Predicting the progression of MCI and Alzheimer's disease on structural brain integrity and other features with machine learning.

Machine learning (ML) on structural MRI data shows high potential for classifying Alzheimer's disease (AD) progression, but the specific contribution of brain regions, demographics, and proteinopathy remains unclear. Using Alzheimer's Disease Neuroimaging Initiative (ADNI) data, we applied an extreme gradient-boosting algorithm and SHAP (SHapley Additive exPlanations) values to classify cognitively normal (CN) older adults, those with mild cognitive impairment (MCI) and AD dementia patients. Features included structural MRI, CSF status, demographics, and genetic data. Analyses comprised one cr

Dietary intake of potassium, vitamin E, and vitamin C emerges as the most significant predictors of cardiovascular disease risk in adults.

Prediction models were developed to assess the risk of cardiovascular disease (CVD) based on micronutrient intake, utilizing data from 90,167 UK Biobank participants. Four machine learning models were employed to predict CVD risk, with performance evaluation metrics including area under the receiver operating characteristic curve (AUC), accuracy, recall, specificity, and F1-score. The eXtreme Gradient Boosting (XGBoost) model was utilized to rank the importance of 11 micronutrients in cardiovascular health. Results indicated that vitamin E, calcium, vitamin C, and potassium intake were associa

Imaging of Intimate Partner Violence, From the AJR Special Series on Emergency Radiology.

Intimate partner violence (IPV) is a highly prevalent public health issue with multiple adverse health effects. Radiologists are well suited to assessing a patient's likelihood of IPV. Recognition of common IPV injury mechanisms and resulting target and defensive injury patterns on imaging and understanding of differences between patients who have experienced IPV and those who have not with respect to use of imaging will aid radiologists in accurate IPV diagnosis. Target injuries often involve the face and neck as a result of blunt trauma or strangulation; defensive injuries often involve an e

Intelligent System for Upper Limb Motor Assessment Using Inertial Sensors and Machine Learning for Telerehabilitation Therapies.

Neurological injuries can lead to a significant disability, particularly in upper limb motor function, impairing patients' ability to perform Activities of Daily Living (ADLs). Telerehabilitation has emerged as a promising solution for remote patient monitoring and rehabilitation. Hence, there is a need to develop technologies to facilitate the patient assessment process remotely. This study presents an intelligent evaluation system for assessing upper limb motor function using three Inertial Measurement Units (IMUs). The upper limb joint trajectories are used as inputs to a machine learning m

Novel Physics-Informed Bayesian Fusion Post-Processor for Enhanced Gait Phase Recognition Using Surface Electromyography.

Myoelectric pattern recognition systems serve as a promising predictive control approach for the lower limbs prostheses and exoskeletons. However, their actual deployment is challenged by the signal stochastic nature that could contaminate the decision stream with physiologically implausible transitions, posing safety and metabolic cost concerns on the potential user. Therefore, this study proposes a novel Physics-Informed Bayesian Fusion (PI-BF) post-processor that embeds biomechanical sequentiality constraints into the posterior probabilistic output of the classifiers to suppress unstable tr

Developing a 4-Helix Bundle Protein Engineering Platform for Isolating Rare-Earth Elements.

Rare-earth elements (REEs) are crucial for sustainable industrial processes, but current extraction methods are environmentally damaging. Our research explores using proteins that bind REEs as a potentially eco-friendly alternative for purification. Lanmodulin (LanM) is a natural protein known for its extremely strong (picomolar) affinity for REEs. However, LanM's disordered structure makes it challenging for engineering. To overcome this, we developed a protein engineering platform based on the stable 4-Helix Bundle (4HB) framework, which has 3 helix-loop-helix regions. We incorporated the co

Predicting Mammal Species Suitable for Chagas Disease Spread Using a Machine-Learning Framework.

Understanding which mammalian species participate in the sylvatic cycle of Trypanosoma cruzi is vital for clarifying the ecological processes that maintain Chagas disease transmission. Although more than 100 mammal species have been reported as naturally infected, the actual diversity of hosts is much broader. In this study, we employed a trait-based, machine-learning framework to evaluate mammals associated with natural infection. Data concerning life-history traits, morphological characteristics, environmental factors, trophic ecology, habitat use, biogeographic distributions, and spatial ov

Using large language models to probe the limits of atom-centered structural descriptors.

