Nucleoside diphosphate kinase from the parasitic nematode Brugia malayi.

Using a reverse transcription-polymerase chain reaction (RT-PCR) procedure that exploited the presence of a conserved 22-nucleotide spliced leader (SL) sequence that is trans-spliced to the 5' end of nematode transcripts, a novel Brugia malayi (Bm) infective-stage SL cDNA expression library was constructed and characterized. The library was immunoscreened with rabbit anti-infective-stage antibodies (Ab) and an immunodominant clone, BmG4-7, was identified and characterized. BmG4-7 contained a full-length cDNA that had significant sequence similarity to nucleoside diphosphate kinase (NDK)-encodi

From parasite to pathology: A novel manifestation of cestodiasis in Octopus maya.

OBJECTIVE: This study aimed to characterize the effects of Prochristianella sp. infection (Cestoda: Trypanorhyncha) in the Octopus maya, with focus on tissue pathology, immune response, and potential implications for host health. METHODS: Molecular analyses of 28S ribosomal DNA were performed to confirm the identity of collected parasites from the buccal mass of octopus hosts. Macro- and microscopic observations of O. maya tissues, hemocyte counts, and histological analyses were conducted to assess the damage caused by the infection. Comparisons were made between naturally infected octopuses

Identification and analysis of the gap region in the 23S ribosomal RNA from Actinobacillus actinomycetemcomitans.

Actinobacillus actinomycetemcomitans is a Gram-negative coccobacillus which can cause certain severe extra-oral infections as well as forms of human periodontal disease such as localized juvenile periodontitis. In contrast to many prokaryotic and eukaryotic species which exhibit an intact 23S ribosomal RNA (rRNA) molecule, examination of six A. actinomycetemcomitans strains--including three serogroup representative strains and two strains from non-human primates--revealed that this micro-organism does not produce an intact 23S ribosomal RNA (rRNA) molecule but, rather, two smaller forms of 1.8

Case Report: Diagnosis and Assessment of Cure Approaches for Acute Schistosomiasis in Pre-School Children.

Acute schistosomiasis (AS) manifests with a broad spectrum of clinical features in pediatric populations. Diagnosis may be difficult in the absence of detectable numbers of eggs. As a result, new approaches may be required to achieve an accurate diagnosis. Optimal praziquantel (PZQ) treatment regimen for young children is debatable. Also, the post-treatment response is still poorly evaluated due to the lack of reliable markers. A group of 6 children (a toddler and 5 pre-school children) and one pre-adolescent were investigated for AS clinical manifestations and followed-up for two years after

Integrative morphological and molecular characterization of Proterodiplostominae metacercariae (Digenea: Diplostomidae) infecting siluriform fishes in the Peruvian Amazon Basin.

Proterodiplostominae Dubois, 1936 is a small but evolutionarily significant lineage of diplostomid trematodes that primarily parasitize crocodilians, with fishes acting as second intermediate or paratenic hosts. In Peru, knowledge of this group has been limited to the record of adult specimens of Proterodiplostomum longum (Brandes, 1888) recovered from Caiman crocodilus Linnaeus, 1758 (Sauropsida: Alligatoridae) in the Loreto region, and molecular evidence linking larval and adult stages has been absent. The present study provides the first integrative morphological and molecular characterizat

High-throughput sequencing of Fasciola spp. shows co-infection and intermediate forms in Balochistan, but only Fasciola gigantica in the Punjab province of Pakistan.

Fasciola gigantica and Fasciola hepatica are digenetic trematodes causing fasciolosis in ruminants. The host and geographical distribution of both Fasciola species are influenced by environmental and climatic conditions favouring survival and development of free-living stages and intermediate hosts, and livestock management practices. The aim of the present study was to describe the host distribution of the two Fasciola species in buffalo, cattle, goats, and sheep in the Balochistan and Punjab provinces of Pakistan. 359 flukes were collected from a total of 32 livers from the four livestock sp

Epidemiology and molecular identification of Anisakis pegreffii (Nematoda: Anisakidae) in the horse mackerel Trachurus trachurus from northern Morocco.

Anisakis infection parameters were studied in horse mackerel (Trachurus trachurus) taken from two areas of northern Morocco (Tetouan and Tangier), which showed a mean prevalence of 54.9%. Identification of the ITS1-5.8S-ITS2 fragment by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) showed A. pegreffii to be the dominant species; no A. simplex s.s. were detected. The presence of A. pegreffii in horse mackerel was not influenced by the sex (P = 0.46) or catch area (Atlantic versus Mediterranean, P = 0.52) of the fish, but was significantly related to their length,

Genetic differentiation of cercariae infrapopulations of the avian schistosome Trichobilharzia szidati based on RAPD markers and mitochondrial cox1 gene.

Avian schistosome Trichobilharzia szidati is a member of the largest genus within the family Schistosomatidae (Trematoda). Population genetic structure of Trichobilharzia spp. schistosomes, causative agents of cercarial dermatitis in humans, has not been studied yet. The knowledge of the genetic structure of trichobilharzian populations is essential for understanding the host-parasite coevolutionary dynamics and epidemiology strategies. Here we examined genetic diversity in three geographically isolated local populations of T. szidati cercariae inhabiting Russia based on nuclear (randomly ampl

Hyper-variable regions in 18S rDNA of Strongyloides spp. as markers for species-specific diagnosis.

