Alcohol is a well-established teratogen that can cause variable physical and behavioral effects on the fetus. The most severe condition in this spectrum of diseases is known as fetal alcohol syndrome (FAS). The differences in maternal and fetal enzymes, in terms of abundance and efficiency, in addition to reduced elimination, allow for alcohol to have a prolonged effect on the fetus. This can act as a teratogen through numerous methods including reactive oxygen species (generated as by products of CYP2E1), decreased endogenous antioxidant levels, mitochondrial damage, lipid peroxidation, disru

AIMS: To examine the causes of variability in the effect of maternal drinking on the foetus, with particular reference to the pattern, frequency and duration of the period of drinking, differences in maternal, foetal and placental metabolism of ethanol/acetaldehyde, and genetic factors.
METHODS: Narrative review of published studies of the pathogenesis of foetal alcohol syndrome (FAS) with emphasis in the development of the central nervous system.
RESULTS: Animal models suggest that acetaldehyde, the primary hepatic oxidative metabolite of ethanol, reaches the foetus either by placental prod

A study was made of the development of the brain from 44 embryos (5-12 weeks) and 1 fetus (14 weeks) obtained from mothers who used alcohol during pregnancy. 16 cases made up the control group. In 34 cases out of 45 (75.5%), deviations in the development of the brain were established and distributed into the three degrees of gravity: mild, moderate and severe. 11 variants of brain pathology are described. Deviations in the formation of brain walls and disorders of the development of the cortical lamina occurred most frequently (70.6 and 57.9%, respectively). In alcoholic mothers, brain patholo

Studies of alcohol consumption, although somewhat inconsistent, have shown a relationship between prenatal alcohol exposure, growth retardation, and morphologic abnormalities. The inconsistency of the observed effects may be a result of differential exposure, exposure at different times during pregnancy, methodologic problems with identification and measurement, or inadequate control for risk factors that covary with alcohol consumption. There have been too few studies to assess accurately the effect of drinking on development beyond the neonatal period. Some studies have found that infants of

Foetal alcohol syndrome (FAS) is the combination of growth retardation, neurological impairment and abnormal facies shown by some infants born to alcoholic women. It seems likely that FAS only indicates a small proportion of the total damage inflicted on the foetus by maternal drinking. Levels of alcohol consumption below the equivalent of 30 ml per day of absolute alcohol probably do not have a significant effect on the foetus. But there is an increased incidence of either growth retardation or behavioural and congenital abnormalities amongst the infants of women who drink heavily in pregnanc

INTRODUCTION: Alcohol consumption during pregnancy can lead to fetal alcohol spectrum disorders. This study developed and validated a questionnaire to assess university students' knowledge regarding the effects of alcohol during pregnancy.
METHODS: We designed an instrument with true-false-I do not know statements. Initially, 45 true statements were formulated and subjected to content validation by 19 experts. Based on the Content Validity Index (CVI), 17 items were selected. The instrument, called the Fetal Alcohol Consequences Test (FACT), was first assessed by 31 university students for th…

Fetal Alcohol Spectrum Disorder, caused by prenatal alcohol exposure, remains a major public health concern, affecting up to 1 in 20 U.S. schoolchildren. Pregnancy is typically not recognized until six weeks of gestation, alcohol exposure often occurs unknowingly during the peri-conceptional period, a highly vulnerable developmental window of oocyte maturation. The placenta is increasingly recognized as a mediator of alcohol-induced neurodevelopmental risk through the placenta-brain axis. We characterize peri-conceptional alcohol exposure on placental development in a mouse PCA model mimicking

Exposure to alcohol during development can result in a constellation of morphological and behavioral abnormalities that are collectively known as Fetal Alcohol Spectrum Disorders (FASDs). At the most severe end of the spectrum is Fetal Alcohol Syndrome (FAS), characterized by growth retardation, craniofacial dysmorphology, and neurobehavioral deficits. Studies with animal models, including rodents, have elucidated many molecular and cellular mechanisms involved in the pathophysiology of FASDs. Ethanol administration to pregnant rodents has been used to model human exposure during the first and…

