Aromatic Interaction-Driven Adaptive Transformation of Macrocyclic Unimolecular Chloride Channels Into Nanopores.

Biological ion channels and pores frequently exhibit dynamic, concentration-dependent adaptive assembly, yet synthetic systems that faithfully replicate this intriguing behavior remain rare. In this study, we present a novel class of tripeptide-appended macrocyclic ion channels that undergo aromatic interaction-driven adaptive transformation from unimolecular chloride channels into higher-order nanopores. At low concentrations, analogues with aromatic tripeptide-functionalized macrocycles (MC-F and MC-W) function as discrete and efficient chloride channels. Upon increasing the concentration, c

Revealing global stoichiometry conservation architecture in cells from Raman spectral patterns.

Cells can adapt to various environments by changing their biomolecular profiles while maintaining physiological homeostasis. What organizational principles in cells enable the simultaneous realization of adaptability and homeostasis? To address this question, we measure Raman scattering light from Escherichia coli cells under diverse conditions, whose spectral patterns convey their comprehensive molecular composition. We reveal that dimension-reduced Raman spectra can predict condition-dependent proteome profiles. Quantitative analysis of the Raman-proteome correspondence characterizes a low-d

Skin adaptation in lower limb amputees assessed through Raman spectroscopy and mechanical characterization.

Following lower limb amputation residuum skin from the lower leg is used to reconstruct the residual limb. Unlike skin on the sole of the foot (plantar skin), leg skin is not inherently load bearing. Despite this, leg skin is required to be load bearing in the prosthetic socket. Current hypotheses propose that lower limb amputee skin can adapt to become load bearing with repeated prosthesis use. Here, we show using confocal Raman spectroscopy, mechanical characterization and cytokine analysis that adaptations occur which actually result in impaired barrier function, higher baseline inflammatio

Gene Duplication and Loss of AANAT in Mammals Driven by Rhythmic Adaptations.

Arylalkylamine N-acetyltransferase (AANAT) plays a crucial role in synchronizing internal biological functions to circadian and circannual changes. Generally speaking, only one copy of AANAT gene has been found in mammals, however, three independent duplications of this gene were detected in several cetartiodactyl lineages (i.e., Suidae, Hippopotamidae, and Pecora), which originated in the middle Eocene, a geological period characterized with the increased climate seasonality. Lineage-specific expansions of AANAT and the associated functional enhancement in these lineages strongly suggest an i

Adaptative mechanism of the equilibrative nucleoside transporter 1 (ENT-1) and blood adenosine levels in elite freedivers.

PURPOSE: Long static or intense dynamic apnoea-like high-altitude exposure is inducing hypoxia. Adenosine is known to participate to the adaptive response to hypoxia leading to the control of heart rate, blood pressure and vasodilation. Extracellular adenosine level is controlled through the equilibrative nucleoside transporter 1 (ENT-1) and the enzyme adenosine deaminase (ADA). The aim of this study was to determine the control of adenosine blood level (ABL) via ENT-1 and ADA during apnoea-induced hypoxia in elite freedivers was similar to high-altitude adaptation. METHODS: Ten freediver cha

Whipworm kinomes reflect a unique biology and adaptation to the host animal.

Roundworms belong to a diverse phylum (Nematoda) which is comprised of many parasitic species including whipworms (genus Trichuris). These worms have adapted to a biological niche within the host and exhibit unique morphological characteristics compared with other nematodes. Although these adaptations are known, the underlying molecular mechanisms remain elusive. The availability of genomes and transcriptomes of some whipworms now enables detailed studies of their molecular biology. Here, we defined and curated the full complement of an important class of enzymes, the protein kinases (kinomes)

Essential Roles of Toll-Like Receptors in Atherosclerosis.

Atherosclerosis is driven by inflammation with an involvement of innate and adaptive immune responses. Toll-like receptors, the well-defined pattern recognition receptors of the immune system, play a central role in macrophage activation. Toll-like receptors recognize pathogen-associated molecular patterns expressed by a wide range of infectious agents and provide a strong link between local innate and adaptive immunity. Activation of these receptors triggers an intracellular signaling cascade mediated through myeloid differentiation factor 88 or toll/interleukin-1 receptor-domain-containing a

Pathogenesis of systemic inflammatory diseases in childhood: "Lessons from clinical trials of anti-cytokine monoclonal antibodies for Kawasaki disease, systemic onset juvenile idiopathic arthritis, and cryopyrin-associated periodic fever syndrome".

Inflammation has often been considered to be a nonspecific response and to play a bridging role in the activation of adaptive immunity. However, it is now accepted that inflammation is the product of an independent innate immune system closely linked to the adaptive immune system. The key mediators of inflammation are inflammatory cytokines, as determined by multiple lines of evidence both in vitro and in vivo. Due to the crucial role of inflammatory cytokines in the pathogenesis of autoimmune disorders, anti-cytokine treatment has been developed as a therapy for rheumatoid arthritis, juvenile

Hypoxic adaptation theory of cancer.

