Multi-omics analysis and experimental verification reveal testicular fatty acid metabolism disorder in non-obstructive azoospermia.

Increasing evidence implicates disruptions in testicular fatty acid metabolism as a contributing factor in non-obstructive azoospermia (NOA), a severe form of male infertility. However, the precise mechanisms linking fatty acid metabolism to NOA pathogenesis have not yet been fully elucidated. Multi-omics analyses, including microarray analysis, single-cell RNA sequencing (scRNA-seq), and metabolomics, were utilized to investigate disruptions in fatty acid metabolism associated with NOA using data from public databases. Results identified ACSL6, ACSBG2, and OLAH as key genes linked to fatty ac

Endocrine disruptors and male infertility: multi-omics identification of key genes in non-obstructive azoospermia.

PURPOSE: Environmental endocrine-disrupting chemicals (EDCs) are increasingly implicated in male infertility, yet the gene-level mechanisms by which EDCs contribute to non-obstructive azoospermia (NOA) remain unclear. This study aimed to identify EDC-related genes that are causally linked to NOA and uncover their potential roles in reproductive dysfunction. METHODS: We integrated transcriptomic analysis of GEO datasets, EDC-associated gene data from the Comparative Toxicogenomics Database (CTD), and Mendelian randomization (MR) to prioritize candidate genes with causal relevance to male infer

Vaginal microecological characterization of women of childbearing age at different altitudes: a multi-omics exploration.

BACKGROUND: Reproductive tract diseases have become a serious public health problem threatening women's health. Vaginal microecological balance in women of childbearing age is essential for the maintenance of reproductive health, and different environmental conditions may have an impact on the composition of the vaginal microbial community and its metabolites. This is particularly true at different altitudes. Changes in environmental factors with increasing altitude may lead to vaginal microecological imbalances that increase the risk of reproductive tract infections and other gynecological di

Iron-lead mixed exposure causes bone damage in mice: A multi-omics analysis.

Excessive intake of essential and toxic metals affects the pathological process of osteoporosis. At present, the effects of single forms of iron (Fe), lead (Pb) and other metals on bone injury have been widely studied. However, these metal elements usually do not exist in the environment in a separate form. They are ingested in various ways and are often found together in the human body. However, the mechanism of bone damage caused by Fe and Pb mixed exposure is still unclear at this stage. At present, the combined analysis of multi-omics is the conventional method to explore the molecular mec

From molecular chaos to precision medicine in the treatment of cardiac hypertrophy: A rational use of natural products by integrating artificial intelligence and multi-omics data.

Given the intricate and multifactorial molecular mechanisms underlying cardiac hypertrophy (CH), conventional pharmacological treatments often face significant challenges in achieving disease reversal. This review presents an innovative approach to treating CH by combining natural products with artificial intelligence (AI) and multi-omics technologies. We provide detailed analysis of the molecular pathways that drive pathological hypertrophy, with a particular emphasis on impaired calcium signaling and the pro-inflammatory-oxidative feedback loop involving ferroptosis and pyroptosis. Human-like adrenal features in Chinese tree shrews revealed by multi-omics analysis of adrenal cell populations and steroid synthesis.

The Chinese tree shrew ( Tupaia belangeri chinensis) has emerged as a promising model for investigating adrenal steroid synthesis, but it is unclear whether the same cells produce steroid hormones and whether their production is regulated in the same way as in humans. Here, we comprehensively mapped the cell types and pathways of steroid metabolism in the adrenal gland of Chinese tree shrews using single-cell RNA sequencing, spatial transcriptome analysis, mass spectrometry, and immunohistochemistry. We compared the transcriptomes of various adrenal cell types across tree shrews, humans, macaq

A multi-omics dataset of the response to early plant polysaccharide ingestion in rabbits.

The transition from a milk-based diet to exclusive solid feeding deeply modifies microbiota-host crosstalk. Specifically, early ingestion of plant polysaccharides would be one of the main nutritional components to drive host-microbiota-interaction. To capture the effects of polysaccharides early-life nutrition (starch vs rapidly fermentable fiber) on the holobiont development, we investigated on the one hand the gut bacteriome and metabolome and on the other hand the transcriptome of two host gut tissues. Rabbit model was used to study post-natal co-development of the gut microbiota and its ho

Evidence Based on an Integrative Analysis of Multi-Omics Data on METTL7A as a Molecular Marker in Pan-Cancer.

