Correlation between insulin-like growth factor (IGF)-binding protein 5 and IGF-I gene expression during brain development.

Insulin-like growth factor (IGF)-binding proteins (IGFBPs) potently modulate the interactions of IGF-I and -II with the IGF-I receptor. Previous studies have shown that IGFBP2 gene expression is localized in astroglia, where it is anatomically and temporally coordinated with neuronal IGF-I expression during postnatal brain development. The present study shows that IGFBP5 gene expression is also highly abundant during brain development and also demonstrates significant spatiotemporal correlation with IGF-I, but exhibits a neuroanatomical distribution that is entirely distinct from IGFBP2. IGFBP

Cochlear Dead Regions in Sporadic Unilateral Vestibular Schwannomas Using the Threshold-Equalizing Noise Test.

BACKGROUND: Vestibular schwannoma (VS) is a benign intracranial neoplasm originating in the Schwann cells of the vestibular nerve. Despite its origin, the most common symptom is sensorineural hearing loss which is presented in more than 90% of patients. The underlying pathophysiology of this hearing loss has not been fully understood. OBJECTIVE: To assess the in vivo function of cochlear inner hair cells and spiral ganglion neurons in VS, cochlear dead regions (DRs) were evaluated via the threshold-equalizing noise (TEN) test in untreated VS patients. METHOD: Untreated patients diagnosed wit

Structural basis of ligand recognition and gating in a heteromeric Deg-3/Des-2 nicotinic acetylcholine receptor.

During evolution, nicotinic acetylcholine receptors (nAChRs) have diversified in subunit composition and ligand selectivity, enabling a conserved ion channel scaffold to support a broad range of signaling functions beyond classical synaptic transmission. Deg-3/Des-2 is a nematode-specific, calcium-permeable heteromeric α-type nicotinic receptor in sensory neurons to mediate chemosensation, nociception, and mechanotransduction, and represents a promising anthelmintic target. Here, we report cryo-electron microscopy structures of Deg-3/Des-2 in its apo, agonist-bound intermediate and agonist-bou

[The role of the inner ear biological barrier in targeted drug delivery for inner ear diseases].

The endolymphatic barrier, composed of tight junctions and ion transporters, regulates the ionic gradient between endolymph and perilymph and plays a pivotal role in maintaining inner ear function. Disruption of this biological barrier can result in ionic imbalance, causing both auditory and vestibular dysfunction. Medication needs to enter the perilymph instead of the endolymph by passing through the perilymphatic barrier to reach the hair cells. Targeted drug delivery has emerged as a promising therapeutic strategy for inner ear diseases, including intratympanic injection, intracochlear infu

Expression and localization of α2A-adrenergic receptor in the rat post-natal developing cochlea.

Lots of adrenergic receptors (ARs) are widely present across the auditory pathways and are positioned to affect auditory and vestibular functions. However, noradrenergic regulation in the cochlea has not been well characterized. In this study, a rat model of noise-induced hearing loss was developed to investigate the expression of α2A-adrenergic receptor (AR) after acoustic trauma, then, we investigated the expression of α2A-AR in the developing rat cochlea using immunofluorescence, qRT-PCR, and Western blotting. We found that the expression of α2A-AR significantly increased in rats exposed to

Developmental alterations of the auditory brainstem centers--pathogenetic implications in Sudden Infant Death Syndrome.

Sudden Infant Death Syndrome (SIDS), despite the success of campaigns to reduce its risks, is the leading cause of infant death in the Western world. Even though the pathogenesis remains unexplained, brainstem abnormalities of the neuronal network that mediates breathing and protective responses to asphyxia, particularly in the arousal phase from sleep, are believed to play a fundamental role. This is the first study to identify, in SIDS, developmental defects of specific brainstem centers involved in hearing pathways, particularly in the cochlear and vestibular nuclei, in the superior olivary

Chronic inhalation of rotenone or paraquat does not induce Parkinson's disease symptoms in mice or rats.

