A critical appraisal of the role of auditory brain stem response and magnetic resonance imaging in acoustic neuroma diagnosis.

Auditory brain stem response (ABR) is the most sensitive and specific audiometric test for acoustic neuroma (AN) detection. The ABR sensitivity rate increases with tumor size; consequently, small tumors have notably low ABR sensitivity for detection. However, larger tumors also can fail recognition by ABR testing. The ABR must be viewed on its own merits compared to magnetic resonance imaging studies for the diagnosis of AN. Where does ABR fit into the screening process for detecting AN today?

Tooth pulp deafferentation is not associated with changes in primary afferent depolarization of facial afferent endings in the brain stem.

Previous studies have demonstrated that tooth pulp deafferentation is associated with statistically significant alterations in the low-threshold facial mechanoreceptive field properties of brain stem neurons in trigeminal (V) subnucleus oralis. A loss of spinal afferent-induced presynaptic inhibition as a consequence of a decrease in primary afferent depolarization (PAD) following spinal nerve deafferentation has been invoked as a mechanism underlying deafferentation-induced somatosensory neuroplasticity. Therefore, this study was initiated to determine if these pulp deafferentation-induced ne…

[Brain stem evoked response audiometry study with a bone conduction receiver in atresia of the auditory canal and microtia].

Conductive hearing loss may be determined from the latency delay of the brainstem auditory evoked response Jewett V, if other reasons that may prolong latency are ruled out. Therefore it is important to determine the bone conduction threshold when examining infants. Two bone vibrators have been chosen, which, with the help of "inverse filtering", provide the possibility of a good sound conduction of the click stimulus and thus a measurement near the hearing threshold. When applying bone vibrators to infants it must be borne in mind, that the sound transfer is reduced because the cranial suture

Aging and presbycusis: effects on 2-deoxy-D-glucose uptake in the mouse auditory brain stem in quiet.

Autoradiography was used to assess the incorporation of [2-14C]deoxy-D-glucose by the auditory brain stem of young and aging mice of the C57BL/6 strain (which demonstrates progressive chronic sensorineural hearing loss) and the CBA strain (which maintains good hearing until late in life). Animals were injected with labeled 2-deoxyglucose and placed in quiet for 45 min; brain stem sections were prepared for autoradiography. The amounts of 2-deoxyglucose incorporated into the anterior ventral cochlear nucleus (AVCN), inferior colliculus (IC), and trigeminal nerve (TN) were densitometrically anal…

Involvement of noradrenergic systems in the effects of conditioning stimulation of the lower brain stem on the lumbar spinal reflex in rats.

The effects of conditioning stimulation at various positions in the pons and the medulla on the lumbar spinal monosynaptic reflex (MSR) were examined in rats. The conditioning stimulation facilitated the MSR. Facilitation with long latency was completely blocked by phenoxybenzamine, chlorpromazine or haloperidol. Phenoxybenzamine and chlorpromazine inhibited the MSR. Haloperidol, on the other hand, augmented the MSR, and this effect became more potent after the brain stem had been transected at the upper part of the medulla oblongata. These results suggest that pontine noradreneric systems hav…

Some remarks on the mode of spread of primary benign brain-stem tumours based on description of three cases.

On the basis of long-standing own experiences and the literature, the author subdivides the tumours mentioned in the title into two types. T … em structures. Signs of cerebral compression in the early phase. Signs of transversal damage of the brain stem are slight and disappear after decompression. Partial removal and subsequent irradiation lead to a far-reaching freedom from symptoms and a socially useful survival over many years. So these tumours should be operated on with subsequent irradiation.

Angiographic disappearance and reappearance of an arteriovenous malformation of the cerebellum and brain stem, and its surgical excision. A case report.

A case of arteriovenous malformation (AVM) of the cerebellum and brainstem at the cerebellopontine angle is reported. Though a postoperative angiogram after the first operation indicated complete excision of the lesion, the patient returned with subarachnoid haemorrhage, and an angiogram indicated reappearance of the lesion. Total excision was carried out at the second operation.

The value of brain stem auditory, visual and somatosensory evoked potentials and blink reflexes in the diagnosis of multiple sclerosis.

Cervical and cortical somatosensory evoked potentials (SEP) following electrical stimulation of the median nerve and blink reflexes (BR) following electrical stimulation of the supraorbital nerve were recorded in 30 normal subjects aged 20-49 years. Subjects aged 40-49 had longer SEP latencies than subjects aged 20-39 years. A total of 29 slightly affected patients with multiple sclerosis (MS) aged 26-49 years, including four patients without clinical signs (suspected MS) and 19 patients with signs indicating only one lesion (possible MS) were examined by low-rate random-stimulated brain stem …

Neural stem cells may be uniquely suited for combined gene therapy and cell replacement: Evidence from engraftment of Neurotrophin-3-expressing stem cells in hypoxic-ischemic brain injury.

Previously, we reported that, when clonal neural stem cells (NSCs) were transplanted into brains of postnatal mice subjected to unilateral hypoxic-ischemic (HI) injury (optimally 3-7 days following infarction), donor-derived cells homed preferentially (from even distant locations) to and integrated extensively within the large ischemic areas that spanned the hemisphere. A subpopulation of NSCs and host cells, particularly in the penumbra, "shifted" their differentiation towards neurons and oligodendrocytes, the cell types typically damaged following asphyxia and least likely to regenerate spon

Intravenous SPION-labeled adipocyte-derived stem cells targeted to the brain by magnetic attraction in a rat stroke model: An ultrastructural insight into cell fate within the brain.