Methyltransferase-like protein 7A (METTL7A), an RNA N6-methyladenosine (m6A) methyltransferase, has attracted much attention as it has been found to be closely associated with various types of tumorigenesis and progression. This study provides a comprehensive assessment of METTL7A from a pan-cancer perspective using multi-omics data. The gene ontology enrichment analysis of METTL7A-binding proteins revealed a close association with methylation and lipid metabolism. We then explored the expression of METTL7A in normal tissues, cell lines, different subtypes and cancers, and found that METTL7A w

A Review of Radiation-Induced Alterations of Multi-Omic Profiles, Radiation Injury Biomarkers, and Countermeasures.

Increasing utilization of nuclear power enhances the risks associated with industrial accidents, occupational hazards, and the threat of nuclear terrorism. Exposure to ionizing radiation interferes with genomic stability and gene expression resulting in the disruption of normal metabolic processes in cells and organs by inducing complex biological responses. Exposure to high-dose radiation causes acute radiation syndrome, which leads to hematopoietic, gastrointestinal, cerebrovascular, and many other organ-specific injuries. Altered genomic variations, gene expression, metabolite concentration

Innovative breakthroughs facilitated by single-cell multi-omics: manipulating natural killer cell functionality correlates with a novel subcategory of melanoma cells.

BACKGROUND: Melanoma is typically regarded as the most dangerous form of skin cancer. Although surgical removal of in situ lesions can be used to effectively treat metastatic disease, this condition is still difficult to cure. Melanoma cells are removed in great part due to the action of natural killer (NK) and T cells on the immune system. Still, not much is known about how the activity of NK cell-related pathways changes in melanoma tissue. Thus, we performed a single-cell multi-omics analysis on human melanoma cells in this study to explore the modulation of NK cell activity. MATERIALS AND

Single-cell multi-omics analysis identifies two distinct phenotypes of newly-onset microscopic polyangiitis.

The immunological basis of the clinical heterogeneity in autoimmune vasculitis remains poorly understood. In this study, we conduct single-cell transcriptome analyses on peripheral blood mononuclear cells (PBMCs) from newly-onset patients with microscopic polyangiitis (MPA). Increased proportions of activated CD14+ monocytes and CD14+ monocytes expressing interferon signature genes (ISGs) are distinctive features of MPA. Patient-specific analysis further classifies MPA into two groups. The MPA-MONO group is characterized by a high proportion of activated CD14+ monocytes, which persist before a

Glycosidase-Derived Arabinoxylan Hydrolyzates Attenuate Intestinal Inflammation via Restructuring the Gut Microbiota-Metabolite Axis.

The highly polymeric structure and complex branching of arabinoxylan (AX) limit its immunomodulatory potential. This study investigated whether enzymatic hydrolysis using xylanase (XYN) and α-l-arabinofuranosidase (ARF) could enhance anti-inflammatory efficacy in DSS-induced colitis. Synergistic ARF-XYN treatment, yielding low-polymerization and debranched oligosaccharides, exhibited superior efficacy in ameliorating colitis symptoms, suppressing pro-inflammatory cytokines (IL-6, TNF-α, IL-1β), and restoring intestinal barrier integrity compared to native AX. Multiomics analyses revealed that …

Genomic and Immune Landscape of Recurrent and/or Metastatic Squamous Cell Carcinoma of the Head and Neck Progressing on Anti-PD-1 Treatment.

Anti-PD-1 therapies improve survival in recurrent/metastatic (R/M) squamous cell carcinoma of the head and neck (SCCHN), but only a minority of patients achieve durable responses. The mechanisms driving resistance to anti-PD-1 in SCCHN remain poorly understood. Using the IMMUcan multiomics workflow, we characterized the molecular and immune profiles of R/M SCCHN progressing on anti-PD-1 treatment and compared them with an anti-PD-1-naïve cohort. Tumor biopsies from patients with anti-PD-1-resistant SCCHN exhibited significantly more EGFR and MYCL amplifications, along with increased MYC pathwa…
