Cardiovascular and endocrine effects of naloxone compared in normotensive and hypertensive patients.

Naloxone, a competitive antagonist of opioid receptors, and placebo (dextrose 5% in water (D5W) were administered on separate days to healthy normotensive (NT) male volunteers and to male patients with essential hypertension (HT). A single-blind, placebo-controlled, cross-over design was employed. Increasing doses of naloxone (0.4, 1.2, 3.6, 10.8, 32.4, 97.2 mg) were given every 30 min as slow i.v. boluses. On a separate day, i.v. boluses of D5W were given according to a similar protocol. Naloxone failed to significantly modify systolic and diastolic blood pressure (BP), heart rate (HR), respi

Neuropeptides and their role in nociception and analgesia.

The authors highlight the potential importance of neuropeptides as clinical tools in the study and treatment of pain. Although many questions remain unanswered, it is encouraging that several clinical and experimental advances have shed new light on the neuropharmacology of nociception and have prompted new hope for more effective treatments of such diverse problems as chronic intractable pain, migraine, and drug dependency. Within the next few years, it is anticipated that further work in this area will lead to better basic and clinical understanding of these important clinical problems.

Intracisternal bombesin induced inhibition of gastric secretion is not mediated through prostaglandin or opioid pathways.

Neuromedin B, a mammalian decapeptide which shares homology with the carboxyl terminal fragment of ranatensin R and bombesin, injected intracisternally (880 pmol) did not modify gastric acid secretion stimulated either by pylorus ligation in conscious rats or by pentagastrin infusion in urethane-anesthetized rats. Intracisternal bombesin (300 pmol) further enhanced PGE2-induced inhibition of gastric acid secretion and suppressed prostaglandin-induced stimulation of gastric secretory volume. Gastric response to intracisternal bombesin was not modified by indomethacin (10 mg/kg) or naloxone pret

Naloxone and TRH in the treatment of shock and trauma: what future roles?

Endogenous opioid peptides are released in response to stressful situations, such as circulatory shock, both as hormones and as central and peripheral neurotransmitters. Naloxone, an opiate antagonist, improves cardiovascular function in a variety of animal models of shock caused by endotoxemia, hemorrhage, anaphylaxis, or spinal trauma. Administration of thyrotropin-releasing hormone (TRH) in supraphysiologic doses also has pressor effects in these shock models. Given acutely after injury, TRH improves recovery in models of spinal trauma; however, the experimental effects of TRH do not involv

Effect of naloxone on gonadotropin secretion before and after testosterone in Klinefelter's syndrome.

A study was performed on eight subjects with Klinefelter's syndrome to assess the relation between gonadal hormones and opioid inhibition of gonadotropin secretion through comparison of their gonadotropin response to naloxone (NAL) (0.3 mg/kg; 1/3 bolus iv. at time 0 and 2/3 iv. for 120 min) before and after testosterone propionate (TP) 100 mg/day im. for 5 days. Under basal conditions, NAL failed to induce a significant change in LH levels. After TP, however, despite unchanged basal LH levels (mean +/- S.E.M.: 27.0 +/- 3.4 vs 21.2 +/- 3.21 microU/ml), LH significantly increased in response to

Characteristics of analgesia induced by noncatecholic phenylethylamine derivatives: possible involvement of endogenous opioid peptides and serotonin in phenylethylamine analog-induced analgesia.

Characteristics of the analgesic action of phenylethylamine derivatives, amphetamine, phenylethylamine (PEA), hydroxyphenylethylamine (OHPEA) and hydroxyphenylalanine (OHF), were examined. Pain threshold of mice was measured by using the hot plate method. OHPEA (50 mg/kg), amphetamine (0.5-8 mg/kg) or PEA (50 mg/kg) produced an analgesic effect in the absence of MAO inhibitor, and the analgesia was reversed by naloxone (5 mg/kg) or reserpine (2 mg/kg x 2). Ten mg/kg of PEA, 250 mg/kg of OHF and 10 mg/kg of OHPEA could not produce detectable analgesia, but they revealed analgesic activity when

Physiological effects of high dose naloxone administration to normal adults.

The intravenous bolus administration of high doses of the opioid receptor antagonist naloxone (in the mg./kg. range) to normal adults produced dose-dependent increases in systolic blood pressure and respiratory rate. No significant alterations in other physiological processes were found although there were trends for increases in oral temperature and pulse rate with increasing dose. These results suggest the involvement of an endogenous opioid system in the tonic regulation of human systolic blood pressure and respiration and the probable inadequacy of doses of naloxone previously utilized in

Increased pressor responsiveness to enkephalin in spontaneously hypertensive rats: the role of vasopressin.

1. The cardiovascular effects of an enkephalin analogue were examined in spontaneously hypertensive and normotensive Wistar-Kyoto rats. (D-Ala2)-methionine enkephalin caused a biphasic increase in blood pressure and an increase in heart rate after intracerebroventricular injection. 2. The initial pressor response to (D-Ala2)-methionine enkephalin was greater in hypertensive than in normotensive rats. No difference was noted between groups during the secondary pressor response. Heart rate increases paralleled the secondary increase in blood pressure. 3. Naloxone pretreatment abolished the secon

Electroencephalographic and behavioral effects of D-ala2-methionine-enkephalinamide and morphine in the rat.