Mesenchymal stem cell therapy after stroke is a promising option investigated in animal models and clinical trials. The intravenous route is commonly used in clinical settings guaranteeing an adequate safety profile although low yields of engraftment. In this report, rats subjected to ischemic stroke were injected with adipose-derived stem cells (ADSCs) labeled with superparamagnetic iron oxide nanoparticles (SPIONs) applying an external magnetic field in the skull to retain the cells. Although most published studies demonstrate viability of ADSCs, only a few have used ultrastructural techniqu

Deciphering the Role of Cancer Stem Cells in Breast Cancer Brain Colonization Using a Novel Patient-Derived Model.

Brain metastases from breast cancer (BCBM) are fatal and lack effective treatments. Their cellular and molecular drivers remain poorly understood, partly due to limited preclinical models that fail to capture patient tumor heterogeneity. Cancer stem-like cells (CSCs) are implicated in metastatic dissemination; however, their specific role in brain metastasis remains unclear. In this study, CSCs are isolated from human BCBM specimens and characterized for stem-like properties, including CD44 and ALDH1 expression, sphere formation, tumorigenicity, and in vitro and in vivo self-renewal. Intra-nip

Alcohol diminishes barrier integrity in human stem cell-derived brain microvascular endothelial cells: Role of reactive oxygen species.

The World Health Organization has linked alcohol consumption to over 200 diseases including neurodegenerative diseases. A dysfunctional blood-brain barrier (BBB) has been found to be influential in a number of brain disorders. The BBB is critical in maintaining homeostasis between the brain vasculature and parenchyma and a loss in barrier integrity would enable otherwise impermeable immune cells, molecules, and inflammatory mediators to reach the brain parenchyma. A subset of studies demonstrated that alcohol could diminish BBB integrity, but it is unclear if this effect translates clinically.

Genetic Modification of Mesenchymal Stem Cell to Overexpress CXCR4 Enhances Treatment Efficacy for Brain Injury After Cardiopulmonary Resuscitation.

AIM: To investigate whether genetically modifying human umbilical cord-derived mesenchymal stem cells (MSC) to overexpress the CXCR4 receptor can enhance their therapeutic efficacy for treating brain injury following cardiac arrest (CA). METHODS: MSC were engineered to overexpress CXCR4 (CXCR4-MSC) via lentiviral transduction. The migration capacity of these cells was tested using in vitro chemotaxis assays. In a rat model of CA/CPR, the homing ability of CXCR4-MSC to the brain was tracked in vivo, and their therapeutic effects on neuronal death and neurological recovery were assessed. The ro

Lab Protocols for Coxsackievirus B3 Infection Using Brain-Like Endothelial Cells Derived from Induced-Pluripotent Human Stem Cells.

Coxsackievirus B3 (CVB3) is an enterovirus known to be a leading cause of viral aseptic meningoencephalitis worldwide. Therefore, the virus may interact with and cross the blood-brain barrier (BBB). The BBB is a highly specialized endothelial barrier that helps protect the brain from foreign bodies and infection, primarily being composed of brain endothelial cells. Despite the fact that CVB3 can be fatal, particularly in young children, the mechanism through which CVB3 is able to penetrate the BBB and gain access to the central nervous system is not fully characterized and is poorly understood

A Novel Human Stem Cell Culture Model for Severe Traumatic Brain Injury Reflecting Sexual Dimorphism in Heterotopic Ossification.

Heterotopic ossification (HO) is a disease characterized by ectopic bone formation, which can occur following severe traumatic brain injury (TBI). However, the underlying mechanisms remain poorly understood. In this study, we established a stem cell model using adipose-derived stem cells (ADSCs) and skeletal stem cells (SSCs) to examine osteogenic factors present in the sera of TBI patients. Incubation of ADSCs and SSCs with osteoinductive medium supplemented with TBI serum significantly enhanced osteogenic differentiation, particularly in male ADSCs and both female and male SSCs, with male SS

Dental Pulp Stem Cells Attenuate Early Brain Injury After Subarachnoid Hemorrhage via miR-26a-5p/PTEN/AKT Pathway.

Subarachnoid hemorrhage (SAH) is a type of hemorrhagic stroke with high morbidity, mortality and disability, and early brain injury (EBI) after SAH is crucial for prognosis. Recently, stem cell therapy has garnered significant attention in the treatment of neurological diseases. Compared to other stem cells, dental pulp stem cells (DPSCs) possess several advantages, including abundant sources, absence of ethical concerns, non-invasive procurement, non-tumorigenic history and neuroprotective potential. Therefore, we aim to investigate whether DPSCs can improve EBI after SAH, and explore the mec

Mesenchymal Stem Cells Expressing Baculovirus-Engineered Brain-Derived Neurotrophic Factor Improve Peripheral Nerve Regeneration in a Rat Model.

BACKGROUND: Peripheral nerve injuries are a major clinical challenge because of their complex nature and limited regenerative capacity. This study aimed to improve peripheral nerve regeneration using Wharton's jelly mesenchymal stem cells (WJ-MSCs) engineered to express brain-derived neurotrophic factor (BDNF) via a baculovirus (BV) vector. The cells were evaluated for efficacy when seeded into acellular nerve grafts (ANGs) in a rat sciatic nerve defect model. METHODS: WJ-MSCs were transfected with recombinant BV to upregulate BDNF expression. Conditioned medium (CM) from these cells was util
