Altered Mitochondrial Homeostasis during Systemic Lupus Erythematosus Impairs Neutrophil Extracellular Trap Formation Rendering Neutrophils Ineffective at Combating Staphylococcus aureus.

Inflammation involves a delicate balance between pathogen clearance and limiting host tissue damage, and perturbations in this equilibrium promote disease. Patients suffering from autoimmune diseases, such as systemic lupus erythematosus (SLE), have higher levels of serum S100A9 protein and increased risk for infection. S100A9 is highly abundant within neutrophils and modulates antimicrobial activity in response to bacterial pathogens. We reasoned that increased serum S100A9 in SLE patients reflects accumulation of S100A9 protein in neutrophils and may indicate altered neutrophil function. In

Hydrogen sulfide of air induces macrophage extracellular traps to aggravate inflammatory injury via the regulation of miR-15b-5p on MAPK and insulin signals in trachea of chickens.

Hydrogen sulfide (H2S) is an environmental contaminant to cause the airway damage. The release of macrophage extracellular traps (METs) is the mechanism of immune protection to harmful stimulation via microRNAs, but excessive METs cause the injury. However, few studies have attempted to interpret the mechanism of an organism injury due to H2S via METs in chickens. Here, we investigated the transcriptome profiles, pathological morphologic changes and METs release from chicken trachea after H2S exposure. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis revealed that…

Protective effects of curcumin on short and long-term particulate matter (PM10/2.5) induced lung damage severity via modulation of neutrophil extracellular traps (NETs) formation in mouse model.

Particulate matter (PM10/2.5) exposure and lung damage have recently been found to be associated with Neutrophil extracellular trap (NETs) formation and NETosis. NETs are the structures released from neutrophils in response to pathogens and during inflammatory response. This study was undertaken to investigate lung damage after short and long-term exposures to PM10/2.5 collected from Varanasi, one of the oldest Indian cities and its association with NETs formation in neutrophils. BALB/c mice were exposed to PM10/2.5(0.5 mg/kg, i.n.) for 2-4 weeks where wet/dry lung tissue weight ratio were sig

Neutrophil Extracellular Traps are Present in Immune-complex-mediated Cutaneous Small Vessel Vasculitis and Correlate with the Production of Reactive Oxygen Species and the Severity of Vessel Damage.

Formation of neutrophil extracellular traps has been implicated in autoimmunity. However, the presence and clinical relevance of neutrophil extracellular traps in immune-complex-mediated cutaneous small and medium vessel vasculitides has not been investigated. This study retrospectively analysed 72 patients with histology-proven hypersensitivity vasculitis (n = 21), IgA vasculitis (n = 22), urticarial vasculitis (n = 22), erythema elevatum diutinum (n = 3) and polyarteritis nodosa (n = 4). Neutrophil extracellular traps were detected in hypersensitivity vasculitis, IgA vasculitis, urticarial v…

miR-301b-3p/ITPKB drives clear cell renal cell carcinoma progression by promoting PARP1/IL8-mediated neutrophil extracellular trap formation.

Clear cell renal cell carcinoma (ccRCC) progression is closely associated with pro-tumorigenic neutrophil extracellular traps (NETs) within the tumor microenvironment; however, the upstream molecular mechanisms remain poorly defined. Through integrated multi-omics analysis of ccRCC cohorts, we identified ITPKB as a key downregulated gene associated with NET formation. Subsequent functional validation, including dual-luciferase reporter assays, confirmed that ITPKB is a direct target of the miR-301b-3p. Mechanistically, the loss of ITPKB prevents the ubiquitination-mediated degradation of PARP1…

Advanced glycation end product (AGE)-modified albumin does not enhance neutrophil extracellular trap formation and minimally impacts endothelial inflammation in large vessels.

