Surface-enhanced Raman scattering-based lateral flow immunoassays (SERS-LFIAs) have emerged as a promising platform for rapid food safety analysis. However, their quantitative reliability is often compromised by matrix-dependent variations during lateral flow, and the influence of probe size on analytical performance remains insufficiently understood. In this work, Au@DTNB immunoprobes with diameters of 15, 30, and 50 nm were prepared to systematically investigate the effect of nanoparticle size on the quantitative performance of SERS-LFIA for aflatoxin B1 (AFB1) detection. All three probe siz

Babassu (Orbignya phalerata) plays a crucial role in several indigenous and quilombo communities in the north and northeast regions of Brazil. Mechanical pressing of nuts yields babassu oil of interest to the cosmetic and pharmaceutical industries. However, the solid fraction from pressing (babassu press cake, BPC) has rarely been explored, thereby devaluing its potential for application in the food industry. Therefore, this study comprehensively characterized BPC, extracted residual lipids with hexane and ethanol, and characterized the extracted babassu oils (BO.H and BO.E) and the correspond

Aflatoxin B1 (AFB1), a common contaminant in broiler feed, impairs hepatic function and poses a significant threat to the poultry industry by disrupting cellular autophagy. Ferulic acid (FA) is a naturally occurring phenolic compound with antioxidant, anti-inflammatory, and autophagy-modulating properties. However, whether FA alleviates AFB1-induced autophagy impairment in broiler liver remains unclear. In this study, an AFB1-induced liver injury model was established. A series of experiments were conducted to evaluate the effects of AFB1 and FA on autophagy-related processes. The results show

Hepatocellular carcinoma (HCC) represents a global public health burden of increasing magnitude, driven by risk factors including viral hepatitis, metabolic dysfunction-associated fatty liver disease and various environmental carcinogens, which may inactivate key tumor suppressor genes or activates oncogenes. Recent studies have linked various environmental exposures with inactivation of tumor suppressor gene CDKN2A (cyclin dependent kinase inhibitor 2A) and its encoded proteins p16INK4a and p14ARF to various molecular pathways leading to hepatocarcinogenesis. Current studies suggest p16INK4a

Mycotoxins pose a major risk to food safety and human health, yet their hepatotoxic mechanisms remain incompletely characterized due to limitations in conventional in vitro models. In this study, we systematically compared mycotoxin-induced hepatotoxicity and metabolomic profiling across two human hepatic models cultured under 2D monolayer and 3D spheroid conditions. The various mycotoxins (Aflatoxin B1, Citrinin, Deoxynivalenol, Ochratoxin A, Patulin, and Zearalenone) exhibit distinct metabolic signatures, thereby serving as an appropriate panel for comprehensively evaluating diverse hepatoto

Galactomannan comes from a wide range of plant resources and has some biological activities, but its bioavailability is limited due to its large molecular weight and complex structure. In this study, three degradation methods (H2O2, ultrasound, and β-mannanase) combined with ethanol fractional precipitation (25 %, 50 %, and 75 %) were used to degrade and separate Gleditsia sinensis galactomannans (GSG), and the physicochemical properties and biological activities of GSG after degradation were analyzed. Comprehensive comparison indicates that H2O2 exhibits had a better degradation effect. After

Metabolic disruption encompasses the processes leading to adverse effects to major metabolic organs, such as liver and pancreas after exposure to e.g., environmental chemicals. As some mycotoxins act as endocrine disruptors, their structural similarity may lead to effects in lipid and glucose metabolism as well. Via systematic literature search, we mapped the potential of mycotoxins to cause metabolic disruption. Our systematic data search involved mycotoxin keywords combined with metabolic disruption keywords. The retrieved 31 studies revealed 24 in vivo studies, and 18 in vitro studies in to

The use of a protein-based electrochemical sensor for rapid and sensitive detection of ochratoxin (OTA) is explored. We have employed a combinatorial approach involving in silico target fishing and molecular docking analysis to identify the protein candidates with high affinity for OTA. Porcine pancreatic α-amylase has emerged as a promising candidate. α-Amylase was immobilized on multi-walled carbon nanotube (MWCNT) and amine modified titanium dioxide nanoparticles (TNP-NH2) nanocomposite coated glassy carbon electrode (GCE) (α-amylase/NH2-TNP + MWCNT/GCE) via inverse NHS-EDC chemistry within

Mycotoxin presence in meat products is principally due to the development of toxigenic moulds during ripening, which concretely affect the dry-cured ones. Ochratoxin A (OTA) is the most prevalent mycotoxin in dry-cured hams and fermented sausages. Apart from collecting information on the main toxigenic moulds in dry-cured meat products, the objective of this review is to compile data about the mycotoxin prevalence and the tools for their mitigation. Although no very extensive studies have been performed on the mycotoxin occurrence in such products, much research has been conducted over the las