Despite several decades of research and prevention efforts, fetal alcohol spectrum disorders (FASD) remain the most common preventable cause of neurodevelopmental disabilities worldwide. Animal and human studies have implicated fetal alcohol-induced alterations in epigenetic programming as a chief mechanism in FASD. Several studies have demonstrated fetal alcohol-related alterations in methylation and expression of imprinted genes in placental, brain, and embryonic tissue. Imprinted genes are epigenetically regulated in a parent-of-origin-specific manner, in which only the maternal or paternal…

Heavy alcohol consumption (more than 80 g per day) in pregnancy is associated with the foetal alcohol syndrome. More moderate consumption is reported to result in increased rates of spontaneous abortion, still birth and congenital malformation as well as growth retardation. There may be functional deficits in early childhood. In the light of current knowledge abstinence from alcohol before and during pregnancy is advisable.

Since the identification of fetal alcohol syndrome (FAS) over 35 years ago, mounting evidence about the impact of maternal alcohol consumption during pregnancy has prompted increased attention to the link between prenatal alcohol exposure (PAE) and a constellation of developmental disabilities that are characterized by physical, cognitive, and behavioral impairments. These disabilities include a continuum of developmental disorders known as fetal alcohol spectrum disorders (FASDs). Longitudinal studies suggest that individuals with FASDs are at a greatly increased risk for adverse long-term ou…

BACKGROUND: Prenatal alcohol exposure can contribute to a wide range of neurodevelopmental impairments in children and adults including behavioral and neuropsychiatric disorders. In rhesus monkeys, we examined whether moderate-level prenatal alcohol exposure would alter acoustic startle responses and prepulse inhibition (PPI) of the acoustic startle. PPI is a highly quantifiable measure of inhibitory neural processes or sensorimotor gating associated with neuropsychiatric disorders.
METHODS: Acoustic startle and PPI of the acoustic startle were tested in 37 adult rhesus monkeys (Macaca mulatt

The facial anomalies associated with fetal alcohol syndrome (FAS), some of which are also present in individuals with less severe forms of the broader category of fetal alcohol spectrum disorders (FASD), are typically identified with the aid of linear distance measurements taken between facial landmarks. Digital facial imaging methods are increasingly being used in syndrome delineation. Distance measurements derived from stereo-photogrammetry and facial surface imaging have been used to study the FAS facial anomalies. Geometric morphometric methods capture the spatial arrangement between landm…

Alcohol constitutes one of the main risk factors for the health of the newborn infant and is also one of the leading causes of cognitive deficit. The distinctive pattern of abnormalities that characterizes fetal alcohol syndrome (FAS) includes: pre- and postnatal growth retardation, cognitive deficit, behavior and language disorders, cerebral malformations (schizencephaly, polymicrogyria, agenesis of the corpus callosum), facial changes (short palpebral fissures, low nasal bridge, anomalies of the auricle, maxillary hypoplasia, cleft lip and palate) and organ anomalies (heart defects, renal an

In 1988 the US Congress passed a law requiring a health warning label on alcoholic beverage containers, to include the message that pregnant women should not drink alcohol. This paper addresses the role that scientific knowledge played in the formation and passage of the alcohol warning label policy. The constellation of birth defects implicated in the fetal alcohol syndrome (FAS) (including fetal alcohol effects) is sketched, and the FAS-related legislative events leading to the law's passage are described. A synopsis of the state of knowledge in 1988 regarding the effects of alcohol on the f

The offspring of serial breedings of voluntary alcohol consuming Sinclair (S-1) miniature swine have been evaluated over the past three years. The first and second litters of 12 dams and the third litter of 6 dams were examined. All study animals had been consuming alcohol for at least 18 months prior to the first breeding. Alcohol consumption was continuous throughout the ninth study. Twelve non-alcohol-consuming sows were used as controls. The results of the study revealed a progressive decrease in mean litter size from controls of 6.66 in the control litters to 1.8 piglets per litter in the