Mapping an atomic structure to a compact set of geometric descriptors is an essential step in any machine-learning application to atomic-scale modeling. A powerful and widely used approach can be understood as a discretization of the histogram of pair distances, triangles, etc., which results in a hierarchy of symmetry-invariant atom-centered descriptors. Unfortunately, the lower rungs on this hierarchy (two, three, four-body clusters) were found to be incomplete, with symmetry-unrelated pairs of structures having exactly the same descriptors. However, all the "degeneracies" reported so far ar

Development and validation of a machine learning based early warning scoring system for high altitude polycythemia.

BACKGROUND: High-altitude polycythemia (HAPC) lacks a lifestyle-focused risk-stratification tool among lifelong high-altitude residents. Here we aimed to develop and validate a novel machine-learning predictive scoring system for HAPC using readily modifiable lifestyle variables in this population. METHODS: In a high altitude cohort (≥4,500 m, n = 1,089), 82 candidate variables were reduced to seven lifestyle predictors via LASSO, Logistic regression, XGBoost and random forest models were trained and compared (10 fold cross validation). RESULTS: Logistic regression achieved the best balance

Development and validation of a machine learning-based prediction model for thrombocytopenia following cardiopulmonary bypass.

Postoperative thrombocytopenia (TP) is a serious complication of cardiopulmonary bypass (CPB). An accurate prediction model is crucial for the early identification of high-risk patients. This study aimed to develop and validate a prediction model for TP following CPB using machine learning (ML) algorithms combined with least absolute shrinkage and selection operator (LASSO) regression for variable selection. Using a large public database, patients admitted to the intensive care unit (ICU) following initial CPB were included. Clinical demographics, outcomes, and biochemical marker data were col

Graph Fourier transform for spatial omics representation and analyses of complex organs.

Spatial omics technologies are capable of deciphering detailed components of complex organs or tissue in cellular and subcellular resolution. A robust, interpretable, and unbiased representation method for spatial omics is necessary to illuminate novel investigations into biological functions, whereas a mathematical theory deficiency still exists. We present SpaGFT (Spatial Graph Fourier Transform), which provides a unique analytical feature representation of spatial omics data and elucidates molecular signatures linked to critical biological processes within tissues and cells. It outperformed

Biodegradation of ciprofloxacin using machine learning tools: Kinetics and modelling.

Recently, the rampant administration of antibiotics and their synthetic organic constitutes have exacerbated adverse effects on ecosystems, affecting the health of animals, plants, and humans by promoting the emergence of extreme multidrug-resistant bacteria (XDR), antibiotic resistance bacterial variants (ARB), and genes (ARGs). The constraints, such as high costs, by-product formation, etc., associated with the physico-chemical treatment process limit their efficacy in achieving efficient wastewater remediation. Biodegradation is a cost-effective, energy-saving, sustainable alternative for r

Multiscale, multi-perspective imaging assisted robotic microinjection of 3D biological structures.

Microinjection is a widely used technique employed by biologists with applications in transgenesis, cryopreservation, mutagenesis, labeling/dye injection and in-vitro fertilization. However, microinjection is an extremely laborious manual procedure, which makes it a critical bottleneck in the field and thus ripe for automation. Here, we present a computer-guided robot that automates the targeted microinjection of Drosophila melanogaster and zebrafish (Danio rerio) embryos, two important model organisms in biological research. The robot uses a series of cameras to image an agar plate containing

Type III secretion system effectors form robust and flexible intracellular virulence networks.

Infections with many Gram-negative pathogens, including Escherichia coli, Salmonella, Shigella, and Yersinia, rely on type III secretion system (T3SS) effectors. We hypothesized that while hijacking processes within mammalian cells, the effectors operate as a robust network that can tolerate substantial contractions. This was tested in vivo using the mouse pathogen Citrobacter rodentium (encoding 31 effectors). Sequential gene deletions showed that effector essentiality for infection was context dependent and that the network could tolerate 60% contraction while maintaining pathogenicity. Desp