SCOPE: Mycotoxins co-contamination of agricultural products poses a serious threat to human and animal health, especially hepatic dysfunction. Zearalenone (ZEN), deoxynivalenol (DON), and aflatoxin B1 (AFB1) are three commonly co-occurring mycotoxins. This study is to determine whether lycopene (LYC) can alleviate hepatic toxicity induced by the co-occurrence of ZEN, DON, and AFB1 in mice.
METHODS AND RESULTS: Eighty 6-week-old male ICR mice are divided into four groups: CON group (solvent control), LYC group (10 mg kg-1 LYC), Co-M group (10 mg kg-1 ZEN + 1 mg kg-1 DON + 0.5 mg kg-1 AFB1), an

Aptamers are promising affinity ligands with considerable applications, such as biosensors, disease diagnosis, therapy, etc. Characterization of aptamer-target binding is important in aptamer selection and aptamer applications. Microscale thermophoresis (MST) is an emerging optical technique for molecular interactions, which monitors fluorescence responses of fluorescent molecules in a microscopic temperature gradient. Harnessing merits in trace sample consumption, high speed, free separation, free immobilization, and ratiometric analysis, MST draws intense wide attention. MST is often applied

Kwashiorkor and marasmus are two clinical syndromes observed in severe acute malnutrition. In this review, we highlighted the differences between these two syndromes by reviewing the data comparing kwashiorkor and marasmus in literature, combined with recent microbiological findings and meta-analysis. Depletion of antioxidants, vitamins and minerals were more severe in kwashiorkor than marasmus. This was consistent with the severe and uncontrolled oxidative stress associated with the depletion of gut anaerobes and the relative proliferation of aerotolerant gut pathogens. This relative prolifer

Oxidative stress is an important molecular mechanism for kidney injury in aflatoxin B1 (AFB1) nephrotoxicity. Nuclear factor erythroid 2-related factor 2 (Nrf2) is a master transcription factor for regulating the cellular oxidative stress response, which has been confirmed in animal models. Lycopene (LYC), a natural carotenoid, has received extensive attention due to its antioxidant effect with the activation of Nrf2. However, the role of LYC in protecting against AFB1-induced renal injury is unknown. To evaluate the chemoprotective effect of LYC on AFB1-induced renal injury, forty-eight male

Hepatocellular carcinoma (HCC) is the most frequent primary liver carcinoma, accounting for about 80% of cases. In spite of advances in modern oncology, this neoplasia still holds the second place in overall cancer mortality. HCC is a multifactor disease: it results from accumulated oncogenic potentials made up of several groups of risk factors, the most significant of which is an infection with hepatotropic viruses. The hepatitis C virus (HCV) is one of the primary causes of morbidity and mortality across the world and affects 1.1% of worldwide population. It has been calculated that on avera

Aspergillus flavus is the second most leading cause of aspergillosis. The ability of A. flavus to adapt within the host environment is crtical for its colonization. Onset of germination of conidia is one of the crucial events; thus, in order to gain insight into A. flavus molecular adaptation while germination, protein profile of A. flavus was obtained. Approximately 82 % of conidia showed germination at 7 h; thus, samples were collected followed by protein extraction and subjected to nLC-Q-TOF mass spectrometer. Q-TOF data were analysed using Protein Lynx Global Services (PLGS 2.2.5) software

The use of integrated approaches in genetic toxicology, including the incorporation of gene expression data to determine the molecular pathways involved in the response, is becoming more common. In a companion article, a genomic biomarker was developed in human TK6 cells to classify chemicals as genotoxic or nongenotoxic. Because TK6 cells are not metabolically competent, we set out to broaden the utility of the biomarker for use with chemicals requiring metabolic activation. Specifically, chemical exposures were conducted in the presence of rat liver S9. The ability of the biomarker to classi

Clay feed additives have been increasingly incorporated into animal diets to prevent aflatoxicosis. Due to the nonselective nature of the binding interaction, many important components of the diets could also be made unavailable because of these feed additives. The anticoccidial monensin (MON) could also be sequestered by these clays. The use of sodium bentonite (Na-B) from a mine in the province of Mendoza, Argentina, was investigated as a sequestering agent to prevent the effects of 100 µg/kg of dietary aflatoxin B(1) (AFB(1)). In vitro studies demonstrated that the above Na-B was a good can

Detection and analysis of DNA adducts resulting from endogenous or exogenous exposures to carcinogens are essential not only for quantifying biologically effective doses but also for establishing relationships between exposure and cancer risk. We have developed and validated a procedure of high sensitivity and specificity based on fluorescence labeling of DNA adducts combined with high-performance liquid chromatography-laser-induced fluorescence detection. The fluorescent dye 4,4-difluoro-5,7-dimethyl-4-bora-3a,4a-diaza-s-indacene-3-propionic acid (BODIPY FL) was used to label the deoxynucleos

Chemoprevention, pharmacological intervention for disease prevention, aims to intervene in pathways that lead to clinical disease before the disease occurs. Cancer chemoprevention is a relatively new field, but for gastrointestinal cancers, clinical trials have highlighted the chemopreventive potential of several agents. For colorectal neoplasia, trials with aspirin and other non-steroidal anti-inflammatory drugs (NSAIDS) and calcium have demonstrated the most significant reductions of risk. In observational studies, NSAIDs also consistently appear to protect against oesophageal and gastric ca
