Hybrid assemblies of fluorescent nanocrystals and membrane proteins in liposomes.

Because of the growing potential of nanoparticles in biological and medical applications, tuning and directing their properties toward a high compatibility with the aqueous biological milieu is of remarkable relevance. Moreover, the capability to combine nanocrystals (NCs) with biomolecules, such as proteins, offers great opportunities to design hybrid systems for both nanobiotechnology and biomedical technology. Here we report on the application of the micelle-to-vesicle transition (MVT) method for incorporation of hydrophobic, red-emitting CdSe@ZnS NCs into the bilayer of liposomes. This met

Irregular structure of the HIV fusion peptide in membranes demonstrated by solid-state NMR and MD simulations.

To better understand peptide-induced membrane fusion at a molecular level, we set out to determine the structure of the fusogenic peptide FP23 from the HIV-1 protein gp41 when bound to a lipid bilayer. An established solid-state (19)F nuclear magnetic resonance (NMR) approach was used to collect local orientational constraints from a series of CF(3)-phenylglycine-labeled peptide analogues in macroscopically aligned membranes. Fusion assays showed that these (19)F-labels did not significantly affect peptide function. The NMR spectra were characteristic of well-behaved samples, without any signs

The case for considering the term 'mitochondrial vesicle' as a misnomer in publications about assisted reproductive technologies (ART) for amphibians.

The term 'mitochondrial vesicle' was first used in 2003 in a description of anuran sperm and persists to this day throughout the literature on assisted reproductive technologies (ART) for amphibians. In the present paper, we argue that the term is inappropriate because the widely accepted definition of a 'vesicle' refers to an integral structure with an enclosing lipid bilayer/membrane. Moreover, there are no electron micrographs that show a vesicular structure encapsulating mitochondria on amphibian sperm heads in the literature. In fact, in 1993, the mitochondria in the anuran sperm head had

[Advances in the Regulation of Follicular Development by Extracellular Vesicles and Non-Coding RNAs].

Extracellular vesicles (EV),nanoscale vesicles encapsulated by phospholipid bilayers, are rich in biological molecules such as nucleic acids, metabolites, proteins, and lipids derived from parental cells. They are mainly involved in intercellular communication, signal transmission, and material transport and affect the functions of target cells.Ovulation disorders account for a higher proportion in the factors causing infertility which demonstrates increasing incidence year by year.Non-coding RNAs participate in a series of physiological and pathological processes of follicular development, playing a …

A structural perspective on GABAA receptor pharmacology.

GABAA receptors are pentameric ligand-gated chloride channels of crucial importance for the vertebrate nervous system physiology. They typically modulate the fast inhibitory neurotransmission, and represent the target receptors for major classes of drugs used in the clinic, such as benzodiazepines and general anesthetics. Recent technological progress in structural biology, in particular single-particle cryo-electron microscopy, has led to fundamental advances in understanding the detailed organization and signalling mechanisms of major GABAA receptor subtypes. This effort culminated with the

Getting membrane proteins on and off the shuttle bus between the endoplasmic reticulum and the Golgi complex.

Secretory proteins exit the endoplasmic reticulum (ER) in coat protein complex II (COPII)-coated vesicles and then progress through the Golgi complex before delivery to their final destination. Soluble cargo can be recruited to ER exit sites by signal-mediated processes (cargo capture) or by bulk flow. For membrane proteins, a third mechanism, based on the interaction of their transmembrane domain (TMD) with lipid microdomains, must also be considered. In this Commentary, I review evidence in favor of the idea that partitioning of TMDs into bilayer domains that are endowed with distinct physic

The conserved Bardet-Biedl syndrome proteins assemble a coat that traffics membrane proteins to cilia.

The BBSome is a complex of Bardet-Biedl Syndrome (BBS) proteins that shares common structural elements with COPI, COPII, and clathrin coats. Here, we show that the BBSome constitutes a coat complex that sorts membrane proteins to primary cilia. The BBSome is the major effector of the Arf-like GTPase Arl6/BBS3, and the BBSome and GTP-bound Arl6 colocalize at ciliary punctae in an interdependent manner. Strikingly, Arl6(GTP)-mediated recruitment of the BBSome to synthetic liposomes produces distinct patches of polymerized coat apposed onto the lipid bilayer. Finally, the ciliary targeting signal

The role of citrullinated proteins suggests a novel mechanism in the pathogenesis of multiple sclerosis.

The pathogenesis of MS is unknown. In our studies, we have demonstrated an important role for citrullinated myelin basic protein (MBP). The accompanying loss of positive charge compromises the ability of MBP to interact with the lipid bilayer. The conversion of arginine to citrulline in brain is carried out by an enzyme peptidyl arginine deiminase (PAD) 2. The amount of PAD 2 in brain was increased in MS normal-appearing white matter. The mechanism responsible for this increase involved hypomethylation of the promoter region in the PAD 2 gene in MS, but no change (compared to normal) was found

Evaluation of the antimicrobial efficiency of three novel chimeric peptides through biochemical and biophysical analyses.

Three chimeric membrane-active antimicrobial peptides (AMPs) were designed from previously characterized parental molecules, namely pandinin-2, ascaphin-8, and maximin-3. The aim of constructing these chimeras was to obtain sequences with improved therapeutic indices or increased activity, while simultaneously investigating the functional roles of key segments of the parental peptides. Chimera-1 was the most active peptide against clinically relevant bacterial species, followed by chimera-2, and chimera-3, respectively, with no clear preference towards Gram-negative or Gram-positive strains. Phospholipid membrane permeabilization and leakage of cell content by surfactin-C15 from novel B. subtilis B-11 strain: a computational and experimental analysis.