Rats were prepared with chronic cortical and temporalis muscle electrodes and bilateral intraventricular (i.vt.) cannulae. The direct and voltage integrated electroencephalogram (EEG) and integrated electromyogram as well as gross behavior were monitored after the i.vt. injection of D-enkephalin (10--240 microgram), morphine (2.5--40 microgram), naloxone (25--50 microgram) or sterile water (10 microliter). EEG high-amplitude slow-frequency waves (EEG slow bursts) occurred after i.vt. morphine or D-enkephalin and were associated with behavioral stupor. The behavioral depressant effect was follo

Functional characterization of opioid receptor ligands by aequorin luminescence-based calcium assay.

A functional assay, based on aequorin-derived luminescence triggered by receptor-mediated changes in intracellular calcium levels, was used to examine relative potency and efficacy of the mu-opioid agonists endomorphin-1, endomorphin-2, morphiceptin, and their position 3-substituted analogs, as well as the delta-agonist deltorphin-II. The results of the aequorin assay, performed on recombinant cell lines, were compared with those obtained in the functional assay on isolated tissue preparations (guinea pig ileum and mouse vas deferens). A range of nine opioid peptide ligands produced a similar

Influence of the endogenous opioid system on high alcohol consumption and genetic predisposition to alcoholism.

There is increasing evidence supporting a link between the endogenous opioid system and excessive alcohol consumption. Acute or light alcohol consumption stimulates the release of opioid peptides in brain regions that are associated with reward and reinforcement and that mediate, at least in part, the reinforcing effects of ethanol. However, chronic heavy alcohol consumption induces a central opioid deficiency, which may be perceived as opioid withdrawal and may promote alcohol consumption through the mechanisms of negative reinforcement. The role of genetic factors in alcohol dependency is we

Effects of a number of short peptides isolated from the brain of the hibernating ground squirrel on the EEG and behavior in rats.

Intracerebroventricular administration of the peptides kyotorphin (Tyr-Arg), neokyotorphin (Thr-Ser-Lys-Tyr-Arg), and Asp-Tyr at doses of 4 and 8 micrograms altered the behavior of rats in a manner similar to that seen after similar administration of brain fractions from hibernating ground squirrels (Citellus undulatus), which contained these peptides; there were increases in orientational reactions, increases in the frequency of stereotypical scratching movements, grooming, yawning, hiccuping, and sneezing. Animals became drowsy after 15-20 min. Peptides and brain fractions also had similar e

Failure to reproduce the in vitro cardiac electrophysiological effects of naloxone in humans.

1. Opioid receptor antagonists such as naloxone have shown antiarrhythmic activity in animal models of coronary artery occlusion. Studies have indicated that these effects are stereospecific but both isomers of naloxone prolong action potential duration and refractoriness in guinea-pig and rabbit isolated ventricular myocardium (Class III effect). 2. This study was performed to identify whether this Class III effect of naloxone could be reproduced in human myocardium in vivo. Twenty patients with coronary artery disease received intravenous racemic naloxone (1-40 micrograms kg-1 min-1). Surfac

Opioid peptides in the rabbit carotid body: identification and evidence for co-utilization and interactions with dopamine.

The rabbit carotid body is a catecholaminergic organ that contains dopamine and norepinephrine in a proportion of nearly 5:1. Chronic (15 days) carotid sinus nerve denervation or superior cervical ganglionectomy did not modify the carotid body dopamine content (5-6 nmol/mg of protein, equivalent to 250 pmol per carotid body), but sympathectomy reduced by approximately 50% the norepinephrine content. The carotid body has also a very high content of opioid activity (250 equivalent pmol of Leu-enkephalin/mg of protein) as measured by a radioreceptor assay that detects preferentially delta-opioid

Morphine and morphiceptin increase the threshold for epinephrine-induced cardiac arrhythmias in the rat through brain mu opioid receptors.

1. To determine whether morphine modulates the development of cardiac arrhythmias through mu opioid receptors by an action within the central nervous system (CNS). Catecholamine-induced ventricular arrhythmias were produced, in the rat, by continuous infusion of epinephrine at incremental doses until the development of fatal arrhythmias, usually ventricular fibrillation. 2. Morphine, 0.1 mg/kg i.v., significantly suppressed (P < 0.05) the development of epinephrine-induced arrhythmias compared with the control group. This was opposed by the mu opioid antagonist naloxone (1 or 2 mg/kg) in a dos

Central and peripheral control of postprandial pyloric motility by endogenous opiates and cholecystokinin in dogs.

The role of endogenous opiates and cholecystokinin (CCK) in the control of postprandial pyloric myoelectric activity was investigated in conscious dogs with chronically implanted intraparietal electrodes at the gastroduodenal junction. Meals consisted of either 20 g/kg of canned food (standard meal) or the same food supplemented with 0.5 mL/kg of arachis oil (fat meal). During the 6 hours after standard and fat meals, the number of pyloric spike bursts, 2-4 seconds in duration, was 61.8 +/- 15.8 and 49.9 +/- 12.7/15 minutes, respectively. Administered 15 minutes before a fat meal, naloxone (50
