Plasma exosomal proteomics: Unveiling novel biomarkers for ischemic stroke in the context of diabetes.

Diabetes elevates the risk of ischemic stroke and poor prognosis. Exosomes, which carry nucleic acids and proteins, transmit stable signals across the blood-brain barrier and serve as promising non-invasive biomarkers for brain injury. Exosome proteomics may thus reveal novel therapeutic strategies to improve outcomes in diabetic stroke patients. Exosomes were isolated from the plasma of acute ischemic stroke patients with and without type 2 diabetes (n = 10 each) and characterized them using electron microscopy, flow cytometry, and nanoparticle tracking analysis. Label-free proteomics and bio

Nuclear miR-3920 expression in hepatocellular carcinoma is associated with stemness marker expression and poorer prognosis.

BACKGROUND: Stemness-related marker expression in hepatocellular carcinomas (HCC) has been associated with aggressive behavior. The association between miRNA expression and HCC stemness remains unclear. AIMS: To identify miRNAs associated with stemness marker expression in HCC and their clinical significance. METHODS: MicroRNA microarray analysis was performed to identify microRNAs that were differentially expressed between EpCAM-positive and EpCAM-negative HCCs. The subcellular localization and expression level of miR-3920 was evaluated by locked nucleic acid-in situ hybridization analysis

ATP-sensitive peptide-based coacervates for intracellular delivery of therapeutic oligonucleotides.

Despite advances in the fields of lipoplexes, metal nanoparticles, and other nucleic acid carriers, intracellular delivery of DNA/RNA therapeutics remains a pressing area of molecular medicine. The existing delivery systems have limitations in terms of stability, efficacy, or toxicity. Peptide-based coacervates have recently emerged as a promising alternative. They offer several advantages, including easy DNA/RNA incorporation, low toxicity, and the ability to penetrate membranes. However, they have one main drawback: inefficient intracellular unpacking. In this study, we present a novel appro

Anti-Inflammatory Effect of miR-197-3p-Loaded Microvesicles in Familial Mediterranean Fever Mouse Model.

Familial Mediterranean Fever (FMF) is an inherited autoinflammatory disease triggered by Mediterranean Fever (MEFV) gene mutations that lead to spontaneous pyrin inflammasome activation and exaggerated pro-inflammatory cytokine secretion. The V726A knock-in (KI) mouse model of FMF mimics most of the clinical and immunologic manifestations of the disease, such as recurrent febrile attacks and systemic inflammation. Here, we explored the proof-of-concept evaluation of miR-197-3p-loaded microvesicles (MVs) as a therapeutic approach in the FMF KI mouse model. MVs were purified and structurally cha

Selective Packaging of miR-22 and miR-29 Orthologues into Extracellular Vesicles of the Porcine Whipworm Trichuris Suis.

Helminths modulate host immunity, and in this respect, the porcine whipworm Trichuris suis has been explored clinically as an immunotherapy for inflammatory diseases. T. suis secretes a complex mixture of excretory/secretory products (ESP), including extracellular vesicles (EVs), nanosized membranous particles with bioactive cargoes of lipids, proteins and nucleic acids that are implicated in intercellular communication within organisms. Here, we characterise the EV protein and miRNA cargo of T. suis to identify potential roles in host-parasite communication. In contrast to EVs from clade III/

Serum/plasma viral DNA: mechanisms and diagnostic applications to nasopharyngeal and cervical carcinoma.

Following reports describing circulating tumor DNA, serum/plasma viral nucleic acid has shown its potential as a new diagnostic target in cancer. In the majority of examples of viral carcinogenesis, the viral genome is consistently present in certain tumors and serves as an effective marker. This article reviews recent findings, proposes possible mechanisms, and examines the potential clinical application of serum/plasma Epstein-Barr virus (EBV) DNA in nasopharyngeal cancer (NPC) and human papillomavirus (HPV) DNA in cervical carcinoma (CC). These tumors share a DNA viral etiology and present

Amplification-Free Ribonucleic Acid Detection via Cas13-Csm6-Horseradish Peroxidase Cascade with Single-Molecule Sensitivity.

