Naturally occurring macrocyclic peptides represent a unique class of compounds that exhibit various biological activities ranging from antibiotics to immunosuppressant. Although the discovery of such macrocyclic peptides had relied on their isolation from living organisms, recent advances in ribosomal peptide synthesis and in display techniques made it possible to use artificially generated macrocyclic peptide libraries for selection of ligands for biologically relevant proteins. In this review, we discuss the technologies and their applications for the discovery of peptide ligands.

Bioelectrical impedance analysis (BIA) was used to assess body composition in rats fed on either standard laboratory diet or on high-fat diet designed to induce obesity. Bioelectrical impedance analysis predictions of total body water and thus fat-free mass (FFM) for the group mean values were generally within 5% of the measured values by tritiated water (3H2O) dilution. The limits of agreement for the procedure were, however, large, approximately +/- 25%, limiting the applicability of the technique for measurement of body composition in individual animals.

PURPOSE: To determine the serial radiographic appearance of the lungs of neonates who underwent partial liquid ventilation with perflubron because of congenital diaphragmatic hernia (CDH) or primary pulmonary hypertension.
MATERIALS AND METHODS: Bedside anteroposterior (AP) and lateral chest radiographs (n = 235) were scored for percentage of lung opacification by perflubron during partial liquid ventilation (PLV) and extracorporeal membrane oxygenation (ECMO). Five neonates participated in the study; four had CDH, and one had primary pulmonary hypertension.

Symptoms as an important sign of the effects of methyl bromide were studied in 56 male workers (37 currently exposed and 19 previously exposed) in a methyl bromide factory. Male F344 rats, 4 weeks old, were given tris (2,3-dibromopropyl)phosphate (TBP) via stomach tube at a dosage level of 100 mg. per kg. of body weight per day. Treatment was given 5 days per week and continued for 4 or 52 weeks. Representative treated and control animals were killed at predetermined time intervals and the histomorphology and ultrastructure of the kidneys investigated. Two new synthetic steroid analogues, (I) 16beta-bromo-3beta,17alpha-dihydroxy-5alpha-pregnane-11,20-dione and (II) 17beta-ureido-1,4-androstadien-3-one have been shown to give kinetic patterns consistent with active-site-directed irreversible inhibition of adult rat testicular microsomal steroid 17alpha-hydroxylase and C17-20 lyase in vitro.
