The role of regulatory peptides in the pathogenesis of cutaneous necrotizing vasculitis.

Cutaneous necrotizing vasculitides (CNV) are a group of clinical disorders characterized by: 1) angiocentric segmental inflammation, 2) cellular infiltrate, 3) red cell extravasation, 4) fibrinoid necrosis of the blood vessels, and possibly 5) nuclear debris. There are many classifications of vasculitides based or on the diameter of the vessels or on the type of the cellular infiltrate. In this paper we outline the major key role played by the endothelium in this pathological process and the possible role of the regulatory peptides in the pathogenesis of CNV. Despite there are not direct yet c

Histometry of splenic microvascular architecture in hairy cell leukaemia.

In this study we investigated the splenic microvascular architecture in hairy cell leukaemia, in order to provide a morphological basis for the haemodynamic modifications occurring in the disease. When compared with controls, the four leukaemic spleens examined showed a set of changes involving both the arterial and the venous system. A real increase in the absolute volume, surface and length of pulp arterial vessels was present. This increase was not so great as the enlargement of the spleen, thus resulting in a reduced density of distribution of arterial vessels in the infiltrated pulp. Enla

Reduced BCL2 level in astrocytes contributes to blood-brain barrier disruption in the striatum of offspring exposed to maternal preeclampsia.

Maternal preeclampsia (PE) contributes to neurodevelopmental disorders. However, the underlying molecular mechanism remains unclear. We investigated the potential biological regulatory mechanisms by establishing an animal model and primary cell culture. Data from the Adolescent Brain Cognitive Development human study revealed that the striatum mediates maternal PE and offspring behavioral disorders. In a rat PE model, offspring exhibited abnormal behaviors and reduced striatal vascular density, with disrupted blood-brain barrier (BBB) and swollen astrocytes. Primary astrocytes and brain microv

HIF-1α ameliorates myocardial apoptosis and promotes angiogenesis in diabetic myocardial infarction.

Diabetic hearts exhibit low hypoxia-inducible factor-1α (HIF-1α) activity, which aggravates cardiomyocyte apoptosis and impairs angiogenesis after infarction. Whether sustained HIF-1α expression can rescue these defects remains unclear. Here, a triple-point mutant that escapes von Hippel-Lindau (VHL)-mediated degradation (HIF-1α-Trip; P402A/P564A/N803A) was delivered to primary cardiomyocytes, cardiac microvascular endothelial cells (CMECs) and to streptozotocin-diabetic mice subjected to permanent left-anterior-descending (LAD) ligation. HIF-1α-Trip restored PI3K/Akt signaling, increased the

Multiple sevoflurane exposures in early development lead to long-term vascular abnormalities in the hippocampus.

Children who undergo multiple exposures to anesthesia for surgeries and procedures are at increased risk of developing neurocognitive impairments. Growing evidence suggests that anesthesia-induced neurotoxicity and acute damage to neural cells underlie these impairments. However, the mechanisms underlying the long-term adverse effects of early anesthetic exposure remain largely unknown. Here, we show that repeated early-life exposures to sevoflurane, a commonly used anesthetic in clinical practice, lead to long-term impairments in hippocampal vascular architecture, characterized by reduced vas

APOE-mediated sex differences in microvascular pathology and AD-associated proteinopathies in the medial temporal lobe.

INTRODUCTION: Cerebral small vessel disease (CSVD) contributes to the development of Alzheimer's disease (AD) dementia and co-occurs with AD-associated proteinopathies. However, how sex modulates the interaction between CSVD and AD-associated proteinopathies in the medial temporal lobe (MTL) remains unclear. METHODS: One hundred fifty-two autopsy cases from the Massachusetts Alzheimer's Disease Research Center were included. Deep-learning and semiquantitative scores were applied to MTL histological sections to obtain quantitative measures of proteinopathies and CSVD (cerebral amyloid angiopat

Extracellular vesicles derived from endothelial progenitor cells modified by Houshiheisan promote angiogenesis and attenuate cerebral ischemic injury via miR-126/PIK3R2.