For more than 70 years, the somatic mutation theory (SMT) has dominated cancer biology, conceptualizing carcinogenesis as the cumulative consequence of genetic mutations, However, expanding molecular and microenvironmental evidence reveals important limitations in this mutation-centric framework. The Hypoxic Adaptation Theory (HAT) reframes carcinogenesis not as a purely mutation-driven process, but as the maladaptive culmination of chronic cellular hypoxia. HAT integrates with SMT by situating mutagenesis within a microenvironmental and evolutionary context rather than opposing it, positionin

Protein lactylation in health and diseases: molecular mechanisms, biological significance, and clinical implications.

Protein lysine lactylation (Kla) is a newly identified post-translational modification (PTM), in which lactyl groups are transferred to specific lysine residues in proteins. As a crucial intermediary between cellular metabolism and epigenetic regulation, Kla immensely increases the functional diversity of the proteome. This intriguing modification extends beyond histones to non-histone proteins, signaling molecules, enzymes, and substrates. In addition to enzymatic L-lactylation utilizing lactate as a lactyl donor and involving enzymes including writers (lactyltransferases), readers (lactylati

Barrier-matched, pharmacodynamic (PD)-gated immunotherapy development in MSS/pMMR colorectal cancer.

Immune checkpoint inhibitors (ICIs) have transformed treatment for deficient mismatch repair/high microsatellite instability (dMMR/MSI-H) colorectal cancer (CRC), but durable benefit remains uncommon in microsatellite-stable/proficient mismatch repair (MSS/pMMR) disease. Negative combination trials may reflect heterogeneous resistance programs tested without adequate biological stratification or prospective pharmacodynamic verification. This narrative and critical Review examines MSS/pMMR CRC through tissue-level immune-desert, immune-excluded, and immune-suppressed contexts, with active liver

Mapping the inter- and intra-genic codon-usage landscape in Homo sapiens.

Although the genetic code is degenerate, codon selection is nonrandom and reflects significant functional constraints. Codon-usage bias (CUB) acts as a layer of post-transcriptional regulation, influencing messenger RNA (mRNA) stability, translation kinetics, and co-translational protein folding. While CUB is well-characterized in unicellular organisms, its regulatory scope and functional consequences in humans remain complex and less defined. Our study offers a comprehensive evaluation of human codon usage. We report that genes exhibiting the strongest codon bias are enriched in high-stoichio

Weight regain after obesity treatment: mechanisms, predictors, and real-world evidence from the IGOBE program.

Weight regain is the defining challenge of obesity treatment and remains insufficiently addressed in current clinical paradigms. Despite substantial initial weight loss achieved through lifestyle, pharmacological, or surgical interventions, most individuals experience partial or complete regain over time, reflecting powerful and persistent biological adaptations that defend body weight. These include reductions in energy expenditure, sustained hormonal changes that promote appetite, and neurobehavioral mechanisms that favor increased energy intake, all of which are further modulated by genetic

System-Level Identification of Heat Stress-Responsive Pathways in Human Saliva.

Environmental heat stress disrupts cellular homeostasis through coordinated molecular responses involving protein quality control, oxidative stress regulation, inflammatory signaling, and water homeostasis. Although saliva plays an essential role in maintaining oral homeostasis, the molecular pathways underlying salivary adaptation to heat stress remain insufficiently characterized. We hypothesized that integrating the human salivary proteome with systems biology approaches would identify a reproducible heat-stress-responsive molecular signature. Human salivary proteomic datasets retrieved fro

Systemic Degree of Perturbation of Plasma Markers Reveals Cumulative Biological Stress Across the Competitive Season in Professional Soccer Players.

BACKGROUND: Fixture congestion has emerged as one of the defining challenges of modern professional soccer, exposing players to repeated high-intensity demands with limited opportunities for recovery. Such cumulative exposure taxes multiple physiological systems and may lead to sustained biological strain throughout the competitive season. However, the integrated systemic impact of congested schedules remains incompletely characterized. OBJECTIVE: We aimed to characterize the systemic physiological impact of cumulative competitive load across a full professional soccer season using an integra

Brain O-GlcNAcylation in Neurodegenerative Diseases: Context-Dependent Mechanisms and Precision Therapeutic Translation.

O-linked β-N-acetylglucosamine modification (O-GlcNAcylation) is a dynamic, nutrient-sensitive post-translational modification that couples hexosamine biosynthesis pathway flux to protein function in neurons and glia. This reversible cycling, catalyzed by O-GlcNAc transferase (OGT) and O-GlcNAcase (OGA), integrates glucose, glutamine, acetyl-CoA, and nucleotide metabolism with synaptic activity, mitochondrial adaptation, transcriptional regulation, proteostasis, and neuroimmune signaling. Dysregulated O-GlcNAc cycling has been implicated in major neurodegenerative diseases, including Alzheimer

Perceptions of Infectability to Disease Moderate the Association between Daily Concerns about Contracting COVID-19 and Satisfaction with Sex.

The behavioral immune system is an evolved adaptation comprised of automatic behavioral, cognitive, and affective reactions that has allowed humans throughout evolutionary history to avoid situations that risk infection by pathogens (e.g., physical proximity to sick people). Although behavioral immune system activation may be functional by helping people avoid such situations, experiencing these automatic reactions during sexual interactions may undermine people's evaluations of those interactions because sex requires close physical contact. We examined whether two sources of behavioral immune
