Induction of early light-inducible protein gene expression in Pisum sativum after exposure to low levels of UV-B irradiation and other environmental stresses.

Plants are constantly subjected to environmental changes and have developed various defence mechanisms to facilitate their continued existence. Pisum sativum plants were exposed to low levels of UV-B radiation and ELIP (early light-inducible proteins) mRNA, with a probable protective function, was rapidly and strongly induced during this type of stress. To our knowledge, this is the only photosynthetic gene that is up-regulated following exposure to UV-B, and this result has to be compared with studies predominantly reporting down-regulation by UV-B of genes encoding proteins localised in the

Molecular identification of the phosphate transporter family 1 (PHT1) genes and their expression profiles in response to phosphorus deprivation and other abiotic stresses in Brassica napus.

Phosphate (Pi) transporters play critical roles in Pi acquisition and homeostasis. However, little is known about these transporters in oilseed rape. Therefore, the aim of the present study was to characterize the members of the PHT1 gene family in allotetraploid Brassica napus and to analyze their expression profiles in response to environmental stresses. In total, 49 PHT1 family members were identified in B. napus, including 27 genes in the A subgenome and 22 in the C subgenome. Most of the PHT1 proteins were predicted to localize to the plasma membrane. Phylogenetic analysis suggested that

Iron sensors and signals in response to iron deficiency.

The transcription of genes involved in iron acquisition in plants is induced under iron deficiency, but our understanding of iron sensors and signals remains limited. Iron Deficiency-responsive Element-binding Factor 1 (IDEF1) and Hemerythrin motif-containing Really Interesting New Gene- and Zinc-finger proteins (HRZs)/BRUTUS (BTS) have recently emerged as candidate iron sensors because of their functions as potent regulators of iron deficiency responses and their iron-binding properties. IDEF1 is a central transcriptional regulator of graminaceous genes involved in iron uptake and utilization

Gibberellin 20-oxidase gene OsGA20ox3 regulates plant stature and disease development in rice.

Gibberellin (GA) 20-oxidase (GA20ox) catalyses consecutive steps of oxidation in the late part of the GA biosynthetic pathway. A T-DNA insertion mutant (17S-14) in rice, with an elongated phenotype, was isolated. Analysis of the flanking sequences of the T-DNA insertion site revealed that an incomplete T-DNA integration resulted in enhanced constitutively expression of downstream OsGA20ox3 in the mutant. The accumulation of bioactive GA(1) and GA(4) were increased in the mutant in comparison with the wild-type plant. Transgenic plants overexpressing OsGA20ox3 showed phenotypes similar to those

Molecular characterization of Oryza sativa WRKY6, which binds to W-box-like element 1 of the Oryza sativa pathogenesis-related (PR) 10a promoter and confers reduced susceptibility to pathogens.

WRKY proteins are transcription factors (TFs) that regulate the expression of defense-related genes. The salicylic acid (SA)-inducible Oryza sativa WRKY6 (OsWRKY6) was identified as a positive regulator of Oryza sativa pathogenesis-related 10a (OsPR10a) by transient expression assays. A physical interaction between OsWRKY6 and W-box-like element 1 (WLE1), which positively regulates OsPR10a/probenazole induced protein 1 expression, was verified in vitro. Several pathogenesis-related (PR) genes were constitutively activated, including OsPR10a, and transgenic rice (Oryza sativa) plants overexpres

Basal Differences in the Transcriptional Profiles of Tomato Roots Associated with the Presence/Absence of the Resistance Gene Mi-1 and Time-Course Changes During the Compatible and Incompatible Interactions with the Root-Knot Nematode Meloidogyne javanica.

The Mi-1 gene of tomato is responsible for the resistance of certain genotypes to root-knot nematodes or RKN (Meloidogyne spp.) and other harmful organisms such as aphids or whiteflies, in a complex cascade of transcriptional changes in which other tomato genes are also involved. The objective of this study was to gain a deeper understanding of the Mi-1-mediated resistance of tomato to Meloidogyne javanica using oligonucleotide microarrays to identify additional plant genes involved in the compatible or incompatible tomato/nematode interactions. Microarray analysis was selected as it has been

Knockout analysis of Arabidopsis transcription factors TGA2, TGA5, and TGA6 reveals their redundant and essential roles in systemic acquired resistance.