Epidemiological studies suggest that some pesticides might constitute a risk factor for Parkinson's disease (PD). However, risk assessment cannot be performed in the current experimental animal models because they use non-natural pathways of pesticide exposure, such as intraperitoneal or intravenous injection, that might bypass body defences. A new model based on daily inoculation of neurotoxins in the nasal cavity of C57BL/6 mice for 30 days was used to evaluate risk of three complex I inhibitors, 1-methyl-4-phenyl1,2,3,6-tetrahydropyridine (MPTP), rotenone and paraquat. These compounds displ

Vulvodynia: still a dramatically neglected condition. A position statement on pathogenetic mechanisms from the Italian Society of Andrology and Sexual Medicine (SIAMS).

PURPOSE: 8–10% of women experience vulvar pain. We aim to summarize the current literature on pathogenetic mechanisms of vulvodynia, and highlight its invisible nature, that often results in misdiagnosis, prolonged diagnostic process and/or inappropriate care. METHODS: A comprehensive search was conducted on the main medical databases, including studies up to April 30, 2025. The statements were internally discussed, and levels of evidence were provided according to the Oxford 2011 LoE criteria. RESULTS: The panel highlights the multifactorial and complex systems underlying vulvodynia, advocati

Skull Vibration Induced afternystagmus: A new clinical indicator of superior canal dehiscence.

Vibrations applied to the cranium induce, in 55%-92% of patients with CT-verified superior canal dehiscence (SCD), a perstimulatory nystagmus most often ipsilaterally beating when the vertex location is stimulated at 100Hz. The skull vibration-induced nystagmus (SVIN) test is a bone-conducted (BC) Tullio phenomenon in patients with a 3rd mobile window syndrome (TMWS). Here, we describe the technical cues and procedures to obtain an afternystagmus regularly observed in 55% of a series of 43 patients with SCD chosen as examples and stimulated at 100Hz and higher frequencies by bone-conducted vib

Sequential sparse autoencoder for dynamic heading representation in ventral intraparietal area.

To navigate in space, it is important to predict headings in real-time from neural responses in the brain to vestibular and visual signals, and the ventral intraparietal area (VIP) is one of the critical brain areas. However, it remains unexplored in the population level how the heading perception is represented in VIP. And there are no commonly used methods suitable for decoding the headings from the population responses in VIP, given the large spatiotemporal dynamics and heterogeneity in the neural responses. Here, responses were recorded from 210 VIP neurons in three rhesus monkeys when the

Hearing loss and vestibular schwannoma: new insights into Schwann cells implication.

Hearing loss (HL) is the most common and heterogeneous disorder of the sensory system, with a large morbidity in the worldwide population. Among cells of the acoustic nerve (VIII cranial nerve), in the cochlea are present the hair cells, the spiral ganglion neurons, the glia-like supporting cells, and the Schwann cells (SCs), which alterations have been considered cause of HL. Notably, a benign SC-derived tumor of the acoustic nerve, named vestibular schwannoma (VS), has been indicated as cause of HL. Importantly, SCs are the main glial cells ensheathing axons and forming myelin in the periphe

Endocrine implications of neurofibromatosis 1 in childhood.

In 1882, von Recklinghausen described a group of patients with multiple tumors arising from the 'endoneurium' of peripheral nerves, and called them 'neurofibromas'. The term von Recklinghausen disease was used up to the end of the 20th century, when the gene of neurofibromatosis (NF1) was cloned on chromosome 17q11.2. The gene product is a cytoplasmic protein termed neurofibromin, regulating proliferation and maturation of both glial and neuronal progenitors during embryogenesis. Loss of neurofibromin function determines the hyperactivation of the proto-oncogene RAS, leading to an increased ri

Cervico-oculo-Acoustic syndrome in a male with consanguineous parents.

