Thioridazine elicits potent antitumor effects in colorectal cancer stem cells.

As a member of the phenothiazine family, thioridazine (THIO) is a potent anti-anxiety and anti-psychotic drug. Recent studies have reported that THIO could suppress the growth of several types of cancer cells. However, the effect of THIO on colorectal cancer stem cells (CSCs) has not been investigated. In the present study, we examined the effect of THIO on viability of CSCs isolated from the human colon cancer cell line HCT116 and its colony-formation ability, along with its stem cell-specific gene expression. The CSCs, EpCAM+ and CD44+ subpopulations from HCT116 cells were isolated using imm

Second hits in the FLCN gene in a hereditary renal cancer syndrome in dogs.

In this study, samples from multifocal renal tumors from two dogs affected with renal cystadenocarcinoma and nodular dermatofibrosis (RCND) were collected for detection of putative second hits in the FLCN gene. Genomic DNA from the samples was typed at the previously identified disease-associated missense mutation and cDNA representing the entire coding region of the FLCN gene was sequenced for mutation detection. Second hits with predicted functional implications for the wild-type FLCN allele were observed in 12 of 17 (71%) of the kidney tumor samples. The type of mutation of the second hits

Characterization of the tumor immune microenvironment in pregnancy-associated breast cancer through multiplex immunohistochemistry and transcriptome analyses.

BACKGROUND: Pregnancy-associated breast cancer (PABC) is breast cancer diagnosed during pregnancy or within 2 years postpartum. Although relatively rare, it is associated with a poor prognosis, and the underlying mechanisms contributing to this unfavorable condition remain incompletely understood. In this study, we investigated tumor microenvironmental features linked to pregnancy and lactation in an effort to elucidate these mechanisms. METHODS: This retrospective study included 26 patients with PABC, 51 patients with breast cancer diagnosed 2-5 years postpartum (post-weaning breast cancer [

Cell-free DNA promotes malignant transformation in non-tumor cells.

Cell-free DNA is present in different biological fluids and when released by tumor cells may contribute to pro-tumor events such as malignant transformation of cells adjacent to the tumor and metastasis. Thus, this study analyzed the effect of tumor cell-free DNA, isolated from the blood of prostate cancer patients, on non-tumor prostate cell lines (RWPE-1 and PNT-2). To achieve this, we performed cell-free DNA quantification and characterization assays, evaluation of gene and miRNA expression profiling focused on cancer progression and EMT, and metabolomics by mass spectrometry and cellular m

Network-based differential gene expression analysis suggests cell cycle related genes regulated by E2F1 underlie the molecular difference between smoker and non-smoker lung adenocarcinoma.

BACKGROUND: Differential gene expression (DGE) analysis is commonly used to reveal the deregulated molecular mechanisms of complex diseases. However, traditional DGE analysis (e.g., the t test or the rank sum test) tests each gene independently without considering interactions between them. Top-ranked differentially regulated genes prioritized by the analysis may not directly relate to the coherent molecular changes underlying complex diseases. Joint analyses of co-expression and DGE have been applied to reveal the deregulated molecular modules underlying complex diseases. Most of these method

Game of clones: decipher lineage plasticity in hormone-driven cancers.

Hormone-dependent cancers such as prostate, breast, and endometrial carcinomas rely on nuclear hormone receptors to sustain lineage identity and growth. Therapies targeting androgen, estrogen, or progesterone signaling are initially effective but ultimately impose selective pressures that drive resistance through lineage plasticity, the ability of tumor cells to abandon their native identity and adopt alternative cellular fates. While the biological consequences of lineage plasticity are increasingly recognized, a major challenge lies in defining the regulatory programs that govern these cell

The Preliminary Study for the Trial of Using Selected miRNAs for the Laboratory Diagnosis of Colorectal Cancer.

Colorectal cancer (CRC) is the second most common cancer in Europe. There is a need to explore and validate new blood-based tumor markers to improve the selection of patients who are likely to benefit from an early, non-invasive diagnosis of CRC. The purpose of this report is to present the test protocol and its verification. The study is planned in four phases. The first trial phase involves collecting material, consisting of healthy tissue, diseased tissue, and plasma, from 120 CRC patients over 50 years old during surgery. This phase also involves identifying microRNAs (miRNAs) and comparin

Identification of an Anoikis-Related Gene Signature to Predict the Prognosis in Patients With Oral Squamous Cell Carcinoma.