AIMS: Advanced glycation end products (AGEs), particularly AGE-modified albumin (AGE-BSA), have been implicated in vascular inflammation and endothelial dysfunction, while neutrophil extracellular traps (NETs) are known to aggravate endothelial injury. Here, we investigated whether AGE-BSA directly promotes endothelial inflammatory and thrombotic activation, modulates neutrophil oxidative stress and NET formation, and may enhance NET-induced endothelial inflammation. MATERIALS, METHODS, AND KEY FINDINGS: In human aortic and human umbilical vein endothelial cells, two different endotoxin-free …

Buyang Huanwu Decoction attenuates vascular aging by suppressing the pathway of neutrophil extracellular trap formation via modulation of the HMGB1/TLR4/p38 signaling pathway.

ETHNOPHARMACOLOGICAL RELEVANCE: Vascular aging is a significant driver of age-related cardiovascular diseases, in which the immune-inflammatory response driven by excessive formation of neutrophil extracellular traps (NETs) is a core process accelerating this progression. Buyang Huanwu Decoction (BHD) is a classic traditional Chinese medicine (TCM) formula widely used for treating cardio-cerebrovascular diseases, but whether it acts through modulating NET-driven vascular aging is unknown. AIM OF THE STUDY: This study aims to investigate the mechanism by which BHD delays vascular aging, focusi…

Recombinant human DNase treatment mitigates extracellular trap mediated damage and improves long-term recovery after spinal cord injury in male mice.

After traumatic spinal cord injury (SCI), inflammation and other reactive processes exacerbate tissue damage and impair long-term motor recovery. Extracellular traps (ETs) are an immune cell effector function first described in neutrophils wherein chromatin is decondensed, decorated with cytotoxic granule enzymes, and expelled from the cell body. Recently, ETs have been linked to poor functional outcomes in SCI; however, translatable agents to prevent ET-mediated damage after SCI have yet to be explored. We assessed recombinant human (rh) DNase (trade name Pulmozyme) as a potential therapeutic…

Inhibition of Peptidyl Arginine Deiminase 4-Dependent Neutrophil Extracellular Trap Formation Reduces Angiotensin II-Induced Abdominal Aortic Aneurysm Rupture in Mice.

Objective: Neutrophil infiltration plays an important role in the initiation and development of abdominal aortic aneurysm (AAA). Recent studies suggested that neutrophils could release neutrophil extracellular traps (NETs), leading to tissue injury in cardiovascular diseases. However, the role of NETs in AAA is elusive. This study aimed to investigate the role and underlying mechanism of NETs in AAA development. Methods and Results: An angiotensin II (Ang II) infusion-induced AAA model was established to investigate the role of NETs during AAA development. Immunofluorescence staining showed th…

Anti-Carbamylated Protein Antibodies Stabilize Carbamylated Histone H3 to Promote Synovial Activation and Neutrophil Extracellular Trap-Mediated Bone Loss in Rheumatoid Arthritis.

OBJECTIVE: Carbamylation, a nonenzymatic post-translational modification, contributes to rheumatoid arthritis (RA) pathogenesis. Anti-carbamylated protein antibodies (anti-CarP) occur in around 50% of patients with RA and associate with greater joint damage. Neutrophil extracellular traps (NETs) are a major source of carbamylated autoantigens. We sought to study how carbamylation and anti-CarP antibodies promote joint pathology. METHODS: NETs were generated with calcium ionophore and analyzed for carbamylation by immunofluorescence, flow cytometry, and mass spectrometry. Enzymatic pathways we…

Magnesium hydride attenuates intestinal barrier injury during hemorrhage shock by regulating neutrophil extracellular trap formation via the ROS/MAPK/PAD4 pathway.

Magnesium hydride (MgH2) is a hydrogen storage material that is known for its high capacity and safety and is capable of releasing hydrogen in a controlled manner when administered orally. This release of hydrogen has been associated with a range of biological effects, including anti-inflammatory properties, antioxidant activity, and protection of the intestinal barrier. Previous research has shown that neutrophil extracellular traps (NETs) play a role in the dysfunction of the intestinal barrier in conditions such as sepsis and critical illnesses. However, it remains unclear as to whether MgH…