BACKGROUND: The cervico-oculo-acoustic syndrome comprises Klippel-Feil anomaly, sensorineural deafness and Duane's retraction syndrome. Polygenic, autosomal dominant, and X-linked inheritance have been hypothesized. The disorder has rarely been reported in males. CASE REPORT: A 42-year-old male, born of consanguineous parents, presented with Duane's syndrome, mixed hearing loss, C2-C3 fusion, neck stiffness, and right facial palsy. A variety of cardiac, neurological and urogenital anomalies occurred in his relatives. The electro-oculographic studies showed impaired abduction and adduction of

Familial dysautonomia (Riley-Day syndrome). Temporal bone findings and otolaryngological manifestations.

Familial dysautonomia, or Riley-Day syndrome, is inherited in an autosomal recessive fashion and occurs almost exclusively in Jewish families. This disorder is characterized by a smooth tongue devoid of fungiform papillae and of taste buds, and is clinically associated with poor taste discrimination. An unsteady gait and dizziness on change in position are also common presenting symptoms. This study reports the histopathological findings of eight temporal bones from four patients with documented familial dysautonomia. For the control series, 13 normal temporal bones were also studied. The most

Neurotologic Findings of Idiopathic Acute Unilateral Audiovestibulopathy.

BACKGROUND AND PURPOSE: Unlike acute unilateral vestibulopathy or vestibular neuritis (AUVP/VN), of which the diagnosis is made based on well-established neurotologic findings, the diagnosis of idiopathic acute unilateral audiovestibulopathy (iAUAV, also known as labyrinthitis) still requires further elucidation. METHODS: We retrospectively reviewed the medical records of patients with first-onset acute audiovestibulopathy (AAVS) in a referral-based university hospital in South Korea between March 2018 and September 2024. The results of video-oculography, video head-impulse, and other neuroto

Dominant foot could affect the postural control in vestibular neuritis perceived by dynamic body balance.

During attacks of vestibular neuritis (VN), patients typically lose postural balance, with resultant postural inclination, gait deviation toward the lesion side, and tendency to fall. In this study, we examined and analyzed static and dynamic postural control during attacks of VN to characterize differences in postural control between right and left VN. Subjects were patients diagnosed with VN at the Department of Otolaryngology, Toho University Sakura Medical Center, and underwent in-patient treatment. Twenty-five patients who had spontaneous nystagmus were assessed within 3days after the ons

Head position and increased head velocity to optimize video head impulse test sensitivity.

This study investigated the effects of head position on gain values during video head impulse tests (vHITs). Different head positions were used for vHIT of the horizontal semicircular canals of 20 healthy controls and 18 patients with unilateral vestibular loss (UVL), with head velocities ranging from 150°/s to 200°/s. Differences in vestibulo-ocular reflex gain in the control and patient groups according to head position (0° and 30° downward pitch) were analyzed. In the unaffected control group, the 30° pitched-down position resulted in a mean gain increase of up to 1.0 in both ears (right ea

Video head impulse testing (VHIT) in the pediatric population.

OBJECTIVES: VHIT is the first vestibular test to evaluate all six semicircular canals. This test has advantages over the rotary chair and caloric tests in evaluating children since it does not require fear-inducing darkness or provocation of dizziness. The goal of this study was to review our initial experience with VHIT in children and adolescents at a pediatric vestibular program. METHODS: Results using the ICS Impulse VHIT device in 33 patients <20 years of age were retrospectively reviewed. Rotary chair testing was used to designate 26 subjects into groups with normal (n=20) and abnormal

Use of mammalian inner ear antigens for the diagnosis of autoimmune sudden loss of vestibular function.

It has been postulated that bilateral sensorineural hearing loss (SNHL) may be the result of an ongoing autoimmune process against the inner ear and a pattern of progressive bilateral SNHL linked to an autoimmune inner ear disorder has been reported. Various attempts have been made to develop an assay to confirm the diagnosis of autoimmune inner ear disease. In this study we used a Western blot assay to determine the presence of IgG antibodies directed against a PO antigen (30 kDa) of the guinea pig in the sera of patients affected by sudden loss of vestibular function (SLVF). Ten patients aff