Achieving a single-molecule level for RNA detection remains a significant challenge in point-of-care settings. We present CCHP, a Cas13-Csm6-horseradish peroxidase three-step enzymatic cascade assay that enables amplification-free RNA detection with a simple colorimetric readout at 450 nm within 1 h under standard conditions. Across diverse viral and non-coding targets, CCHP spans a dynamic range of 10-107 copies per reaction, showing strong agreement with RT-qPCR on serial dilutions (|r| ≈ 0.95). In a small blinded feasibility comparison using extracted RNA from infected versus uninfected cel

Droplet digital PCR: An effective method for monitoring and prognostic evaluation of minimal residual disease in JMML.

Juvenile myelomonocytic leukaemia (JMML) is a rare myeloproliferative neoplasm requiring haematopoietic stem cell transplantation (HSCT) for potential cure. Relapse poses a significant obstacle to JMML HSCT treatment, as the lack of effective minimal residual disease (MRD)-monitoring methods leads to delayed interventions. This retrospective study utilized the droplet digital PCR (ddPCR) technique, a highly sensitive nucleic acid detection and quantification technique, to monitor MRD in 32 JMML patients. The results demonstrated that ddPCR detected relapse manifestations earlier than tradition

Inactivation of feline calicivirus and adenovirus type 40 by UV radiation.

Little information regarding the effectiveness of UV radiation on the inactivation of caliciviruses and enteric adenoviruses is available. Analysis of human calicivirus resistance to disinfectants is hampered by the lack of animal or cell culture methods that can determine the viruses' infectivity. The inactivation kinetics of enteric adenovirus type 40 (AD40), coliphage MS-2, and feline calicivirus (FCV), closely related to the human caliciviruses based on nucleic acid organization and capsid architecture, were determined after exposure to low-pressure UV radiation in buffered demand-free (BD

Peptide additives reprogram the lipid nanoparticle corona and enhance gene delivery in a serum-free environment for lung epithelium.

Lipid nanoparticles (LNPs) have become a clinical standard for systemically-administered nucleic acid drugs and vaccines, but the LNP pipeline for locally-delivered LNP therapies remains much less mature. Local delivery in lung represents a particularly compelling application space for DNA-LNP therapeutics, as local gene therapies could support sustained epithelial recovery and functional restoration in various lung diseases. However, locally-delivered DNA-LNPs face multiple barriers, including the limited availability of serum components that often support conventional LNP activity, and the a

Global knowledge mapping and emerging research trends in the convergence of rheumatoid arthritis and exosomes: A CiteSpace-based visual analysis.

Rheumatoid arthritis (RA) is an autoimmune disorder characterized by persistent synovial inflammation and progressive joint destruction, with a rising global prevalence. By 2050, the age-standardized incidence rate is projected to reach 16.78 per 1,00,000 women and 7.85 per 1,00,000 men. In recent years, exosomes (EXOs) - membrane-bound vesicles (30-150 nm in diameter) that mediate intercellular communication by shuttling proteins, nucleic acids, and lipids - have emerged as promising targets for unraveling RA pathogenesis and developing novel therapies. As key carriers of biological signals,

Drug delivery of antisense molecules to the brain for treatment of Alzheimer's disease and cerebral AIDS.

Antisense oligonucleotides (ODNs) and peptide nucleic acids (PNAs) are potential therapeutics for eradication of malignancies, viral infections, and other pathologies. However, ODNs and PNAs in general are unable to cross cellular membranes and blood-tissue barriers, such as the blood-brain barrier (BBB), which is only permeable to lipophilic molecules of molecular weight <600 Da. Cellular delivery systems based on conjugates of streptavidin (SA) and the OX26 monoclonal antibody directed to the transferrin receptor may be employed as a universal carrier for the transport of mono-biotinylated p

Biochemical components of chicken primordial germ cells are modified by cryopreservation: original analysis by Fourier Transform Infrared (FTIR) microspectroscopy.

Chicken primordial germ cells (cPGCs) are a highly valuable resource for preserving chicken genetics and poultry biodiversity, as these diploid stem cells, derived from both male and female genotypes, can be maintained in long-term in vitro culture and are still capable of producing germline-transmitting progeny. However, the cryopreservation process, which guarantees that these cells can be used to maintain genetic resources, raises various questions about their quality after thawing. We assessed cryo-induced biological alterations in cPGCs derived from Lueng Hang Khao (LK), a Thai native chi
