A syngeneic murine glioma model recapitulates key molecular, immunologic, and functional features of human glioblastoma to enable therapeutic development.

Glioblastoma (GBM) is the most common and deadly primary brain tumor in adults, characterized by diffuse infiltrative lesions, distinct oncogenic alterations, and a suppressive tumor immune microenvironment (TIME). These traits, however, are not fully recapitulated in current syngeneic murine glioma models, thereby limiting the pre-clinical development of novel therapies. Here we developed murine GBM models with patient-specific mutations that shared striking resemblance with human disease. Mosaic analysis with dual-recombinase mediated cassette exchange (MADR) and CRISPR-Cas9 were leveraged t

Exploring the Neurotherapeutic Potential of Bacillus-derived Macrolactins as NMDAR Antagonists for Alzheimer's Disease: Network Pharmacology and Molecular Modeling Insights.

INTRODUCTION: The N-methyl-D-aspartate receptor (NMDAR) plays a critical role in regulating excitatory glutamatergic neurotransmission and synaptic plasticity. However, excessive NMDAR activation can lead to increased calcium ion influx, resulting in excitotoxicity- a key contributor to neurodegenerative diseases. Although current NMDAR inhibitors exist, their clinical use is limited due to adverse effects. METHODS: This study employed computational screening of Bacillus-derived macrolactins to identify potential NMDAR antagonists. Molecular docking simulations were performed using AMDock v1.

Multiscale Numerical Modelling and Structural Design of Bulk Heterojunction Nanocomposites for Organic Photovoltaics: From Molecular Interfaces to Device Optimization.

Bulk heterojunction (BHJ) active layers in organic photovoltaics (OPVs) are nanostructured composites in which electron-donating and electron-accepting semiconductors form interpenetrating phases for exciton dissociation and charge transport. The power conversion efficiency (PCE) of these organic-organic nanocomposites is governed by structural features spanning multiple length scales: molecular packing and energy-level alignment at donor/acceptor (D/A) interfaces, phase-separation morphology and crystallite connectivity, and thin-film optical and charge-transport characteristics. Rational des

Imaging spatial transcriptomics reveals molecular patterns underlying accumulation of p-Ser129 α-synuclein in a transgenic mouse model.

In Parkinson's disease and dementia with Lewy bodies, aggregated and phosphorylated α-synuclein appears in select neurons throughout cortical and subcortical regions, but little is currently known about why certain populations are selectively vulnerable. Here, using imaging spatial transcriptomics (IST) coupled with downstream immunofluorescence for α-synuclein phosphorylated at Ser129 (pSyn) in the same tissue sections, we identified neuronal subtypes in the cortex and hippocampus of transgenic human α-synuclein-overexpressing mice that preferentially developed pSyn accumulation. Additionally

Computational profiling of vernomenin from Vernonia amygdalina via network pharmacology and molecular modeling: a preliminary assessment for fever targeting.

This study aimed to computationally evaluate the anti-fever potential of vernomenin from Vernonia amygdalina and its interactions with critical molecular targets. Network pharmacology was employed to screen bioactive compounds and identify fever-related targets (SIRT1, TP53, HDAC1), followed by molecular docking and 100-ns molecular dynamics (MD) simulations to assess binding affinities, interaction modes, and complex stability. Network analysis highlighted vernomenin as uniquely linked to all three targets, with the strongest binding affinity to SIRT1. MD simulations showed stable dynamics of

Identification of small-molecule TNF-α inhibitor candidates using machine learning-guided screening and multiscale molecular modelling.

Lumbar disc herniation (LDH) is a major cause of chronic low back pain, in which tumor necrosis factor-alpha (TNF-α) plays a central role in inflammation and pain signaling. While biologic TNF-α inhibitors have shown therapeutic benefit, their systemic administration, high cost, and limited penetration into the avascular disc environment restrict their clinical utility. In this study, we present a multiscale computational framework to identify small-molecule TNF-α inhibitor candidates. A curated dataset of experimentally validated TNF-α inhibitors was used to train supervised machine learning

Assessment of passive immunity transfer in dairy calves using integrated immunological and molecular approaches in a cow-calf dyad model.

BACKGROUND: In bovine species, the epitheliochorial placenta prevents prenatal transfer of immunoglobulins, rendering calves dependent on colostrum intake for passive immunity. Efficient passive immunity transfer (PIT) is critical for neonatal health, yet conventional assessments primarily focus on immunoglobulin quantification without evaluating structural integrity. OBJECTIVE: This pilot study aimed to comprehensively evaluate PIT in a cohort of Holstein calves using an integrated approach combining field-based diagnostic tests, quantitative protein assays, and molecular validation techniqu

Atmospheric Particulate Matter as a Carrier of Pb, Cd, and Ni: From Environmental Transfer and Bioaccessibility to Molecular Toxicity and Predictive Modeling.

