An Escherichia coli Expressed Multi-Disulfide Bonded SARS-CoV-2 RBD Shows Native-like Biophysical Properties and Elicits Neutralizing Antisera in a Mouse Model.

A large-scale Escherichia coli (E. coli) production of the receptor-binding domain (RBD) of the SARS-CoV-2 could yield a versatile and low-cost antigen for a subunit vaccine. Appropriately folded antigens can potentially elicit the production of neutralizing antisera providing immune protection against the virus. However, E. coli expression using a standard protocol produces RBDs with aberrant disulfide bonds among the RBD's eight cysteines resulting in the expression of insoluble and non-native RBDs. Here, we evaluate whether E. coli expressing RBD can be used as an antigen candidate for a su

Differential antigen requirements for protection against systemic and intranasal vaccinia virus challenges in mice.

The development of a subunit vaccine for smallpox represents a potential strategy to avoid the safety concerns associated with replication-competent vaccinia virus. Preclinical studies to date with subunit smallpox vaccine candidates, however, have been limited by incomplete information regarding protective antigens and the requirement for multiple boost immunizations to afford protective immunity. Here we explore the protective efficacy of replication-incompetent, recombinant adenovirus serotype 35 (rAd35) vectors expressing the vaccinia virus intracellular mature virion (IMV) antigens A27L a

Immunopotentiating reconstituted influenza virosomes as a novel antigen delivery system.

Using immunopotentiating, reconstituted influenza virosomes (IRIV) as a delivery vehicle, a number of vaccines have been developed. In humans, IRIV-based vaccines containing hepatitis A and influenza antigens have been found to possess enhanced immunogenicity compared to alum-adsorbed vaccine for hepatitis A or commercial subunits or whole virion influenza vaccines. These vaccines were safe and did not engender any anti-phospholipid antibodies against the liposome components of the IRIV. Hepatitis B, tetanus toxoid and diphtheria toxoid, and nucleic acids have also been incorporated into IRIVs

Lipid nanoparticle-encapsulated DNA vaccine encoding African swine fever virus p54 antigen elicits robust immune responses in pigs.

African swine fever virus (ASFV) is one of the most significant viral pathogens affecting swine production worldwide. While several live attenuated ASF vaccines have been approved for clinical application in certain countries, there is a concern that the vaccine viruses might revert to virulence. Subunit vaccines containing one or a few viral immunogens provide a safer alternative. DNA plasmids are highly stable, easy to produce in large quantities at low cost, and safe for use in animals. However, unencapsulated DNA vaccines often exhibited low immunogenicity, largely due to the inefficient c

Baculovirus Vector-Based Varicella-Zoster Virus Vaccine as a Promising Alternative with Enhanced Safety and Therapeutic Functions.

Varicella-zoster virus (VZV) poses lifelong risks, causing varicella and herpes zoster (HZ, shingles). Currently, varicella and HZ vaccines are predominantly live attenuated vaccines or adjuvanted subunit vaccines utilizing VZV glycoprotein E (gE). Here, we propose our vaccine candidates involving a comparative analysis between recombinant baculoviral vector vaccines (AcHERV) and a live attenuated vaccine strain, vOka. AcHERV vaccine candidates were categorized into groups encoding gE only, VZV glycoprotein B (gB) only, or both gE and gB (gE-gB) as AcHERV-gE, AcHERV-gB, and AcHERV-gE-gB, respe

Vaccines against Tuberculosis: Where Are We Now?

Tuberculosis (TB) is among the top 10 leading causes of death in low-income countries. Statistically, TB kills more than 30,000 people each week and leads to more deaths than any other infectious disease, such as acquired immunodeficiency syndrome (AIDS) and malaria. TB treatment is largely dependent on BCG vaccination and impacted by the inefficacy of drugs, absence of advanced vaccines, misdiagnosis improper treatment, and social stigma. The BCG vaccine provides partial effectiveness in demographically distinct populations and the prevalence of multidrug-resistant (MDR) and extensively drug-

Polymerized porin as a novel delivery platform for coronavirus vaccine.

Coronavirus disease 2019 (COVID-19), caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), seriously threatens human life and health. The correct folding and polymerization of the receptor-binding domain (RBD) protein of coronavirus in Escherichia coli may reduce the cost of SARS-CoV-2 vaccines. In this study, we constructed this nanopore by using the principle of ClyA porin polymerization triggered by the cell membrane. We used surfactants to "pick" the ClyA-RBD nanopore from the bacterial outer membrane. More importantly, the polymerized RBD displayed on the ClyA-RBD polyme

Soil-transmitted helminthic vaccines: Where are we now?

It has been tested and proven that vaccination is still the best strategy to combat infectious diseases. However, to date, there are still no vaccines against human soil-transmitted helminthic diseases, despite their high prevalence globally, particularly in developing countries and rural areas with tropical climates and poor sanitation. The development of vaccines against helminths is riddled with obstacles. Helminths have a complex life cycle, multiple stages within the same host with stage-specific antigen expression, and the ability to regulate host immune reactions to evade the immune res

Advances in Infectious Disease Vaccine Adjuvants.

Vaccines are one of the most significant medical interventions in the fight against infectious diseases. Since their discovery by Edward Jenner in 1796, vaccines have reduced the worldwide transmission to eradication levels of infectious diseases, including smallpox, diphtheria, hepatitis, malaria, and influenza. However, the complexity of developing safe and effective vaccines remains a barrier for combating many more infectious diseases. Immune stimulants (or adjuvants) are an indispensable factor in vaccine development, especially for inactivated and subunit-based vaccines due to their decr

Immune Response to SARS-CoV-2 Vaccines.