Surfactin-C15 produced by novel Bacillus subtilis B-11 strain has the potential to inhibit phytopathogens by permeabilizing their phospholipid cell membranes at the water/bilayer interface. This permeabilization leads to the disintegration of cell membranes, thus inhibiting growth, replication, and pathogenicity of phytopathogens. Model dipalmitoyl phosphocholine (DPPC) vesicles for pathogenic membranes were prepared by liposomal assays and used as representatives for phospholipid bilayer cell membrane. Results show that the hydrophobic fatty acid tail of surfactin-C15 binds with the hydrophob

Influence of Cholesterol on the Insertion and Interaction of SARS-CoV-2 Proteins with Lipid Membranes.

Cholesterol is an essential sterol in cell membranes that regulates organization and fluidity. This biomolecule has been identified as one of the critical factors in the internalization process of various viruses in human cells. Therefore, understanding these mechanisms is crucial for a deeper comprehension of viral pathogenicity in the search for practical therapeutic approaches against viral diseases. The biochemical and biophysical processes related to these diseases are highly complex. For this reason, studying model systems capable of mimicking the interaction of lipid membranes with chol

Association of alpha-synuclein and mutants with lipid membranes: spin-label ESR and polarized IR.

Alpha-synuclein is a presynaptic protein, the A53T and A30P mutants of which are linked independently to early-onset familial Parkinson's disease. The association of wild-type alpha-synuclein with lipid membranes was characterized previously by electron spin resonance (ESR) spectroscopy with spin-labeled lipids [Ramakrishnan, M., Jensen, P. H., and Marsh, D. (2003) Biochemistry 42, 12919-12926]. Here, we study the interaction of the A53T and A30P alpha-synuclein mutants and a truncated form that lacks the acidic C-terminal domain with phosphatidylglycerol bilayer membranes, using anionic phosp

3D nanoscale imaging of amyloid-β oligomer interactions with extracellular vesicles by cryo-ET.

Central to Alzheimer's disease pathology are prefibrillar oligomer assemblies of amyloid-β (Aβ) peptide. A widely discussed hypothesis proposes that amyloid-β oligomers insert into neuronal lipid membranes, disrupting their integrity and causing a loss of cellular homeostasis in Alzheimer's disease. This membrane disruption is believed to be a major source of Aβ-induced neurotoxicity. Cryo electron tomography (cryo-ET) has facilitated 3D nanoscale imaging of Aβ-membrane interactions under near-native conditions. Analyses of small extracellular vesicles (sEVs) reveals that Aβ oligomers includin

Limitations and challenges in the characterization of extracellular vesicles from stem cells and serum.

Exosomes are a subpopulation of nanosized extracellular vesicles (EVs), formed by a lipid bilayer and naturally secreted by cells. They transport RNA, microRNAs, bioactive proteins, and lipids and play an important role in intercellular communication. Exosomes are a promising alternative cell-free therapy in regenerative medicine, immunotherapy, and drug delivery. The implementation of new exosomes treatments requires knowledge of its concentration, purity, and full characterization. However, comparing different studies is highly challenging due to the lack of validated methodologies for isola

Anti-PEG antibodies compromise the integrity of PEGylated lipid-based nanoparticles via complement.

PEGylation of lipid-based nanoparticles and other nanocarriers is widely used to increase their stability and plasma half-life. However, either pre-existing or de novo formed anti-PEG antibodies can induce hypersensitivity reactions and accelerated blood clearance through binding to the nanoparticle surfaces, leading to activation of the complement system. In this study, we investigated the consequences and mechanisms of complement activation by anti-PEG antibodies interacting with different types of PEGylated lipid-based nanoparticles. By using both liposomes loaded with different (model) dru

Self-assembly of laminin induced by acidic pH.

The supramolecular architecture of the basement membrane is provided by two enmeshed networks of collagen IV and laminin. The laminin network is maintained exclusively by interactions among individual laminin molecules and does not depend on the presence of other extracellular matrix components. Laminin polymers can be obtained in vitro either in solution or in association with the surface of bilayers containing acidic lipids. In this work, we have tested the hypothesis that the negative charges present on acidic lipids establish an acid microenvironment that is directly responsible for induci

How cholesterol modulates LL-37 function: A biophysical study in eukaryotic-like membrane systems.

The limited selectivity of antimicrobial peptides (AMPs) can lead to unintended damage to healthy eukaryotic cells, a process strongly influenced by membrane physicochemical properties. Cholesterol (CHO) is a key regulator of lipid packing, membrane order, and bilayer mechanical stability; however, its role in modulating AMP-membrane interactions remain incompletely understood. Here, we examine how CHO alters peptide-membrane interactions at the molecular level using eukaryotic membrane models composed of phosphatidylcholine, sphingomyelin, and increasing CHO fractions. Simulating Gas Permeation Through the Central Pore of AQP5.

Aquaporins (AQPs), a family of integral membrane proteins that serve as water channels, have been hypothesized to facilitate the permeation of metabolic gases (O2 and CO2) via their central pore (CP) formed in the middle of AQP tetramers. Our examination of aquaporin-5 (AQP5), an isoform expressed in lung epithelial cells, using atomistic molecular dynamics (MD) simulations suggests that gas can indeed permeate through the protein. Free energy calculations by implicit ligand sampling (ILS) predict that gas molecules favorably partition into the hydrophobic interior of the CP. Following up, we