Atmospheric particulate matter (PM) is a heterogeneous carrier of toxic metals whose environmental fate and biological effects depend on particle size, source-related composition, chemical form, solubility, and bioaccessibility. Lead (Pb), cadmium (Cd), and nickel (Ni) are of particular concern because atmospheric transport and deposition connect air pollution with persistent contamination of soils, vegetation, waters, sediments, food, and feed, followed by human and animal exposure. This review integrates evidence across a source-to-effect continuum encompassing emission, atmospheric transpor

Design, Synthesis, and Antimalarial Evaluation of Novel Quinazolin-4(3H)‑one Derivatives with Molecular Modeling Insights into Target Selectivity.

Malaria is a tropical disease caused by protozoa of the genus Plasmodium and is responsible for several deaths worldwide. Current therapies are limited by the high incidence of adverse effects and the rising prevalence of parasite drug resistance, underscoring the need for research into new chemical scaffolds that can overcome these limitations and for the identification of novel drug targets to advance antimalarial development. In this context, quinazolines show promising potential as new antimalarials. Therefore, in this study, a new series of quinazolin-4-(3H)-one analogs was synthesized an

Expression Profiling and Molecular Modeling Analysis of Cyp51C 14α-Demethylase Associated with Azole Resistance in Clinical Aspergillus flavus Isolates.

Invasive infections caused by Aspergillus flavus are more common in tropical and subtropical countries. The emergence of azole resistance in A. flavus complicates the management of aspergillosis, as azoles are the first-line and empirical therapy. The aim of this study was to investigate the molecular mechanisms underlying azole resistance in A. flavus, focusing on the cyp51C gene. We screened 34 molecularly confirmed A. flavus isolates obtained from patients with invasive aspergillosis for cyp51C gene expression by real-time RT-qPCR and for mutations by PCR sequencing. In silico identification and validation of vaccine and drug targets in Coccidioides posadasii through integrated genomic, proteomic, and molecular modeling approaches.

Coccidioides posadasii is a pathogenic fungus endemic to the arid regions of the Americas, causing coccidioidomycosis, a debilitating respiratory infection. Coccidioides infection is caused by breathing arthroconidia generated by mycelia that grow in desert soil. Arthroconidia develop inside the lung into massive, multinucleated spherules that are pathognomonic. Despite its significant impact on public health, there remains a paucity of research investigating novel therapeutic approaches to combat this disease. Addressing this research gap, our study employs a multi-faceted approach integratin

Unimodal vs Multimodal Learning: A Systematic Evaluation of Fusion Strategies and Model Design for Molecular Property Prediction and Uncertainty Quantification.

Accurate prediction of molecular properties is fundamental to environmental chemistry, yet remains challenging when experimental data are limited. Multimodal fusion provides a promising strategy for integrating complementary molecular representations; however, the relative contributions of molecular representation, fusion strategy, and learning algorithm to the predictive accuracy and uncertainty remain poorly understood. Five molecular modalities (RDKit descriptors, Mol2Vec embeddings, graph neural network embeddings, SMILES representations, and MS2 fragmentation spectra) were evaluated by us

Molecular insights, resistance profiling, and in silico structural modeling of extended-spectrum beta-lactamase-producing E. coli in camels.

The global surge of extended-spectrum beta-lactamase (ESBL)-producing Escherichia coli (E. coli) imposes an unnoticed but significant threat, fueling antimicrobial resistance (AMR). Being a neglected host, camels suffering from subclinical mastitis (SCM) caused by ESBL-producing E. coli present a crucial one-health hazard. The current study investigated the prevalence, associated risk factors, in silico structural modeling, and antimicrobial resistance profiling of ESBL-producing E. coli strains isolated from SCM in camels. A total of 384 milk samples were evaluated by the California mastitis

Identification of molecular subtypes and prognostic risk model of glucocorticoid-related lncRNAs in bladder cancer to evaluate prognosis and immunological characteristics.

BACKGROUND: Bladder cancer (BC) heterogeneity presents significant challenges in prognosis and personalized therapy. Long non-coding RNAs (lncRNAs) have been increasingly considered to be critical regulatory elements in tumor biology, and glucocorticoids exert complex effects on tumor biology. This investigation aimed to recognize glucocorticoid-related lncRNAs (GR-lncRNAs) and construct a prognostic signature for BC, elucidating their roles in subtype identification, prognosis, and immune characteristics. METHODS: We retrieved messenger RNA (mRNA) expression profiles, mutation information, a

Integrated analysis and single-cell sequencing of mitochondrial metabolism related gene molecular subtype and diagnostic model in ulcerative colitis.

Ulcerative colitis (UC) is a chronic inflammatory bowel disease that seriously affects the life expectancy of patients. Although increasingly sophisticated combinations of drugs can alleviate symptoms, 10-20% of patients still do not respond well. Therefore, it is necessary to further explore the pathogenesis and potential biomarkers of UC. Many clues have suggested the important value of mitochondrial metabolism in UC, but its role and related targets need to be further explored. By public database data, we identified differentially expressed mitochondrial metabolism related genes (MMRG) in U