BACKGROUND: Anoikis plays an important role in cancer metastasis. Nevertheless, research that focuses on the prognostic values of anoikis-related genes (ARGs) in oral squamous cell carcinoma (OSCC) is still scarce. METHODS: The ARG-related gene signature was developed via LASSO regression using the TCGA-OSCC cohort. OSCC patients in the TCGA_OSCC and GSE41613 datasets were grouped into high- and low-ARG score (H-score, L-score) groups, and the overall survival (OS) between the 2 groups was analysed. Meanwhile, functional enrichment, CIBERSORT and drug sensitivity were conducted based on the d

Identification and prognostic analysis of genes related to CTNNB1 mutations in hepatocellular carcinoma.

BACKGROUND: Mutations in CTNNB1 are recognized oncogenic drivers of hepatocellular carcinoma (HCC); however, the downstream effector molecules and their prognostic significance remain incompletely defined. In this study, we developed a mutant CTNNB1 gene signature (MCGS) to predict CTNNB1 mutation status and to evaluate the prognostic relevance of the constituent genes. METHODS: Hydrodynamic tail vein injection mouse models of HCC were generated by co-expressing Sleeping Beauty transposon/transposase and mutant CTNNB1 with either AKT-c-Myc or AKT-NRASG12V, followed by RNA sequencing of tumor

Identification of polyamine metabolism-related gene signatures for prognostic and immunological stratification in glioma.

Polyamine metabolism plays an essential role in glioma progression and the tumor microenvironment (TME). However, its prognostic and immunotherapeutic significance remains incompletely understood. We integrated transcriptomic and clinical data from The Cancer Genome Atlas (TCGA) to construct a prognostic risk model via Least Absolute Shrinkage and Selection Operator (LASSO) regression. This model was externally validated using the GSE108474 and GSE43378 cohorts. Multivariable Cox regression was employed to identify key prognostic predictors, while consensus clustering was applied to stratify p

The RNA methylation modification as an immunometabolic regulatory hub in pancreatic cancer: from mechanistic insights to clinical translation perspectives.

RNA methylation represents the most abundant post-transcriptional modification in RNA and has recently emerged as a central topic in molecular biology. It precisely regulates various biological functions by influencing key processes like RNA splicing, translation, transport, and stability. Pancreatic cancer remains one of the most aggressive and lethal malignancies, with its high mortality largely attributed to late diagnosis and limited therapeutic options. Recent research has shown that RNA methylation is intimately linked to tumor immunity and is vital in forming the tumor microenvironment

Analysis and validation of abnormal signaling pathways and immune cell infiltration characteristics in digestive system cancers based on peroxisome-related genes.

BACKGROUND: Although emerging evidence suggests a role for peroxisomes in tumorigenesis, their functions in digestive cancers remain unclear. This study aims to investigate the association between peroxisomes and digestive tract tumors. METHODS: To systematically investigate peroxisomal functions in digestive cancers, we first constructed and validated tumor-specific prognostic signatures based on peroxisome-related genes (PRGs) through univariate Cox, least absolute shrinkage and selection operator (LASSO), and multivariate Cox regression analyses. We then characterized the tumor immune micr

MicroRNA-based strategies to overcome the immunotherapy barrier in pancreatic ductal adenocarcinoma.

Pancreatic ductal adenocarcinoma (PDAC) is a highly lethal malignancy that continues to pose a major clinical challenge, primarily due to the difficulty of early detection and the limited efficacy of existing therapeutic approaches. Immunotherapy, which has revolutionized the treatment of many other cancers, has shown limited success in PDAC, largely because of the complex and immunosuppressive features of its tumor microenvironment (TME). Consequently, strategies aimed at remodeling or modulating the TME have emerged as promising avenues for enhancing the therapeutical potential of immunother

MicroRNAs in esophageal squamous cell carcinoma.

Esophageal squamous cell carcinoma (ESCC) is an aggressive malignancy requiring improved early diagnosis and prognostic assessment. MicroRNAs (miRNAs), small non-coding RNAs that regulate gene expression, have emerged as promising biomarkers for ESCC. We conducted a comprehensive literature search in PubMed for studies published up to 2025. While miRNA profiles in ESCC tissues show significant associations with lymphatic dissemination, tumor progression, and patient survival, clinical translation remains limited by issues of sensitivity, specificity, and standardization. This review summarizes
