The Impact of Vipassana Meditation on Health and Well-Being: A Systematic Review of Current Evidence.

Vipassana meditation, an ancient Buddhist-derived practice that emphasises insight through non-judgmental observation of sensations and thou … and fewer migraine days; and behavioural improvements encompassed enhanced executive functions and memory consolidation, with stronger effects noted in intensive retreats and among experienced meditators, although evidence was limited by small sample sizes, moderate to high risk of bias, and absence of blinding. In conclusion, moderate evidence supports the benefits …

Slow synaptic plasticity from the hippocampus underlies gradual mapping and fragmentation of novel spaces by grid cells.

Animals construct internal "cognitive maps" of the world during navigation in spatial and non-spatial domains, with grid cells in the medial entorhinal cortex (MEC) playing a key role. This requires associating internal position estimates with external cues to reduce spatial uncertainty over time. However, how grid cell representations evolve in novel spaces to support map formation is unclear. To address this question, we longitudinally imaged calcium dynamics of grid cells over 10 days as mice learn operant tasks in novel virtual linear tracks. We observe that spatial tuning of grid cells is

Lamotrigine Restores Impairments in Theta Rhythms and LTP as Early Biomarkers of Aβ1-42-Induced Hippocampal Network Dysfunction.

Neuronal excitation/inhibition (E/I) imbalance, epileptiform activity, and synaptic dysfunction are present in individuals diagnosed with dementia, including sporadic Alzheimer's disease (sAD), even at early stages. These abnormalities are associated with altered neuronal oscillatory activity, as seen in EEG recordings of sAD patients and transgenic rodents. Hippocampal theta oscillations are crucial for sensorimotor integration, memory consolidation, and network coordination, and depend on synaptic and ionic mechanisms, including the Ih current, mediated by hyperpolarization-activated cyclic …

Ripple activity in the dentate gyrus of dishinibited hippocampus-entorhinal cortex slices.

Fast oscillations at approximately 200 Hz, termed ripples, occur in the hippocampus and cortex of several species, including humans, and are thought to play a role in physiological (e.g., sensory information processing or memory consolidation) and pathological (e.g., seizures) processes. Blocking gamma-aminobutyric acid type A (GABA(A)) receptor-mediated inhibition represents one of the most often used models of epileptiform discharge. Here we found that bath application of the GABA(A) receptor antagonist picrotoxin (50 microM) to mouse hippocampus-entorhinal cortex slices induced spontaneous …

Thymic Composition Predicts Radiation Pneumonitis in Locally Advanced NSCLC.

INTRODUCTION: Durvalumab consolidation after concurrent chemoradiotherapy (cCRT) is standard for unresectable stage III NSCLC; however, radiation pneumonitis (RP) remains a major treatment-limiting toxicity. Lung dosimetry incompletely explains the risk of toxicity. Recent studies have established radiographic thymic health as clinically relevant in cancer, but its relationship to treatment-related toxicity remains unclear. We evaluated whether percent thymic tissue (pTT), an interpretable CT-derived measure of residual thymic tissue, is associated with severe pneumonitis after cCRT in NSCLC.

Cannabidiol and brain function: current knowledge and future perspectives.

Cannabidiol (CBD) is a naturally occurring non-psychoactive cannabinoid found in Cannabis sativa, commonly known as cannabis or hemp. Although currently available CBD products do not meet the safety standards of most food safety authorities to be approved as a dietary supplement or food additive, CBD has been gaining widespread attention in recent years due to its various potential health benefits. While primarily known for its therapeutic effects in managing epileptic seizures, psychosis, anxiety, (neuropathic) pain, and inflammation, CBD's influence on brain function has also piqued the inte

REM sleep dysregulation in depression: state of the art.

Disturbances of sleep are typical for most depressed patients and belong to the core symptoms of the disorder. Since the 1960s polysomnographic sleep research has demonstrated that besides disturbances of sleep continuity, depression is associated with altered sleep architecture, i.e., a decrease in slow wave sleep (SWS) production and disturbed rapid eye movement (REM) sleep regulation. Shortened REM latency (i.e., the interval between sleep onset and the occurrence of the first REM period), increased REM sleep duration and increased REM density (i.e., the frequency of rapid eye movements per

Long-term effects of brief acute stress on cellular signaling and hippocampal LTP.