Four hyper-variable regions (HVR-I to -IV) found in 18S ribosomal DNA sequences were compared among 34 isolates of 15 species of the genus Strongyloides to evaluate their diagnostic value. HVR-I to -III were short, and plural species exhibit the same nucleotide arrangement. Meanwhile, HVR-IV had 23 to 39 nucleotides, showing species-specific arrangements, except Strongyloides ransomi and Strongyloides venezuelensis, which had the same nucleotide sequence in HVR-IV but were readily distinguished by the difference in HVR-I and -III. Isolates of Strongyloides stercoralis from humans of USA, Japan

Detection of Paragonimus heterotremus eggs in experimentally infected cats by a polymerase chain reaction-based method.

A polymerase chain reaction (PCR) procedure for the detection of Paragonimus heterotremus eggs in stool samples was developed and compared with Stoll's egg count method. The primers were designed on the basis of a previously constructed pPH-13-specific DNA probe, which produced an approximate 0.5-kb amplified product. This PCR method could detect as few as 5 eggs in 0.6 g of artificially inoculated feces of a healthy control cat or as little as 1 x 10(-4) ng of P. heterotremus genomic DNA. The assay had 100% sensitivity in all infected cats. The method did not yield an approximate 0.5-kb produ

Neutrophil extracellular traps induced by Haemonchus contortus excretory-secretory proteins varies among goats, gerbils, and mice.

BACKGROUND: Previous studies indicated that infection with Haemonchus contortus is host-specific (goat: susceptible host; gerbil: paratenic host; mouse: resistant host). Neutrophils play an essential role in host defense against parasitic infection through phagocytic engulfment, reactive oxygen species (ROS) generation, and neutrophil extracellular traps (NETs) formation. NETs are large web-like complexes consisting of a DNA scaffold decorated with various proteins components, including histones, myeloperoxidase, and elastase. They are released through both ROS-dependent and ROS-independent pa

Development and field evaluation of a multiplex qPCR assay for environmental DNA detection of Schistosoma mekongi and its intermediate snail host Neotricula aperta in the Mekong River Basin.

BACKGROUND: Schistosoma mekongi remains an important cause of intestinal schistosomiasis in the lower Mekong River Basin. Sensitive environmental surveillance tools are needed to support transmission monitoring, particularly where conventional snail surveys are seasonally constrained. This study developed and evaluated a multiplex quantitative PCR (qPCR) assay for simultaneous detection of S. mekongi and its intermediate snail host, Neotricula aperta, in aquatic environmental DNA (eDNA) samples. METHODS: Singleplex and multiplex qPCR assays targeting S. mekongi and N. aperta were compared to

Comprehensive study based on mtDNA signature (nad1) providing insights on Echinococcus granulosus s.s. genotypes from Pakistan and potential role of buffalo-dog cycle.

Pakistan has long been considered neglected endemic region for Echinococcus granulosus. Limited surveillance studies have failed to epidemiologically draw complete picture on geographical presence and etiological agents of cystic echinococcosis. Amidst such lacunae, current study explored main transmission routes of this disease through molecular characterization of hydatid cyst isolates obtained from sheep (n = 35), goats (n = 26), cattle (n = 30) and buffalo (n = 30) from the four provinces of Pakistan. Two strains of E. granulosus sensu stricto, G1/G3, and their haplotypes were observed to

Redescription of Cichlidogyrus philander (Monogenea, Ancyrocephalidae) using scanning electron microscopy (SEM) and molecular analysis.

The sclerotized structures of monogeneans have traditionally been studied by light microscopy and different staining techniques. Recently, enzymatic digestion followed by scanning electron microscopy (SEM) has enabled the examination of structural details not visible with light microscopy. In order to obtain better, and more accurate, morphological information on sclerotized structures not affected by mounting medium or cover slip pressure, the sclerites of Cichlidogyrus philander Douëllou, 1993 (Monogenea, Ancyrocephalidae), collected from Pseudocrenilabrus philander (Weber, 1897), were redes

Molecular Characterization of Nippostrongylus brasiliensis (Nematoda: Heligmosomatidae) from Mus musculus in India.

Mus musculus (Rodentia: Muridae) has generally been infected with a rodent hookworm Nippostrongylus brasiliensis. In this report, we present morphological and molecular identification of N. brasiliensis by light and scanning electron microscopy and PCR amplification of mitochondrial cytochrome c oxidase subunit 1 (cox1) gene and the protein sequences encoded by cox1 gene, respectively. Despite the use of N. brasiliensis in many biochemistry studies from India, their taxonomic identification was not fully understood, especially at the species level, and no molecular data is available in GenBank

M aritrema corai n. sp. (Digenea: Microphallidae) from the white ibis Eudocimus albus (Linnaeus) (Aves: Threskiornithidae) in Mexico.

M aritrema corai n. sp. is described based on material from the intestine of the white ibis Eudocimus albus (L.) (Threskiornithidae) in Mexico. The new species can be distinguished morphologically from all congeners by the unique combination of the following morphological features: a very long cirrus sac attenuated distally [cirrus sac to body length ratio 1:0.90-1.29 (mean 1:1.07)]; a large, elongate-oval seminal receptacle, located dorsally between the cirrus sac and ovary; and long, filiform, unarmed, evaginable cirrus.