Arabidopsis nonexpresser of pathogenesis-related (PR) genes (NPR1) is the sole positive regulator that has been shown to be essential for the induction of systemic acquired resistance. In npr1 mutant plants, salicylic acid (SA)-mediated PR gene expression and pathogen resistance are abolished completely. NPR1 has been shown to interact with three closely related TGA transcription factors-TGA2, TGA5, and TGA6-in yeast two-hybrid assays. To elucidate the biological functions of these three TGA transcription factors, we analyzed single and combined deletion knockout mutants of TGA2, TGA5, and TGA

Two ferric uptake regulator proteins in Ralstonia pseudosolanacearum strain OE1-1 function cooperatively in response to the extracellular iron level under ferrous iron-rich conditions.

UNLABELLED: Soil-borne Gram-negative bacteria in the Ralstonia solanacearum species complex (RSSC) cause bacterial wilt symptoms in diverse crop plants. Although iron is an important metal ion for most organisms, the regulatory effect of iron on soil-borne RSSC gene expression remains unknown. Thus, we identified two ferric uptake regulator (Fur) proteins, Fur1 and Fur2, in R. pseudosolanacearum strain OE1-1. All four RSSC phylotype strains had two Fur homologs, a conserved Fur (Fur1) and an alternative Fur (Fur2). We also analyzed the transcriptomes of a fur1 deletion mutant, fur2 deletion mu

Stomatal XVE: an inducible system for cell-stage-specific gene expression and editing in the stomatal lineage.

Stomatal development has emerged as a valuable model for studying developmental processes. Examining gene function along the stomatal lineage often requires gene perturbation in a controlled and cell-stage-specific manner, but this remains tedious without a dedicated genetic tool. Here, we describe Stomatal XVE, a modular, two-component XVE-based inducible system that enables user-controlled gene overexpression and Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR)-based knockout at defined stomatal cell stages in Arabidopsis thaliana. The system consists of a collection of est

A Simple RUBY Reporter System in Nicotiana benthamiana for Studying Inositol Pyrophosphate Dynamics.

In recent years, there has been substantial progress in the development of methods to analyze inositol phosphates (InsPs) and inositol pyrophosphates (PP-InsPs). However, many of these techniques are labor- and cost-intensive and can usually only be carried out by laboratories specialized in InsPs/PP-InsPs analysis. In this chapter, we present a simple method that exploits the fact that phosphorylation and/or dephosphorylation of certain InsP/PP-InsP species induces the activation of promoters driving the expression of genes involved in phosphate starvation response (PSR). By linking PSR-induc

Editing a gibberellin receptor gene improves yield and nitrogen fixation in soybean.

Soybean is an important source of oil, protein, and feed. However, its yield is far below that of major cereal crops. The green revolution increased the yield of cereal crops partially through high-density planting of lodging-resistant semi-dwarf varieties, but required more nitrogen fertilizers, posing an environmental threat. Genes that can improve nitrogen use efficiency need to be integrated into semi-dwarf varieties to avoid the overuse of fertilizers without the loss of dwarfism. Unlike cereal crops, soybean can assimilate atmospheric nitrogen through symbiotic bacteria. Here, we created

A detailed comparative in silico and functional analysis of ccd1 gene in maize gives new insights of its expression and functions.

BACKGROUND: Biofortified maize with enhanced carotenoid content was developed to combat vitamin A deficiency. However, it was observed that during storage, carotenoids present in maize grain get degraded and it has been reported that carotenoid cleavage dioxygenase1 (ccd1) is responsible for this degradation. METHODS AND RESULTS: In our current study, comprehensive in-silico analysis deciphered a complete overview of the ccd1 gene in maize including the gene structures, phylogeny, chromosomal locations, promoter analysis, conserved motifs and interacting protein partners. In addition to these

PtoNF-YC9-SRMT-PtoRD26 module regulates the high saline tolerance of a triploid poplar.