Angiogenesis following cerebral ischemia is crucial for restoring blood supply to the ischemic region. Extracellular vesicles (EVs) derived from endothelial progenitor cells (EPCs) offer potential therapeutic benefits in the treatment of cerebral ischemia. Houshiheisan (HSHS) has been shown to improve clinical outcomes in ischemic stroke patients, reduce cerebral ischemic damage in rats, and protect endothelial cells. However, the potential effects of HSHS-modified EPC-derived EVs (EVsHSHS) for cerebral ischemia remain unexplored. This study investigated the impact of EVsHSHS on angiogenesis u

Intraoperative evaluation of local cerebral hemodynamic change by laser speckle contrast imaging for predicting postoperative cerebral hyperperfusion during STA-MCA bypass in adult patients with moyamoya disease.

Cerebral hyperperfusion (CHP) occurred frequently after direct superficial temporal artery-middle cerebral artery (STA-MCA) bypass surgery for moyamoya disease (MMD). We analyzed cortical microvascular density (CMD) and the change of cerebral blood flow (LΔCBF) using intraoperative laser speckle contrast imaging (LSCI) on 130 hemispheres of 95 consecutive adult patients with MMD. The demographic characteristics, cortical hemodynamic sources, bypass methods, intraoperative blood flow data, and relative CBF changes on single-photon emission computed tomography (SPECT) examination (SΔrCBF) were c…

Bone Metabolism Alterations in Systemic Sclerosis: An Insight into Bone Disease in SSc: From the Radiographic Findings to their Potential Pathogenesis and Outcome.

Previous research has shown conflicting reports about the effect of systemic sclerosis (SSc) on bone metabolism, especially considering bone mineral density (BMD), bone microarchitecture, and risk of fracture. The objective of this review is to analyze data from previous articles to investigate the differences in BMD and fracture risk between SSc and non-SSc populations and to discuss potential underlying mechanisms. The main factors investigated have been BMD (mean and standard deviation), t-scores and z-scores at the lumbar spine, femoral neck, and total hip measured by dual-energy X-ray abs

Attenuation of post-infarction remodeling in rats by sustained myocardial growth hormone administration.

Prevention of left ventricular remodeling is an important therapeutic target post-myocardial infarction. Experimentally, treatment with growth hormone (GH) is beneficial, but sustained local administration has not been thoroughly investigated. We studied 58 rats (322 ± 4 g). GH was administered via a biomaterial-scaffold, following in vitro and in vivo evaluation of degradation and drug-release curves. Treatment consisted of intra-myocardial injection of saline or alginate-hydrogel, with or without GH, 10 min after permanent coronary artery ligation. Echocardiographic and histologic remodeling

Three-Dimensional Maps of the Lenticulostriate Artery Territory.

BACKGROUND AND OBJECTIVES: Accurate knowledge of the cerebral vascular territories is foundational to stroke care. Yet, precise digital reference maps are not widely accessible, especially for subcortical structures. To address this shortcoming, we constructed 3-dimensional vascular territory maps of the MCA perforators-the lenticulostriate arteries (LSAs). METHODS: Nineteen LSA infarcts were demarcated on DWI scans and then normalized onto a standard 3-dimensional template brain. Normalized infarct volumes were then superimposed to create infarct density maps (heatmaps). RESULTS: LSA territ

Ginsenoside CK cooperates with bone mesenchymal stem cells to enhance angiogenesis post-stroke via GLUT1 and HIF-1α/VEGF pathway.

The transplantation of bone marrow mesenchymal stem cells (MSCs) in stroke is hindered by the restricted rates of survival and differentiation. Ginsenoside compound K (CK), is reported to have a neuroprotective effect and regulate energy metabolism. We applied CK to investigate if CK could promote the survival of MSCs and differentiation into brain microvascular endothelial-like cells (BMECs), thereby alleviating stroke symptoms. Therefore, transwell and middle cerebral artery occlusion (MCAO) models were used to mimic oxygen and glucose deprivation (OGD) in vitro and in vivo, respectively. Ou
