle when implantation takes place. The molecular mechanisms underlying their action include the modulati

ZNF207-driven PRDX1 lactylation and NRF2 activation in regorafenib resistance and ferroptosis evasion.

Regorafenib (RGF) is a critical second-line therapy for advanced hepatocellular carcinoma (HCC) following disease progression on sorafenib; however, the rapid onset of RGF resistance poses a significant barrier to enhancing patient outcomes. In this study, CRISPR/Cas9 screening in RGF-treated HCC cells identified Zinc Finger Protein 207 (ZNF207) as a primary driver of resistance. Further analysis revealed that ZNF207 promotes resistance by inducing antioxidant responses that inhibit ferroptosis, a form of iron-dependent cell death. Mechanistically, ZNF207 facilitates the lactylation of peroxir

Critical role of complement in antibody mediated rejection in kidney transplantation.

Antibody-mediated rejection (AMR) represents a major challenge in kidney transplantation, significantly contributing to tissue injury and graft failure. AMR is primarily driven by donor-specific alloantibodies (DSAs), which recognize and bind to specific target antigens present within the transplanted kidney tissue. Upon binding, these DSAs commonly initiate activation of the complement system within the graft. The activation of the complement cascade sets off a powerful inflammatory response characterized by the recruitment and activation of immune cells, endothelial damage, and subsequent ti

NRF2 activation resolves neuropathic pain via dual antioxidant and NaV1.7-Suppressive mechanisms.

Neuropathic pain remains a major clinical challenge, with current analgesics often proving ineffective and frequently refractory. Nociceptive responses are protective, while dysregulation of pain-resolving mechanisms drives the pain to be pathological. Enhancing resolving and protective signaling pathways is critical for treating neuropathic pain, yet such knowledge remains limited. In this study, we revealed the antinociceptive effects of Nrf2 in sensory neurons under both physiological and pathological conditions. Notably, insufficient activation of endogenous Nrf2 underlies the transition f

Primary cilia in growth plates orchestrate long bone development.

The growth plate plays a crucial role in long bone development and elongation, housing stem cells that contribute to bone formation. This study investigates the role of primary cilia, specialized organelles that regulate stem cell fate, in the development and repair of long bones. Here, we report the presence of primary cilia in all zones of the growth plate, particularly during embryonic development. Spatial transcriptomics and analysis of conditional knockout (CKO) mice identified that primary cilia mediate critical developmental signaling pathways within the growth plate. Disruption of prim

Methyl Donors, Epigenetic Alterations, and Brain Health: Understanding the Connection.

Methyl donors such as choline, betaine, folic acid, methionine, and vitamins B6 and B12 are critical players in the one-carbon metabolism and have neuroprotective functions. The one-carbon metabolism comprises a series of interconnected chemical pathways that are important for normal cellular functions. Among these pathways are those of the methionine and folate cycles, which contribute to the formation of S-adenosylmethionine (SAM). SAM is the universal methyl donor of methylation reactions such as histone and DNA methylation, two epigenetic mechanisms that regulate gene expression and play r

Role of Transcriptional and Epigenetic Regulation in Lymphatic Endothelial Cell Development.

The lymphatic system is critical for maintaining the homeostasis of lipids and interstitial fluid and regulating the immune cell development and functions. Developmental anomaly-induced lymphatic dysfunction is associated with various pathological conditions, including lymphedema, inflammation, and cancer. Most lymphatic endothelial cells (LECs) are derived from a subset of endothelial cells in the cardinal vein. However, recent studies have reported that the developmental origin of LECs is heterogeneous. Multiple regulatory mechanisms, including those mediated by signaling pathways, transcrip

Host Immune Response to Clinical Hypervirulent Klebsiella pneumoniae Pulmonary Infections via Transcriptome Analysis.

Klebsiella pneumoniae (K. pneumoniae), especially those with hypervirulence, is becoming a global concern and posing great threat to human health. Studies on individual immune cells or cytokines have partially revealed the function of the host immune defense against K. pneumoniae pulmonary infection. However, systematic immune response against K. pneumoniae has not been fully elucidated. Herein, we report a transcriptome analysis of the lungs from a mouse pneumonia model infected with a newly isolated K. pneumoniae clinical strain YBQ. Total RNA was isolated from the lungs of mice 48 hours pos