BACKGROUND: Sensing and responding to stresses determine the tolerance of plants to adverse environments. The triploid Chinese white poplar is widely cultivated in North China because of its adaptation to a wide range of habitats including highly saline ones. However, its triploid genome complicates any detailed investigation of the molecular mechanisms underlying its adaptations. RESULTS: We report a haplotype-resolved genome of this triploid poplar and characterize, using reverse genetics and biochemical approaches, a MYB gene, SALT RESPONSIVE MYB TRANSCRIPTION FACTOR (SRMT), which combines

Functional analysis of the role of hydrogen sulfide in the regulation of dark-induced leaf senescence in Arabidopsis.

There is growing evidence that hydrogen sulfide (H2S) is involved in many physiological processes in plants, but the role of H2S in dark-induced leaf senescence remains unknown. In this work, we found that H2S not only inhibited chlorophyll degradation but also caused the accumulation of photoreactive pheide a in detached leaves under extended darkness. Despite this, transcript levels of senescence-associated genes (SAGs) were less affected in H2S-treated detached leaves compared with those in H2S-untreated detached leaves. Furthermore, cell death/rapid bleaching occurred in both H2S-treated d

The CHD3 remodeler PICKLE promotes trimethylation of histone H3 lysine 27.

CHD3 proteins are ATP-dependent chromatin remodelers that contribute to repression of developmentally regulated genes in both animal and plant systems. In animals, this repression has been linked to a multiple subunit complex, Mi-2/NuRD, whose constituents include a CHD3 protein, a histone deacetylase, and a methyl-CpG-binding domain protein. In Arabidopsis, PICKLE (PKL) codes for a CHD3 protein that acts during germination to repress expression of seed-associated genes. Repression of seed-associated traits is promoted in pkl seedlings by the plant growth regulator gibberellin (GA). We underto

Autonomous differentiation of transgenic cells requiring no external hormone application: the endogenous gene expression and phytohormone behaviors.

The ectopic overexpression of developmental regulator (DR) genes has been reported to improve the transformation in recalcitrant plant species because of the promotion of cellular differentiation during cell culture processes. In other words, the external plant growth regulator (PGR) application during the tissue and cell culture process is still required in cases utilizing DR genes for plant regeneration. Here, the effect of Arabidopsis BABY BOOM (BBM) and WUSCHEL (WUS) on the differentiation of tobacco transgenic cells was examined. We found that the SRDX fusion to WUS, when co-expressed wit

Agrochemical-responsive gene expression control systems for modulating plant development and metabolism.

Precise temporal control of gene expression is critical for studying plant biology and engineering complex crop traits. Current systems enable chemically inducible regulation but rely on costly or agriculturally impractical inducers and lack the flexibility needed to regulate combinations of native loci and transgenes. In this work, we elucidate the design rules for control systems, based on Cas9 and Cre recombinase fused to the ecdysone receptor (EcR), which respond to a widely used agrochemical methoxyfenozide (MF). First, we validated the function of both circuits in transient assays and ex

Genome-wide identification of the WRKY gene family in apricot (Prunus armeniaca L.) and functional characterization of PaWRKY57 in salt stress tolerance.

Soil salinization severely restricts the growth, yield, and industrial development of apricot (Prunus armeniaca L.). WRKY transcription factors play crucial roles in plant responses to abiotic stresses. However, the genome-wide characteristics of the WRKY family in apricot and their functional mechanisms in salt tolerance remain unclear. In this study, we identified 58 PaWRKY genes in apricot and characterized their phylogeny, chromosomal distribution, and gene structure. Based on transcriptomic and phylogenetic analyses, PaWRKY57 (homologous to AtWRKY33) was selected as a key salt-responsive

Effect of the overexpression of the GGP1 gene on cell wall remodelling and redox state in the tomato fruit.

Tomato fruit ripening is a complex physiological process that involves morphological, anatomical, biochemical, and molecular alterations. One of these changes occurring during ripening is the softening of the fruit, which is attributed to modifications in the biosynthesis and degradation of individual cell wall components, i.e. polysaccharides and proteoglycans. In addition, ripening is affected by redox processes, and interplay of the reactive oxygen species (ROS) and specific antioxidants, enzymes, ascorbate, and phenolic compounds. The present study aims to determine the impact of the overe