In a previous study, we reported that a brief exposure to swim stress transforms an electrically induced, protein synthesis-independent early long-term potentiation (early LTP) into a protein synthesis-dependent late LTP ["reinforcement of LTP" in the hippocampal dentate gyrus (DG)] (Korz and Frey, 2003). This transformation depends on activation of mineralocorticoid receptors (MRs) by corticosterone, and on intact basolateral amygdala (BLA) function. Here, we demonstrate that a brief swim experience results in lasting changes in levels of hippocampal cellular signaling molecules that are know

Post-translational modifications in CD8+ T cell exhaustion: mechanisms, networks, and epigenetic locking.

CD8+ T cells are core effector cells of adaptive immunity. However, in chronic infection and tumor microenvironments, they often lose their effector functions. Under persistent antigenic and metabolic stress, CD8+ T cells may progressively acquire a stable dysfunctional state termed T cell exhaustion. Post-translational modifications (PTMs) constitute a rapid and dynamic regulatory system that targets four core functional modules: membrane receptor signaling, transcription factors, metabolic enzymes, and epigenetic regulators. These modules are interconnected through a multi-layered PTM networ

Advancing Scoliosis Treatment with Patient-Specific Functionally Graded NiTi-SMA Rods: Key Considerations and Development Objectives.

This review develops a materials-to-clinic framework for patient-specific, functionally graded (FG) NiTi shape memory alloy (SMA) rods as a complementary paradigm for scoliosis correction that targets durable alignment with motion preservation. The article synthesizes the thermomechanical basis of NiTi (thermoelastic martensitic transformation, near constant superelastic plateau, and hysteretic damping) while leveraging additive manufacturing (AM) capabilities to spatially program transformation temperatures (e.g., Af), effective stiffness, and geometric inertia along the rod. Does anodal cerebellar tDCS boost transfer of after-effects from throwing to pointing during prism adaptation?

Prism Adaptation (PA) is a useful method to study the mechanisms of sensorimotor adaptation. After-effects following adaptation to the prismatic deviation constitute the probe that adaptive mechanisms occurred, and current evidence suggests an involvement of the cerebellum at this level. Whether after-effects are transferable to another task is of great interest both for understanding the nature of sensorimotor transformations and for clinical purposes. However, the processes of transfer and their underlying neural substrates remain poorly understood. Transfer from throwing to pointing is know

Effects of bright light exposure on human fear conditioning, extinction, and associated prefrontal activation.

Bright light (BL) not only regulates human emotion and circadian physiology but can also directly modulate emotional memories. Impaired fear extinction and enhanced fear acquisition and consolidation are hallmarks of fear-circuitry disorders; thus, we tested whether BL facilitates fear extinction and inhibits fear acquisition. We randomly exposed 29 healthy humans to high- (9000 lx) or low-intensity light (<500 lx) for 15 min, near the nadir of the phase response to light, in a single-blind manner. Simultaneously with the light exposure, subjects performed fear extinction training and second f

Remodeling mechanisms and intervention strategies of the oral mucosal immune barrier.

The oral mucosa serves as the primary line of defense against exogenous pathogens and antigens, and its barrier function is critical for both local oral health and systemic well-being. The oral mucosal barrier forms a multilevel defense system consisting of physical, chemical, microbial, and cellular barriers, and its dynamic remodeling relies on an intricate network among epithelial cells, immune cells and the microbiota. Epithelial cells act as immune sentinels to coordinate immune responses, while innate and adaptive immune cells mediate immediate defense, precise regulation and immune memo

Mechanisms involved in corticotropin-releasing factor-induced excitation of entorhinal neurons and facilitation of glutamate release at the perforant pathway.

The entorhinal cortex (EC) is closely related to emotional control, consolidation and recall of memories, Alzheimer's disease, schizophrenia, and temporal lobe epilepsy. Corticotropin-releasing factor (CRF) is also implicated in these physiological functions and pathological disorders. Although both CRF peptide and CRF2 receptors are expressed in the EC, their roles and underlying cellular and molecular mechanisms in the EC have not been determined. Here, we found that activation of CRF2 receptors excited layer II principal neurons in the medial EC of rats and mice aged 18 to 28 days via eleva

Behavioral Consequences of Hippocampal 5-HT7 Receptors Blockade in Stressed Rats.

Serotonin (5-HT) has long been involved in response to stress and its effect may be, in part, mediated by 5-HT1a and 5-HT7 receptor subtypes in different brain structures. Both pre- and post-synaptic activation of 5-HT1a receptor, respectively, in the rat median raphe nucleus (MnRN) and hippocampus, lead to adaptation to acute inescapable stressors such as restraint and forced swim. 5-HT7 receptor (5HT7r), a stimulatory G-protein coupled receptor, has also been investigated as a possible candidate for mediating stress response. In the MnRN, activation of 5-HT7r has antidepressant effects, whil