COVID-19 vaccines have been developed to confer immunity against the SARS-CoV-2 infection. Prior to the pandemic of COVID-19 which started in March 2020, there was a well-established understanding about the structure and pathogenesis of previously known Coronaviruses from the SARS and MERS outbreaks. In addition to this, vaccines for various Coronaviruses were available for veterinary use. This knowledge supported the creation of various vaccine platforms for SARS-CoV-2. Before COVID-19 there are no reports of a vaccine being developed in under a year and no vaccine for preventing coronavirus

Mucosal Vaccination with Cyclic Dinucleotide Adjuvants Induces Effective T Cell Homing and IL-17-Dependent Protection against Mycobacterium tuberculosis Infection.

Tuberculosis consistently causes more deaths worldwide annually than any other single pathogen, making new effective vaccines an urgent priority for global public health. Among potential adjuvants, STING-activating cyclic dinucleotides (CDNs) uniquely stimulate a cytosolic sensing pathway activated only by pathogens. Recently, we demonstrated that a CDN-adjuvanted protein subunit vaccine robustly protects against tuberculosis infection in mice. In this study, we delineate the mechanistic basis underlying the efficacy of CDN vaccines for tuberculosis. CDN vaccines elicit CD4 T cells that home t

Vaccine technology: looking to the future.

The impact of vaccination on the health of the world's peoples has been considerable. With the possible exception of clean water, no other development has had such a major effect on the reduction of mortality and on population growth. During the last 200 years, vaccination has controlled nine major diseases and has led to the eradication of one, ie smallpox. However, in many instances the exact mechanisms of successful vaccines are not fully understood. Almost all of the vaccines in use today are of three types: live attenuated micro-organisms, inactivated whole micro-organisms, or split or su

Effects of three commercial vaccines against porcine parvovirus 1 in pregnant gilts.

Porcine parvovirosis is a common and important cause of reproductive failure in naïve dams. Even though vaccination is generally effective at preventing disease occurrence, the homology between the vaccine and challenge strains has been recently suggested to play a role in protection. Therefore, the purpose of this study was to evaluate and compare the efficacy of three currently available commercial vaccines against porcine parvovirus genotype 1 (PPV1) in an experimental model using pregnant gilts. Seventy-seven PPV1-negative gilts were included in the trial and randomly allocated to four gro

A VLP-Based Vaccine Candidate Protects Mice against Japanese Encephalitis Virus Infection.

Japanese encephalitis virus (JEV) is the leading cause of epidemic encephalitis in Asia, and vaccination is the most effective way to prevent JE. Although several licensed vaccines were widely used, there is still a demand for developing safer, cheaper, and more effective JE vaccines. In the current study, a virus-like particle (VLP) vaccine candidate containing the envelope structural protein of JEV expressed by the Pichia pastoris was assembled in vitro. It elicited a robust humoral and cellular immune response in mice model, conferring immunodeficient mice complete protection against lethal

Vaccination strategies against Zika virus.

The epidemic emergence of Zika virus (ZIKV) in 2015-2016 has been associated with congenital malformations and neurological sequela. Current efforts to develop a ZIKV vaccine build on technologies that successfully reduced infection or disease burden against closely related flaviviruses or other RNA viruses. Subunit-based (DNA plasmid and modified mRNA), viral vectored (adeno- and measles viruses) and inactivated viral vaccines are already advancing to clinical trials in humans after successful mouse and non-human primate studies. Among the greatest challenges for the rapid implementation of i

Expression and immunogenicity of novel subunit enterovirus 71 VP1 antigens.

Hand, foot, and mouth disease (HFMD) is a common viral illness in young children. HFMD is caused by viruses belonging to the enterovirus genus of the picornavirus family. Recently, enterovirus 71 (EV71) has emerged as a virulent agent for HFMD with severe clinical outcomes. In the current report, we conducted a pilot antigen engineering study to optimize the expression and immunogenicity of subunit VP1 antigen for the design of EV71 vaccines. DNA immunization was adopted as a simple technical approach to test different designs of VP1 antigens without the need to express VP1 protein in vitro fi

Type B hepatitis: a review of current prospects for a safe and effective vaccine.

Several subunit vaccines against hepatitis B virus (HBV) have been developed and either are being evaluated or soon will be evaluated for their ability to prevent HBV infection in humans. Most of these preparations consist of highly purified, 22-nm spherical particles of hepatitis B surface antigen (HBsAg) that have been extracted from the plasma of chronic carriers of HBV and inactivated with formalin. Extensive testing in humans and chimpanzees showed these vaccines to be free of residual HBV or other harmful agents and to be capable of stimulating the production of protective antibodies to

Pre-Clinical Development of Two DNA Vaccines Encoding MrkA of Klebsiella pneumoniae: Effect of Plasmid Backbone on Immunogenicity and Protective Efficacy Against Sepsis.

Development of vaccines against Klebsiella pneumoniae has potential to prevent infections in both community and hospital settings and reduce associated mortality and morbidity worldwide. DNA vaccines represent a promising approach given their stability, safety, design flexibility and cost-effectiveness for rapid production. In this study, two plasmid backbones, namely the commercial pVAX1 and the newly designed PLASMIVAX (containing additional CpG motifs), were tested to develop two DNA vaccines targeting the MrkA fimbrial subunit from K. pneumoniae. MrkA is highly conserved among K. pneumonia
